CCR5A32 mutation does not influence the susceptibility to HCV infection, severity of liver disease and response to therapy in patients with chronic hepatitis C

AIM： To study whether CCR5△32 mutation was associated with viral infection and severity of liver disease. METHODS： Two hundred and fifty two histologically proven, chronic HCV patients （mean age： 41±14 years; M/F： 164/88） were genotyped. PCR based genotyping of 32 bp deletion at the CCR5 locus was done. Fourhundred and eight matched healthy controls were studied to assess susceptibility to HCV infection. To assess correlation of immune gene polymorphism with severity of HCV related liver disease, patients with chronic HCV infection were divided into those with a fibrosis score of ≤ 2 （mild） or 〉 2 （severe） and histological activity index （HAI） of ≤ 5 or 〉 5. For correlation between CCR5△32 mutations and response to therapy, 129 patients who completed therapy were evaluated. RESULTS： The majority （89.4%） of the patients were infected with genotype 3. The frequency of homozygous CCR5△32 mutants was comparable to HCV patients as compared to the healthy controls （0.7% vs 0%, P = 0.1）. Further more, the frequency of CCR5△32 mutation was comparable in patients with mild or severe liver disease.（P = NS）. There was also no association observed with response to therapy and CCR5△32 mutation. CONCLUSION： CCR5△32 mutation does not have a role in disease susceptibility, severity or response to therapy in patients with chronic hepatitis C infection.

Aim:The aim of this study was to compare and investigate the tissue thickness at the maxillary and mandibular central incisors utilizing the probe visibility method and traditional way of measuring gingival thickness(gt)by endodontic ***:In 200 selected patients,the gt was measured at both the maxillary and mandibular anterior using an endodontic file after local infiltration at the mid-distance from the gingival margin to the mucogingival *** checkpoint of transition from thin to thick periodontal phenotype was determined by probe visibility method in the following age groups:Group A:21-30 years;Group B:31-40 years;Group C:41-50 years;and Group D:51-60 ***:gt varied from 0.3 to 1.9 *** translucency was observed up to the thickness of 0.7 *** 0.8 mm of gt,there was no probe ***:According to the present research,the gingiva appears transparent up to a thickness of around 0.7 *** checkpoint of transition between thin and thick periodontal phenotypes at both the maxillary and mandibular anterior was associated with a gt of 0.8 mm.

Analysis of frequency shifting in seismic signals using Gabor-Wigner transform

A hybrid time-frequency method known as Gabor-Wigner transform (GWT) is introduced in this paper for examining the time-frequency patterns of earthquake damaged buildings. GWT is developed by combining the Gabor transform (gt) and Wigner-Ville distribution (WVD). gt and WVD have been used separately on synthetic and recorded earthquake data to identify frequency shifting due to earthquake damages, but gt is prone to windowing effect and WVD involves ambiguity function. Hence to obtain better clarity and to remove the cross terms (frequency interference), gt and WVD are judiciously combined and the resultant GWT used to identify frequency shifting. Synthetic seismic response of an instrumented building and real-time earthquake data recorded on the building were investigated using GWT. It is found that GWT offers good accuracy for even slow variations in frequency, good time-frequency resolution, and localized response. Presented results confirm the efficacy of GWT when compared with gt and WVD used separately. Simulation results were quantified by the Renyi entropy measures and GWT shown to be an adequate technique in identifying localized response for structural damage detection.

Optimal PV Array Configuration for Extracting Maximum Power Under Partial Shading Conditions by Mitigating Mismatching Power Losses

The grid-connected or standalone PV central inverter architecture is comprised of several PV modules which are connected in different ways to form the PV *** power generation capability of the PV array is primarily affected by partial shading conditions(PSC).Due to PSCs,the power output of the PV array is dramatically reduced,and mismatching losses are induced in the PV *** on the extent of these problems,multiple peaks also appear in the power-voltage(P-V)curve,which makes it very difficult to track the global maximum power point(GMPP).The main objective of this research paper is to model and simulate the series(S),series-parallel(SP),bridge-link(BL),honey-comb(HC),total-cross-tied(TCT)and proposed triple-tied(TT)solar PV array configurations under various partial shading *** performance of all PV configurations is evaluated under a uniform approach,considering eight different shading *** performance of the considered PV configurations is analyzed in terms of their mismatching power losses,fill factors,efficiency,global maximum power points(GMPPs),local maximum power points(LMPPs),voltages and currents at GMPPs,open circuit voltage and short circuit *** above-mentioned PV configurations are modeled and simulated in a Matlab/Simulink environment by considering the KC-200gt module parameters.

Assessment of glacier stored water in Karakoram Himalaya using satellite remote sensing and field investigation

Karakoram Himalaya(KH) has continental climatic conditions and possesses largest concentration of glaciers outside the polar regions. The melt water from these glaciers is a major contributor to the Indus river. In this study, various methods have been used to estimate the ice volume in the Karakoram Range of glaciers such as Coregistration of Optically Sensed Images and Correlation(COSI-Corr) method and Area-Volume relations. Landsat 8 satellite data has been used to generate the ice displacement, velocity and thickness map. Our study for 558 Karakoram glaciers revealed that the average ice thickness in Karakoram is 90 m. Ground Penetrating Radar(GPR) survey has been conducted in one of the KH glacier i.e. Saser La glacier and the collected GPR data is used for the validation of satellite derived thickness map. GPR measured glacier thickness values are found comparable with satellite estimated values with RMSE of 4.3 m. The total ice volume of the Karakoram glaciers is estimated to be 1607±19 km3(1473±17 gt), which is equivalent to 1473±17 km3 of water equivalent. Present study also covers the analysis of glacier surface displacement, velocity and ice thickness values with reference to glacier mean slope.

Biocompatible silver nanoparticles: An investigation into their protein binding efficacies, anti-bacterial effects and cell cytotoxicity studies

Green synthesis of silver nanoparticles(AgNPs)has garnered tremendous interest as conventional methods include the use and production of toxic chemicals,products,by-products and *** this regard,the synthesis of AgNPs using green tea(gt)extract and two of its components,(-)-epigallocatechin gallate(EGCG)and(+)-catechin(Ct)as capping/stabilizing agents,is *** synthesized AgNPs showed antibacterial activity against the bacterial strains Staphylococcus aureus and Escherichia coli,along with anticancer activity against HeLa *** administering nanoparticles to the body,they come in contact with proteins and results in the formation of a protein corona;hence we studied the interactions of these biocompatible AgNPs with hen egg white lysozyme(HEWL)as a carrier *** quenching mechanism was accountable for the quenching of HEWL fluorescence by the *** binding constant(Kb)was found to be higher for EGCG-AgNPs((2.309±0.018)×104 M-1)than for gt-AgNPs and Ct-AgNPs towards ***-AgNPs increased the polarity near the binding site while Ct-AgNPs caused the opposite effect,but gt-AgNPs had no such observable *** dichroism studies indicated that the AgNPs had no such appreciable impact on the secondary structure of *** key findings of this research included the synthesis of AgNPs using gt extract and its constituent polyphenols,and showed significant antibacterial,anticancer and protein-binding ***-OH groups of the polyphenols drive the in situ capping/stabilization of the AgNPs during synthesis,which might offer new opportunities having implications for nanomedicine and nanodiagnostics.

Efficient Gene Targeting in Maize Using Inducible CRISPR-Cas9 and Marker-free Donor Template

CRISPR-Cas9 is a powerful tool for generating targeted mutations and genomic ***,precise gene insertion or sequence replacement remains a major hurdle before application of CRISPR-Cas9 technology is fully realized in plant ***,we report high-frequency,selectable marker-free intra-genomic gene targeting(gt)in *** shock-inducible Cas9 was used for generating targeted double-strand breaks and simultaneous mobilization of the donor template from pre-integratad *** construct was designed such that release of the donor template and subsequent DNA repair activated expression of the selectable marker gene within the donor *** approach generated up to 4.7%targeted insertion of the donor sequence into the target locus in TO plants,with up to 86%detected donor template release and 99%mutation rate being observed at the donor loci and the genomic target site,*** previous in planta or intra-genomic homologous recombination reports in which the original chimeric gt plants required extensive progeny screening in the next generation to identify non-chimeric gt individuals,our method provides non-chimeric heritable gt in one generation.

Novel CMS lines in pigeonpea[Cajanus cajan(L.)Millspaugh]derived from cytoplasmic substitutions,and their effective restoration and deployment in hybrid breeding

The availability of stable cytoplasmic male sterile(CMS or A) lines coupled with a robust restoration system(R lines) is an essential prerequisite for efficient hybrid ***-enabled hybrid technology holds immense potential to enhance the long-stagnant productivity of pigeonpea. In the present investigation, cytoplasmic substitutions were made in the nuclear backgrounds of early-maturing pigeonpea varieties or lines. Three new CMS lines(ICPL 88039 A, Pusa 992 A, and DPP 3-2A) resulted from genetic crosses involving cytoplasmic donors from A2(gt 288A) and A4(ICPA 2089) categories. In addition to visual inspection of anthers, pollen-staining techniques and scanning electron microscopy(SEM)analysis were used to confirm pollen sterility. Further, given the relevance of the plant mitochondrial genome to CMS manifestation, 25 mitochondrion-specific DNA markers were assayed on these newly developed A lines and isogenic maintainer(B) lines. DNA polymorphism between Pusa 992 A and Pusa 992 B as revealed by the nad7a_del marker confirmed the successful combination of sterilizing cytoplasm(A4) and nonrestoring nuclear background(Pusa 992). Such cytoplasm-specific DNA markers are required for A2-CMS as well. Further, to assess restoration ability, potential restorers were crossed with these CMS lines, and as a consequence, promising A × R combinations exhibiting 100%pollen fertility could be identified. In parallel, we also analyzed the inheritance patterns underlying fertility restoration using ICPL 88039A-derived F2 and BC1F1populations,and established a monogenic dominant model to explain the phenomenon of A2-CMS restoration. In summary, we report the successful development of new CMS lines and describe their effective deployment in hybrid breeding of pigeonpea.

The Onset of Buoyancy and Surface Tension Driven Convection in a Ferrofluid Layer by Influence of General Boundary Conditions

This paper investigated the buoyancy and surface tension-driven ferro-thermal-convection (FTC) in a ferrofluid (FF) layer due to influence of general boundary conditions. The lower surface is rigid with insulating to temperature perturbations, while the upper surface is stress-free and subjected to general thermal boundary condition. The numerically Galerkin technique (gt) and analytically regular perturbation technique (RPT) are applied for solving the problem of eigenvalue. It is analyzed that increasing Biot number, decreases the magnetic and Marangoni number is to postponement the onset. Additionally, magnetization nonlinearity parameter has no effect on FTC in the non-existence of Biot number. The results under the limiting cases are found to be in good agreement with those available in the literature.

Experimental Evaluation of Juniper Network's Netscreen-5gt Security Device against Layer4 Flood Attacks

Cyber attacks are continuing to hamper working of Internet services despite increased use of network secu-rity systems such as firewalls and Intrusion protection systems (IPS). Recent Distributed Denial of Service (DDoS) attacks on Dec 8th, 2010 by Wikileak supporters on Visa and Master Card websites made headlines on prime news channels all over the world. Another famous DDoS attacks on Independence Day weekend, on July 4th, 2009 were launched to debilitate the US and South Korean governments’ websites. These attacks raised questions about the capabilities of the security systems that were used in the network to counteract such attacks. Firewall and IPS security systems are commonly used today as a front line defense mechanism to defend against DDoS attacks. In many deployments, performances of these security devices are seldom evaluated for their effectiveness. Different security devices perform differently in stopping DDoS attacks. In this paper, we intend to drive the point that it is important to evaluate the capability of Firewall or IPS secu-rity devices before they are deployed to protect a network or a server against DDoS attacks. In this paper, we evaluate the effectiveness of a security device called Netscreen 5gt (or NS-5gt) from Juniper Networks under Layer-4 flood attacks at different attack loads. This security device NS-5gt comes with a feature called TCP-SYN proxy protection to protect against TCP-SYN based DDoS attacks, and UDP protection feature to protect against UDP flood attacks. By looking at these security features from the equipments data sheet, one might assume the device to protect the network against such DDoS attacks. In this paper, we con-ducted real experiments to measure the performance of this security device NS-5gt under the TCP SYN and UDP flood attacks and test the performance of these protection features.
