AGK2 pre-treatment protects against thioacetamide-induced acute liver failure via regulating the MFN2-PERK axis and ferroptosis signaling pathway

Background:Acute liver failure(ALF)is an unpredictable and life-threatening critical illness.The pathological characteristic of ALF is massive necrosis of hepatocytes and lots of inflammatory cells infiltration which may lead to multiple organ failure.Methods:Animals were divided into 3 groups,normal,thioacetamide(TAA,ALF model)and TAA+AGK2.Cultured L02 cells were divided into 5 groups,normal,TAA,TAA+mitofusin 2(MFN2)-siRNA,TAA+AGK2,and TAA+AGK2+MFN2-siRNA groups.The liver histology was evaluated with hematoxylin and eosin staining,inositol-requiring enzyme 1(IRE1),activating transcription factor 6β(ATF6β),protein kinase R(PKR)-like endoplasmic reticulum kinase(PERK)and phosphorylated-PERK(p-PERK).C/EBP homologous protein(CHOP),reactive oxygen species(ROS),MFN2 and glutathione peroxidase 4(GPX4)were measured with Western blotting,and cell viability and liver chemistry were also measured.Mitochondriaassociated endoplasmic reticulum membranes(MAMs)were measured by immunofluorescence.Results:The liver tissue in the ALF group had massive inflammatory cell infiltration and hepatocytes necrosis,which were reduced by AGK2 pre-treatment.In comparison to the normal group,apoptosis rate and levels of IRE1,ATF6β,p-PERK,CHOP,ROS and Fe2+in the TAA-induced ALF model group were significantly increased,which were decreased by AGK2 pre-treatment.The levels of MFN2 and GPX4 were decreased in TAA-induced mice compared with the normal group,which were enhanced by AGK2 pretreatment.Compared with the TAA-induced L02 cell,apoptosis rate and levels of IRE1,ATF6β,p-PERK,CHOP,ROS and Fe2+were further increased and levels of MFN2 and GPX4 were decreased in the MFN2-siRNA group.AGK2 pre-treatment decreased the apoptosis rate and levels of IRE1,ATF6β,p-PERK,CHOP,ROS and Fe2+and enhanced the protein expression of MFN2 and GPX4 in MFN2-siRNA treated L02 cell.Immunofluorescence observation showed that level of MAMs was promoted in the AGK2 pre-treatment group when compared with the TAA-induced group in both mice and L02 cells.Conclusions:The d

DEM investigation of strength and critical state behaviours of granular materials under true triaxial loadings

This paper investigates the strength and critical state(CS)behaviours of granular materials via DEM simulations of true triaxial drained tests under three different loading modes including constant b(in-termediate stress ratio)tests with constantσ'1(major principal stress),constant p(mean pressure)and constantσ'3(minor principal stress)respectively.To this end,a series of samples are generated with the same particle size distribution,and with the confining stresses ranging from 100 kPa to 900 kPa.The CS is achieved for all samples.Both the macroscopic behaviours and the microscopic behaviours are examined and compared considering different loading modes,confining stresses and intermediate stress ratios(b).The critical state lines(CSLs)are found to be unique and independent of the loading modes,but dependent on the b values.The CSLs with b=0 and b=1 form the two boundaries of CSLs respectively beyond which CSLs under all other b tests cannot go beyond.Six different strength criteria are examined and compared in terms of both peak and CS failures.The Mohr-Coulomb strength criterion is found to be only suitable for axisymmetric loading conditions.The Lade-Duncan criterion is only suitable for describing peak strengths,which is dependent on the loading modes and confining stresses.The Satake criterion and Matsuoka-Nakai criterion are the more appropriate strength criteria for describing CS failures,indicating that the CS values of both the Satake parameter and the Matsuoka-Nakai parameter describe an inherent property that characterizes the CS failure for a given type of soil.The CS mechanical coordination number is fitted by a curved line for a given b value,which is unique regardless of the loading modes.The peak and CS values of both major and minor principal fabric tensors decrease with increasing b values,while they increase with increasing b values for the intermediate principal fabric tensor.

Non-small cell lung cancer(NSCLC)is a malignant tumor that poses a serious threat to human health.Adenocarcinoma(LUAD)and squamous cell carcinoma(LUSC)are two common types of NSCLC.They originate from glandular and squamous cells of the bronchial epithelium,respectively.Identifying gene changes during tumorigenesis is conducive to the selection of suitable treatment methods for patients.CD98 is a heterodimeric transmembrane glycoprotein,which is composed of a heavy chain and a light chain.1 cD98 heavy chain(CD98hc),also known as 4F2hc or SLC3A2,is an 85kDa type ll transmembraneglycoprotein,which consists of a cytoplasmic region(NH2 terminal),a single chain transmembrane region,and a huge extracellular region(COOH terminal).1 CD98hc can combine with the CD98 light chain,LAT1(SLC7A5),to form CD98 protein through disulfide bonds.1 SLC7A5 is a 12 times transmembrane helix bundle protein composed of 501-535 amino acid residues1.

Emergent symmetry in quantum phase transition:From deconfined quantum critical point to gapless quantum spin liquid

The emergence of exotic quantum phenomena in frustrated magnets is rapidly driving the development of quantum many-body physics,raising fundamental questions on the nature of quantum phase transitions.Here we unveil the behaviour of emergent symmetry involving two extraordinarily representative phenomena,i.e.,the deconfined quantum critical point(DQCP)and the quantum spin liquid(QSL)state.Via large-scale tensor network simulations,we study a spatially anisotropic spin-1/2 square-lattice frustrated antiferromagnetic(AFM)model,namely the J1x-J1y-J2 model,which contains anisotropic nearestneighbor couplings J1x,J1y and the next nearest neighbor coupling J2.For small J1y/J1x,by tuning J2,a direct continuous transition between the AFM and valence bond solid phase is observed.With growing J1y/J1x,a gapless QSL phase gradually emerges between the AFM and VBS phases.We observe an emergent O(4)symmetry along the AFM–VBS transition line,which is consistent with the prediction of DQCP theory.Most surprisingly,we find that such an emergent O(4)symmetry holds for the whole QSL–VBS transition line as well.These findings reveal the intrinsic relationship between the QSL and DQCP from categorical symmetry point of view,and strongly constrain the quantum field theory description of the QSL phase.The phase diagram and critical exponents presented in this paper are of direct relevance to future experiments on frustrated magnets and cold atom systems.

BACKGROUND Rectal mucinous adenocarcinoma(MAC)is a rare pathological type of rectal can-cer with unique pathological features and a poor prognosis.It is difficult to diag-nose and treat early because of the lack of specific manifestations in some aspects of the disease.The common metastatic organs of rectal cancer are the liver and lung;however,rectal carcinoma with metastasis to subcutaneous soft tissue is a rare finding.CASE SUMMARY In this report,the clinical data,diagnosis and treatment process,and postope-rative pathological features of a patient with left waist subcutaneous soft tissue masses were retrospectively analyzed.The patient underwent surgical treatment after admission and recovered well after surgery.The final pathological diagnosis was rectal MAC with left waist subcutaneous soft tissue metastasis.CONCLUSION Subcutaneous soft tissue metastasis of rectal MAC is rare,and it can suggest that the tumor is disseminated,and it can appear even earlier than the primary ma-lignant tumor,which is occult and leads to a missed diagnosis and misdiagnosis clinically.When a subcutaneous soft tissue mass of unknown origin appears in a patient with rectal cancer,a ma-lignant tumor should be considered.

Effect of BaO and MgO on structure and properties of aluminate slag with various CaO/Al_(2)0_(3)ratios

To provide theoretical guidance for performance stability control of low-reactive mold fluxes,the effects of BaO and MgO on the structure and properties of aluminate slag with various CaO/Al_(2)0_(3)(C/A)ratios were investigated using the Fourier transform infrared spectrometer,Raman spectroscope,hemispherical melting point instrument,rotational viscometer and X-ray diffractometer.The results indicated that with BaO and MgO addition,the structure polymerization was first weakened and then enhanced at C/A of 1.1,and the transition contents corresponded to 8 wt.%BaO and 2 wt.%MgO,respectively,while the structure polymerization decreased continuously at C/A of 1.3.Since the viscosity change was well consistent with the structure evolution,the polymerization degree played a more prominent role in the slag viscosity than superheat degree when the melting temperature difference was within 40℃.The break temperature decreased initially and then increased with augment of BaO and MgO at C/A of 1.1,while it manifested a decrease trend with BaO addition,and it decreased obviously but then turned to increase with MgO increment at C/A of 1.3.The crystallization phase and crystallization ratio kept stable with BaO increment,while the crystallization ratio rose greatly with MgO promoting LiAlO_(2)precipitation at C/A of 1.1.The crystal types of all experimental slags were mainly Ca_(12)Al_(14)O_(33)and CaF_(2)at C/A of 1.3,and the precipitation of crystalline phase BaAl2O4 demonstrated a rising trend,while that of Ca_(12)Al_(14)O_(33)gradually declined with BaO augment.

Occurrence of dark septate endophytes in Phragmites australis in the Baiyang Lake and their resistance to Cd stress

Heavy metal pollution poses a serious hazard to human health,and microbial remediation of heavy metals in soil has been widely studied.A group of ascomycetes classified as dark septate endophytes(DSEs)colonize plant roots and benefit host plants under abiotic stress conditions.In this study,Phragmites australis,a common remediation plant in the Baiyang Lake in North China,was investigated.Soils and roots of P.australis were collected in typical heavy metal-contaminated sites,and the species diversity and community structure of DSEs in P.australis roots were studied.In addition,DSE strains were isolated,cultured,and tested for their tolerance to Cd stress.The results showed that DSEs occurred extensively in P.australis roots,forming typical dark septate hyphae,with a total colonization rate of 19.7%–83.1%.Morphological and internal transcribed spacer sequencing analyses were used to identify 10 species within 9 genera of DSE fungi.Among these fungi,6 strains with considerable resistance to Cd stress were identified.The biomasses of Poaceascoma helicoides,Alternaria doliconidium,and Acrocalymma vagum strains increased as the Cd levels increased.These results can not only help to understand plant-DSE interactions in wetland environments,but also provide a theoretical basis for making full use of DSE fungi to alleviate heavy metal contamination in soil.

In this paper,we consider the B?hmer–Vandersloot(BV)model of loop quantum black holes obtained from the improved dynamics approach.We adopt the Saini–Singh gauge,in which it was found analytically that the BV spacetime is geodesically complete.We show that black/white hole horizons do not exist in this geodesically complete spacetime.Instead,there exists only an infinite number of transition surfaces,which always separate trapped regions from anti-trapped ones.Comments on the improved dynamics approach adopted in other models of loop quantum black holes are also given.

OCTA characteristics in non-arteritic central retinal artery occlusion and correlation with visual acuity

AIM:To observe the retinal and choroidal circulations in patients with non-arteritic permanent central retinal artery occlusion(NA-CRAO)via optical coherence tomography angiography(OCTA)and analyze their correlation with visual acuity.METHODS:Sixty-two eyes with clinically confirmed acute NA-CRAO were included in the study and divided into:A type(mild n=29),B type(moderate n=27)and C type(severe n=6)based on the degree of visual loss,retinal edema,and arterial blood flow delay in fundus fluorescence angiography(FFA).Contralateral healthy eyes were used as the control group.Best-corrected visual acuity(BCVA),slit lamp microscopy,indirect ophthalmoscopy,fundus color photography,OCTA,and FFA were performed.Spearman’s correlation analysis was used to determine the correlations between retinal and choroidal vessels and visual acuity.RESULTS:There were no statistically significant differences in age,gender,and intraocular pressure among the three types and the control group(P>0.05).Vessel density in deep capillary plexus(VD-DCP)significantly decreased(P<0.05)in all three types of NA-CRAO patients compared to the control group.Vessel density in superficial vascular plexus(VD-SVP)significantly decreased(P<0.05)in type A patients and choriocapillaris flow area significantly decreased(P<0.05)in type B and type C patients compared to the control group;while outer retinal flow areas significantly increased in the type A(P<0.05)and decreased in type C patients(P<0.05).The retinal thickness significantly increased in type C group(P<0.05).The VD-SVP at fovea in the type A was significantly lower than both of type B and C.The VD-SVP at nasal parafovea in type A and B was significantly lower than type C(P<0.05).The logMAR BCVA of type A was significantly better than that of type B and C groups(P<0.05).Spearman’s correlation analysis showed that the logMAR BCVA was positively correlated with VD-SVP at fovea(r=0.679,P=0.031)and nasal parafovea(r=0.826,P=0.013).CONCLUSION:OCTA is valuable for assessing retinal ischemia,and evaluatin

Electroencephalogram findings in 10 patients with post-stroke epilepsy:A retrospective study

BACKGROUND Post-stroke epilepsy is a common and easily overlooked complication of acute cerebrovascular disease.Long-term seizures can seriously affect the prognosis and quality of life of patients.Electroencephalogram(EEG)is the simplest way to diagnose epilepsy,and plays an important role in predicting seizures and guiding medication.AIM To explore the EEG characteristics of patients with post-stroke epilepsy and improve the detection rate of inter-seizure epileptiform discharges.METHODS From January 2017 to June 2020,10 patients with post-stroke epilepsy in our hospital were included.The clinical,imaging,and EEG characteristics were collected.The stroke location,seizure type,and ictal and interictal EEG manifestations of the patients with post-stroke epilepsy were then retrospectively analyzed.RESULTS In all 10 patients,epileptiform waves occurred in the side opposite to the stroke lesion during the interictal stage;these manifested as sharp wave,sharp-wave complex,or spike discharges in the anterior head lead of the side opposite to the lesion.CONCLUSION In EEG,epileptiform waves can occur in the side opposite to the stroke lesion in patients with post-stroke epilepsy.
