Piperlongumine Inhibits Zika Virus Replication In vitro and Promotes Up-Regulation of HO-1 Expression, Suggesting An Implication of Oxidative Stress

Owing to the widespread distribution of mosquitoes capable of transmitting Zika virus, lack of clinical vaccines and treatments, and poor immunity of populations to new infectious diseases, Zika virus has become a global public health concern. Recent studies have found that Zika virus can continuously infect human brain microvascular endothelial *** cells are the primary components of the blood–brain barrier of the cerebral cortex, and further infection of brain tissue may cause severe damage such as encephalitis and fetal pituitary disease. The present study found that a biologically active base, piperlongumine(PL), inhibited Zika virus replication in human brain microvascular endothelial cells, Vero cells, and human umbilical vein endothelial cells. PL also significantly increased heme oxygenase-1(HO-1) gene expression, while silencing HO-1 expression and using the reactive oxygen species scavenger, N-acetylcysteine, attenuated the inhibitory effect of PL on Zika virus replication. These results suggest that PL induces oxidative stress in cells by increasing reactive oxygen species. This, in turn, induces an increase in HO-1 expression, thereby inhibiting Zika virus replication. These findings provide novel clues for drug research on the prevention and treatment of Zika virus.

A high resolution simulation of climate change over China

Multi-decadal high resolution climate change simulations over East Asia were performed by using The Abdus Salam International Centre for Theoretical Physics (ICTP) Regional Climate Model (RegCM3), nested within the NASA/NCAR global model FvGCM/CCM3. Two sets of simulations were conducted at 20-km grid spacings, one for present day (1961-1990) and one for the future climate (2071-2100, IPCC A2 scenario). Simulations of present climate conditions over China by RegCM3 and FvGCM were compared against observations to assess the model performance. Results showed that both models repro- duced the observed spatial structure of 500 hPa height, surface air temperature and precipitation. Compared with FvGCM, RegCM3 provided increasing spatial detail of surface variables. Furthermore, RegCM3 improved the simulation of monsoon precipitation over the region. Changes in the mean temperature and precipitation were analyzed and compared between the two models. Significant warming in the end of the 21st century was simulated by both models in December-January-February (DJF), June-July-August (JJA), and the annual mean. In DJF, greater warming was simulated by FvGCM over Northeast and Northwest China, as well as the Tibetan Plateau, compared with RegCM. In JJA, RegCM3 simulated greater warming over northern China, Inner Mongolia, Northwest China, and the Tibetan Plateau. Simulated changes in DJF precipitation showed similar spatial patterns between the two models. In JJA, while FvGCM projected a prevailing increase of monsoon precipitation over China, which is in agreement with other global models, RegCM3 projected extended areas of decreased precipitation. Changes in the variability for annual mean temperature and precipitation also are presented.

Generally, nutrient cycle is closely related to the element distribution in biomass and the popula- tion dynamics in ecosystems. Carbon, nitrogen and phosphorus in Japanese anchovy （Engraulis japonicus） of different body lengths from the Huanghai Sea （Yellow Sea） were determined to better understand their variability and reasons during its life history. The mean content was 45.12%, 10.12% and 2.02% for C, N and P, respectively. Significant differences in C, N and P elemental composition were found among different sizes, which could be explained by varying proportions of storage compounds in whole body fish, and varying degrees of ossification. Considering abun- dant resources in Japanese anchovy, it was an important P-pool in the cycle of P. Moreover, the excreted N/P ratio was significantly different in fishes of different sizes, especially at high gross growth efficiency. In the past two decades, overexploitation tended to cause smaller body length in the community structure of anchovy, which presumably changed the nutrient cycle in food webs of the Huanghai Sea. Exptrapolation of the results indicates that Japanese anchovy may be important for conveying nutrient in the Huanghai Sea.

Thermal adaptation plays a fundamental role in shaping the distribution and abundance of insects,and heat shock proteins（Hsps）play important roles in the temperature adaptation of various *** better understand the temperature tolerance of the indigenous ZHJ2-biotype of whitefly Bemisia tabaci species complex,we obtained complete cDNA sequences for hsp90,hsp70,and hsp20 and analyzed their expression profiles under different high temperature treatments by real-time quantitative polymerase chain *** high temperature tolerance of *** ZHJ2-biotype was determined by survival rate after exposure to different high temperatures for 1 *** results showed that after 41°C heat-shock treatment for 1 h,the survival rates of ZHJ2 adults declined significantly and the estimated temperature required to cause 50% mortality（LT50）is 42.85°C for 1 *** for onset（Ton）or maximal（Tmax）induction of hsps expression in *** ZHJ2-biotype were 35 and 39°C（or 41°C）.Compared with previous studies,indigenous ZHJ2-biotype exhibits lower heat temperature stress tolerance and Ton（or Tmax）than the invasive B-biotype.

Insects are ectotherms and their ability to resist temperature stress is limited. The immediate effects of sub-lethal heat stress on insects are well documented, but longer- term effects of such stresses are rarely reported. In this study, survival, development and reproduction of the whitefly, Bemisia tabaci （Gennadius）（Hemiptera： Aleyrodidae） biotype B, were compared over five consecutive generations at 27, 31 and 35℃ and for one generation at 37 ℃. Both temperature and generation significantly affected the fitness of the whitefly. These impacts were more dramatic with increasing generations and temperatures. Among the experimental temperatures, the most favorable for development and reproduction were 27 ℃ and 31 ℃. At 27 ℃, survival, development and fecundity were all stable over these five generations. At 31 ℃, immature survival rate was the highest in the fifth generation, but female fecundities decreased in the fourth and fifth generations. At 35 ℃, egg hatching rate, immature survival rate and female fecundity decreased significantly in the fourth and fifth generations. At 37 ℃, survival ofB. tabaci was not adversely affected, but female fecundity at 37 ℃ was less than 10% of that at 27 ℃ or 31 ℃. These results demonstrate that the lethal high temperature for B. tabaci is over 37 ℃, and the whitefly population continued expanding in the five generations at 35 ℃. The ability orB. tabaci biotype B to survive high temperature stress will play an important role in its population extension under global warming.

In this paper, a diffusive predator-prey system of Holling type functional III is considered. For one hand, we considered the possibility of the occurrence of Turing patterns of the system. Our results show that there is no Turing patterns found in the system. On the other hand, we performed detailed Hopf bifurcation analysis to the systems, and showed that the system have multiple oscillatory patterns. Moreover, we also derived the conditions to determine the Hopf bifurcation direction and the stability of the bifurcating periodic solutions. Computer simulations are included to support our theoretical analysis.
