Epistasis is a ubiquitous phenomenon in genetics,and is considered to be one of main factors in current efforts to unveil missing heritability of complex *** data is crucial for evaluating epistasis detection tools in genome-wide association studies(gWAS).Existing simulators normally suffer from two limitations:absence of support for high-order epistasis models containing multiple single nucleotide polymorphisms(SNPs),and inability to generate simulation SNP data *** this study,we proposed a simulator SimHOEPI,which is capable of calculating penetrance tables of high-order epistasis models depending on either prevalence or heritability,and uses a resampling strategy to generate simulation data *** of SimHOEPI are the preservation of realistic minor allele frequencies in sampling data,the accurate calculation and embedding of high-order epistasis models,and acceptable simulation time.A series of experiments were carried out to verify these properties from different *** results show that SimHOEPI can generate simulation SNP data independently with high-order epistasis models,implying that it might be an alternative simulator for gWAS.

Background: Rhinoplasty is a complex surgical procedure that requires critical analysis and precise design before surgery, making it a challenging operation for both the surgical team and medical educators. This study aimed to evaluate the impact of 3D design involvement on learning curves and to establish a more effective method for rhinoplasty ***: Surgeons who participated in an educational program were divided into two groups. The experimental group was involved in the 3D design before the operation, and the control group was asked to review the rhinoplasty atlas. A self-assessment questionnaire was used to evaluate the learning curve of the eight rhinoplasty procedures for each surgeon, and the overall satisfaction rate data were also ***: The self-assessment scores in both groups showed an increasing trend from the first to the eighth operation. The mean scores of the experimental group were significantly higher than those of the control group at the fifth operation(P=0.01). The satisfaction rate of the experimental group(91.7%) was higher than that of the control group(54.5%).Conclusion: The 3D imaging system can improve the learning curve and satisfaction rate of rhinoplasty education,proving that it is an easy and effective tool for medical education.

Enhancement of the light-absorption response and utilization of the photogenerated carriers represent a robust strategy for the design of high-performance *** this work,grafting Co_(x)P nanoclusters onto S-scheme heterojunction of W18O49/ZnIn2S4(WO/ZIS-CoxP)with strong response to the ultraviolet-visible-near infrared ray(UV-vis-NIR)region has been achieved,which possesses efficient electron-transfer-channel,and boosts charge-separation and transport *** as-prepared WO/ZIS-Co_(x)P yields an impressive solar-driven hydrogen production rate of 45 mmol·g^(-1)·h^(-1).The increased photocatalytic performance is attributed to the synergistic effect of the composite catalyst:(1)The local surface plasmon resonance-induced“hot electron”injection of W_(18)O_(49)significantly increases the electron density;(2)the engineered S-scheme directional electron transfer promotes charge separation and enhances the reducing capability of photoexcited electrons;and(3)Co_(x)P as electron-trap site for accelerating surface proton reduction *** work provides a platform to impart nonprecious co-catalyst for engineering S-scheme heterojunction,serving a class of efficient solar-driven photocatalyst towards hydrogen production.

Background: Early detection of esophageal squamous cell carcinoma (ESCC) can considerably improve the prognosis of patients. Aberrant cell-free DNA (cfDNA) methylation signatures are a promising tool for detecting ESCC. However, available markers based on cell-free DNA methylation are still inadequate. This study aimed to identify ESCC-specific cfDNA methylation markers and evaluate the diagnostic performance in the early detection of ***: We performed whole-genome bisulfite sequencing (WgBS) for 24 ESCC tissues and their normal adjacent tissues. Based on the WgBS data, we identified 21,469,837 eligible Cpg sites (Cpgs). By integrating several methylation datasets, we identified several promising ESCC-specific cell-free DNA methylation markers. Finally, we developed a dual-marker panel based on methylatedKCNA3 andOTOP2, and then, we evaluated its performance in our training and validation ***: The ESCC diagnostic model constructed based onKCNA3 andOTOP2 had an AUC of 0.91 [95% CI: 0.85–0.95], and an optimal sensitivity and specificity of 84.91% and 94.32%, respectively, in the training cohort. In the independent validation cohort, the AUC was 0.88 [95% CI: 0.83–0.92], along with an optimal sensitivity of 81.5% and specificity of 92.9%. The model sensitivity for stage I–II ESCC was 78.4%, which was slightly lower than the sensitivity of the model (85.7%) for stage III–IV ***: The dual-target panel based on cfDNA showed excellent performance for detecting ESCC and might be an alternative strategy for screening ESCC.

Objective Intestinal fibrosis is a refractory complication of inflammatory bowel disease(IBD).Tumor necrosis factor ligand-related molecule-1A(TL1A)is important for IBD-related intestinal fibrosis in a dextran sodium sulfate(DSS)-induced experimental colitis *** study aimed to explore the effects of TL1A on human colonic *** A trinitrobenzene sulfonic acid(TNBS)-induced experimental colitis model of LCK-CD2-TL1A-gFP transgenic(Tg)or wild-type(WT)mice was established to determine the effect and mechanism of TL1A on intestinal *** human colonic fibroblast CCD-18Co cell line was treated concurrently with TL1A and human peripheral blood mononuclear cell(PBMC)*** proliferation and activation of CCD-18Co cells were detected by BrdU assays,flow cytometry,immunocytochemistry and Western *** metabolism was tested by Western blotting and real-time quantitative polymerase chain reaction(RT-qPCR).Results The level of collagen metabolism in the TNBS+ethyl alcohol(EtOH)/Tg group was greater than that in the TNBS+EtOH/WT *** growth factor-β1(TgF-β1)and p-Smad3 in the TNBS+EtOH/Tg group were upregulated as compared with those in the TNBS+EtOH/WT *** proliferation of CCD-18Co cells was promoted by the addition of human PBMC supernatant supplemented with 20 ng/mL TL1A,and the addition of human PBMC supernatant and TL1A increased CCD-18Co proliferation by 24.4%at 24 ***1A promoted cell activation and increased the levels of COL1A2,COL3A1,and TIMP-1 in CCD-18Co *** of CCD-18Co cells with TL1A increased the expression of TgF-β1 and *** TL1A promotes TgF-β1-mediated intestinal fibroblast activation,proliferation,and collagen deposition and is likely related to an increase in the TgF-β1/Smad3 signaling pathway.

Cooperation among enterprises can bring overall and individual performance improvement,and a smooth coordination method is ***,due to the lack of customized coordination methods,cooperation in the downstream oil supply chain cannot be carried out *** paper intends to propose a multi-party coordination method to promote cooperation between oil shippers and pipeline operator by optimizing oil transportation,oil substitution and pipeline pricing *** integrated game-theoretic modeling and analysis approach is developed to characterize the operation behaviors of all stakeholders in the downstream oil supply *** proposed mixed integer nonlinear programming model constrains supply and demand capacity,transportation routes,oil substitution rules and pipeline freight *** transformation and price discretization are introduced for model linear *** experiments are carried out in the oil distribution system in South *** results show that compared to the business-as-usual scheme,the new scheme saves transportation cost by 3.48%,increases pipeline turnover by 5.7%,and reduces energy consumption and emissions by 7.66%and 6.77%.It is proved that the proposed method improves the revenue of the whole system,achieves fair revenue distribution,and also improves the energy and environmental benefits of the oil supply chain.

In this paper,high-energy Ne ions were used to irradiate Zr_(63.5)Cu_(23)Al_(9)Fe_(4.5) metallic glass(Mg)and crystalline W to investigate their difference in mechanical response after *** results showed that with the irradiation dose increased,the tensile micro-strain increased,nano-hardness increased from 7.11 gPa to 7.90 gPa and 8.62 gPa,Young’s modulus increased,and H3/E2 increased which indicating that the plastic deformability decreased in crystalline *** the same irradiation conditions,the Zr_(63.5)Cu_(23)Al_(9)Fe_(4.5) Mg still maintained the amorphous structure and became more disordered despite the longer range and stronger displacement damage of Ne ions in Zr_(63.5)Cu_(23)Al_(9)Fe_(4.5) Mg than in crystalline *** the irradiation hardening and embrittlement behavior of crystalline W,Zr_(63.5)Cu_(23)Al_(9)Fe_(4.5) Mg showed the gradual decrease in hardness from 6.02 gPa to 5.89 gPa and 5.50 gPa,the decrease in modulus and the increase in plastic deformability with the increasing ***,the irradiation softening and toughening phenomenon of Zr_(63.5)Cu_(23)Al_(9)Fe_(4.5) Mg could provide new ideas for the design of nuclear materials.

Magnesium ion batteries(MIBs)are a potential field for the energy storage of the future but are restricted by insufficient rate capability and rapid capacity ***-sodium hybrid ion batteries(MSHBs)are an effective way to address these ***,we report a new type of MSHBs that use layered sodium vanadate((Na,Mn)V_(8)O_(20)·5H_(2)O,Mn-NVO)cathodes coupled with an organic 3,4,9,10-perylenetetracarboxylic diimide(PTCDI)anode in Mg^(2+)/Na^(+)hybrid *** electrochemical cycling,Mg^(2+)and Na^(+)co-participate in the cathode reactions,and the introduction of Na^(+)promotes the structural stability of the Mn-NVO cathode,as cleared by several ex-situ ***,the Mn-NVO cathode presents great specific capacity(249.9 mA h g^(−1)at 300 mA g^(−1))and cycling(1500 cycles at 1500 mA g^(−1))in the Mg^(2+)/Na^(+)hybrid ***,full battery displays long lifespan with 10,000 cycles at 1000 mA g^(−1).The rate performance and cycling stability of MSHBs have been improved by an economical and scalable method,and the mechanism for these improvements is discussed.

The autotetraploid Carassius auratus(4nRR,4n=200,RRRR)is derived from whole-genome duplication of Carassius auratus red var.(RCC,2n=100,RR).In the current study,we demonstrated that chromatophores and pigment changes directly caused the coloration and variation of 4nRR skin(red in RCC,brownish-yellow in4nRR).To further explore the molecular mechanisms underlying coloration formation and variation in 4nRR,we performed transcriptome profiling and molecular functional verification in RCC and *** revealed that scarb1,associated with carotenoid metabolism,underwent significant down-regulation in *** editing of this candidate pigment gene provided clear evidence of its significant role in RCC ***,we identified four divergent scarb1 homeologs in 4nRR:two original scarb1 homeologs from RCC and two duplicated ***,three of these homeologs possessed two highly conserved alleles,exhibiting biased and allelespecific expression in the ***,after precise editing of both the original and duplicated scarb1homeologs and/or alleles,4nRR individuals,whether singly or multiply mutated,displayed a transition from brownishyellow skin to a cyan-gray ***,the proportional areas of the cyan-gray regions displayed a gene-dose *** findings illustrate the subfunctionalization of duplicated scarb1,with all scarb1genes synergistically and equally contributing to the pigmentation of *** is the first report concerning the functional differentiation of duplicated homeologs in an autopolyploidfish,substantiallyenrichingour understanding of coloration formation and change within this group of organisms.
