It is widely known that intestinal capacities such as the enzymatic hydrolysis of carbohydrates,lipids and proteins,and the subsequent absorption of the hydrolyzed *** evolutionary matched to dietary loads and feeding *** this study,we demonstrate that the protein expression of apically located sodium-dependent glucose cotransporter-1(SgLT-1)throughout rat ontogeny is daily adjusted to afford glucose uptake when the load of this metabolically essential monosaccharide in the intestinal lumen is *** jejunal expression of SgLT-1 protein in 14 one-day-old suckling pups was found to increase at dark and early light phase(Pg21-d-old and juvenile 28-d-old rats,the cotransporter expression was high throughout the entire day(PgLT-1 protein(Pght and dark phase when the highest intestinal glucose load was *** our knowledge,these results are the first reporting the daily profile of SgLT-1 expression during rat early developmental stage and may contribute to understand the biological significance of a well-established molecular capacity to deal with the crucial increase of glucose load in the diet during the weaning process.

The occurrence of obesity has increased across the whole world. Many epidemiological studies have indicated that obesity strongly contributes to the development of cancer, cardiovascular diseases, type 2 diabetes, liver diseases and other disorders, accounting for a heavy burden on the public and on health-care systems every year. Excess energy uptake induces adipocyte hypertrophy, hyperplasia and formation of visceral fat in other non-adipose tissues to evoke cardiovascular disease, liver diseases. Adipose tissue can also secrete adipokines and inflammatory cytokines to affect the local microenvironment,induce insulin resistance, hyperglycemia, and activate associated inflammatory signaling pathways. This further exacerbates the development and progression of obesity-associated diseases. Although some progress in the treatment of obesity has been achieved in preclinical and clinical studies, the progression and pathogenesis of obesity-induced diseases are complex and unclear. We still need to understand their links to better guide the treatment of obesity and associated diseases. In this review, we review the links between obesity and other diseases, with a view to improve the future management and treatment of obesity and its co-morbidities.

Comprehensive evaluation of the metabolic effects of porcine CRTC3 overexpression on subcutaneous adipocytes with metabolomic and transcriptomic analyses

Background:Meat quality is largely driven by fat deposition,which is regulated by several genes and signaling *** cyclic adenosine monophosphate(cAMP)-regulated transcriptional coactivator 3(CRTC3)is a coactivator of cAMP response element binding protein(CREB)that mediates the function of protein kinase A(PKA)signaling pathway and is involved in various biological processes including lipid and energy ***,the effects of CRTC3 on the metabolome and transcriptome of porcine subcutaneous adipocytes have not been studied ***,we tested whether porcine CRTC3 expression would be related to fat deposition in Heigai pigs(a local fatty breed in China)and Duroc×Landrace×Yorkshire(DLY,a lean breed)pigs in *** effects of adenovirus-induced CRTC3 overexpression on the metabolomic and transcriptomic profiles of subcutaneous adipocytes were also determined in vitro by performing mass spectrometry-based metabolomics combined with RNA sequencing(RNA-seq).Results:Porcine CRTC3 expression is associated with fat deposition in *** addition,CRTC3 overexpression increased lipid accumulation and the expression of mature adipocyte-related genes in cultured porcine subcutaneous *** to the metabolomic analysis,CRTC3 overexpression induced significant changes in adipocyte lipid,amino acid and nucleotide metabolites in *** RNA-seq analysis suggested that CRTC3 overexpression alters the expression of genes and pathways involved in adipogenesis,fatty acid metabolism and glycerophospholipid metabolism in ***:We identified significant alterations in the metabolite composition and the expression of genes and pathways involved in lipid metabolism in CRTC3-overexpressing *** results suggest that CRTC3 might play an important regulatory role in lipid metabolism and thus affects lipid accumulation in porcine subcutaneous adipocytes.

Silk fibroin(SF)can be used to construct various stiff material interfaces to support bone *** essential preparatory step is to partially transform SF molecules from random coils toβ-sheets to render the material water ***,the influence of the SF conformation on osteogenic cell behavior at the material interface remains ***,three stiff SF substrates were prepared by varying theβ-sheet content(high,medium,and low).The substrates had a comparable chemical composition,surface topography,and *** adsorbed fibronectin was used as a model cellular adhesive protein,the stability of the adsorbed protein-material interface,in terms of the surface stability of the SF substrates and the accompanying fibronectin detachment resistance,increased with the increasingβ-sheet content of the SF ***,(i)larger areas of cytoskeleton-associated focal adhesions,(ii)higher orders of cytoskeletal organization and(iii)more elongated cell spreading were observed for bone marrow-derived mesenchymal stromal cells(BMSCs)cultured on SF substrates with high ***β-sheet contents,along with enhanced nuclear translocation and activation of YAP/TAZ and ***,osteogenic differentiation of BMSCs was stimulated on highβ-sheet *** results indicated that theβ-sheet content influences osteogenic differentiation of BMSCs on SF materials in vitro by modulating the stability of the adsorbed protein-material interface,which proceeds via protein-focal adhesion-cytoskeleton links and subsequent intracellular *** findings emphasize the role of the stability of the adsorbed protein-material interface in cellular mechanotransduction and the perception of stiff SF substrates with differentβ-sheet contents,which should not be overlooked when engineering stiff biomaterials.

An efficient synthesis of novelα-aminophosphonates by the reaction of aldehydes and amines with triethyl phosphite in the presence of the easily available,inexpensive,and nontoxic catalyst thiamine hydrochloride（VB1）.This method affords theα-aminopho-sphonates under the influence of ultrasound irradiation in aqueous medium,in short reaction times（4-6 min）,high yields（85-95%）, with improved *** process is green,mild,inexpensive and excellent yields are the main compensation of this procedure.

In this case report we describe the relationship between ferritin levels and hepcidin in a patient with alcohol-related spur cell anemia who underwent liver *** demonstrate a reciprocal relation-ship between serum or urinary hepcidin and serum ferritin,which indicates that inadequate hepcidin production by the diseased liver is associated with elevated serum *** ferritin level falls with increasing hepcidin production after *** inflammatory indices(IL6)nor erythropoietin appear to be related to hepcidin expression in this *** suggest that inappropriately low hepcidin production by the cirrhotic liver may contribute substantially to elevated tissue iron stores in cirrhosis and speculate that hepcidin replacement in these patients may be of therapeutic benefi t in the future.

Data collection: Rigaku AFC-5R diffractometer,(MoKα, λ=0.71069), ω /2θ scans. θmax=25°, empirical absorption correction, 4658 measured reflections, 4387 independent, 1743 observed reflections [I≥3σ(I)]. Microbial profile and antibiotic sensitivity pattern in bile cultures from endoscopic retrograde cholangiography patients

AIM： To identify the frequency of bacterial growth, the most commonly grown bacteria and their antibiotic susceptibility, and risk factors for bacterial colonization in bile collected from patients with different biliary dis- eases. METHODS： This prospective study was conducted be- tween April 2010 and August 2011. Patients with vari- ous biliary disorders were included. Bile was aspirated by placing a single-use, 5F, standard sphincterotome catheter into the bile duct before the injection of con- trast agent during endoscopic retrograde cholangio- pancreaticography （ERCP）. Bile specimens were trans- ported to the microbiology laboratory in blood culture bottles within an anaerobic transport system. Bacteria were cultured and identified according to the standard protocol used in our clinical microbiology laboratory. The susceptibilities of the organisms recovered were identified using antimicrobial disks, chosen according tothe initial gram stain of the positive cultures RESULTS： Ninety-one patients （27% male, mean age 53.7 4- 17.5 years, range： 17-86 years） were included in the study. The main indication for ERCP was benign biliary disease in 79 patients and malignant disease in 12 patients. The bile culture was positive for bacterial growth in 46 out of 91 （50.5%） patients. The most frequently encountered organisms were gram-negative bacteria including Escherich/a coli （28.2%）, Pseudo- monas （17.3%） and Stenotrophomonas rnaltophilia （15.2%）. There were no significant differences between patients with malignant and benign disease （58% vs 49%, P = 0.474）, patients with acute cholangitis and without acute cholangitis （52.9% vs 50%, P = 0.827）, patients who were empirically administered antibiotics before intervention and not administered （51.4% vs 60.7%, P = 0.384）, with regard to the bacteriobilia. We observed a large covering spectrum or low resistance to meropenem, amikacin and imipenem. CONCLUSION： We did not find a significant risk fac- tor for bacteriobilia in

Surrogate Measures to Assess Mobility of People as a Resilience Indicator in Disaster Management: An Exploratory Study in Southeastern Sri Lanka

Understanding social resilience can assist in the formulation of disaster management policies to help communities better prepare for, respond to, and recover from disasters. However, direct social resilience measurement methods such as household surveys are not always a practical option as they are a time-and resource-exhaustive process. Existing measures mainly utilize publicly available census data, which often provide a poor and outdated assessment of current social resilience status. Another limitation includes a failure to capture multiple facets of indicators that are process-oriented and dynamic in nature such as mobility of people. These challenges can be addressed by employing a surrogate approach. Surrogates are alternative measures to depict the target indicator. The surrogate approach can capture key facets of a target indicator, which can be used as potential measures for the target indicator. A framework to conceptualize the surrogate approach is presented, and operationalized using a case study approach on the southeastern coast of Sri Lanka to identify surrogates to measure mobility of people as a resilience indicator. Six higher-order themes were identi-fied as potential surrogates to measure mobility of people in a disaster context. The approach proposed to methodically identify potential surrogates and their measurement protocols can help to improve the current knowledge base and understanding of complex interrelationships of social resilience.
