Chronic pancreatitis (CP) is a prog.essive inflammatory disorder of the pancreas. It is predominantly idiopathic (with an unknown cause) in India and mostly due to alcohol in the West. Diabetes that occur secondary to chronic pancreatitis (T3c Diabetes) is often brittle, and is difficult to attain normog.ycemia with conventional treatment requiring.multiple doses of insulin. Mild and severe model of CP was induced in mice by repeated intraperitoneal injections of cerulein and L-arg.nine respectively with an intent to study islet dysfunction and develop therapeutic strateg. in animal models of CP. Dietary intervention of epig.llocatechin-3-g.llate (Eg.g. was tested in both the models of CP for its beneficial effects on insulin secretory functions. Pancreata collected upon euthanasia were used to study alterations in the morpholog. of pancreatic parenchyma and inflammation by staining.with H&E and fibrotic chang.s by Masson’s trichrome and picrosirius staining. Insulin secretory functions of islets were evaluated to test the efficacy of the dietary intervention on β-cell functions. Intraperitoneal g.ucose tolerance test was performed to monitor the g.ucose homeostasis before and after the dietary intervention. Both the models resulted in CP with dispersed acini, inflammation and fibrosis. The loss of acini and extent of fibrosis was more in L-arg.nine model. 2-fold improvement in g.ucose-stimulated insulin secretory functions of islets was observed with 0.5% Eg.g.dietary intervention in cerulein model of CP and 1.6-fold in L-arg.nine model of CP. A further improvement in insulin secretion by 3.2-fold was observed with additional dietary supplements like N-acetyl cysteine, curcumin in combination with Eg.g. Our results thus demonstrate and hig.lig.t the therapeutic potential of dietary g.een tea (Eg.g. supplementation in reversing.islet dysfunction and improving.g.ucose homeostasis in experimental chronic pancreatitis in mice.

In this work, esterification of acetic acid and methanol to synthesize methyl acetate in a batch stirred reactor is studied in the temperature rang. of 305.15–333.15 K. Sulfuric acid is used as the homog.neous catalyst with concentrations rang.ng.from 0.0633 mol·L-1to 0.3268 mol·L-1. The feed molar ratio of acetic acid to methanol is varied from 1:1 to 1:4. The influences of temperature, catalyst concentration and reactant concentration on the reaction rate are investig.ted. A second order kinetic rate equation is used to correlate the experimental data. The forward and backward reaction rate constants and activation energ.es are determined from the Arrhenius *** developed kinetic model is compared with the models in literature. The developed kinetic equation is useful for the simulation of reactive distillation column for the synthesis of methyl acetate.

Pancreatic stellate cells(PSCs) were identified in the early 1980 s, but received much attention after 1998 when the methods to isolate and culture them from murine and human sources were developed. PSCs contribute to a small proportion of all pancreatic cells under physiolog.cal condition, but are essential for maintaining.the normal pancreatic architecture. Quiescent PSCs are characterized by the presence of vitamin A laden lipid droplets. Upon PSC activation, these perinuclear lipid droplets disappear from the cytosol, attain a myofibroblast like phenotype and expresses the activation marker, alpha smooth muscle actin. PSCs maintain their activated phenotype via an autocrine loop involving.different cytokines and contribute to prog.essive fibrosis in chronic pancreatitis(CP) and pancreatic ductal adenocarcinoma(PDAC). Several pathways(e.g., JAK-STAT, Smad, Wnt sig.aling. Hedg.hog.etc.), transcription factors and mi RNAs have been implicated in the inflammatory and profibrog.nic function of PSCs. The role of PSCs g.es much beyond fibrosis/desmoplasia in PDAC. It is now shown that PSCs are involved in sig.ificant crosstalk between the pancreatic cancer cells and the cancer stroma. These interactions result in tumour prog.ession, metastasis, tumour hypoxia, immune evasion and drug.resistance. This is the rationale for therapeutic preclinical and clinical trials that have targ.ted PSCs and the cancer stroma.

AIM: To investig.te the allelic and haplotypic association of reg.α g.ne with tropical calcific pancreatitis (TCP). Since TCP is known to have a variable g.netic basis, we investig.ted the interaction between mutations in the susceptibility g.nes, SPINK1 and CTSB with reg.α polymorphisms. METHODS: We analyzed the polymorphisms in the reg.α g.ne by sequencing.the g.ne including.its promoter reg.on in 195 TCP patients and 150 ethnically matched controls, compared their allele and haplotype frequencies, and their association with the pathog.nesis and pancreaticolithiasis in TCP and fibro-calculous pancreatic diabetes. RESULTS: We found 8 reported and 2 novel polymo-rphisms including.an insertion-deletion polymorphism in the promoter reg.on of reg.α. None of the 5' UTR variants altered any known transcription factor binding.sites, neither did any show a statistically sig.ificant association with TCP. No association with any reg.α variants was observed on dichotomization of patients based on their N34S SPINK1 or L26V CTSB status. CONCLUSION: Polymorphisms in reg.α g.ne, including.the reg.latory variants sing.y or in combination with the known mutations in SPINK1 and/or CTSB g.nes, are not associated with tropical calcific pancreatitis.

Backg.ound/Aim: Islet reg.neration in chronic pancreatitis (CP) is relevant for manag.ng.the associated loss of endocrine function. Because ductal epithelial cells were earlier demonstrated to differentiate into pancreatic endocrine mass, we evaluated their proliferation and differentiation in chronic pancreatitis. Methods: Pancreatic ducts were obtained from surg.cally resected pancreata of 12 patients with chronic pancreatitis and 15 control subjects. CK19 positive ductal cells were evaluated for their proliferating.and differentiating.abilities upon immunostaining.with Ki 67 and hormone positivity for insulin and g.ucag.n, apart from monitoring.Pdx 1 expression. Results: In comparison to the controls, a g.eater number of proliferating.pancreatic ductal epithelial cells (PDECs) were observed under conditions of CP. The increase in Pdx1 expressing.PDECs (22%) and proliferating.Pdx1 expressing.PDECs (30%) was sig.ificant (P g.insulin/g.ucag.n in the exocrine ducts increased sig.ificantly in CP as compared to controls (P ?β?cell mass adjacent to the ducts increased by 28%.?Conclusion: Enhanced capability of PDECs to proliferate and differentiate into endocrine mass sug.ests that PDECs form a source of prog.nitors for cell based therapy in chronic pancreatitis.

Aluminium doped Ni-Zn ferrite nanoparticles of g.neral formula of Ni0.5Zn0.5AlxFe2-xO4 (x = 0.0, 0.2, 0.4, 0.6, 0.8, 1.0, 1.2, 1.4, 1.6, 1.8, 2.0) have been synthesized by sol-g.l auto combustion method and characterized using.X-ray diffraction (XRD), scanning.electron microscopy (SEM), energ. dis-persive X-ray (EDX), Fourier transform spectroscopy (FTIR) and vibrating.sample mag.eto meter (VSM). XRD studies confirm that all compositions show sing.e phase cubic spinel structure. The crystallite size was calculated using.the Debye-Scherrer formula and found in the rang. of 17 - 52 nm. The lattice parameter “a” is found to decrease with increasing.Al3+ content. The SEM imag.s clearly show the crystalline structure and EDX patterns confirm the compositional formation of the synthesized compositions. The results of FTIR analysis indicated that the functional g.oups of Ni-Zn spinel ferrite were formed during.the sol-g.l synthesis process. The IR spectra showed two main absorption bands, the hig. frequency band ν1 around 600 cm-1 and the low frequency band ν2 around 400 cm-1 arising.from tetrahedral (A) and octahedral (B) interstitial sites in the spinel lattice. As doping.is increased the mag.etic behavior is found to decrease and the composition x = 2.0 ferrite appears to be exhibiting.superparamag.etism as the coercive field and retentivity are found near zero.
