Expression of the Viral Antigen VP60 in Transgenic Potatoes and its Effect on the Nutritional Composition of Tubers

Recombinant plant-derived pharmaceuticals have been investigated for the last two decades and some products will soon be brought to market. Since veterinary pharmaceuticals seem to be the front-runners of plant-derived vaccines, we selected one model subunit vaccine, the structural capsid protein VP60 against rabbit haemorrhagic disease, and ana-lyzed the expression of three different sequences representing the vp60 open reading frame in potato plants. The gen-eration of antigenic VP60 molecules in the leaf and tuber tissue of potato was tremendously enhanced by replacing virus-derived sequences with plant-optimized codons. In order to identify potentially undesirable alterations in the composition of these genetically modified food components, we studied their nutrient composition and nutritional value in comparison to two parental conventional breeding varieties (Albatros and Desiree). The largest differences in nutrient composition were found between the two conventional breeds and between conventional Desiree and its near-isogenic genetically modified potato plant, indicating that genetic modification as well as conventional breeding can influence nutrient composition. Nevertheless, most parameters of nutritional value seemed to be more affected by conventional breeding than by genetic modification.

Numerical simulation of blood flow and interstitial fluid pressure in solid tumor microcirculation based on tumor-induced angiogenesis

A coupled intravascular-transvascular-interstitial fluid flow model is developed to study the distributions of blood flow and interstitial fluid pressure in solid tumor microcirculation based on a tumor-induced microvascular network. This is generated from a 2D nine-point discrete mathematical model of tumor angiogenesis and contains two parent vessels. Blood flow through the microvascular network and interstitial fluid flow in tumor tissues are performed by the extended Poiseuille＇s law and Darcy＇s law, respectively, transvascular flow is described by Starling＇s law; effects of the vascular permeability and the interstitial hydraulic conductivity are also considered. The simulation results predict the heterogeneous blood supply, interstitial hypertension and low convection on the inside of the tumor, which are consistent with physiological observed facts. These results may provide beneficial information for anti-angiogenesis treatment of tumor and further clinical research.

Abstract The expression of early developmental markers such as doublecortin （DCX） and the polysialylated-neural cell adhesion molecule （PSA-NCAmIp has been used to identify immature neurons within canonical neurogenic niches. Additionally, DCX/PSA-NCAm ＋ immature neurons reside in cortical layer II of the paleocortex and in the paleo- and entorhinal cortex of mice and rats, respectively. These cells are also found in the neocortex of guinea pigs, rabbits, some afrotherian mammals, cats, dogs, non-human primates, and humans. The population of cortical DCX/PSA-NCAm ＋ immature neurons is generated prenatally as conclusively demonstrated in mice, rats, and guinea pigs. Thus, the majority of these cells do not appear to be the product of adult proliferative events. The immature neurons in cortical layer II are most abundant in the corlLices of young individuals, while very few DCX/PSA-NCAm ＋ cortical neurons can be detected in aged mammals. maturation of DCX/PSA-NCAm ＋ cells into glutamatergic and gABAergic neurons has been proposed as an explanation for the age-dependent reduction in their population over time. In this review, we compile the recent information regarding the age-related decrease in the number of cortical DCX/PSA-NCAm ＋ neurons. We compare the distribution and fates of DCX/PSA-NCAm ＋ neurons among mammalian species and speculate their impact on cognitive function. To respond to the diversity of adult neurogenesis research produced over the last number of decades, we close this review by discussing the use and precision of the term ＂adult non-canonical neurogenesis.＂

The monitoring of urban slopes employing in-situ equipment seeks to avoid the loss of human lives midst emergency events like landslides. Cemaden (Brazilian National Centre for monitoring and Early Warnings of Natural Disasters) has a pilot network of soil moisture and rain gauge stations located in urban slopes to understand, prevent, and mitigate landslides. Due to the large volume of data produced by this system there was a need for tools for data retrieval and visualization. This paper describes the Solam (Environmental Data Observation System) software which generates automated graphs that enable researchers to analyze and comprise data from soil moisture sensors and rain gauge stations. Those results allow to research moisture variations in six different sensor levels (from 0.5 m to 3 m deep) and precipitation indexes (hourly and accumulated rainfall) for the established period. This analysis will provide information to set critical environmental thresholds on base recorded landslides.

Calculations of the free energy difference between two states at quantum mechanical(Qm) level directly are generally prohibitively expensive, because some intermediates states are usually required in order to increase the overlap in phase space between two adjacent states. Fortunately, free energy is a state function, of which the difference is path-independent. Therefore, in a more practical way the intermediate states can be described by molecular mechanics(mm). The free energy difference between two adjacent states can be estimated using the Bennett Acceptance Ratio(BAR), which has been shown to give the minimum variance with fixed number of samples. To further reduce the computational expense by refraining from sampling at Qm level for the end states, Boresch proposed a method termed Non-Boltzmann Bennett’s acceptance ratio(NBB), which combines BAR and energy reweighting. However, we showed that the most efficient way to calculate the free energy difference between two Qm states is BAR+TP, which gives the minimum variance of the results. In this scheme, the free energy differences between mm states are estimated using BAR, and at both ends a thermodynamic perturbation from mm to Qm states are applied. In addition, the Qm expense in this scheme is only half of that in NBB. We also show that defining the biasing potential as the difference of the solute-solvent interaction energy, instead of the total energy, can converge to the calculated solvation free energies much faster but possibly to different values. It has also been discovered in this study that BLYP yields better results than mP2 and some later functionals such as B3LYP, m06-2X, and ωB97X-D. This reweighting scheme can also be applied to protein-ligand binding free energy calculations and the corresponding work is in progress.
