Gender-Biased Regulation of Human IL-17-Producing Cells <i>in Vitro</i>by Peptides Corresponding to Distinct <i>HLA-DRB</i>1 Allele-Coded Sequences

Rheumatoid arthritis (RA) is associated with an HLA-DRB1-coded sequence motif called “shared epitope” (SE). To explore potential mechanisms of RA susceptibility, we analyze in vitro effect of peptides bearing different HLA-DR4 sequences on human peripheral blood-derived cells. Three 15-mer peptides were used: 65-79*0401 (HLA-DRB1*04:01- coded sequence SE motif, QKRAA);65-79*0402 (HLA-DRB1*04:02-coded sequence SE-negative motif, DERAA);65-79*0403 (HLA-DRB1*04:03-coded sequence SE-negative motif, QRRAE). We found that CD4 TH17 cells are regulated by peptide treatment with gender bias. In male-derived T cells, all peptide treatments significantly reduced TH17 cell differentiation in vitro when compared to no peptide treatment, and to female samples. TH17 differentiation in samples not treated with peptides, either in the presence or absence of TH17-polarizing cytokines, was higher in males than in females;however, in unfractionated PBMC after treatment with TH17 polarizing cytokines, IL-17A-positive cells were more abundant in females than in males. In addition, SE-positive females showed a significantly higher percentage of IL-17A-positive cells compared to SE-negative females. In conclusion, donor’s SE status and gender may both influence TH17 immune polarization.

＜正＞Embryonic stem cells（ESCs） can self - renew and are pluripotent,meaning that they can proliferate indefinitely in culture while maintaining the ability to differentiate into all cell types,including the cardiomyocytes. During myocardial infarction,significant number of cardiomyocytes is *** can be a potential source of cardiomyocytes for cell replacement *** myocardial infarction,proinflammatory cytokines, including interleukin 1 -beta（IL-1 β ）,interleukin 6（IL-6）,interleukin 10（IL - 10） and interleukin 18（IL- 18） are *** aims of the present study were to investigate the effects of these cytokines on the cardiac differentiation of ESCs,and the mechanisms underlying their effects on cardiac differentiation. The changes of intracellular reactive oxygen species（ROS） level upon cytokine treatment in differentiating embryoid bodies（EBs） were examined by confocal ***-1 β ,IL - 10 and IL - 18 were found to increase the intracellular ROS level in differentiating *** western blotting,ROS were found to enhance cardiac differentiation by increasing the expression of cardiac transcription factors（TFs）,consistent with previous *** addition,ROS was also found to post-translationally modify and activate the cardiac TFs. ROS was found to increase the expression of active phosphorylated TFs while decrease the expression of inactive phosphorylated *** present results showed that some proinflammatory cytokines increased ROS generation in differentiating EBs while ROS increased cardiac differentiation by both increasing the expression and activity of cardiac *** studies would be required to investigate if these cytokines increase cardiac differentiation through increasing ROS.

Objective To investigate the in vivo immunomodulatory and anti-tumor mechanisms of the combined treatment of novel Four-Herb formula(4HF)and doxorubicin in triple-negative breast cancer(TNBC).Methods Murine-derived triple-negative mammary carcinoma cell line,4T1 cells,was cultured and inoculated into mouse mammary ***-six mice were randomly assigned into 6 groups(n=11 in ench):naive,control,LD 4HF(low dose 4HF),HD 4HF(high dose 4HF),LD 4HF+D(low dose and doxorubicin),and D(doxorubicin).Apart from the naive group,each mouse received subcutaneous inoculation with 5×10^(5)4T1 cells resuspended in 100µL of normal saline in the mammary fat *** from the day of tumor cell inoculation,tumors were grown for 6 *** LD and HD groups received daily oral gavage of 658 and 2,630 mg/kg 4HF,*** LD 4HF+D group received daily oral gavage of 658 mg/kg 4HF and weekly intraperitoneal injection of doxorubicin(5 mg/kg).The D group received weekly intraperitoneal injections of doxorubicin(5 mg/kg).The treatment naive mice received daily oral gavage of 0.2 mL double distilled water and 0.1 mL normal saline via intraperitoneal injection once a *** control group received daily oral gavage of 0.2 mL double-distilled *** treatment period was 30 *** the end of treatment,mice organs were harvested to analyze immunological activities via immunophenotyping,gene and multiplex analysis,histological staining,and gut microbiota *** Mice treated with the combination of 4HF and doxorubicin resulted in significantly reduced tumor and spleen burdens(P<0.05),altered the hypoxia and overall immune lymphocyte landscape,and manipulated gut microbiota to favor the anti-tumor immunological ***,immunosuppressive genes,cytokines,and chemokines such as C-C motif chemokine 2 and interleukin-10 of tumors were significantly downregulated(P<0.05).4HF-doxorubicin combination treatment demonstrated synergetic activities and was most effective

目的病人针灸治疗时,漏针可能造成严重后果。为增强针灸安全性,本研究将开发一个基于深度学习的自动云端数针系统。方法本研究利用拍有针灸针的手机图片为曾预先训练的定向目标检测模型(Oriented R-CNN)进行转移学习,以开发一种能够自动计算图片中针灸针数量的目标检测算法。我们首先拍摄了一个包含590张图片的训练集以及一个包含1025张图片的验证集。随后在带有NVIDIA Tesla T4图形处理器(GPU)的Google Colab环境中部署了MMRotate工具箱为模型进行训练。我们将训练的模型与Telegram bot手机小程序界面集成,并评估模型的准确率、精确率和召回率。本小程序中云端数针系统的速度则以端到端处理时间计算。结果在每张图片20根针的验证集情景中,我们的定向检测模型在准确率、精确率和召回率上分别达到96.49%、99.98%和99.84%,平均端到端处理时间为1.535秒。结论这项云服务系统在速度、准确性和可靠性上的提升展示了深度学习物体检测技术改进针灸实践的潜力。

Background:Although overnight fasting is recommended prior to endoscopic retrograde cholangiopancreatography(ERCP),the benefits and safety of high-carbohydrate fluid diet(CFD)intake 2 h before ERCP remain *** study aimed to analyze whether high-CFD intake 2 h before ERCP can be safe and accelerate patients’***:This prospective,multicenter,randomized controlled trial involved 15 tertiary ERCP centers.A total of 1330 patients were randomized into CFD group(n=665)and fasting group(n=665).The CFD group received 400 mL of maltodextrin orally 2 h before ERCP,while the control group abstained from food/water overnight(>6 h)before *** ERCP procedures were performed using deep sedation with intravenous *** investigators were blinded but not the *** primary outcomes included postoperative fatigue and abdominal pain score,and the secondary outcomes included complications and changes in metabolic *** outcomes were analyzed according to a modified intention-to-treat ***:The post-ERCP fatigue scores were significantly lower at 4 h(4.1±2.6 vs.4.8±2.8,t=4.23,P<0.001)and 20 h(2.4±2.1 vs.3.4±2.4,t=7.94,P<0.001)in the CFD group,with least-squares mean differences of 0.48(95%confidence interval[CI]:0.26-0.71,P<0.001)and 0.76(95%CI:0.57-0.95,P<0.001),*** 4-h pain scores(2.1±1.7 vs.2.2±1.7,t=2.60,P=0.009,with a least-squares mean difference of 0.21[95%CI:0.05-0.37])and positive urine ketone levels(7.7%[39/509]vs.15.4%[82/533],χ^(2)=15.13,P<0.001)were lower in the CFD *** CFD group had significantly less cholangitis(2.1%[13/634]vs.4.0%[26/658],χ^(2)=3.99,P=0.046)but not pancreatitis(5.5%[35/634]vs.6.5%[43/658],χ^(2)=0.59,P=0.444).Subgroup analysis revealed that CFD reduced the incidence of complications in patients with native papilla(odds ratio[OR]:0.61,95%CI:0.39-0.95,P=0.028)in the multivariable ***:Ingesting 400 mL of CFD 2 h before ERCP is safe,with a reduction in post-ERCP fatigue,
