Ethanol extract of Herpetospermum caudigerum Wall ameliorates psoriasis-like skin inflammation and promotes degradation of keratinocyte-derived ICAM-1 and CXCL9

Objective:To explore whether the ethanol extract of Herpetospermum caudigerum Wall(EHC),a Xizang medicinal plant traditionally used for treating liver diseases,can improve imiquimod-induced psoriasis-like skin inflammation.Methods:Immunohistochemistry and immunofluorescence staining were used to determine the effects of topical EHC use in vivo on the skin pathology of imiquimod-induced psoriasis in mice.The protein levels of interferon-γ(IFN-γ),tumor necrosis factor-a(TNF-a),and interleukin-17A(IL-17A)in mouse skin samples were examined using immunohistochemical staining.In vitro,IFN-γ-induced HaCaT cells with or without EHC treatment were used to evaluate the expression of keratinocyte-derived intercellular cell adhesion molecule-1(ICAM-1)and chemokine CXC ligand 9(CXCL9)using Western blotting and reverse transcription-quantitative polymerase chain reaction.The protein synthesis inhibitor cycloheximide and proteasome inhibitor MG132 were utilized to validate the EHC-mediated mechanism underlying degradation of ICAM-1 and CXCL9.Results:EHC improved inflammation in the imiquimod-induced psoriasis mouse model and reduced the levels of IFN-γ,TNF-a,and IL-17A in psoriatic lesions.Treatment with EHC also suppressed ICAM-1 and CXCL9 in epidermal keratinocytes.further mechanistic studies revealed that EHC suppressed keratinocyte-derived ICAM-1 and CXCL9 by promoting ubiquitin–proteasome-mediated protein degradation rather than transcriptional repression.Seven primary compounds including ehletianol C,dehydrodiconiferyl alcohol,herpetrione,herpetin,herpetotriol,herpetetrone and herpetetrol were identified from the EHC using ultra-performance liquid chromatography-quadrupole-time of flight-mass spectrometry.Conclusion:Topical application of EHC ameliorates psoriasis-like skin symptoms and improves the inflammation at the lesion sites.

Objective: Psoriasis is a common chronic inflammatory skin disease that is prone to recurrence, and the proinflammatory factor, cysteine-rich protein 61(Cyr61), is important in its pathophysiology. Long-term clinical practice has shown that Sancao Formula(SC), a Chinese herbal compound, is effective in the treatment of psoriasis, but the precise mechanism remains unknown. In this study, we investigate the mechanism by which SC extract alleviates imiquimod(IMQ)-induced psoriasis.Methods: The expression of Cyr61 in psoriatic lesions and normal healthy skin was detected using immunohistochemical analysis to investigate the biological role of Cyr61 in models of psoriatic inflammation. A psoriatic mouse model was established by topical application of IMQ, and the effect of topical application of SC extract was evaluated using the psoriasis area and severity index(PASI) score,hematoxylin-eosin staining, and histopathological features of the skin. Next, a HaCaT cell inflammation model was established using interferon-γ(IFN-γ), and the effect of SC extract on the mRNA and protein levels of Cyr61 and intercellular cell adhesion molecule-1(ICAM-1) was confirmed using Western blot and quantitative real-time polymerase chain reaction analyses.Results: Immunohistochemical staining showed that the expression of Cyr61 in psoriatic lesions was higher than that in normal skin samples(78.26% vs 41.18%, P < 0.05), and the number of Cyr61-positive cells in psoriatic lesions was also significantly higher than in normal skin(18.66 ± 2.51 vs4.33 ± 1.52, P < 0.05). Treatment in mice with IMQ-induced psoriasis showed that SC extract could significantly improve the inflammatory phenotype, PASI score(10.875 ± 0.744 vs 3.875 ± 0.582, P < 0.05),and pathological features compared with those in IMQ model group;SC treatment was also associated with decreased levels of Cyr61 and ICAM-1. Multi-physical fields distribution in billet during helical electromagnetic stirring:A numerical simulation research

Electromagnetic stirring is one of the widely applied techniques to modify the quality of casting billets.Different from conventional rotate stirring,the helical stirring is more professional in assisting multi-dimensional flow of molten metal and eliminating solidification defects.In this study,the single-winding helical stirring(SWHS)was introduced,offering advantages such as smaller volume and lower electromagnetic shielding compared to traditional helical stirring methods.Following a comprehensive numerical simulation,the stirring parameters of SWHS were adjusted to yoke inclination angle of 43°and frequency of 12 Hz.The higher electromagnetic force and flow velocity in drawing direction,as well as the lower temperature gradient induced by the SWHS,are positive factors for homogeneous solidification of billet.The experimental results on Al-8%Si alloy and 0.4%C-1.1%Mn steel demonstrate that compared to rotate stirring,the SWHS process can induce better billet quality and is more effective in accelerating the equiaxed expansion and reducing element segregation.The SWHS process can enhance the equiaxed ratio of the billet by 58.3%and reduce segregation degree of carbon element by 10.97%.Consequently,SWHS holds great promise as a potential approach for improving the quality of continuous casting billets.

A 76-year-old man presented with a two-year history of a slow-growing keratotic tumor on the edge aspect of his right hand ring finger.There was no history of trauma prior to the onset of the lesion.Over the two-year period,the benign projecting tumor has grown to the size of 0.5 x 0.4 x 0.3 cm with occasional pain while be squeezed.The lesion surface is smooth,and the basal epidermal ridges surrounding the node.The patient was scheduled for an excision treatment.The histopathology revealed hyperkeratosis,parakeratosis and acanthosis with thick interwoven bundles of the collagen forming a central core.On the basis of these clinical and histopathologic signs,the tumor was diagnosed as acquired digital fibrokeratoma(ADFK).There has been no recurrence for 6 months since the surgical excision.

A li_(3)P nanoparticle dispersion strengthened ultrathin li metal electrode for high energy density rechargeable batteries

Achievement of lithium(li)metal anode with thin thickness(e.g.,≤30µm)is highly desirable for rechargeable high energy density batteries.However,the fabrication and application of such thin li metal foil electrode remain challenging due to the poor mechanical processibility and inferior electrochemical performance of metallic li.Here,mechanico-chemical synthesis of robust ultrathin li/li_(3)P(LLP)composite foils(~15µm)is demonstrated by employing repeated mechanical rolling/stacking operations using red P and metallic li as raw materials.The in-situ formed li+-conductive li_(3)P nanoparticles in metallic li matrix and their tight bonding strengthen the mechanical durability and enable the successful fabrication of free-standing ultrathin li metal composite foil.Besides,it also reduces the electrochemical li nucleation barrier and homogenizes li plating/stripping behavior.When matching to high-voltage liCoO_(2),the full cell with a low negative/positive(N/P)capacity ratio of~1.5 offers a high energy density of~522 W·h·kg^(-1) at 0.5 C based on the mass of cathode and anode.Taking into account its facile manufacturing,potentially low cost,and good electrochemical performance,we believe that such an ultrathin composite li metal foil design with nanoparticle-dispersion-strengthened mechanism may boost the development of high energy density li metal batteries.

Catalytic anode surface enabling in situ polymerization of gel polymer electrolyte for stable li metal batteries

Employing quasi-solid-state gel polymer electrolyte(GPE)instead of the liquid counterpart has been regarded as a promising strategy for improving the electrochemical performance of li metal batteries.However,the poor and uneven interfacial contact between li metal anode and GPE could cause large interfacial resistance and electrochemical li stripping/plating inhomogeneity,deteriorating the electrochemical performance.Herein,we proposed that the functional component of composite anode could work as the catalyst to promote the in situ polymerization reaction,and we experimentally realized the integration of polymerized-dioxolane electrolyte and li/li_(22)Sn_(5)/liF composite electrode with low interfacial resistance and good stability by in situ catalyzation polymerization.Thus,the reaction kinetics and stability of metallic li anode were significantly enhanced.As a demonstration,symmetric cell using such a GPE-li/li_(22)Sn_(5)/liF integration achieved stable cycling beyond 250 cycles with small potential hysteresis of 25 mV at 1 mA·cm^(−2)and 1 mAh·cm^(−2),far outperforming the counterpart regular GPE on pure li.Paired with liNi0.5Co0.3Mn0.2O2,the full cell with the GPE-li/li_(22)Sn_(5)/liF integration maintained 85.7%of the original capacity after 100 cycles at 0.5 C(1 C=200 mA·g^(−1)).Our research provides a promising strategy for reducing the resistance between GPE and li metal anode,and realizes li metal batteries with enhance electrochemical performance.

Atrophie blanche is a clinically common cutaneous disease which tends to affect women instead of men.It mostly presents with purpura rashes,polymorphic painful ulcer and leave over atrophic scars after recovery.Lower limbs are the most easily affected sites and this disease is easy to relapse again and again.Due to the low incidence of this disease,clinical doctors lack related diagnostic and treating experience about it,which leads to frequent misdiagnosis and misuse of drugs.All of these has brought great pain to patients and financial burden to society.further study and comprehensive literature review is needed to provide more guidance and introduction of Atrophie blanche.

3D Burgers equation is an important model for turbulence.It is natural to expect the long-time behaviour for this hydrodynamics equation.However,there is no result about the long-time behaviour for this deterministic model.Surprisingly,if the system is perturbed by stochastic noise,we establish the existence and uniqueness of invariant measure for 3D stochastic Burgers equation.

A Review of Computing with Spiking Neural Networks

Artificial neural networks(ANNs)have led to landmark changes in many fields,but they still differ significantly fromthemechanisms of real biological neural networks and face problems such as high computing costs,excessive computing power,and so on.Spiking neural networks(SNNs)provide a new approach combined with brain-like science to improve the computational energy efficiency,computational architecture,and biological credibility of current deep learning applications.In the early stage of development,its poor performance hindered the application of SNNs in real-world scenarios.In recent years,SNNs have made great progress in computational performance and practicability compared with the earlier research results,and are continuously producing significant results.Although there are already many pieces of literature on SNNs,there is still a lack of comprehensive review on SNNs from the perspective of improving performance and practicality as well as incorporating the latest research results.Starting from this issue,this paper elaborates on SNNs along the complete usage process of SNNs including network construction,data processing,model training,development,and deployment,aiming to provide more comprehensive and practical guidance to promote the development of SNNs.Therefore,the connotation and development status of SNNcomputing is reviewed systematically and comprehensively from four aspects:composition structure,data set,learning algorithm,software/hardware development platform.Then the development characteristics of SNNs in intelligent computing are summarized,the current challenges of SNNs are discussed and the future development directions are also prospected.Our research shows that in the fields of machine learning and intelligent computing,SNNs have comparable network scale and performance to ANNs and the ability to challenge large datasets and a variety of tasks.The advantages of SNNs over ANNs in terms of energy efficiency and spatial-temporal data processing have been more fully exploited.And the developme

Introduction:Inverse psoriasis is a rare type of psoriasis,especially in children,characterized by special pathogenic sites.Different with vulgaris psoriasis,it usually affects flexural sites like groins,axillas and externalias.As it has a relatively low morbidity,its confused pathogenic sites usually make it hard for doctors to diagnose correctly.Here,we introduce the case of a 9-year-old boy who presented with inverse psoriasis.According to our former clinical experience,we made an accurate diagnosis and the following treatment achieved a significant effect.Similar cases once occurring in the literature on various aspects of inverse psoriasis will also be described.
