Inhibitory effect of norcantharidin on the growth of human gallbladder carcinoma GBC-SD cells in vitro

BACKGROUND: Gallbladder carcinoma is a lethal malignant neoplasm with dismal surgical results. Unfortunately, the adjuvant therapies for gallbladder carcinoma such as chemotherapy and radiotherapy are also disappointing. We reported that norcantharidin (NCTD), a demethylated form of cantharidin, which is an active ingredient of the Chinese medicine Mylabris, was used against human gallbladder carcinoma GBC-SD cells. In the present study, we further studied the mechanism underlying the inhibitory effect of NCTD on growth of human gallbladder carcinoma GBC-SD cells in vitro. METHODS: Human gallbladder carcinoma GBC-SD cells were grown in cell culture and divided into a NCTD group and a control group. The inhibitory effect of NCTD on growth of GBC-SD cells was investigated by evaluation of proliferation, cell cycle, apoptosis and morphological changes of the cells. Cell proliferation was assessed by tetrazolium-based colorimetric assay. The induction of cell cycle arrest and apoptosis was measured by flow cytometry. The morphological changes of the cells were observed by light- and electron-microscopy. To elucidate the anticancer mechanism of NCTD, expression of the proliferation-related gene proteins PCNA, Ki-67, cyclin-D-1 and p27 and the apoptosis-related gene proteins Bcl-2, Bax and Survivin were determined by the streptavidin-biotin complex method and RT-PCR. RESULTS: NCTD inhibited the proliferation of GBCSD cells in a dose- and time-dependent manner, with an IC50 of 56.18 mu g/ml at 48 hours. The flow cytometric profiles revealed that NCTD (at the IC50 for 48 hours) significantly increased the proportion of cells in G(2)/M phase and significantly decreased the proportion of cells in S phase, with a significantly increased rate of cell apoptosis. BACKGROUND: Gallbladder carcinoma, a lethal malignant neoplasm with poor prognosis, has dismal results of surgical resection and chemoradiotherapy. We previously reported that norcantharidin (NCTD) is useful against growth, proliferation, and invasion of human gallbladder carcinoma GBC-SD cells in vitro. In this study, we further studied the inhibitory effect of NCTD on the growth of xenografted tumors of human gallbladder carcinoma in nude mice in vivo and the underlying mechanisms. METHODS: The tumor xenograft model of human gallbladder carcinoma in nude mice in vivo was established with subcutaneous GBC-SD cells. The experimental mice were randomly divided into control, 5-fu, NCTD, and NCTD+5-fu groups which were given different treatments. Tumor growth in terms of size, growth curve, and inhibitory rate was evaluated. Cell cycle, apoptosis, and morphological changes of the xenografted tumors were assessed by flow cytometry and light/electron microscopy. The expression of the cell cycle-related proteins cyclin-D1 and p27 as well as the apoptosis-related proteins Bcl-2, Box, and survivin were determined by the streptavidin-biotin complex (SABC) method and RT-PCR. RESULTS: NCTD inhibited the growth of the xenografted tumors in a dose- and time-dependent manner. Tumor volume decreased (5.61+/-0.39 vs. 9.78+/-0.61 cm(3), P=0.000) with an increased tumor inhibitory rate (42.63% vs. 0%, P=0.012) in the NTCD group compared with the control group. The apoptosis rate increased (15.08+/-1.49% vs. 5.49+/-0.59%, P=0.0001) along with a decreased percentage of cells in S phase (43.47+/-2.83% vs. 69.85+/-1.96%, P=0.0001) in the NTCD group compared with the control group. The morphological changes of apoptosis such as nuclear shrinkage, chromatin aggregation, chromosome condensation, and typical apoptosis bodies in the xenografted tumor cells induced by NCTD were observed by light and electron microscopy. Large-scale toppling slope under water level fluctuation of reservoir:A case of Yunnan Province,China

Landslides induced by reservoir inundation are common in Southwest China,negatively influencing hydropower *** hydropower station dam was constructed in the upper reaches of the Lancang River,accordingly causing the water level at the Lajinshengu slope to increase by 30 m.A tension crack with a visible depth of 8 m was observed in the upper sector of the Lajinshengu slope after reservoir impoundment for 170 *** the following days,numerous cracks appeared on the surface of the slope,and the maximum displacement of the slope reached 3.22 ***,a large-scale active deformation body within the Lajinshengu slope formed with an area of 2.62×10^(5)m^(2)and a volume of 1.65×10^(7)m^(3).Detailed field investigations,on-site monitoring,and centrifugal model tests were carried out to analyze the surface features,deformation characteristics,and failure mechanism of the Lajinshengu *** results show that the slope is an ancient landslide,divided into two parts(*** A and zone B)by the *** B is a traction landslide caused by the displacement of zone *** longterm inundation weakens the soft rock at the slope foot,intensifying the toppling of bedrock and consequently triggering the sliding of the overburden in zone *** failure mode of the Lajinshengu slope is a typical case of toppling-sliding failure,and the underlying rock toppling drives the overlying *** addition,early identification methods for toppling deformation covered by overburdened soil were proposed based on monitoring data and deformation signs.

Characterization and degradation of β-Sn particles in thermal aged Pb-rich solder joint for low-temperature co-fired ceramic(LTCC) applications

High-lead solder joints are still playing an indispensable role in military and space ***, in-depth characterization of high-lead solder joints and the underlying degradation mechanisms remain unexplored. This research first performed aging tests on Sn10Pb90 solder joints, the shear strength at room and elevated temperatures gradually reduced, and the resistance increased. Here, a two-layered Ni–Sn intermetallic compound(IMC) structure was identified using transmission electron microscopy(TEM), which could be attributed to the change of Sn content in the solder. Moreover, the internal annealing twin of a Sn particle was discovered,which could be attributed to creeping induced by thermal expansion coefficient(CTE) difference between Sn and *** analysis of partial and whole annealing twins was conducted through high-resolution TEM(HRTEM).Finally, four degradation mechanisms were *** of the IMC layer would result in increased brittleness and resistivity. For particle coarsening, apart from diminishing the ductility and toughness of the solder joint, it would also accelerate the creeping rate by weakening the phase boundary strength. Regarding voids and cracks induced by phase boundary sliding, wedgeshaped cracking and pore-shaped cracking were discovered and their formation was analyzed. Most importantly, the consumption of Sn resulted in a depletion of wettable layer,leading to the formation of Pb streams and isolated IMC islands, also known as the spalling and delamination of IMCs. Pb diffusion followed a spiral path, which was mutually influenced by orientation misfit and concentration gradient. Study on Static Deflection Model of MEMS Capacitive Microwave Power Sensors

In this paper, a static deflection model of MEMS cantilever beam is proposed, which can better study the force deformation of micro-electro-mechanical system(MEMS) cantilever beam and the output characteristics of capacitive microwave power sensor. The deflection curve is used to describe the deformation of the cantilever beam and then the overload power and sensitivity of this power sensor are derived. It is found that the overload power decreases with the beam length, and increases with the initial height of beam. The sensitivity increases with the beam length, and has a linear growth relationship with the measuring electrode width. A MEMS dual-channel microwave power sensor is designed, fabricated and measured. At a microwave signal frequency of 10 GHz, the sensitivity of the sensor is measured to be 0.11 V/W for the thermoelectric detection channel and 65.17 fF/W for the capacitive detection channel. The sensitivity calculated by the lumped model is 92.93 fF/W, that by the pivot model is 50.88 fF/W,and that by the deflection model proposed in this work is 75.21 fF/W. Therefore, the theoretical result of the static deflection model is more consistent with the measured result and has better accuracy than the traditional lumped model and pivot model.

A self-centering and stiffness-controlled MEMS accelerometer

This paper presents a high-performance MEMS accelerometer with a DC/AC electrostatic stiffness tuning capability based on double-sided parallel plates(DSPPs).DC and AC electrostatic tuning enable the adjustment of the effective stiffness and the calibration of the geometric offset of the proof mass,respectively.A dynamical model of the proposed accelerometer was developed considering both DC/AC electrostatic tuning and the temperature *** on the dynamical model,a self-centering closed loop is proposed for pulling the reference position of the forceto-rebalance(FTR)to the geometric center of *** self-centering accelerometer operates at the optimal reference position by eliminating the temperature drift of the readout circuit and nulling the net electrostatic tuning *** stiffness closed-loop is also incorporated to prevent the pull-in instability of the tuned low-stiffness accelerometer under a dramatic temperature ***-time adjustments of the reference position and the DC tuning voltage are utilized to compensate for the residue temperature drift of the proposed *** a result,a novel controlling approach composed of a self-centering closed loop,stiffness-closed loop,and temperature drift compensation is achieved for the accelerometer,realizing a temperature drift coefficient(TDC)of approximately 7μg/℃ and an Allan bias instability of less than 1μg.

Clinical observation on 90 cases of primary dysmenorrhea treated by buccal acupuncture therapy:a randomized controlled study

OBJECTIVE:To explore the short-term and long-term efficacy of buccal acupuncture therapy in the treatment of patients with primary dysmenorrhea(PD).METHODS:A total of 90 patients with PD who were admitted to the Second Affiliated Hospital of Zhejiang University of Traditional Chinese Medicine from January 2020 to December 2021 were included in this study and randomly divided into three groups,including the observation group,the control group,and the placebo group,with 30 patients in each *** in the observation group were treated according to the syndrome types of Traditional Chinese Medicine,those in the control group were treated with Saridon,and those in the placebo group received a *** visual analog scale(VAS)scores of these patients were evaluated at different time points(before treatment,5 min after needle retention,30 min after needle retention,1 h after treatment,2 h after treatment,1 d after treatment,and 2 d after treatment).The Cox Menstrual Symptom Scale(CMSS)scores were assessed before the treatment and after three courses of ***,these two scales were also evaluated during ***,the pain score and adverse reactions of these patients were assessed during ***:After treatment,the VAS and CMSS scores in the observation group decreased significantly(P<0.05)5 and 30 min after needle retention and after ***:Buccal acupuncture therapy is verified to have short-term efficacy in the treatment of *** addition,this therapy has an immediate analgesic effect and long-term efficacy in reducing the severity and frequency of PD.

A hybrid memory architecture supporting fine-grained data migration

High-frequency Resonance Analysis and Impedance Reshaping Control of MMC-HVDC System Based on Frequency Coupling Impedance Model

In recent years, high-frequency resonance (HFR) events occurred in several modular multilevel converter based high-voltage direct current (MMC-HVDC) projects. The time delay of an MMC-HVDC system is the critical factor that induces HFR. The frequency coupling affects the impedance characteristics of an MMC and further deteriorates system stability. Therefore, in this paper, a multi-input multi-output admittance model of an MMC-HVDC system is developed to analyze its frequency characteristics. The effects of current loop, power loop, phase-locked loop, and operating point on the MMC frequency coupling degree are analyzed in detail. Meanwhile, to further suppress HFR in the MMC-HVDC system, an enhanced impedance reshaping control strategy based on the equivalent single-input single-output impedance model is proposed. Finally, the accuracy of the enhanced impedance model and the effectiveness of the impedance reshaping control are verified by electromagnetic transient simulations in PSCAD.

Low-temperature NH_(3) abatement via selective oxidation over a supported copper catalyst with high Cu^(+) abundance

Selective catalytic NH_(3)-to-N_(2) oxidation(NH_(3)-SCO)is highly promising for abating NH_(3) emissions slipped from stationary flue gas after-treatment *** practical application,however,is limited by the non-availability of low-cost catalysts with high activity and N_(2) ***,using defect-rich nitrogen-doped carbon nanotubes(NCNT-AW)as the support,we developed a highly active and durable copper-based NH_(3)-SCO catalyst with a high abundance of cuprous(Cu^(+))*** obtained Cu/NCNT-AW catalyst demonstrated outstanding activity with a T_(50)(*** temperature to reach 50% NH_(3) conversion)of 174℃ in the NH_(3)-SCO reaction,which outperformed not only the Cu catalyst supported on N-free O-functionalized CNTs(OCNTs)or NCNT with less surface defects,but also those most active Cu catalysts in open *** kinetics measurements and temperature-programmed surface reactions using NH_(3)as a probe molecule revealed that the NH_(3)-SCO reaction on Cu/NCNT-AW follows an internal selective catalytic reaction(i-SCR)route involving nitric oxide(NO)as a key *** to mechanistic investigations by X-ray photoelectron spectroscopy,Raman spectroscopy,and X-ray absorption spectroscopy,the superior NH_(3)-SCO performance of Cu/NCNT-AW originated from a synergy of surface defects and ***,surface defects promoted the anchoring of Cu O nanoparticles on N-containing sites and,thereby,enabled efficient electron transfer from N to Cu O,increasing significantly the fraction of SCR-active Cu^(+)sites in the *** study puts forward a new idea for manipulating and utilizing the interplay of defects and N-dopants on carbon surfaces to fabricate Cu^(+)-rich Cu catalysts for efficient abatement of slip NH_(3)emissions via selective oxidation.
