4-c.umarate ： c.enzyme A Ilgase （4c.） Is one of the key enzymes In phenylpropanoid metabolism leading to series of phenollc., Inc.uding water-soluble phenolic.ac.ds, whic. are important c.mpounds determining the medic.nal quality of Danshen （Salvia miltiorrhiza Bunge）, a traditional c.inese medic.nal herb. To Investigate the func.ion of 4c. in the biosynthesis of water-soluble phenolic.ac.d in Danshen, we have c.oned two c.NAs （Sm4c.1 and Sm4c.2） enc.ding divergent 4c. members by applying nested reverse transc.lptlon-polymerase c.ain reac.ion （RT-Pc.） with degenerate primers followed by 5′/3′rapid amplific.tion of c.NA ends （RAc.）（Note, these sequenc. data have been submitted to the GenBank database under ac.ession numbers AY237163 and AY237164）. Either of the c.ding regions was inserted into a pRSET vec.or and a kinetic.assay was performed with purified rec.mbinant proteins. The substrate utilization profile of Sm4c.1 was distinc. from that of Sm4c.2. The Km values of Sm4c.1 and Sm4c.2 to 4-c.umarlc.ac.d were （72.20±4.10） and （6.50±1.45） μmol/L, respec.ively. These results, In c.njunc.ion with Northern blotting and other information, imply that Sm4c.2 may play an Important role in the biosynthesis of watersoluble phenolic.c.mpounds, whereas Sm4c.1 may play a minor role in the pathway. Southern blotting analysis suggested that both Sm4c.1 and Sm4c.2 genes are present as a single c.py and are loc.ted at different sites In the genome.

We foc.s on the c.assific.tion of pointed p^3-dimensional Hopf algebras H over any algebraic.lly c.osed field of prime c.arac.eristic.p 〉 0. In partic.lar, we c.nsider c.rtain c.ses when the group of grouplike elements is of order p or p^2 that is, when H is pointed but is not c.nnec.ed nor a group algebra. The struc.ures of the assoc.ated graded algebra gr H are c.mpletely desc.ibed as bosonizations of graded braided Hopf algebras over group algebras, and most of the lifting struc.ures of H are given. This work provides many new examples of （parametrized） non-c.mmutative, non-c.c.mmutative finite- dimensional Hopf algebras in positive c.arac.eristic.

c.mparative Genomic.Study Reveals a Transition from TA Ric.ness in Invertebrates to Gc.Ric.ness in Vertebrates at c.G Flank-ing Sites:An Indic.tion for c.ntext-Dependent Mutagenic.ty of Methylated c.G Sites

Vertebrate genomes are c.arac.erized with c.G defic.enc., partic.larly for Gc.oor regions. The Gc.c.ntent-related c.G defic.enc. is probably c.used by c.ntext-dependent deamination of methylated c.G sites. This hypothesis was examined in this study by c.mparing nuc.eotide frequenc.es at c.G flanking positions among invertebrate and vertebrate genomes. The finding is a transition of nuc.eotide preferenc. of 5＇ T to 5＇ A at the invertebrate-vertebrate boundary, indic.ting that a large number of c.G sites with 5＇ Ts were depleted bec.use of global DNA methylation developed in vertebrates. At genome level, we investigated c.G observed/expec.ed （obs/exp） values in 500 bp fragments, and found that higher c.G obs/exp value is shown in Gc.poor regions of invertebrate genomes （exc.pt sea urc.in） but in Gc.ric. sequenc.s of vertebrate genomes. We next c.mpared Gc.c.ntent at c.G flanking positions with genomic.average, showing that the Gc.c.ntent is lower than the average in invertebrate genomes, but higher than that in vertebrate genomes. These results indic.te that although 5＇ T and 5＇ A are different in induc.ng deamination of methylated c.G sites, Gc.c.ntent is even more important in affec.ing the deamination rate. In all the tests, the results of sea urc.in are similar to vertebrates perhaps due to its frac.ional DNA methylation. c.G defic.enc. is therefore suggested to be mainly a result of high mutation rates of methylated c.G sites in Gc.poor regions.
