Counter-regulatory phosphatases TNAP and NPP1 temporally regulate tooth root cementogenesis

Cementum is critical for anchoring the insertion of periodontal ligament fibers to the tooth root. Several aspects of cementogenesis remain unclear, including differences between acellular cementum and cellular cementum, and between cementum and bone. Biomineralization is regulated by the ratio of inorganic phosphate （Pi） to mineral inhibitor pyrophosphate （PPi）, where local Pi and PPi concentrations are controlled by phosphatases including tissue-nonspecific alkaline phosphatase （TNAP） and ectonucleotide pyrophosphatase/phosphodiesterase 1 （NPP1）. The focus of this study was to define the roles of these phosphatases in cementogenesis. TNAP was associated with earliest cementoblasts near forming acellular and cellular cementum. With loss of TNAP in the Alpl null mouse, acellular cementum was inhibited, while cellular cementum production increased, albeit as hypomineralized cementoid. In contrast, NPP1 was detected in cementoblasts after acellular cementum formation, and at low levels around cellular cementum. Loss of NPP1 in the Enppl null mouse increased acellular cementum, with little effect on cellular cementum. Developmental patterns were recapitulated in a mouse model for acellular cementum regeneration, with early TNAP expression and later NPP1 expression. In vitro, cementoblasts expressed Alpl gene/protein early, whereas Enppl gene/protein expression was significantly induced only under mineralization conditions. These patterns were confirmed in human teeth, including widespread TNAP, and NPP1 restricted to cementoblasts lining acellular cementum. These studies suggest that early TNAP expression creates a low PPi environment promoting acellular cementum initiation, while later NPP1 expression increases PPi, restricting acellular cementum apposition. Alterations in PPi have little effect on cellular cementum formation, though matrix mineralization is affected.

Tissue engineering is an emerging field of science that focuses on creating suitable conditions for the regeneration of tissues. The basic components for tissue engineering involve an interactive triad of scaffolds, signaling molecules, and cells. In this context,stem cells(SCs) present the characteristics of selfrenewal and differentiation capacity, which make them promising candidates for tissue engineering. Although they present some common markers, such as cluster of differentiation(CD)105, CD146 and STRO-1, SCs derived from various tissues have different patterns in relation to proliferation, clonogenicity, and differentiation abilities in vitro and in vivo. Tooth-derived tissues have been proposed as an accessible source to obtain SCs with limited morbidity, and various tooth-derived SCs(TDSCs) have been isolated and characterized, such as dental pulp SCs, SCs from human exfoliated deciduous teeth, periodontal ligament SCs, dental follicle progenitor cells, SCs from apical papilla, and periodontal ligament of deciduous teeth SCs. however, heterogeneity among these populations has been observed, and the best method to select the most appropriate TDSCs for regeneration approaches has not yet been established. The objective of this review is to outline the current knowledge concerning the various types of TDSCs, and discuss the perspectives for their use in regenerative approaches.

Investigation of Galactic open cluster remnants： the case of NGC 7193

Galactic open clusters （OCs） that survive the early gas-expulsion phase are gradually de- stroyed over time by the action of disruptive dynamical processes. Their final evolutionary stages are characterized by a poorly populated concentration of stars called an open cluster remnant （OCR）. This study is devoted to assessing the real physical nature of the OCR candidate NGC 7193. GMOS/Gemini spectroscopy of 53 stars in the inner target region were obtained to derive radial velocities and at- mospheric parameters. We also employed photometric and proper motion data. The analysis method consists of the following steps：（i） analysis of the statistical resemblance between the cluster and a set of field samples with respect to the sequences defined in color-magnitude diagrams （CMDs）; （ii） a 5-dimensional iterative exclusion routine was employed to identify outliers from kinematical and posi- tional data; （iii） isochrone fitting to thes× （J - Ks） CMD of the remaining stars and the dispersion of spectral types along empirical sequences in the （J - h） × （h - Ks） diagram were checked. A group of stars was identified for which the mean heliocentric distance is compatible with that obtained via isochrone fitting and whose metallicities are compatible with each other. Fifteen of the member stars observed spectroscopically were identified together with another 19 probable members. Our results indi- cate that NGC 7193 is a genuine OCR, of a once very populous OC, for which the following parameters were derived： d = 501±46 pc, t = 2.5±1.2 Gyr, （[Fe/h]） = -0.17±0.23 and E（B-V） = 0.05±0.05. Its luminosity and mass functions show depletion of low mass stars, confirming the OCR is in a dynam- ically evolved state.

Severity of Leaf Rust and Brown Eyespot in Genotypes of Coffea arabica *** with high Plant Density

This study investigated the severity of leaf rust and brown eyespot in genotypes of Coffea arabica L. cultivated with high plant density in the region of Caparaó-ES. The experiment was conducted in a competition field, cultivated with high plant density (8333 plants per hectare), following a randomized block design, with 16 genotypes and four replications. The plants were evaluated during consecutive harvests to study two complete reproductive cycles (from 2010 to 2012). Data were obtained for the phenologicalstages of flowering, graining, maturation and vegetative rest of each cycle. The severity of leaf rust (hemileia vastratrix) and brown eyespot (Cercospora coffeicola) was evaluated using descriptive scales. It was observed that the genotypes are able to keep a considerable level of resistance to the leaf rust and brown eyespot when cultivated with increased density. The genotypes presented variability regarding the severity of the leaf rust and brown eyespot, indicating the existence of differential levels of resistance between them. For cultivation with high plant density, the genotypes Katipó, Paraíso MG h419-1, h419-3-3-7-16-4-1-1, Araponga MG1, Catucaí Amarelo 24/137, Catiguá MG2, Sacramento MG1, Pau-Brasil MG1, Catiguá MG3, Oeiras MG 6851 and Tupi present higher level of resistance for leaf rust. In addition, the genotypes Paraíso MG h419-1, Catiguá MG2, Pau-Brasil MG1, Catiguá MG3, Oeiras MG 6851, Tupi, Catuaí IAC 44, Catuaí IAC 81 and Catuaí IAC 144 present higher level of resistance for brown eyespot.

AIM To describe racial/ethnic differences in treatment and survival among liver cancer patients in a populationbased cancer *** Invasive cases of primary hepatocellular carcinoma, n = 33270, diagnosed between January 1, 1988-December 31, 2012 and reported to the California Cancer Registry were analyzed by race/ethnicity, age, gender, geographical region, socio-economic status, time period of diagnosis, stage, surgical treatment, and survival. The main outcome measures were receipt of surgical treatment, and cause-specific and all-cause *** After adjustment for socio-demographic characteristics, time period, and stage of disease, compared to Whites, Laotian/hmong [odds ratio(OR) = 0.30, 95%CI: 0.17-0.53], Cambodian(OR = 0.65, 95%CI: 0.45-0.96), AIAN(OR = 0.66, 95%CI: 0.46-0.93), Black(OR = 0.76, 95%CI: 0.67-0.86), and hispanic(OR = 0.78, 95%CI: 0.72-0.84) patients were less likely, whereas Chinese(OR = 1.58, 95%CI: 1.42-1.77), Koreans(OR = 1.45, 95%CI: 1.24-1.70), Japanese(OR = 1.41, 95%CI: 1.15-1.72), and Vietnamese(OR = 1.26, 95%CI: 1.12-1.42) were more likely to receive surgical treatment. AIM:To evaluate the association between helicobacter pylori(***) infection and MLh1 and MGMT methylation and its relationship with microsatellite instability(MSI).METhODS:The methylation status of the MLh1 and MGMT promoter region was analysed by methylation specific methylation-polymerase chain reaction(MSPPCR) in gastric biopsy samples from uninfected or ***-infected children(n = 50),from adults with chronic gastritis(n = 97) and from adults with gastric cancer(n = 92).MLh1 and MGMT mRNA expression were measured by real-time PCR and normalised to a constitutive gene(β actin).MSI analysis was performed by screening MSI markers at 4 loci(Bat-25,Bat-26,D17S250 and D2S123) with PCR;PCR products were analysed by single strand conformation polymorphism followed by silver *** analyses were performed with either the χ 2 test with Yates continuity correction or Fisher's exact test,and statistical significance for expression analysis was assessed using an unpaired Student's ***:Methylation was not detected in the promoter regions of MLh1 and MGMT in gastric biopsy samples from children,regardless of *** infection *** MGMT promoter was methylated in 51% of chronic gastritis adult patients and was associated with *** infection(P his region was methylated in 66% of gastric cancer patients,and the difference in the percentage of methylated samples between these patients and those from ***-infected chronic gastritis patients was statistically significant(P h1 methylation frequencies among ***-infected and non-infected chronic gastritis adult patients were 13% and 7%,*** observed methylation of the MLh1 promoter(39%) and increased MSI levels(68%) in samples from gastric cancer patients in comparison to samples from ***-infected adult chronic gastritis patients(P he frequency of promoter methylation for both genes was higher in gastric cancer sam
