Manipulation of bicarbonate concentration in sperm capacitation media improves in vitro fertilisation output in porcine species

Background: The in vivo concentration of bicarbonate(HCO3-), one of the essential sperm capacitating effectors,varies greatly in the different environments sperm go through from cauda epididymis to the fertilisation site. On the contrary, porcine in vitro sperm capacitation and fertilisation media usually contains a standard concentration of25 mmol/L, and one of the main problems presented is the unacceptable high incidence of polyspermy. This work hypothesised that by modifying the HCO3-concentration of the medium, the output of in vitro sperm capacitation and fertilisation could be increased.Results: Once exposed to the capacitation medium, the intracellular pH(pHi) of spermatozoa increased immediately even at low concentrations of HCO3-, but only extracellular concentrations of and above 15 mmol/L increased the substrates protein kinase A phosphorylation(pPKAs). Although with a significant delay, 15 mmol/L of HCO3-stimulated sperm linear motility and increased other late events in capacitation such as tyrosine phosphorylation(Tyr-P) to levels similar to those obtained with 25 mmol/L. This information allowed the establishment of a new in vitro fertilisation(IvF)system based on the optimization of HCO3-concentration to 15 mmol/L, which led to a 25.3% increment of the viable zygotes(8.6% in the standard system vs. 33.9%).Conclusions: Optimising HCO3-concentrations allows for establishing an IvF method that significantly reduced porcine polyspermy and increased the production of viable zygotes. A concentration of 15 mmol/L of HCO3-in the medium is sufficient to trigger the in vitro sperm capacitation and increase the fertilisation efficiency in porcine.

A new device for deep cervical artificial insemination in gilts reduces the number of sperm per dose without impairing final reproductive performance

Background: The aim of this study was to evaluate the reproductive performance of a new artificial insemination(AI) device specifically designed for gilts(Deep cervical AI, Dp-CAI) by means of which the sperm is deposited deeply in the cervix(8 cm more cranial than in traditional cervical insemination-CAI). New AI techniques have arisen in recent decades in the porcine industry, such as post-cervical artificial insemination(PCAI), which involves depositing the sperm in the body of the uterus [through a catheter(outer tube)-cannula(inner tube)] rather than by CAI. Although the PCAI method has been successfully applied in farm conditions to reduce sperm doses without impairing the reproductive performance, this technique has limitations in gilts mainly because of the difficulty involved in introducing the inner cannula through the cranial part of the cervix. For this reason, the Dp-CAI method described herein may be considered as an alternative to CAI and PCAI methods in gilts.Results: Gilts were divided in two experimental groups: 1) Dp-CAI: gilts(n = 1166) inseminated using 1.5 × 109 sperm/45 m L;2) CAI(as a control group): gilts(n = 130) inseminated using 2.5 × 10~9 sperm/85 mL. The Dp-CAI method was successfully applied in 88.90% of the gilts, with no differences detected between gilts with 1 or 2 previous oestrus cycles, although the catheter could be introduced more deeply in 2 oestrus gilts(P vice that could not be introduced increased(at the moment of insemination), so the success rate of the Dp-CAI device fell, as did the total number of piglets born. When the reproductive output in CAI and Dp-CAI was compared, none of the parameters analysed [pregnancy and farrowing rates(%), and number of piglets born(total and live)] showed significant differences.Conclusions: The use of the Dp-CAI technique provides a new AI method as an alternative to CAI and PCAI for pigs. Background Pediatric cancer survivors are at increased risk of muscle weakness and low areal bone mineral density(aBMD).However,the prevalence of muscle strength deficits is not well documented,and the associations of muscle strength with aBMD are unknown in this population.Therefore,this study aimed to investigate the prevalence of upper-and lower-body muscle strength deficits and to examine the associations of upper-and lower-body muscle strength with age-,sex,and race-specific aBMD Z-scores at the total body,total hip,femoral neck,and lumbar spine.Methods This cross-sectional study included 116 pediatric cancer survivors(12.1±3.3 years old,mean±SD;42.2%female).Upper-and lower-body muscle strength were assessed by handgrip and standing long jump test,respectively.Dual‑energy X‑ray absorptiometry was used to measure aBMD(g/cm2).Associations between muscle strength and aBMD were evaluated in multivariable linear regression models.Logistic regression was used to evaluate the contribution of muscle strength(1-decile lower)to the odds of having low aBMD(Z-score≤1.0).All analyses were adjusted for time from treatment completion,radiotherapy exposure,and body mass index.Results More than one-half of survivors were within the 2 lowest deciles for upper-(56.9%)and lower-body muscle strength(60.0%)in comparison to age-and sex-specific reference values.Muscle strength deficits were associated with lower aBMD Z-scores at all sites(B=0.133–0.258,p=0.001–0.032).Each 1-decile lower in upper-body muscle strength was associated with 30%–95%higher odds of having low aBMD Z-scores at all sites.Each 1-decile lower in lower-body muscle strength was associated with 35%–70%higher odds of having low aBMD Z-scores at total body,total hip,and femoral neck.Conclusion Muscle strength deficits are prevalent in young pediatric cancer survivors,and such deficits are associated with lower aBMD Z-scores at all sites.These results suggest that interventions designed to improve muscle strength in this vulnerable population may have the added ben

Stress tensors and quantification of fracture patterns to analyze connectivity and potential fluid flow in a mesa landform of the Northern Andes

This work applies stress tensors inversions and quantification of fracture patterns along the Mesa de Los Santos, in the Eastern Cordillera of Colombia, to better understand the potential fluid flow. It thus contributes to the conceptual hydrogeological model. The area was subdivided into three blocks, separated by the NW Potreros and the Los Santos faults, having minor inner faults of different orientations. This separation facilitates the analysis of the fractures measured in the field, which in general show high dip angles and a conjugate geometry in the northern block, tension fractures(Mode I) in the central block, and a random distribution in the southwestern block. WinTensor treatment of slickensides yielded a maximum horizontal stress(SHmax) of 111o, which coincides with the WNW-ESE tensor observed from the conjugate and tension joints. We then used Frac Pa Q to generate interpolation maps of fracture intensity and density. The maps show the highest values in the central block and the lowest in the northern block,where the precipitation is higher, causing intensive rock weathering and homogenization of the fracture planes. Although the highest values of connectivity by line are found to the south of the mesa, we suggest the possibility of greater flow from the recharge zone(NE) along bedding planes and open NW-SE fractures.

The emerging role of mesenchymal stem cell-derived extracellular vesicles to ameliorate hippocampal NLRP3 inflammation induced by binge-like ethanol treatment in adolescence

Our previous studies have reported that activation of the NLRP3(NOD-,LRR-and pyrin domain-containing protein 3)-inflammasome complex in ethanol-treated astrocytes and chronic alcohol-fed mice could be associated with neuroinflammation and brain damage.Mesenchymal stem cell-derived extracellular vesicles(MSC-Evs)have been shown to restore the neuroinflammatory response,along with myelin and synaptic structural alterations in the prefrontal cortex,and alleviate cognitive and memory dysfunctions induced by binge-like ethanol treatment in adolescent mice.Considering the therapeutic role of the molecules contained in mesenchymal stem cell-derived extracellular vesicles,the present study analyzed whether the administration of mesenchymal stem cell-derived extracellular vesicles isolated from adipose tissue,which inhibited the activation of the NLRP3 inflammasome,was capable of reducing hippocampal neuroinflammation in adolescent mice treated with binge drinking.We demonstrated that the administration of mesenchymal stem cell-derived extracellular vesicles ameliorated the activation of the hippocampal NLRP3 inflammasome complex and other NLRs inflammasomes(e.g.,pyrin domain-containing 1,caspase recruitment domain-containing 4,and absent in melanoma 2,as well as the alterations in inflammatory genes(interleukin-1β,interleukin-18,inducible nitric oxide synthase,nuclear factor-kappa B,monocyte chemoattractant protein-1,and C–X3–C motif chemokine ligand 1)and miRNAs(miR-21a-5p,miR-146a-5p,and miR-141-5p)induced by binge-like ethanol treatment in adolescent mice.Bioinformatic analysis further revealed the involvement of miR-21a-5p and miR-146a-5p with inflammatory target genes and NOD-like receptor signaling pathways.Taken together,these findings provide novel evidence of the therapeutic potential of MSC-derived Evs to ameliorate the hippocampal neuroinflammatory response associated with NLRP3 inflammasome activation induced by binge drinking in adolescence.

This work shows the synthesis,characterization and evaluation of dense-ceramic membranes made of Ce_(0.85)Gd_(0.15)O_(2-δ)-LaNiO_(3)(CG-LN)composites,where the fluorite-perovskite ratio(CG:LN)was varied as follows:75:25,80:20 and 85:15 wt.%.Supports were initially characterized by XRD,SEM and electrical conductivity(using vacuum and oxygen atmospheres),to determine the composition,microstructural and ionic-electronic conductivity properties.Later,supports were infiltrated with an eutectic carbonates mixture,producing the corresponding dense dual-phase membranes,in which CO_(2)permeation tests were conducted.Here,CO_(2)permeation experiments were performed from 900 to 700℃,in the presence and absence of oxygen(flowed in the sweep membrane side).Results showed that these composites possess high CO_(2)permeation properties,where the O_(2)addition significantly improves the ionic conduction on the sweep membrane side.Specifically,the GC80-LN20 composition presented the best results due to the following physicochemical characteristics:high electronic and ionic conductivity,appropriate porosity,interconnected porous channels,as well as thermal and chemical stabilities between the composite support and carbonate phases.

Personality Trait Detection via Transfer Learning

Personality recognition plays a pivotal role when developing user-centric solutions such as recommender systems or decision support systems across various domains,including education,e-commerce,or human resources.Tra-ditional machine learning techniques have been broadly employed for personality trait identification;nevertheless,the development of new technologies based on deep learning has led to new opportunities to improve their performance.This study focuses on the capabilities of pre-trained language models such as BERT,RoBERTa,ALBERT,ELECTRA,ERNIE,or XLNet,to deal with the task of personality recognition.These models are able to capture structural features from textual content and comprehend a multitude of language facets and complex features such as hierarchical relationships or long-term dependencies.This makes them suitable to classify multi-label personality traits from reviews while mitigating computational costs.The focus of this approach centers on developing an architecture based on different layers able to capture the semantic context and structural features from texts.Moreover,it is able to fine-tune the previous models using the MyPersonality dataset,which comprises 9,917 status updates contributed by 250 Facebook users.These status updates are categorized according to the well-known Big Five personality model,setting the stage for a comprehensive exploration of personality traits.To test the proposal,a set of experiments have been performed using different metrics such as the exact match ratio,hamming loss,zero-one-loss,precision,recall,F1-score,and weighted averages.The results reveal ERNIE is the top-performing model,achieving an exact match ratio of 72.32%,an accuracy rate of 87.17%,and 84.41%of F1-score.The findings demonstrate that the tested models substantially outperform other state-of-the-art studies,enhancing the accuracy by at least 3%and confirming them as powerful tools for personality recognition.These findings represent substantial advancements in personality recognition,making th

Social networks of reintroduced Przewalski's horses in the Great Gobi B Strictly Protected Area (Mongolia)

Analyzing social networks is a powerful tool for understanding the ecology of social species.While most studies focus on the role of each group member,few compare groups with different characteristics.The current population of Przewalski's horses Equus ferus przewalskii at the Great Gobi B Strictly Protected Area(Mongolia)includes groups of wild-born and captive-bred individuals with different experiences with the area(acclimatizing,long-term reintroduced,and wild-born),therefore serving as an ideal natural behavioral lab.We filmed 11 groups for 141.5 hours in summer 2018(July),late spring 2019(May,June),and autumn 2019(September,October).Affiliative and agonistic interactions were recorded,and social networks were created.We tested the influence of origin,experience,season,sex,age,relative time belonging to the group,relat-edness,and dominance rank on different network indices at the individual and group levels.We found that groups with greater experience in the area are generally better connected than members of the newly formed groups.However,these strong networks were created by wild-born individuals with very low interaction rates.On the contrary,inexperienced groups composed of captive-bred individuals displayed many inter actions but created weak social networks.The results show a trend of behavioral transition from acclimatizing through long-term reintroduced to wild-born groups,supporting that the newly formed groups of released Przewalski's horses need time to display the typical social behavior patterns of wild-born individuals.Long-term monitoring of released Przewalski's horses is recommended to promote the success of this reintroduction program.

Structured-illumination reflectance imaging for the evaluation of microorganism contamination in pork: Effects of spectral and imaging features on its prediction performance

Structured-illumination reflectance imaging provides a new means for food quality detection. This original work investigated the capability of Structured-illumination reflectance imaging technique coupled with multivariate chemometrics to evaluate the microbial contamination in pork inoculated with Pseudomonas fluorescens and Brochothrix thermosphacta during storage at different temperatures. The prediction performances based on different spectrum and the textural features of direct component and amplitude component images demodulated from the SIRI pattern images， as well as their data fusion were comprehensively compared. Based on the full wavelength spectrum (420 – 700) nm of amplitude component images， the orthogonal signal correction coupled with support vector machine regression provided the best predictions of the number of Pseudomonas fluorescens and Brochothrix thermosphacta in pork， with the Rp2values of 0.870 and 0.906， respectively. Besides， the prediction models based on the amplitude component or direct component image textural features and the data fusion models using spectrum and textural features from direct component and amplitude component images cannot significantly improve their prediction accuracy. Consequently， structured-illumination reflectance imaging can be further considered as a potential technique for the rapid evaluation of microbial contaminations in pork meat.

Influenza pandemics are unpredictable recurrent events with global health,economic,and social consequences.The objective of this review is to provide an update on the latest developments in early diagnosis and specific treatment of the disease and its complications,particularly with regard to respiratory organ failure.Despite advances in treatment,the rate of mortality in the intensive care unit remains approximately 30%.Therefore,early identification of potentially severe viral pneumonia is extremely important to optimize treatment in these patients.The pathogenesis of influenza virus infection depends on viral virulence and host response.Thus,in some patients,it is associated with an excessive systemic response mediated by an authentic cytokine storm.This process leads to severe primary pneumonia and acute respiratory distress syndrome.Initial prognostication in the emergency department based on comorbidities,vital signs,and biomarkers(e.g.,procalcitonin,ferritin,human leukocyte antigen-DR,mid-regional proadrenomedullin,and lactate)is important.Identification of these biomarkers on admission may facilitate clinical decision-making to determine early admission to the hospital or the intensive care unit.These decisions are reached considering pathophysiological circumstances that are associated with a poor prognosis(e.g.,bacterial co-infection,hyperinflammation,immune paralysis,severe endothelial damage,organ dysfunction,and septic shock).Moreover,early implementation is important to increase treatment efficacy.Based on a limited level of evidence,all current guidelines recommend using oseltamivir in this setting.The possibility of drug resistance should also be considered.Alternative options include other antiviral drugs and combination therapies with monoclonal antibodies.Importantly,it is not recommended to use corticosteroids in the initial treatment of these patients.Furthermore,the implementation of supportive measures for respiratory failure is essential.Current recommendations are limited,heterogeneous,an
