To date,no vaccines or effective drugs have been approved to prevent or treat COVID-19 and the current standard care relies on supportive ***,based on the fast and global spread of the virus,urgent investigations are warranted in order to develop preventive and therapeutic *** this regard,treatments addressing the immunopathology of SARS-CoV-2 infection have become a major ***,while a rapid and well-coordinated immune response represents the first line of defense against viral infection,excessive inflammatory innate response and impaired adaptive host immune defense may lead to tissue damage both at the site of virus entry and at systemic *** studies highlight relevant changes occurring both in innate and adaptive immune system in COVID-19 *** particular,the massive cytokine and chemokine release,the so-called“cytokine storm”,clearly reflects a widespread uncontrolled dysregulation of the host immune *** the prospective of counteracting cytokine storm is compelling,a major limitation relies on the limited understanding of the immune signaling pathways triggered by SARS-CoV-2 *** identification of signaling pathways altered during viral infections may help to unravel the most relevant molecular cascades implicated in biological processes mediating viral infections and to unveil key molecular players that may be ***,given the key role of the immune system in COVID-19,a deeper understanding of the mechanism behind the immune dysregulation might give us clues for the clinical management of the severe cases and for preventing the transition from mild to severe stages.

Background:Platinum chemotherapy(CT)remains the backbone of systemic therapy for patients with smallcell lung cancer(SCLC).The nucleotide excision repair(NER)pathway plays a central role in the repair of the DNA damage exerted by platinum *** in this repair mechanism may affect patients’*** and Methods:We conducted a retrospective analysis of data from 38 patients with extensive disease(ED)-SCLC who underwent platinum-CT at the Clinical Oncology Unit,Careggi University Hospital,Florence(Italy),from 2015 to *** expression analysis and single nucleotide polymorphism(SNP)characterization of three NER pathway genes—namely ERCC1,ERCC2,and ERCC5—were performed on patient tumor ***:Overall,elevated expression of ERCC genes was observed in SCLC patients compared to healthy *** with low ERCC1 and ERCC5 expression levels exhibited a better median progression-free survival(mPFS=7.1 vs.4.9 months,p=0.39 for ERCC1 and mPFS=6.9 vs.4.8 months,p=0.093 for ERCC5)and overall survival(mOS=8.7 vs.6.0 months,p=0.4 for ERCC1 and mOS=7.2 vs.6.2 months,p=0.13 for ERCC5).Genotyping analysis of five SNPs of ERCC genes showed a longer survival in patients harboring the wild-type genotype or the heterozygous variant of the ERCC1 rs11615 SNP(p=0.24 for PFS and p=0.14 for OS)and of the rs13181 and rs1799793 ERCC2 SNPs(p=0.43 and p=0.26 for PFS and p=0.21 and p=0.16 for OS,respectively)compared to patients with homozygous mutant ***:The comprehensive analysis of ERCC gene expression and SNP variants appears to identify patients who derive greater survival benefits from platinum-CT.

Hepatocellular carcinoma(HCC)is a leading cause of cancer-related mortality worldwide,with advanced stages posing significant treatment ***-though hepatic arterial infusion chemotherapy(HAIC)has emerged as a promising modality for treating advanced HCC,particularly in Asian clinical practice,its adoption in Western medicine remains limited due to a lack of large-scale randomized controlled *** editorial reviews and comments on the meta-analysis conducted by Zhou et al,which evaluates the efficacy and safety of HAIC and its combination strategies for advanced *** authors performed a comprehensive meta-analysis of various clinical trials and cohort studies comparing HAIC and its combinations to other first-line treatments,such as sorafenib and transarterial chemoembolization(TACE).In this work,HAIC showed significantly better results regarding overall survival and progression-free survival compared to sorafenib or TACE alone and their *** in combination with lenvatinib,ablation,programmed cell death 1 inhibitors,and radiotherapy further enhanced patient outcomes,indicating a synergistic *** editorial focuses on the critical role of multimodal treatment strategies in managing advanced *** advocates for a paradigm shift towards integrated treatment approaches to enhance survival rates and improve the quality of life in patients with advanced HCC.

There is a need to develop interventions to slow or reverse the degeneration of dopamine neurons in Parkinson’s disease after *** that preclinical and clinical studies suggest benefits of dietary n-3 polyunsaturated fatty acids,such as docosahexaenoic acid,and exercise in Parkinson’s disease,we investigated whether both could synergistically interact to induce recovery of the dopaminergic ***,mice received a unilateral stereotactic injection of 6-hydroxydopamine into the striatum to establish an animal model of nigrostriatal *** weeks after lesion,animals were fed a docosahexaenoic acid-enriched or a control diet for the next 8 *** this period,the animals had access to a running wheel,which they could use or *** acid treatment,voluntary exercise,or the combination of both had no effect on(i)distance traveled in the open field test,(ii)the percentage of contraversive rotations in the apomorphine-induction test or(iii)the number of tyrosine-hydroxylase-positive cells in the substantia nigra pars ***,the docosahexaenoic acid diet increased the number of tyrosine-hydroxylase-positive terminals and induced a rise in dopamine concentrations in the lesioned *** to docosahexaenoic acid treatment or exercise alone,the combination of docosahexaenoic acid and exercise(i)improved forelimb balance in the stepping test,(ii)decreased the striatal DOPAC/dopamine ratio and(iii)led to increased dopamine transporter levels in the lesioned *** present results suggest that the combination of exercise and docosahexaenoic acid may act synergistically in the striatum of mice with a unilateral lesion of the dopaminergic system and provide support for clinical trials combining nutrition and physical exercise in the treatment of Parkinson’s disease.

Potable Water under Pressure: Effects of the Syrian Crisis and Recent Drought on Northwest Syria

The study investigates the impact of the Syrian crisis and the recent drought on the potable water situation in Northwest Syria, comparing various aspects of water availability and quality before and after 2011. 380 key-informants were surveyed, including water-well owners, well-digging companies, water-trucking suppliers, agricultural pharmacies, and service offices within local councils. The surveys covered all nine districts in northwestern Syria across the Aleppo and the Idleb governorates. The survey findings reveal significant shifts in water sources, an almost halving in water availability and per capita consumption, and a notable decrease in water quality. Coping mechanisms include random well drilling and reduced hygiene practices. Water pumping stations face challenges with functionality, and there is a shift towards clean energy sources, particularly solar energy. Challenges identified by the survey respondents include drought, fuel costs, and damaged water networks. The study highlights the urgency of addressing the potable water crisis in Northwest Syria and suggests specific interventions to enhance water sustainability and governance.

The porous-fluid interface encompasses a region bridging the flow inside a porous medium and a free-flowing *** the context of volume-averaged simulations,it can be described by a set of gradually changing parameters defining the porous medium,mainly porosity and *** this paper,both the permeability and the porous-induced drag force are evaluated a-priori,by explicitly filtering a set of Particle-Resolved Simulations(PRS)of the flow in the channel partially occupied by the porous *** porous matrices are considered and the influence of the geometry and filtering parameters on the macroscopic quantities is ***,the focus is placed on the requirements for the kernel type and size to perform filtering accurately,and their impact on the distribution of permeability at the *** performance of the typically used models for the permeability is compared to the explicitly filtered ***,a new model for permeability and the drag force is introduced,taking into account the information about the fltering size and non-uniformity of the velocity *** model greatly improves the prediction of velocity at the porous-fluid interface and serves as a proof of concept that a successful porous drag model should strive to include information about both parameters.
