Soil-derived fungi represent an insufficiently tapped reservoir for discovering new and bioactive natural products(NPs),and despite an ever-increasing number of unknown NPs have been discovered over the past few decades,much of the hidden biosynthetic potential is still in an urgent need to be disclosed.In this research,a chemical inves-tigation was performed on a wetland soil-derived fungus Aspergillus calidoustus TJ403-EL05,leading to the isolation of a total of fourteen drimane sesquiterpenoids(1-14),incorporating three new ones,namely ustusols F-H(1-3).Their structures,comprising absolute configurations,were completely authenticated by widespread spectroscopic data,quantum chemical 13C NMR and ECD calculations,and X-ray crystallography experiments.Compound 14 exhibited moderate anti-inflammatory activity by inhibiting the LPS-induced NO release(IC_(50)=25.6μM).

Hybeanones A and B(1 and 2),two highly oxygenated and rearranged polycyclic polyprenylated acylphloroglucinols(PPAPs),were isolated from the aerial parts of Hypericum beanii.Their structures comprising absolute configurations were elucidated by spectroscopic analysis,single-crystal X-ray diffraction,and quantum chemical calculations.Compounds 1 and 2 are defined by a newly assembled cyclopentanone unit fused to a tricyclic γ-lactone unit via a ketone carbonyl.The breakage of C-1/C-2 linkage via retro-Claisen reaction,attack from C-1 to C-19,and Baeyer-Villiger oxidation at C-1/C-23 bond were presumed to be the key steps in the assembly of 1 and 2.The isolates 1 and 2 showed potential acetylcholinesterase(AchE)inhibitory activities,with IC50 values of 21.34±1.48 and 18.79±2.36μmol/L,respectively.

Aberrant NF-κB activation in odontoblasts orchestrates inflammatory matrix degradation and mineral resorption

Inflammation-associated proteinase functions are key determinants of inflammatory stromal tissues deconstruction.As a specialized inflammatory pathological process,dental internal resorption(IR)includes both soft and hard tissues deconstruction within the dentin-pulp complex,which has been one of the main reasons for inflammatory tooth loss.Mechanisms of inflammatory matrix degradation and tissue resorption in IR are largely unclear.In this study,we used a combination of Cre-loxP reporter,flow cytometry,cell transplantation,and enzyme activities assay to mechanistically investigate the role of regenerative cells,odontoblasts(ODs),in inflammatory mineral resorption and matrices degradation.We report that inflamed ODs have strong capabilities of matrix degradation and tissue resorption.Traditionally,ODs are regarded as hard-tissue regenerative cells;however,our data unexpectedly present ODs as a crucial population that participates in IR-associated tissue deconstruction.Specifically,we uncovered that nuclear factor-kappa b(NF-κB)signaling orchestrated Tumor necrosis factorα(TNF-α)-induced matrix metalloproteinases(Mmps)and Cathepsin K(Ctsk)functions in ODs to enhance matrix degradation and tissue resorption.Furthermore,TNF-αincreases Rankl/Opg ratio in ODs via NF-κB signaling by impairing Opg expression but increasing Rankl level,which utterly makes ODs cell line 17IIA11(A11)become Trap^(+) and Ctsk^(+) multinucleated cells to perform resorptive actions.Blocking of NF-κB signaling significantly rescues matrix degradation and resorptive functions of inflamed ODs via repressing vital inflammatory proteinases Mmps and Ctsk.Utterly,via utilizing NF-κB specific small molecule inhibitors we satisfactorily attenuated inflammatory ODs-associated human dental IR in vivo.Our data reveal the underlying mechanisms of inflammatory matrix degradation and resorption via proteinase activities in IR-related pathological conditions.

Nanofibers with high specific surface area and chemical stability have broad prospects in the applications of adsorption.However,the adsorption capacity is limited by the scarcity of adsorption groups and storage space.Herein,the activated carbon-hybridized and amine-modified nanofibers are prepared by integrating activated carbon(AC)and polyacrylonitrile(PAN)via electrospinning method and the subsequent amination,which could provide additional storage space and adsorption groups for ultrahigh adsorption capability.Thus,the obtained amine-rich porous PAN nanofibers(APAN/AC)readily realized the ultrahigh adsorption capacity for metal ions and dyes in wastewater.Specifically,the adsorption capacity of APAN/AC nanofibers were 284 mg·g^(-1) for Cr(VI)and 248 mg·g^(-1) for methyl orange,which were almost 2 and 4 times than that of amine-modified nanofibers(APAN)and carbon-hybridized nanofibers(PAN/AC),respectively.Moreover,the AC inhibited the chain mobility of polymer matrix and thereby endowing APAN/AC nanofibers with excellent recyclability.The adsorption capability retained 80%after nine adsorption-desorption cycles.The adsorption kinetics and corresponding mechanism were further explored.This strategy combines the advantages of polymer nanofibers and AC,opening a new avenue for developing next-generation absorbent materials.

OBJECTIVE: To investigate the effects of Yiqibushenhuoxue decoction on stable chronic obstructive pulmonary disease(COPD) by observing its influences on patients' quality of life and airway inflammation.METHODS: Seventy patients with stable COPD were randomly divided into a treatment group(n=35) treated with Yiqibushenhuoxue decoction plus Seretide and a control group(n=35) treated with Seretide only.The dosage of Yiqibushenhuoxue decoction was 100 mL each time, twice a day, and the dosage of Seretide was salmeterol 50 μg/fluticasone 250 μg twice a day. Both groups were treated for 12 weeks. Before and after the treatment, St George's respiratory disease questionnaire(SGRQ)scores,forced expiratory volume,and forced expiratory volume in 1 second/forced vital capacity(FEV1/FVC)were measured.RESULTS:The SGRQ scores in both groups were significantly lower than those before treatment(P0.05).CONCLUSION: Yiqibushenhuoxue decoction could significantly decrease the SGRQ scores in patients with stable COPD, which suggests that it is able to improve patient symptoms.

Orbitofrontal cortex action of 5-hydroxytryptamine and its receptor in an acute forced swimming stress-induced depression model

BACKGROUND： The orbitofrontal cortex （OFC） is a brain region closely associated with emotion. 5-hydroxytryptamine （5-HT） has been shown to be involved in human depression. OBJECTIVE： To investigate OFC actions and mechanisms of 5-HT and 5-HT1A receptor （5-HT1AR） in stress-induced depression.DESIGN, TIME AND SETTING： A randomized, controlled, animal experiment was performed at Laboratory of Neurobiology, College of life Science, Shaanxi Normal University between May 2006 and March 2008. MATERIALS： 5-HT, p-chlorophenylalanine （PCPA, an inhibitor to tryptophan hydroxylase） and spiperone （5-HT1AR antagonist） were provided by Sigma, USA; rabbit anti-rat 5-HT1AR antibody was provided by Tianjin Haoyang Biological Manufacture. METHODS： A total of 40 male Sprague Dawley rats, aged 3 months, were randomly divided into five groups： control, model, 5-HT, spiperone ＋ 5-HT, and PCPA, with 8 rats in each group. Except for control group, rats in the other four groups were used to establish depression models by forced swimming for 15 minutes. At 30 minutes before forced swimming test, 0.5 pL of 5-HT （12.5 pg/pL）, PCPA （20 pg/pL）, spiperone （1.3 pg/pL）＋ 5-HT （12.5 pg/pL, 10 minutes later）, and saline were respectively injected into the OFC of 5-HT, PCPA, spiperone ＋ 5-HT, and model groups, respectively. The control group received a saline microinjection into the OFC.MAIN OUTCOME MEASURES： Forced swimming and open field tests were employed to measure animal behaviors, and immunohistochemistry was used to analyze 5-HT1AR expression in the OFC, cingulate cortex, and piriform cortex. RESULTS：（1） Compared with the model group, 5-HT microinjection into the OFC prominently reduced immobility time in the forced swimming test and rearing in open field test （P 〈 0.05）; locomotion and grooming in open field test were increased, although there was no significance （P 〉 0.05）. Furthermore, following PCPA microinjection into the OFC （PCPA ＋ forced swimming stress）, immobility time in forced swimming test increased dramatically （P〈 0

Photosynthetic species belonging to Symbiodiniaceae play important ecological roles in coral reef ecosystems. Nine phylogenetically supported Symbiodiniaceae clades(each containing multiple genetically distinct molecular types) have been detected in reef-associated animals and in seawater or sediments near coral reef areas. Although major advances in research on Symbiodiniaceae diversity have been made, information on the global-scale diversity and distribution characteristics of Symbiodiniaceae clades/types is still limited or must be expanded because most studies on Symbiodiniaceae have mainly focused on a limited number of hosts associated with Symbiodiniaceae or coral reef areas. Here, the diversity and distribution characteristics of Symbiodiniaceae data derived from reef-associated animals and seawater or sediments near coral reef areas were analyzed using 3899 qualified Symbiodiniaceae internal transcribed spacer 2(ITS2) sequences retrieved from the GenBank database in May 2017. These sequences were annotated as Symbiodiniaceae ITS2 types via sequence alignment against database of Symbiodiniaceae ITS2 types. Results showed that photosynthetic Symbiodiniaceae ITS2 sequences could be annotated as 119 Symbiodiniaceae ITS2 types within nine clades(A–I), and Symbiodiniaceae ITS2 types mainly inhabited coral reefs between 30?N and 30?S. Symbiodiniaceae ITS2 types could be associated with multiple host taxa. Interestingly, these results implied that the distribution of Symbiodiniaceae ITS2 types was distinct in the Atlantic Ocean, the Red Sea, the Mediterranean Sea, and the Indo-Pacific Ocean. Although future field research should be performed to verify our results, this study provided a broad perspective of the diversity of Symbiodiniaceae clades/types on a global scale.

Photoelectrochemical(PEC) biosensors have shown great promise in bioanalysis and diagnostic applications in recent years. In this work, the CuO/Cu2O nanowire array(CuO/Cu2O Nanowire) supported on copper foam was prepared as a photocathode for detection of tyrosinase though quinone-chitosan conjugation chemistry method. The in-situ generated quinones that were the catalytic product of tyrosinase acted as electron acceptors, which were captured by the chitosan deposited on the surface of the electrode. Direct immobilization of electron acceptor on the electrode surface improved the photocurrent conversion efficiency and thus sensitivity. The as-prepared biosensor can realize a rapid response in a wide linear range of 0.05 U/mL to 10 U/mL with the detection limit as low as 0.016 U/mL of tyrosinase. The current work provides a new perspective to design and develop highly sensitive and selective PEC biosensor.

Purpose The magnet power supply system of HEPS mainly includes the linear magnet power supply,the booster power supply,the transportation line magnet power supply and the storage ring magnet power supply.According to the physical design requirements,the output current of the booster magnet power supply is a current with a DC bias period of 1 Hz,and the waveform can be a sine wave or a trapezoidal wave(Jiang et al.Sci Sin Phys Mech Astron 44(10):1075-1094,2014).The dipole magnet of the booster should be powered in series,the inductance value is relatively large(about 160 mH),and the power of the power supply reaches several hundred kilowatts.Methods The three-phase PWM rectifier controls the orderly turn-on of power electronic devices and takes the input voltage phase of the power grid as the command current phase to make the input current waveform follow the power grid voltage,so as to achieve the purpose of improving the power factor.In this paper,the PWM rectifier is modeled and simulated,and based on the simulation,an experimental prototype is built.Results The experimental results of the power supply prototype of the PWM rectifier show that the PWM rectifier can achieve high power factor operation on the grid side(Akagi and Sato,in:Control and performance of a flywheel energy storage system based on a doubly-fed induction generator-motor for power conditioning,1999).Conclusions The PWM rectifier can be used in the power supply of the booster and has a very good application prospect.
