Viral vectored vaccines:design,development,preventive and therapeutic applications in human diseases

Human diseases,particularly infectious diseases and cancers,pose unprecedented challenges to public health security and the global *** development and distribution of novel prophylactic and therapeutic vaccines are the prioritized countermeasures of human *** all vaccine platforms,viral vector vaccines offer distinguished advantages and represent prominent choices for pathogens that have hampered control efforts based on conventional vaccine ***,viral vector vaccines remain one of the best strategies for induction of robust humoral and cellular immunity against human *** viruses of different families and origins,including vesicular stomatitis virus,rabies virus,parainfluenza virus,measles virus,Newcastle disease virus,influenza virus,adenovirus and poxvirus,are deemed to be prominent viral vectors that differ in structural characteristics,design strategy,antigen presentation capability,immunogenicity and protective *** review summarized the overall profile of the design strategies,progress in advance and steps taken to address barriers to the deployment of these viral vector vaccines,simultaneously highlighting their potential for mucosal delivery,therapeutic application in cancer as well as other key aspects concerning the rational application of these viral vector *** and accurate technological advances in viral vector vaccines would consolidate their position as a leading approach to accelerate breakthroughs in novel vaccines and facilitate a rapid response to public health emergencies.

Acute liver failure in Chinese children:a multicenter investigation

BACKGROUND: Currently, no documentation is available regarding Chinese children with acute liver failure(ALF). This study was undertaken to investigate etiologies and outcomes of Chinese children with ***: We retrospectively enrolled 32 pediatric patients with ALF admitted in five hospitals in different areas of China from January 2007 to December 2012. The coagulation indices, serum creatinine, serum lactate dehydrogenase,blood ammonia and prothrombin activity were analyzed; the relationship between these indices and mortality was evaluated by multivariate ***: The most common causes of Chinese children with ALF were indeterminate etiology(15/32), drug toxicity(8/32),and acute cytomegalovirus hepatitis(6/32). Only 1 patient(3.13%)received liver transplantation and the spontaneous mortality of Chinese children with ALF was 58.06%(18/31). Patients who eventually died had higher baseline levels of international normalized ratio(P=0.01), serum creatinine(P=0.04), serum lactate dehydrogenase(P=0.01), blood ammonia(P<0.01) and lower prothrombin activity(P=0.01) than those who *** analysis showed that the entry blood ammonia was the only independent factor significantly associated with mortality(odds ratio=1.069, 95% confidence interval 1.023-1.117,P<0.01) and it had a sensitivity of 94.74%, a specificity of 84.62%and an accuracy of 90.63% for predicting the death. Based on the established model, with an increase of blood ammonia level,the risk of mortality would increase by 6.9%.CONCLUSIONS: The indeterminate causes predominated in the etiologies of ALF in Chinese children. The spontaneous mortality of pediatric patients with ALF was high, whereas the proportion of patients undergoing liver transplantation was significantly low. Entry blood ammonia was a reliable predictor for the death of pediatric patients with ALF.

We study the ultrafast solvation dynamics of protein-precipitant *** subtilisin carlsberg(SC)was mixed with several polyethylene glycol(PEG)precipitants for protein ***-resolved emission spectra from single intrinsic tryptophan residue(Trp-113)are recorded to construct solvation correlation *** precipitant concentrations with various crystallization effects,we observe drastically different solvation relaxation *** differences in solvation dynamics are correlated with the local protein structural integrity and water-network stability upon interaction with the *** solvation dynamics at the protein surface is proposed as a new perspective to study precipitant-protein interactions.

We study the semi-leptonic and non-leptonic B weak decays that are governed by the B→D(*)*** branching ratios,CP asymmetries(CPAs)and polarization fractions of non−leptonic decays are investigated in the factorization approximation(FA).The B→D(*)form factors are estimated in the Salpeter *** estimation on branching ratios is in general agreement with existing experimental *** CPAs and polarizations,comparisons among the FA results,the perturbative QCD predictions and experimental data are presented.

Magnetized magma intrusions being sources of a weak and a strong lunar magnetic anomaly revealed by 3D distribution of magnetization

In this work,we aim to investigate the origin of the magnetic carriers in the lunar crust and the intensity of the ancient dynamo *** magnetization and depth range of magnetic carriers are studied under a weak and a strong magnetic anomaly in Mare Tranquillitatis and in Oceanus Procellarum,respectively,where the surface ages are 3.6 and 3.3 billion years.A sophisticated three-dimensional amplitude inversion software program from a geophysical survey is used to reconstruct the distributions of magnetization in the lunar *** no globally measured surface magnetic field exists for the Moon,a crustal magnetic anomaly model with a grid resolution of 0.2°is *** depth range of the magnetic source is fixed by the boundary identified by a relative criterion,which is 20%of the recovered maximum *** central burial depths of the magnetic carriers are approximately 15 km and 25 km under Reiner Gamma and Mare Tranquillitatis,*** volumes of the two magnetic sources are at scales of 104 and 105 km3,*** aforementioned differences may imply a hotter crust under Reiner Gamma than Mare Tranquillitatis by 3.3 billion *** results support the view that the magma intrusions magnetized by an ancient magnetic field could be the origin of magnetic anomalies under Reiner Gamma and Mare *** with previous works,the maximum magnetization of 3 A/m under Reiner Gamma supports the intensity of the field being several microteslas.

Design of antibacterial peptide-like conjugated molecule with broad spectrum antimicrobial ability

Increasing multidrug-resistant （MDR） superbugs emerge worldwide causing a public health crisis. Consequently, it is urgent to find new antibiotics with efficient broad-spectrum antimicrobial activity. By virtue of versatility in molecular design, a new peptide-like cell membrane-broken molecule, oligo-（7,7＇-bifluoren-benzo[c][1,2,5]thiadiazole）（OFBT） possessing a conjugated backbone and eight pendant guanidyl moieties was designed and synthesized. OFBT exhibits favorable broad-spectrum of antirnicrobial activity to pathogens including Gram-negative and Gram-positive bacteria, and fungi with the minimum inhibitory concentration （MIC） below 3.0 μM. Moreover, OFBT exhibits high selectivity for pathogens over human cells to make it a promising broad spectrum antimicrobial agent.

Mechanism of intram olecular charge transfer in DNA helix as probed by the use of the fluorescent 2-aminopurine

As a structural analogue of adenine, 2-aminopurine (2Ap) is often used as a fluorescent probe to study the intramolecular charge transfer reaction in DNA. We have designed and synthesized a series of model DNA helix with the variation in the distance between the 2Ap probe and the GGG sequence, and have investigated, by means of picosecond time-resolved fluorescence spectroscopy, the effect of the length of the bridge (consisting of a number of inosines, I) separating the electron donor (???GGG???) and the acceptor (2Ap) on the charge transfer dynamics. The fluorescence dynamics of 2Ap exhibited three exponential decay components, the one with a time constant of a few hundred picoseconds is assigned to the intramolecular charge transfer from GGG to 2Ap. Within 2.4 nm of the donor-acceptor separation, the rate of charge transfer decreased exponentially upon increasing the separation, from which the decay factor β is determined to be 1.3 nrrf-1. Beyond 2.4 nm, however, the rate started toincrease, this abnormal behavior of charge transfer is interpreted in terms of the match of elec- tronic energies between the l-bridge and the donor/acceptor couple.

Chronic hepatitis B virus(HBV)infection(CHB)is a public health concern *** therapies utilizing nucleos(t)ide analogs(NA)have not resulted in a complete cure for ***,patients on long-term NA treatment often develop low-level viremia(LLV).Persistent LLV,in addition to causing the progression of liver disease or hepatocellular carcinoma,may shed light on the current plight of NA ***,we review the literature on LLV,NA treatment,and various doses of entecavir to find a strategy for improving the efficacy of this antiviral *** LLV patients,three therapeutic options are available,switching to another antiviral monotherapy,interferon-αswitching therapy,and continuing *** real-world clinical practice,entecavir overdose has been used in antiviral therapy for CHB patients with NA refractory and persistent LLV,which encouraged us to conduct further in-depth literature survey on dosage and duration related entecavir *** studies of pharmacodynamics and pharmacokinetics show that entecavir has the maximal selected index for safety,and has great potential in inhibiting HBV replication,in all of the *** the particular section of the drug approval package published by the United States Food and Drug Administration,entecavir doses 2.5-20 mg/d do not increase adverse events,and entecavir doses higher than 1.0 mg/d might improve the antiviral *** literature survey led us to two suggestions:(1)Increasing entecavir dose to 1.0 mg/d for the treatment of NA naïve patients with HBV DNA>2×106 IU/mL is feasible and would provide better prognosis;and(2)Further research is needed to assess the long-term toxic effects of higher entecavir doses(2.5 and 5.0 mg/d),which may prove beneficial in treating patients with prior NA treatment,partial virological response,or LLV state.

Synthesis and cytotoxic activity of novel curcumin analogues

Five novel curcumin analogues bearing different substituents at 4-position of phenyl group were synthesized. Their structures were confirmed by NMR and HRMS spectrum. Their cytotoxic activities against six tumor cell lines were tested by the standard MTT assay in vitro. The results indicated that four analogues （1A-1C, 1E） with solubilizing moieties showed selective potent cytotoxicity against HepG2, HeLa and CT26 cell lines, and analogue 1A and 1C exhibited more potent cytotoxicity than curcumin against CT26 cell line. It was suggested that introduction of appropriate substituents to 4-position of phenyl group might be a potential option for structural modification of curcumin.
