LAPTM4B,which is a proto-oncogene,plays important roles in cancer progression,including clinical stages and pathologic grades.[1]The LAPTM4B gene encodes the lysosomal-associated protein transmembrane-4 beta(LAPTM4B)protein and LAPTM4B is cloned from hepatocellular carcinoma(HCC).[2]There is increasing evidence that LAPTM4B is a novel oncogene,which is over-expressed in many human solid tumors.[3]Furthermore,the up-regulation of LAPTM4B is associated with chemotherapy resistance,as well as poor clinical outcomes and prognoses.[4,5]LAPTM4B also promotes cell survival,growth,proliferation,metastasis,tolerance to metabolic stress,and autophagy.[6]Thus,the LAPTM4B protein may represent an excellent candidate for individualized treatment and tumor monitoring.

Expression of γ-synuclein in colorectal cancer tissues and its role on colorectal cancer cell line HCT116

AIM: To investigate the expression pattern of γ-synuclein in colorectal cancer (CRC) tissues, and to study the effects of γ-synuclein on CRC cell line HCT116 biological features in ***: The expression pattern of γ-synuclein was determined in 54 CRC tissues and 30 tumor-matched nonneoplastic adjacent tissues (NNAT) 5 cm away from the tumor via real-time quantitative reverse transcription PCR (RT-PCR) and immunohistochemistry. The relationship between γ-synuclein protein expression and clinicopathological factors of CRC tissues was analyzed. Three small interfering RNA (siRNA) targeting γ-synuclein mRNA plasmids were constructed and transfected into the CRC cell line HCT116. The stable cell lines were selected with G-418 for 28 d, and the biological features of these cells were examined by cell growth curve, soft agar assay, and cell migration and invasion assays in vitro. RESULTS: The expression of γ-synuclein mRNA and protein was much higher in CRC tissue samples than in NNAT samples (P = 0.02, P = 0.036). There was a significant correlation between the γ-synuclein protein expression and clinical stage and lymph node involvement of CRC (P = 0.02, P = 0.033). In functional analysis we found that down-regulation of γ-synuclein expression in HCT116 cells could inhibit the growth, colony formation rate, and migration and invasion ability of HCT116 ***: Increased expression of γ-synuclein in CRC tissues and the biological effects of reduced γ-synuclein expression on HCT116 cells suggest that γ-synuclein may play a positive role in the progression of CRC.

Sodium butyrate attenuates high-fat diet-induced steatohepatitis in mice by improving gut microbiota and gastrointestinal barrier

AIM To investigate whether gut microbiota metabolite sodium butyrate (NaB) is an effective substance for attenuating non-alcoholic fatty liver disease (NAFLD) and the internal mechanisms. METHODS Male C57BL/6J mice were divided into three groups, normal control were fed standard chow and model group were fed a high-fat diet (HFD) for 16 wk, the intervention group were fed HFD for 16 wk and treated with NaB for 8 wk. Gut microbiota from each group were detected at baseline and at 16 wk, liver histology were evaluated and gastrointestinal barrier indicator such as zonula occluden-1 (ZO-1) were detected by immunohistochemistry and realtime-PCR, further serum or liver endotoxin were determined by ELISA and inflammation-or metabolism-associated genes were quantified by real-time PCR. RESULTS NaB corrected the HFD-induced gut microbiota imbalance in mice, while it considerably elevated the abundances of the beneficial bacteria Christensenellaceae, Blautia and Lactobacillus. These bacteria can produce butyric acid in what seems like a virtuous circle. And butyrate restored HFD induced intestinal mucosa damage, increased the expression of ZO-1 in small intestine, further decreased the levels of gut endotoxin in serum and liver compared with HF group. Endotoxin-associated genes such as TLR4 and Myd88, pro-inflammation genes such as MCP-1, TNF-alpha, IL-1, IL-2, IL-6 and IFN-gamma in liver or epididymal fat were obviously downregulated after NaB intervention. Liver inflammation and fat accumulation were ameliorated, the levels of TG and cholesterol in liver were decreased after NaB intervention, NAS score was significantly decreased, metabolic indices such as FBG and HOMA-IR and liver function indicators ALT and AST were improved compared with HF group. CONCLUSION NaB may restore the dysbiosis of gut microbiota to attenuate steatohepatitis, which is suggested to be a potential gut microbiota modulator and therapeutic substance for NAFLD.

This paper deals with the structural analysis problem of dynamic lumped process high-index differential algebraic equations(DAE)*** existing graph theoretical method depends on the change in the relative position of underspecified and overspecified subgraphs and has an effect to the value of the differential index for complex *** this paper,we consider two methods for index reduction of such models by differentiation:Pryce’s method and the symbolic differential elimination algorithm *** can remedy the above *** and comparison of these methods are given via a class of fundamental process simulation *** particular,the efficiency of Pryce’s method is illustrated as a function of the number of tanks in process ***,a range of nontrivial problems are demonstrated by the symbolic differential elimination algorithm and fast prolongation.

Co-Benefits of CO2 and NOx Emission Control in China's Cement Industry

Over the past 30 years, China’s cement industry has experienced rapid development. In this study the authors estimate the emissions trend, emissions control policies, and costs of the policies in China’s cement industry under various economic growth scenarios. First, the authors develop a bottom-up energy system — multi-pollutant abatement planning(MAP) model for China’s cement industry based on the existing productivity, a set of retrofitting options and new investments, alternative fuels, and various available emission control technologies. Second, the authors identify key drivers of cement demand to develop scenarios for future cement demand(2012–2030) and corresponding output peak time under high/low economic growth conditions. Third,the authors consider three scenarios including current policies without carbon control(BAU),moderately low carbon scenario(MLC), and radically low carbon scenario(RLC). The scenarios are being built up with different emission control goals and also compared by costs with estimation of marginal abatement cost curve for cement industry. Finally, based on the estimates the authors suggest a cost-efficient green/low carbon development roadmap for China’s cement sector,considering best available technological options and policy instruments. The study estimates the benefits of co-controlling air pollutants and CO2emissions, and proposes an innovative mechanism to deal with air pollution and climate change.

Chromosome painting of telomeric repeats reveals new evidence for genome evolution in peanut

Interspecific hybridization is an important approach to improve cultivated peanut varieties. Cytological markers such as tandem repeats will facilitate alien gene introgression in peanut. Telomeric repeats have also been frequently used in chromosome research. Most plant telomeric repeats are（TTTAGGG）n that are mainly distributed at the chromosome ends, although interstitial telomeric repeats（ITRs） are also commonly identified. In this study, the telomeric repeat was chromosomally localized in 10 Arachis species through sequential GISH（genomic in situ hybridization） and FISH（fluorescence in situ hybridization） combined with 4＇,6-diamidino-2-phenylindole（DAPI） staining. Six ITRs were identified such as in the centromeric region of chromosome Bi5 in Arachis ipa？nsis, pericentromeric regions of chromosomes As5 in A. stenosperma, Bho7 in A. hoehnei and Av5 in A. villosa, nucleolar organizer regions of chromosomes As3 in A. stenosperma and Adi3 in A. diogoi, subtelomeric regions of chromosomes Bho9 in A. hoehnei and Adu7 in A. duranensis, and telomeric region of chromosome Es7 in A. stenophylla. The distributions of the telomeric repeat, 5S r DNA, 45 S r DNA and DAPI staining pattern provided not only ways of distinguishing different chromosomes, but also karyotypes with a higher resolution that could be used in evolutionary genome research. The distribution of telomeric repeats, 5S r DNA and 45 S r DNA sites in this study, along with inversions detected on the long arms of chromosomes Kb10 and Bho10, indicated frequent chromosomal rearrangements during evolution of Arachis species.

Objective：To elucidate the mechanism of Chinese tuina in treating sciatic nerve crush injury,and to detect the levels of tissue plasminogen activator（t PA）and plasminogen activator inhibitor-1（PAI-1）,which is thought to play an important role in nerve ***：Thirty-two adult male Sprague-Dawley rats were subjected to sciatic nerve crush injury and 16 rats（sham-operated group）went through a sham *** group was given no treatment while tuina group received tuina therapy since day 7 *** treatment was performed once a day and lasted for 20 *** sciatic functional index was examined every 5days during the treatment *** rats＇gastrocnemius muscles were evaluated for changes in mass and immunohistochemistry techniques were performed to detect the levels of tP A and ***：Tuina therapy improved the motor function of sciatic nerve injured rats（P〈0.05）,however,it did not increase muscle volume（P〈0.05）.Tuina downregulated the levels of t PA and PAI-1（P〈0.05）.Conclusions：The present study implies that tuina treatment could accelerate rehabilitation of peripheral nerve injury.

Effect of Silicon Addition on Microwave Synthesis of Al_(4)SiC_(4)

Al_(4)SiC_(4)was prepared using Al powder,silicon carbide powder,and graphite as raw materials,and silicon as the additive by microwave *** of silicon additions(0,5%,10%and 15%,by mass)on the phase composition,microstructure,apparent porosity and bulk density of the Al_(4)SiC_(4)sample prepared with different aluminum additions(20%,25%,by mass)were *** the developed Al_(4)SiC_(4)samples were applied to MgO-C bricks to investigate the effect on the oxidation resistance and mechanical properties of the MgO-C *** results show that Al_(4)SiC_(4)can be synthesized at lower temperatures using the microwave sintering *** sample with 20%aluminum addition has a lower Al_(4)SiC_(4)content than the one with 25%*** the silicon addition is 10%and the aluminum addition is 25%,the sample appears needlelike system,the porosity is only 28.69%,and the sample is *** developed Al_(4)SiC_(4)was added into MgO-C bricks as the antioxidant,which greatly increased the oxidation resistance and the mechanical properties of the MgO-C bricks.

Association between red blood cell increase and gut microbiota induced by high-fat diet

OBJECTIVE To investigate the underlyingmechanism on the association of red blood cell and gut microbiota in rats induced by High-Fat Diet(HFD).METHODS A total of 36 male SpragueDawley rats(180±20 g) were randomly divided into two *** control group(n=10) was given a normal chow diet(10% calories of fat),and the High-fat diet group(n=26) was given a HFD(60% *** of fat).We recorded body weight,length and detected serum glucose,serum lipids and insulin *** two *** fresh arterial blood was collected during the experiments and blood gases were measured immediately(Radiometer Medical ApS,Denmark).Thehematocrit(Hct) and partial pressure of oxygen(pO_2) were detected by the sensor cassette,following themanufacturer′s *** *** method was conductivity measurements and current method,*** feces from *** were analyzed by 16 S rRNA gene high-throughput sequencing(Illumina Miseq,USA).RESULTS According to the insulin resistance(IR),body weight and body length,the model group was divided into two small groups.(1) IR group,in which IR,body weight and body length were higher than the control group(P<0.05).(2) un-IR group,body weight and body length were higher than the control group(P<0.05),but the IR was not significantly *** addition,the levels of hematocrit(Hct),***(ctHb) and check total blood oxygen content(ctO_2) showed significantly increased in the IR group when compared with the control group(P<0.05),however,the pO_2 was not statistically ***,we identified that the genus Lactobacillus was moderate positive correlation with Hct,ctHb and ctO_2(P<0.05).Compared with the control group,the relative abundance of the *** was significantly lower in IR group(P<0.05).CONCLUSION The high-fat diet induced rats′ local tissue hypoxia under the red blood cell increasing,oxygen partial pressure constant and the reduction of Lactobacillus′ abundance might be caused by aerobic oxid
