Hepatic carcinoma(HC)is the sixth most frequently occurring malignancies and the third leading cause of cancer death *** bromide(ym155)is a small molecule inhibitor of survivin,which has broad-spectrum anticancer therapeutic effects in various xenograft ***,several-day continuous infusion is required to achieve greater antitumor efficacy because of rapid elimination from the blood ***,a SMMC-7721 cancerous cyto-membrane-cloaked drug delivery system(DDS)(named as iM7721@GQD-ym),was developed for co-encapsulation of ym155 and graphene quantum dots(GQDs).Cytomembrane coating endowed iM7721@GQD-ym with effective targeting for homologous HC cells,excellent biocompatibility and favorable immunocompatibility for in vivo *** decoration of iRGD peptide further enhanced its tumor targeting activity by iRGD-integrin *** addition,under the irradiation of near-infrared ray(NIR),GQDs can directly kill tumors through photothermal effect and cause cell membrane rupture,accurately releasing ym155 at tumor *** physicochemical properties,in vivo and ex vivo anti-tumor efficacy,and mechanisms of iM7721@GQD-ym nanoparticles(NPs)were systematically investigated in this *** experimental results clearly indicate that the versatile biomimetic DDS holds great potential for the treatment of HC,which merits further investigation in both pre-clinical and clinical studies.

The Kela-2 gas field, found in the Kuqa Depression of the Tarim Basin, northwestern China, is a large-sized dry gas field (C1 /C1-5 =0.992 0.999) and characterized by ultra-high pressure (pressure factor up to 2.0 2.2). The pyrolysis experiment was carried out under isothermal gold-tube closed system, with samples collected from the Jurassic coal, Jurassic mudstone and Triassic mudstone in the Kuqa Depression. The result of gas yield showed that the Middle and Lower Jurassic source rocks have higher gas generation potential than the Triassic source rocks. The kinetic modeling of gas generation and methane carbon isotope fractionation suggested that the Kela-2 gases belong to the products of high-over mature stages and were mainly derived from the Middle and Lower Jurassic coal-bearing strata. The Triassic source rocks made a minor contribution to the Kela-2 gases. The Kela-2 gases chiefly generated from coal-bearing source rocks with R o values from 1.3% to 2.5%, and thus primarily accumulated after 5 Ma.

Characteristics of Patients Sponsored by the Internet Platforms of Medical Mutual Aid

The morbidity and mortality of life-threatening diseases such as cancers are increasing year by year in China. Although the national medical insurance has been improved a lot, it still falls short of expecting “universal medical insurance”. To help the families of patients with cancer and other serious diseases to resolve the unaffordable medical cost, several internet platforms of “Peer to Peer Medical Mutual Aid” have gradually emerged recently. In the form of low threshold insurance combined with public support, these platforms provide subsidies to families of patients suffering from serious financial burden and then partly compensate them for the limit of national medical insurance. This article summarized the characteristics of declared patients sponsored by several medical mutual aid internet platforms. The results showed that the sponsored patients were mostly cancer patients (90.1%), aged 21 - 60 (88.4%), and the average amounts of financial support had a trend to decrease with the increase of age. In particular, the reported cases of thyroid cancer were increased dramatically recently, however, with a lowest average mutual aid fund. Given these data, it is urgent to list out a detailed rule for the use of national medical insurance on the treatment of cancer, to heighten the healthcare system for the occupational population, age from 21 to 60, and to establish the credit system for Medicare and Medicaid services.

目的:观察在脱细胞生物瓣支架上种植自体内皮细胞后完成体外构建生物组织工程瓣的可行性。方法:实验于2002-12/2004-12在中国医学科学院阜外心血管病医院先心病研究室和解放军军事医学科学院全军造血干细胞中心实验室完成。①骨髓样品取自军事医学科学院附属医院全军造血干细胞移植中心的健康成人献髓员,从正常成人骨髓中分离骨髓基质细胞进行定向培养和扩增其中的内皮细胞。②十二烷基硫酸钠脱去成人主动脉带瓣管道表面的内皮细胞后作为支架材料;脱细胞的同种生物瓣支架在无菌条件下,剪切成0.8cm×0.8cm的大小,种子细胞选择人骨髓基质细胞体外定向培养的内皮细胞,高密度(>105cm-2)种植于瓣膜支架上静态培养20d。③分别在静态培养的第7,14,20天结束时取瓣膜片用0.5%硝酸银染色行立体显微镜和扫描电镜观察、摄片。瓣膜被内皮细胞覆盖的面积用百分数表示,以确定其再内皮化率。结果:①5.0×105个骨髓基质细胞体外定向培养的内皮细胞共传8代,最后获得了6.0×109个内皮细胞,扩增了约1.2×104倍,可以满足高密度种植内皮细胞的需求。②0.03%十二烷基硫酸钠完全脱去了同种生物瓣表面的内皮细胞,而瓣膜的细胞外基质成份在脱细胞前后均无明显变化,其形态学结构保持良好。③内皮细胞与支架复合体静态培养第7,14,20天,再内皮化率分别为65%、85%和90%。结论:骨髓基质细胞定向培养的内皮细胞,经扩增后是瓣膜组织工程研究中种子细胞的又一来源;静态培养条件下基本实现了生物组织工程瓣体外构建的目标,为进一步脉动流培养以及动物实验提供了材料基础。

AIM:To investigate ym155’s effect on retinal pigment epithelium(RPE)cells’viability and the potential regulatory ***:Human immortalized RPE cell lines(ARPE-19 cell line)were processed with ym155 and epidermal growth factor(EGF).ARPE-19 cell viability was detected by methyl thiazolyl tetrazolium assay,and apoptosis was tested by flow cytometry ***-19 cell proliferation was assessed with bromodeoxyuridine tagged incorporation assay,and migration ability was evaluated via a wound-healing *** growth factor receptor(EGFR)/MAPK pathway proteins were tested via *** localization was examined by immunofluorescence ***:ym155 suppressed ARPE-19 cells’viability in a time and concentration-dependent manner.A high dose of ym155 caused a small amount of ARPE-19 cell ***155 significantly diminished the ARPE-19 cells’proliferative and migrative ***155 downregulated total EGFR and phosphorylated external signalregulated protein kinase(ERK),and it up-regulated the phosphorylation of P38 MAPK and c-Jun N-terminal kinase(JNK).ym155 induced endocytosis of EGFR in ARPE-19 ***155 also attenuated EGF-induced ARPE-19 cells’proliferative and migrative ***,ym155 significantly decreased the expression of phosphorylated EGFR and ERK after treated by ***:ym155 inhibits RPE cell survival,the cell proliferative and migrative capacity,and it effectuates a small amount of cell death through the EGFR/MAPK signaling ***155 might,therefore,be an agent to prevent and treat abnormal RPE cell survival in proliferative vitreoretinopathy.

目的:利用功能性磁共振成像探讨大脑中与汉语名词、动词、形容词词组加工的脑区。方法:实验于2004-05/2006-11在北京安贞医院与中科院心理研究所以及江苏省语言科学与神经认知重点实验室完成。①选择以汉语为母语的正常被试8人,均为右利手。利用基于T2*成像的EPI成像技术记录被试默念汉语名动形词组时大脑局部的血氧增减,实验材料选取汉语最基本的三类实词组成词组,利用组块设计通过E-Prime软件通过投影投射到磁体内部的反光镜上,被试平躺在磁体内,透过反光镜默读实验材料。②实验分成两个序列,每个序列包含9个组块,每个组块包含一类刺激,每类刺激共4个组块。实验机器是SimensSonata1.5T超导型磁共振设备,分别对被试进行3个序列扫描,包括定位像,功能像和三维全脑像。其中功能像扫描重复时间为100ms/层,单次扫描20层,每层厚5mm,层间隔为1.5mm,观察视野为220mm×220mm,分辨率为64×64象素,在扫描结束后利用AFNI软件对两个序列数据进行叠加处理,通过名动形三类词构成的词组之间脑区的差异分析不同词类加工的对应脑区。结果:6名被试进入结果分析。默念汉语词组激活了大脑的左额上、中、下回以及内侧,左颞叶上、中回,扣带回以及右脑等许多区域,这些区域在不同任务中有区别也有重叠,特别值得注意的是,有名词参与的词组与没有名词参与的词组相比脑区激活差异明显,有名词参与的词组中,前扣带回附近,右额叶下回,双侧额叶前部等部位有明显激活。结论:汉语中名词重叠词组要大于动词重叠词组和形容词重叠词组,名动词组要大于名形词组和动形词组。扣带回在与名词相关的词组加工中起重要作用,不同类型的汉语词组加工在大脑中依赖不同的区域。
