Alveolar bone regeneration has been strongly linked to macrophage polarization.M1 macrophages aggravate alveolar bone loss,whereas M2 macrophages reverse this process.Berberine(BBR),a natural alkaloid isolated and refined from Chinese medicinal plants,has shown therapeutic effects in treating metabolic disorders.In this study,we first discovered that culture supernatant(CS)collected from BBR-treated human bone marrow mesenchymal stem cells(HBMSCs)ameliorated periodontal alveolar bone loss.CS from the BBR-treated HBMSCs contained bioactive materials that suppressed the M1 polarization and induced the M2 polarization of macrophages in vivo and in vitro.To clarify the underlying mechanism,the bioactive materials were applied to different animal models.We discovered macrophage colony-stimulating factor(M-CSF),which regulates macrophage polarization and promotes bone formation,a key macromolecule in the CS.Injection of pure M-CSF attenuated experimental periodontal alveolar bone loss in rats.Colony-stimulating factor 1 receptor(CSF1R)inhibitor or anti-human M-CSF(M-CSF neutralizing antibody,Nab)abolished the therapeutic effects of the CS of BBR-treated HBMSCs.Moreover,AKT phosphorylation in macrophages was activated by the CS,and the AKT activator reversed the negative effect of the CSF1R inhibitor or Nab.These results suggest that the CS of BBR-treated HBMSCs modulates macrophage polarization via the M-CSF/AKT axis.Further studies also showed that CS of BBR-treated HBMSCs accelerated bone formation and M2 polarization in rat teeth extraction sockets.Overall,our findings established an essential role of BBR-treated HBMSCs CS and this might be the first report to show that the products of BBR-treated HBMSCs have active effects on alveolar bone regeneration.

Despite extensive efforts in designing and preparing switchable underwater adhesives,it is not easy to regulate the underwater adhesion strength locally and remotely.Here,we design and synthesize photoreversible copolymer of poly[dopamine methacrylamide-co-methoxyethyl-acrylate-co-7-(2-methacryloyloxyethoxy)-4-methylcoumarin].Due to the dynamic formation and breaking of chemical crosslinking networks within the smart adhesives,the material shows widely tunable adhesion strength from∼150 to∼450 kPa and long-range reversible maneuverability under orthogonal 254 and 365 nm ultraviolet light stimulation via the coumarin dimerization and cycloreversion.Moreover,the adhesive exhibits good circulation performance and stability in an acid–base environment.It also demonstrated that the bolt can be coated with the smart adhesive material for on-demand bonding.This design principle opens the door to the development of remotely controllable high-performance smart underwater adhesives.

Sleep has attracted extensive attention due to its significance in health.However,its association with erectile dysfunction(ED)is insufficiently investigated.To investigate the potential causal links between sleep traits(insomnia,sleep duration,and chronotype)and ED,this study was performed.The single-nucleotide polymorphisms(SNPs)associated with insomnia,sleep duration,and chronotype were retrieved from previous genome-wide association studies(GWAS).A conventional two-sample Mendelian randomization(MR)was used to estimate the causal links between sleep traits and ED.The summary statistics of ED were from individuals of European ancestry(6175 cases vs 217630 controls).As shown by the random effect inverse-variance-weighting(IVW)estimator,genetically predicted insomnia was causally associated with a 1.15-fold risk of ED(95%confidence interval:1.07-1.23,P0.05).These findings were consistent with the results of sensitivity analyses.Based on genetic data,this study provides causal evidence that genetically predicted insomnia increases the risk of ED,whereas sleep duration and chronotype do not.

Modern pollen analysis is the basis for revealing the palaeovegetation and palaeoclimate changes from fossil pollen spectra.Many studies pertaining to the modern pollen assemblages on the Tibetan Plateau have been conducted,but little attention has been paid to pollen assemblages of surface lake sediments.In this study,modern pollen assemblages of surface lake sediments from 34 lakes in the steppe and desert zones of the Tibetan Plateau are investigated and results indicate that the two vegetation zones are dominated by non-arboreal pollen taxa and show distinctive characteristics.The pollen assemblages from the desert zone contain substantially high relative abundance of Chenopodiaceae while those from the steppe zone are dominated by Cyperaceae.Pollen ratios show great potential in terms of separating different vegetation zones and to indicate climate changes on the Tibetan Plateau.The Artemisia/Chenopodiaceae ratio and arboreal/non-arboreal pollen ratio could be used as proxies for winter precipitation.Artemisia/Cyperaceae ratio and the sum of relative abundance of xerophilous elements increase with enhanced warming and aridity.When considering the vegetation coverage around the lakes,hierarchical cluster analysis suggests that the studied sites can be divided into four clusters:meadow,steppe,desert-steppe,and desert.The pollen-based vegetation classification models are established using a random forest algorithm.The random forest model can effectively separate the modern pollen assemblages of the steppe zone from those of the desert zone on the Tibetan Plateau.The model for distinguishing the four vegetation clusters shows a weaker but still valid classifying power.It is expected that the random forest model can provide a powerful tool to reconstruct the palaeovegetation succession on the Tibetan Plateau when more pollen data from surface lake sediments are included.

Background Surgery plays an important role in the treatment of neuroblastoma.Perioperative complications may impact the course of neuroblastoma treatment.To date,comprehensive analyses of complications and risk factors have been lacking.Methods Patients with retroperitoneal neuroblastoma undergoing tumor resection were retrospectively analyzed between 2014 and 2021.The data collected included clinical characteristics,operative details,operative complications and postopera-tive outcomes.Risk factors for perioperative complications of retroperitoneal neuroblastoma were analyzed.Results A total of 571 patients were enrolled in this study.Perioperative complications were observed in 255(44.7%)patients.Lymphatic leakage(28.4%),diarrhea(13.5%),and injury(vascular,nerve and organ;7.5%)were the most frequent compli-cations.There were three operation-related deaths(0.53%):massive hemorrhage(n=1),biliary tract perforation(n=1)and intestinal necrosis(n=1).The presence of image-defined risk factors(IDRFs)[odds ratio(OR)=2.09,P<0.01],high stage of the International Neuroblastoma Risk Group staging system(INRGSS)(OR=0.454,P=0.04),retroperitoneal lymph node metastasis(OR=2.433,P=0.026),superior mesenteric artery encasement(OR=3.346,P=0.003),and inferior mesenteric artery encasement(OR=2.218,P=0.019)were identified as independent risk factors for perioperative complications.Conclusions Despite the high incidence of perioperative complications,the associated mortality rate was quite low.Perio-perative complications of retroperitoneal neuroblastoma were associated with IDRFs,INRGSS,retroperitoneal lymph node metastasis and vascular encasement.Patients with high-risk factors should receive more serious attention during surgery but should not discourage the determination to pursue total resection of neuroblastoma.

Excessive accumulation of chloride(Cl^(-))in the aboveground tissues under saline conditions is harmful to crops.Increasing the exclusion of Cl^(-) from shoots promotes salt tolerance in various crops.However,the underlying molecular mechanisms remain largely unknown.In this study,we demonstrated that a type A response regulator(ZmRR1)modulates Cl^(-) exclusion from shoots and underlies natural variation of salt tolerance in maize.ZmRR1 negatively regulates cytokinin signaling and salt tolerance,likely by interacting with and inhibiting His phosphotransfer(HP)proteins that are key mediators of cytokinin signaling.A naturally occurring non-synonymous SNP variant enhances the interaction between ZmRR1 and ZmHP2,conferring maize plants with a salt-hypersensitive phenotype.We found that ZmRR1 undergoes degradation under saline conditions,leading to the release of ZmHP2 from ZmRR1 inhibition,and subsequently ZmHP2-mediated signaling improves salt tolerance primarily by promoting Cl^(-) exclusion from shoots.Furthermore,we showed that ZmMATE29 is transcriptionally upregulated by ZmHP2-mediated signaling under highly saline conditions and encodes a tonoplast-located Cl^(-) transporter that promotes Cl^(-) exclusion from shoots by compartmentalizing Cl^(-) into the vacuoles of root cortex cells.Collectively,our study provides an important mechanistic understanding of the cytokinin signaling-mediated promotion of Cl^(-) exclusion from shoots and salt tolerance and suggests that genetic modification to promote Cl^(-) exclusion from shoots is a promising route for developing salt-tolerant maize.

Aluminum and Activated Alumina Powder Additions on Microwave Synthesis of Al_(4)SiC_(4)

Aiming at improving the properties of magnesia carbon materials,silicon aluminum carbide(Al_(4)SiC_(4))containing materials were prepared using industrial aluminum powder,silicon carbide powder,and graphite as raw materials,and activated alumina powder as an additive,mixing thoroughly,pressing into cylinders and then firing at 1200℃for 30 min in a carbon embedded atmosphere by the microwave method.The effects of the aluminum powder addition(20%and 24%,by mass)and activated alumina powder addition(0,3%,5%and 7%,by mass)on the microwave synthesis of Al_(4)SiC_(4) as well as the effect of the obtained Al_(4)SiC_(4) containing material on the properties of magnesia carbon bricks were studied.The results show that:compared with the samples with 20%aluminum powder,those with 24%aluminum powder generate more Al_(4)SiC_(4).With the activated alumina powder addition increasing from 0 to 7%,the amount of Al_(4)SiC_(4) generated increases first and then decreases.Compared with the sample without activated alumina powder,the samples with activated alumina powder show lower bulk density and higher apparent porosity.With the activated alumina powder addition increasing from 3%to 7%,the bulk density of the samples increases first and then decreases,while the apparent porosity of the samples shows an opposite trend.The optimal additions are 24%aluminum powder and 5%activated alumina powder,and Al_(4)SiC_(4) synthesized in this sample has a hexagonal plate structure.With the synthesized Al_(4)SiC_(4) containing material added,the magnesia carbon brick has slightly increased cold modulus of rupture,basically the same modulus of elasticity and improved oxidation resistance.

OBJECTIVE To evalu ate the role of resting magnetocardiography in identifying seve re coronary artery stenosis in patients with suspected coronary artery disease.METHODS A total of 513 patients with angina symptoms were included and divided into two groups based on the extent of coronary artery disease determined by angiography:the non-severe coronary stenusis group(<70% stenosis) and the severe coronary stenosis group(≥70% stenosis).The diagnostic model was constructed using magnetic field map(MFM) parameters,either individually or in combination with clinical indicators.The performance of the models was evaluated using receiver operating characteristic curves,accuracy,sensitivity,specificity,positive predictive value(PPV) and ne gative predictive value(NPV).Calibration plots and decision curve analysis were performed to investigate the clinical utility and performance of the models,respectively.RESULTS In the severe coronary stenosis group,QR_MCTDd,S_MDp,and TT_(MA)C_(50) were significantly higher than those in the non-severe coronary stenosis group(10,46±10.66 vs,5.11±6.07,P <0.001;7.2±8.64 vs.4.68±6.95,P=0.003;0.32±57.29 vs.0.26±57.29,P <0.001).While,QR_MV_(amp),R_(MA),and T_(MA) in the severe coronary stenosis group were lower(0.23±0.16 vs.0.28±0.16,P<0.001;55.06±48.68 vs.59.24±53.01,P<0.001;51.67±39.32 vs. 60.45±51.33,P <0.001).Seven MFM parameters were integrated into the model,resulting in an area under the curve of 0.810(95% CI:0.765-0.855).The sensitivity,specificity,PPV,NPV,and accurecy were 71.7%,80.4%,93.3%,42.8 %,and 73.5%;respectevely.The combined model exhibited an area under the curve of 0.845(95% CI:0.798-0.892).The sensitivity,specificity,PPV,NPV,and accuracy were 84.3%,73.8%,92.6%,54.6%,and 82.1%;respectively.Calibration curves demonstrate d excellent agreement between the nomogram prediction and actual observation.The decision curve analysis showed that the c ombine d model provided greater net benefit compared to the magnetocardingraphy model.CONCLUSIONS The novel quantitative MFM parameters

Objective: Definitive chemoradiotherapy(dCRT) is the standard treatment for unresectable locally advanced esophageal cancer. However, this treatment is associated with substantial toxicity, and most malnourished or elderly patients are unable to complete this therapy. Therefore, there is a need for a more suitable radiotherapy combination regimen for this population. This study was aimed to evaluate the efficacy and safety of a combination regimen comprising chemotherapy with nimotuzumab and S-1 and concurrent radiotherapy for patients with fragile locally advanced esophageal cancer with a high Nutritional Risk Screening 2002(NRS-2002) score.Methods: Eligible patients with unresectable esophageal carcinoma who had an NRS-2002 score of 2 or higher were enrolled. They were treated with S-1 and nimotuzumab with concurrent radiotherapy, followed by surgery or definitive radiotherapy. The primary endpoint was the locoregional control(LRC) rate.Results: A total of 55 patients who met the study criteria were enrolled. After completion of treatment, surgery was performed in 15 patients and radiotherapy was continued in 40 patients. The median follow-up period was 33.3[95% confidence interval(95% CI), 31.4-35.1)] months. The LRC rate was 77.2%(95% CI, 66.6%-89.4%) at 1 year in the entire population. The overall survival(OS) rate and event-free survival(EFS) rate were 57.5% and 51.5% at 3 years, respectively. Surgery was associated with better LRC [hazard ratio(HR)=0.16; 95% CI,0.04-0.70; P=0.015], OS(HR=0.19; 95% CI, 0.04-0.80; P=0.024), and EFS(HR=0.25; 95% CI, 0.08-0.75;P=0.013). Most adverse events were of grade 1 or 2, and no severe adverse events occurred.Conclusions: For malnourished or elderly patients with locally advanced esophageal cancer, radiotherapy combined with nimotuzumab and S-1 is effective and has a good safety profile.
