Principles of integrity and ethical practice in research have been reinforced and honed over decades.In medical and biomedical fields,they have been shaped by challenges that include endemic,emerg-ing,and re-emerging diseases[1],as well as the scientific responses to each disease as it occurs.These responses include clinical and community prevention trials,vaccine and drug trials for persons with life-threatening diseases,applied and basic research with dangerous microbial agents,and many others.Unexpected fatal pandemics caused by novel agents-for which neither drugs nor vaccines exist-present additional urgent challenges for scientists,defined here to mean biomedical,medical,and pub-lic health scientists and practitioners.In addition to conducting research,scientists are also often tasked with communicating research findings,not only to those who make policy decisions,but also to the media and the public.

The Singapore Statement on Research Integrity,artic-ulated during the 2nd World Conference on Research Integrity in 2010,encompasses principles that apply to all research endeavours,including the various disci-plines that comprise the field of forensic science.(Singapore Statement on Research Integrity(2010)(https://***/guidance/singapore-statement)).Cases of research misconduct and breaches of research integ-rity in any field undermine the trustworthiness of research and make it difficult if not impossible for others to rely upon and replicate research results.Moreover,problems in the research environment,whether in the academic,public,or private sector,can include bullying and harassment,discrimination,abuse of power and corruption,as well as competitive pres-sures related to employment and status.

Soft tissue sarcomas(STS)are a heterogenous group of rare malignancies of mesenchymal origin,affecting both children and adults.The majority of STS have a poor prognosis and advanced stage at the time of diagnosis.Standard treatments for STS largely constitute tumour resection with chemotherapy and/or radiotherapy,and there has been little significant advancement in the application of novel therapies for treatment of these tumours.The current multimodal approach to therapy often leads to long-term side effects,and for some patients,resistance to cytotoxic agents is associated with local recurrence and/or metastasis.There is,therefore,a need for novel therapeutic strategies for the treatment of STS.Recent advances in epigenetics have implicated the histone methyltransferase,EZH2,in the development and progression of diseases such as breast cancer,lymphoma and more recently STS.Here we will review the current literature for EZH2 in STS,including high expression of EZH2 in STS and correlation of this with specific features of malignancy(metastasis,histological grade,and prognosis).The effects of targeting EZH2 using RNA interference and small molecule inhibitors will also be reviewed and the potential for the use of EZH2 inhibition in therapeutic strategies for STS patients will be discussed.

microRNAs是一类短的,高度保守的非编码RNA,可调节基因的表达。MiR-3031作为一个功能性miRNA,参与调节奶山羊乳腺发育和泌乳等多种重要的生物学过程。本试验旨在研究POU2F3和miR-3031在奶山羊乳腺上皮细胞中的靶向关系及其功能,为奶山羊乳腺发育和泌乳提供理论依据。通过RT-qPCR,Western blot和构建双荧光素酶报告载体验证miR-3031与POU2F3的靶向调节作用。生物信息学分析表明,POU2F3的3’UTR区包含miR-3031的结合位点。同时,miR-3031降低了PsiCHECKTM2-POU2F3 3’UTR-wt的双荧光素酶活性。RT-qPCR和Western blot结果显示,与对照组NC相比,miR-3031 mimics显著降低POU2F3的mRNA和蛋白水平的表达;结果表明POU2F3是miR-3031的靶基因。转染miR-3031 24h、48h后CCK8检测miR-3031对奶山羊乳腺上皮细胞活力的影响。结果显示,miR-3031 mimic显著提高奶山羊乳腺上皮细胞的活力。转染siPOU2F3 24h后EDU检测奶山羊乳腺上皮细胞增殖的情况,结果显示,干扰POU2F3显著促进奶山羊乳腺上皮细胞的增殖。将siPOU2F3和si-NC转染到乳腺上皮细胞中,进一步研究POU2F3对奶山羊乳腺上皮细胞凋亡的作用机理。流式结果表明,siPOU2F3显著抑制奶山羊乳腺上皮细胞凋亡。同时,siPOU2F3显著抑制Bcl-2和Bax蛋白的表达,上调Bcl-2/Bax的比率;同时,检测AKT和mTOR的磷酸化水平。结果表明,siPOU2F3显著上调了AKT、mTOR的磷酸化水平和PTEN的蛋白水平以及显著下调PI3K蛋白的表达。综上所述,miR-3031靶向POU2F3通过AKT/mTOR信号通路促进奶山羊乳腺上皮细胞增殖,抑制凋亡。
