The centrosome is a membrane-less organelle consisting of a pair of barrel-shaped centrioles and pericentriolar material and functions as the major microtubule-organizing center and signaling hub in animal cells.The past decades have witnessed the functional complexity and importance of centrosomes in various cellular processes such as cell shaping,division,and migration.In addition,centrosome abnormalities are linked to a wide range of human diseases and pathological states,such as cancer,reproductive disorder,brain disease,and ciliopathies.Herein,we discuss various functions of centrosomes in development and health,with an emphasis on their roles in germ cells,stem cells,and immune responses.We also discuss how centrosome dysfunctions are involved in diseases.A better understanding of the mechanisms regulating centrosome functions may lead the way to potential therapeutic targeting of this organelle in disease treatment.

Background:The Omicron(B.1.1.529)SARS-COV-2 variant has raised serious concerns because of its unprecedented rapid rate of spreading and the fact that there are 36 mutations in the spike protein.Since the vaccine-induced neutralizing antibody targets are the spike protein,this may lead to the possibility of vaccine-induced hu-moral immunity escape.Methods:We measured the neutralizing activity in vitro for Omicron and compared this with wild type(WH-09)and Delta variants in human and monkey sera from different types of immunity.The monkey sera samples were collected at 1 and 3 months post three-dose inactivated(PiCoVacc)and recombinant protein(ZF2001)vaccination.Human sera were collected from 1 month post three-dose inactivated vaccination.Results:In inactivated vaccine sera,at 1/3 months post three-dose,geometric mean titers(GMTs)of neutralization antibody(NAb)against the Omicron variant were 4.9/5.2-fold lower than those of the wild type.In recombinant protein vaccine sera,GMTs of NAb against Omicron were 15.7/8.9-fold lower than those of the wild type.In human sera,at 1 month post three-dose inactivated vaccination,GMTs of NAb against Omicron were 3.1-fold lower than those of the wild type.Conclusion:This study demonstrated that despite a reduction in neutralization titers,cross-neutralizing activity against Omicron and Delta variants was still observed after three doses of inactivated and recombinant protein vaccination.

Comparison of the replication and neutralization of different SARS-CoV-2 Omicron subvariants in vitro

Background:New Omicron subvariants are emerging rapidly from BA.1 to BA.4 and BA.5.Their pathogenicity has changed from that of wild-type(WH-09)and Omicron variants have over time become globally dominant.The spike proteins of BA.4 and BA.5 that serve as the target for vaccine-induced neutralizing antibodies have also changed compared to the previous subvariants,which is likely to cause immune es-cape and the reduction of the protective effect of the vaccine.Our study addresses the above issues and provides a basis for formulating relevant prevention and control strategies.Methods:We collected cellular supernatant and cell lysates and measured the viral titers,viral RNA loads,and E subgenomic RNA(E sgRNA)loads in different Omicron subvariants grown in Vero E6 cells,using WH-09 and Delta variants as a reference.Additionally,we evaluated the in vitro neutralizing activity of different Omicron sub-variants and compared it to the WH-09 and Delta variants using macaque sera with different types of immunity.Results:As the SARS-CoV-2 evolved into Omicron BA.1,the replication ability in vitro began to decrease.Then with the emergence of new subvariants,the replication ability gradually recovered and became stable in the BA.4 and BA.5 subvariants.In WH-09-inactivated vaccine sera,geometric mean titers of neutralization antibodies against different Omicron subvariants declined by 3.7~15.4-fold compared to those against WH-09.In Delta-inactivated vaccine sera,geometric mean titers of neutrali-zation antibodies against Omicron subvariants declined by 3.1~7.4-fold compared to those against Delta.Conclusion:According to the findings of this research,the replication efficiency of all Omicron subvariants declined compared with WH-09 and Delta variants,and was lower in BA.1 than in other Omicron subvariants.After two doses of inactivated(WH-09 or Delta)vaccine,cross-neutralizing activities against various Omicron subvariants were seen despite a decline in neutralizing titers.

This study was designed to investigate the sensitivity of SARS-CoV-2 to different temperatures,to provide basic data and a scientific basis for the control of COVID-19 epidemic.The virus was dispersed in 1 mL basal DMEM medium at a final concentration of 103.2 TCID 50/mL and then incubated at 4,22,30,35,37,38,39 and 40°C for up to 5 days.The infectivity of residual virus was titrated using the Vero E6 cell line.The results showed that the virus remained viable for 5 days at 4°C,and for 1 day only at 22 and 30°C.We found that the infectivity of the virus was completely lost after less than 12 hours at 37,38 and 39°C,while at 40°C,the inactivation time of the virus was rapidly reduced to 6 hours.We show that SARS-CoV-2 is sensitive to heat,is more stable at lower temperatures than higher temperature,remains viable for longer at lower temperatures,and loses viability rapidly at higher temperatures.

Diseases caused by foodborne pathogens are one of the main burdens of public health and seriously hinder global social and economic development.In this research,a novel lateral flow strip was successfully constructed for the simultaneous detection of Gram-positive and Gram-negative bacteria based on the fluidity and color aggregation effect of Au nanoparticles(AuNPs).The lateral flow strip first combined ampicillin(AMP)antigens with AuNPs to form AuNPs-AMP antigens as the first recognition molecule.Then vancomycin specifically recognizing Gram-positive bacteria,and aptamers specifi-cally recognizing Gram-negative bacteria were used as the second recognition molecules.Finally,Gram-positive bacteria and Gram-negative bacteria were detected rapidly using this sandwich mode.The strip can rapidly test the samples within 5 min.Using a lateral flow strip detector,the limit of detection was 4 CFU/mL,and the recovery rates in honey samples were 78.2-88.6%.The lateral flow strip constructed can detect foodborne pathogens quickly,accurately and e fficiently,which is of great significance for food safety detection.

Soft robotics has been receiving increasing attention due to its flexibility and adaptability offered by embodied intelligence. A soft robot may undergo complex motions including stretching,contraction,bending,twisting,and their intricate combinations.Among these basic motions,bending plays a central role when a robot accomplishes tasks such as locomotion,grasping and manipulation. Although a rich repertoire of bending mechanisms has been reported,a systematic and rational design framework is still lack. In this paper,we provide a novel design strategy for soft bending actuators which allows integral modeling and design optimization. Nowadays,metamaterials are emerging as a new tool for soft robots,by encoding the desired complex motions directly within the material architectures,leading to conformable monolithic systems. We combine pneumatic actuators and flexible metamaterials to provide an alternative solution to soft bending actuators,with advantages of compact design,large bending motion,and convenient fabrication. A regular pneumatic chamber is embedded inside auxetic and non-auxetic metamaterials,and bending is generated when inflated. We carry out dimensionless analysis to identify the key design variables. To provide insight into design optimization,we develop a computation framework by modeling metamaterial structures with beam elements and the inner chamber with shell elements as an integral part,allowing efficient simulation of the coupled system. We systematically investigate how the bending angle varies with the key design variables and find the optimal design parameters.The experimental results are well in line with the simulation,and a remarkable bending motion of 0.43°/mm is achieved.

Protein post-translational modification （PTM） by ubiquitination has been observed during many aspects of plant growth, development, and stress responses. The ubiquitin-proteasome system precisely regulates phytohormone signaling by affecting protein activity, localization, assembly, and interaction ability. Absci- sic acid （ABA） is a major phytohormone, and plays important roles in plants under normal or stressed growth conditions. The ABA signaling pathway is composed of phosphatases, kinases, transcription fac- tors, and membrane ion channels. It has been reported that multiple ABA signaling transducers are sub- jected to the regulations by ubiquitination. In particular, recent studies have identified different types of E3 ligases that mediate ubiquitination of ABA receptors in different cell compartments. This review focuses on modulation of these components by monoubiquitination or polyubiquitination that occurs in the plasma membrane, endomembranes, and from the cytosol to the nucleus; this implies the existence of retrograde and trafficking processes that are regulated by ubiquitination in ABA signaling. A number of single-unit E3 ligases, components of multi-subunit E3 ligases, E2s, and specific subunits of the 26S proteasome involved in ABA signal regulation are discussed. Dissecting the precise functions of ubiquitination in the ABA pathway may help us understand key factors in the signaling of other phytohormones regulated by ubiqui- tination and other types of PTMs.

The formability of the material determines the amount of available deformation before failure and thus is important for the production of various structural components in industries. The workability of materiMs is commonly evaluated by different forms of failure mod- els during sheet metal forming （SMF） processes. In order to provide a whole picture about the prediction models for SMF failure, necking-related formability and ductile fracture-related forma- bility studies in SMF processes are systematically summarized, the applicability and limitation of each model are highlighted, and the link between forming limit diagram and ductile fracture criterion is pointed out, Conclusions about some critical issues on failure in SMF are made.

According to the research by the Food and Agriculture Organization of the United Nations(http://***/3/i5199e/i5199e.pdf),>1 billion hectares of land are affected by saline-alkaline conditions,which severely affects plant growth and crop yield,posing great threats to global food security and ecosystems.Saline-alkaline land can be classified into three different types—salinity,alkalinity,and alkaline-salinity,depending on the salt components in the soil.To date,the mechanism of plant tolerance to salt stress has been extensively studied,while the understanding of the plant alkaline stress response is still very preliminary,which impedes the breeding of salt–alkali-tolerant crops by using modern molecular breeding techniques.
