a.niotic membrane of huma. pla.enta.is a.source of a.unda.t mesenchyma. stem cell (ha.SC) which ma.es it a.potentia. source of a.logeneic multipotent cell for bone hea.ing. However, much ha. to be explored a.out its isola.ion procedure a.d the osteogenic differentia.ion potentia.. The a.ms of this study a.e to esta.lish the procurement procedure of huma. a.niotic membrane, the isola.ion a.d culture of ha.SC, the MSC phenotypic cha.a.teriza.ion, a.d the in vitro osteogenic differentia.ion of ha.SC. Results of the study a.e a. follows. The qua.ity of huma. a.niotic membrane would be best if procured from Ca.sa.ea. opera.ion under highly a.eptic condition to a.oid funga. a.d ba.teria. conta.ina.ion on the culture. Isola.ion procedure using modified Soncini protocol yielded la.ge a.ount of MSC with high prolifera.ive ca.a.ity in culture medium. Cha.a.teriza.ion of ha.SC showed tha. the ma.ority of the ta.get cells exhibited specific MSC ma.kers (CD105 a.d CD90) with a.sma.l number of these cells expressing CD45, the ma.ker of hema.opoeitic cells. The in vitro osteogenic differentia.ion of ha.SC followed by a.iza.in Red sta.ning showed tha. osteobla.tic differentia.ion wa. detected in a.significa.tly high number of cells. This study concludes tha. ha.SCs isola.ed from huma. a.niotic membrane ha.e the ca.a.ity for in vitro osteogenesis which ma.es them be one of the potentia. a.logeneic stem cells for a.plica.ion in ma.illofa.ia. bone reconstruction.

Experiments on ma.illofa.ia. bone tissue engineering showed the promising result;however, its hea.ing mecha.isms a.d effectiveness ha. not been fully understood. The a.m of this study is to compa.e the bone hea.ing mecha.ism a.d osteogenic ca.a.ity between bovine bone minera. loa.ed with ha.SC a.d a.togenous bone gra.t in the reconstruction of critica. size ma.dibula. bone defect. Critica. size defects were ma.e a. the ma.dible of 45 New Zea.a.d white ra.bits reconstructed with BBM-ha.SC, BBM a.one, a.d a.G, respectively. a. the end of first, second, a.d twelfth weeks, five ra.bits from ea.h experimenta. week were sa.rificed for histology a.d immunohistochemistry sta.ning. Expressions of va.cula. endothelia. growth fa.tor (VEGF), bone mor-phogenic proteins-2 (BMP2), Runx2 a.d the a.ount of a.giogenesis were a.a.yzed in the first a.d second week groups, while expressions of Runx2, osteoca.cin, colla.en type-I fibres, tra.ecula. a.ea.a.d bone incorpora.ion were a.a.yzed in the twelfth week groups. The result showed tha. expressions of VEGF, BMP2 a.d Runx2 a. well a. the a.ount of a.giogenesis were higher in a.G compa.ed with BBM-ha.SC group in the first a.d second weeks of hea.ing. The result of twelfth week of hea.ing showed tha. expressions of Runx2 a.d osteoca.cin a. well a. the thickness of colla.en type-I fibres were significa.tly higher in BBM-ha.SC compa.ed to a.G group, while there wa. no sta.istica.ly difference in tra.ecula. a.ea.a.d bone incorpora.ion between BBM-ha.SC a.d a.G group. This study concluded tha. ea.ly hea.ing a.tivities were higher in a.to-genous bone gra.t tha. in BBM-ha.SC, while osteogenic a.tivities in the la.e sta.e of hea.ing were higher in BBM-ha.SC compa.ed to a.togenous bone gra.t. It wa. a.so concluded tha. the osteo-genic ca.a.ity of BBM-ha.SC wa. compa.a.le to a.togenous bone gra.t in the reconstruction of critica. size defect in the ma.dible.

Investiga.ing the function of combining induced ra. monocytes-derived bone ma.row-ha.mopoietic stem cell (ra. BM-HSCs) with LPS a.d ra. bone ma.row-mesenchyma. stem cell (ra. BM-MSCs) wa. to a.a.yze the a.celera.ion of homing process mecha.ism in injured pa.crea.. Mononuclea.ed stem cells were isola.ed from a.pira.ed whole ra. BM using ficoll a.d cultured in α-MEM complete growth medium in 10 cm petridish. a.ter two da.s, a.herent cells a.ter wa.hing twice in petridish were a.ded α-MEM growth medium a.d then mesenchyma. cells were cha.a.terized using CD105 ma.ker in third pa.sa.e a.d la.eled PKH26. Then ha.mopoietic stem cells (HSCs) were isola.ed with ma.netic bea.s CD34+ a.d differentia.ed in vitro, a.d then induced monocytes with LPS. a.ima. experiment used 28 ma.e Wista. ra.s, a.d divided them into 4 groups. a.ter transpla.ta.ion combined, both cells between monocyte derived HSc (mHSCs) a.d ra. BM-MSC were a.a.yzed expression of pa.r box gen 4 (Pa.4), pa.crea.ic a.d duodena. homeobox (Pdx1), C-peptide using immunohistochemistry, then secretion of insulin a.d C-peptide a.a.yzed using indirect ELISa. Results showed tha. the expressions of Pa.4, Pdx1, C-peptide found in the surfa.e membrane cell of pa.crea.ic cell, a.d secreted C-peptide a.d insulin were shown significa.t (P ranspla.ted group 2, 3 a.d 4, but in group 3 were transpla.ted with combined cells more domina.t tha. non-combined cells. Conclusions suggested tha. combining of induced monocytes-derived HSCs a.d ra. BM-MSCs ha. a.celera.ed homing MSCs into injured pa.crea.ic tissue.

This study explored the novel stra.egy of hypoxic preconditioning of Bone Ma.row Mesenchyma. Stem Cells (BM-MSCs) before intra.vitrea. transpla.ta.ion to improve neuroprotective effects of Retina. Ga.glion Cells (RGCs) in a.ute Gla.coma.Models. The methods of this resea.ch were isola.ed mesenchyma. stem cells from the bone ma.row of a.ult wild-type Spra.ue-Da.ley (SD) ra.s. BM-MSCs were cultured under normoxic or hypoxic (1% oxygen for 24 hours) conditions. Normoxic or hypoxic BM-MSCs were transpla.ted intra.itrea.ly 1 week a.ter ocula. hypertension induction by a.utely increa.ing IOP to 100 - 120 mmHg for 60 minutes. Ra.s were killed 4 weeks a.ter transpla.ted. a.optosis wa. exa.ined by tunnel a.sa. a.d expression Brn3b (Brn3b = RGCs ma.ker) by immunohistochemica. a.a.ysis of the retina. Results showed tha. transpla.ta.ion of hypoxic preconditioning BM-MSCs in a.ute gla.coma.models resulted in a.significa.t a.optosis decrea.ing (p a.d a. significa.t increa.ing in RGCs (p a. well a. enha.ced mor-phologic a.d functiona. benefits of stem cell thera.y versus normoxic BM-MSCs transpla.ta.ion. Conclusions: Hypoxic preconditioning enha.ces the ca.a.ity of BM-MSCs transpla.ta.ion to improve neuroprotective effects of RGCs in a.ute Gla.coma.Models.

The a.ms of this study were to find: 1) Huma. Umbilica. Cord Mesenchyma. Stem Cells (hUCMSCs) phenotypic cha.a.teriza.ion, 2) The in-vitro osteogenic differentia.ion potentia. of hUCMSCs, 3) The cytotoxicity of gela.in solvent to hUCMSCs using 3-[4,5-dimethylthia.ol-2-yl]-2,5-diphenyl tetra.olium bromide (MTT) a.sa.. a. a.result, through cha.a.teriza.ion of hUCMSCs, the ma.ority of ta.get cells expressed specific MSCs ma.kers, Cellula. Differentia.ion (CD)73, sma.ler number of subpopula.ion expressed CD90 with only minima. subpopula.ion expressed CD105 a.d a.l nega.ive MSCs ma.kers. Osteobla.tic differentia.ion wa. found in a.significa.tly high number of cells when in vitro osteogenic differentia.ion of hUCMSCs with a.iza.in Red sta.ning wa. done.
