Agave americana *** were exposed to different concentrations of ethyl methanesulphonate(EMS)0,15,30,45 and 60 mM and to different times of exposure(2 and 4 h).The viability and capacity of shoot formation were shown to be affected when the callus were exposed to high concentrations(30–60 mM).Only the callus exposed to 15 mM EMS presented shoot formation;the exposure time of two hours produced the largest quantity of shoots regenerated per callus(21 shoots/callus).In order to generate somaclonal variants resistant to Fusarium oxysporum,a selection pressure was applied through of a culture filtrate(CF)of 100 ppm of the *** was made in callus obtained in the treatment with 15 mM EMS during 2 h of *** CF caused oxidation and necrosis in 71.25%of the callus;however,they were capable of generating shoots(3.5 shoots/callus).Molecular markers type RAPD,ISSR and DAMD were used to evaluate the genetic variation arising from the mutations caused by EMS on control plants and 16-month-old somaclonal *** polymorphic information content(PIC)for each one of the initiating groups was:0.28(DAMD),0.09(ISSR)and 0.14(RAPD).DAMD revealed a greater percentage of polymorphism than RAPD and *** bands were detected in the somaclonal *** indicated that the EMS caused genetic variation in the regenerated plants conferring resistance to them against Fusarium oxysporum.

In recent years,there has been a growing interest in exploring alternative treatments for bone *** tissue engineering has turned its attention to plant extracts containing osteogenic flavonoids as potential promoters of bone *** our study,we specifically investigated the extract of Petroselinum crispum,a plant known for its abundance of osteogenic flavonoids such as apigenin,quercetin,and *** objective was to compare the total flavonoid content(TFC)and their accumulation in different *** obtained hydrolyzed aqueous extracts from the leaves of parsley plants(grown for 12 weeks in the field),weekly embryogenic cultures,and suspension cell *** TFC,measured in mg quercetin equivalent per gram of dry weight(QE mg/g dw),was found to be 53.81±0.97 for 52 g dw of leaves(1 kg plant fw),31.10±1.52 for 420 g dw of embryogenic cultures,and 11.80±0.76 for 210 g dw of cell ***,only the leaves and embryogenic cultures showed significant accumulation of osteogenic flavonoids,with apigenin levels of 1.8±0.3 and 0.32±0.02 mg/g dw,and kaempferol levels of 1.0±0.18 and 0.2±0.01 mg/g dw,*** findings indicate that embryogenic cultures have the potential to produce up to 40%more apigenin in 12 weeks compared to 1 kg of parsley plant,thanks to their superior biomass reproductive *** results present a promising biotechnological approach for generating extracts enriched with bioactive compounds,which can be further utilized in in vitro osteogenic testing.

The effect of water deficit was determined on both in vitro and soil seedling as well as in cells in suspension of Agave americana *** order to do the establishment of cells,the formation of callus was induced;for it two auxins were evaluated:2,4-dichlorophenoxyacetic acid(2,4-D)and 4-mino-3,5,6-trichloropicolinic acid(picloram)at three concentrations(0.25,0.5 and 0.75 mg L−1)in three explants(leaf,root and meristems)cultured in MS semisolid *** callogenesis response was related to the type and section of the explant,as well as the regulator used,and a cell suspension was established using 0.5 mg L−1 naphthaleneacetic acid(NAA)+0.5 mg L−1 Benzylaminopurine(BAP).Seedlings were exposed to polyethyleneglycol(15%and 30%w/v)with a water potential of−0.87 and−2.67 MPa,respectively,under soil *** stress was applied through restricted *** weight,root system growth,and chlorophyll concentration were some of the parameters that were affected by the effect of water deficit on *** *** concentration values were significantly decreased by 15 at 30%PEG(19.6 SPAD units)compared to the control *** in vitro plants,the highest concentration of proline was found in the roots,being the treatment with 30%polyethylene glycol where the highest concentration of this osmoregulator was obtained(62.5 mg g−1 DW).Under restricted irrigation conditions,an increase in proline concentration was observed both in the aerial part(2.2μg 100 g−1 DW)and in the root system(1.8μg 100 g−1 DW).However,the concentrations found were approximately ten times greater,less than those found under in vitro ***,the accumulation of proline can be considered an indicator of stress in Agave Americana *** in vitro.

The red palm weevil（RPW; Rhynchophorus ferrugineus） is spreading worldwide and severely harming many palm species. However, most studies on RPW focused on insect biology, and little information is available about the plant response to the attack. In the present experiment, we used metabolomics to study the alteration of the leaf metabolome of Phoenix canariensis at initial（1^（st） stage） or advanced（2^（nd） stage）attack by RPW compared with healthy（unattacked） *** leaf metabolome significantly varied among treatments. At the 1^（st） stage of attack, plants showed a reprogramming of carbohydrate and organic acid metabolism; in contrast, peptides and lipid metabolic pathways underwent more changes during the 2^（nd） than 1^（st） stage of attack. Enrichment metabolomics analysis indicated that RPW attack mostly affected a particular group of compounds rather than rearranging plant metabolic pathways. Some compounds selectively affected during the 1^（st） rather than 2^（nd） stage（e.g. phenylalanine; tryptophan; cellobiose;xylose; quinate; xylonite; idonate; and iso-threonate; cellobiotol and arbutine） are upstream events in the phenylpropanoid,terpenoid and alkaloid biosynthesis. These compounds could be designated as potential markers of initial RPW attack. However,further investigation is needed to determine efficient early screening methods of RPW attack based on the concentrations of these molecules.

Diabetes mellitus and associated chronic hyperglycemia enhance the risk of acute ischemic stroke and lead to worsened clinical outcome and increased mortality. However, post-stroke hyperglycemia is also present in a number of non-diabetic patients after acute ischemic stroke, presumably as a stress response. The aim of this review is to summarize the main effects of hyperglycemia when associated to ischemic injury in acute stroke patients, highlighting the clinical and neurological outcomes in these conditions and after the administration of the currently approved pharmacological treatment, i.e. insulin. The disappointing results of the clinical trials on insulin(including the hypoglycemic events) demand a change of strategy based on more focused therapies. Starting from the comprehensive evaluation of the physiopathological alterations occurring in the ischemic brain during hyperglycemic conditions, the effects of various classes of glucose-lowering drugs are reviewed, such as glucose-like peptide-1 receptor agonists, DPP-4 inhibitors and sodium glucose cotransporter 2 inhibitors, in the perspective of overcoming the up-to-date limitations and of evaluating the effectiveness of new potential therapeutic strategies.

Condensing osteitis of the clavicle in children

AIM: To confirm the rarity of this disorder and then to evaluate the effects of antibiotic treatment alone and assess whether this could produce a complete remission of symptoms in children and ***: We made a retrospective review of all cases of condensing osteitis of the clavicle in children and adolescents between January 2007 and January 2016. Outpatient and inpatient medical records, with radiographs, magnetic resonance imaging, triphasic bone scan and computed tomography scans were retrospectively reviewed. All the patients underwent biopsy of the affected clavicle and were treated with intra venous(IV) antibiotics followed by oral ***: Seven cases of condensing osteitis of the clavicle were identified. All the patients presented with swelling of the medial end of the clavicle, and 5 out of 7 reported persisting pain. The patients' mean age at presentation was 11.5 years(range 10.5-13). Biopsy confirmed the diagnosis in all cases. All the patients completed the treatment with IV and oral antibiotics. At last follow-up visit none of the patients complained of residual pain; all had a clinically evident reduction in the swelling of the medial end of the affected clavicle. The mean follow-up was 4 years(range 2-7).CONCLUSION: Our findings show that condensing osteitis of the clavicle is a rare condition. Biopsy is needed to confirm diagnosis. The condition should be managed with IV and oral antibiotics. Aggressive surgery should be avoided.

The introduction of direct-acting antivirals(DAAs)has revolutionized the natural history of chronic hepatitis C virus(HCV)infection,as the high rates of sustained virologic response(SVR)have been associated with improved survival in both cirrhotic and noncirrhotic patients(1).However,the risk of hepatocellular carcinoma(HCC)after DAA treatment has been a hot topic in the literature of the last few years,especially after two initial studies reporting unexpectedly high rates of both HCC occurrence and recurrence after SVR(2,3).
