Effects of Tinospora tuberculata leaf methanol extract on seedling growth of rice and associated weed species in hydroponic culture

The study was conducted to evaluate the responses of rice and rice weed seedlings （barnyardgrass and weedy rice） at the three-leaf stage toTinospora tuberculata leaf methanol extract （3.12, 6.25 and 12.5 g L–1） under hydroponic culture. It shows that the leaf methanol extract had various degrees effects depending on target plant species and each tested index （biomass, root length, shoot length, transpiration volume, chlorophyla, chlorophylb and carotenoid contents）. The effec-tive concentration of the leaf extract capable of reducing 50% of rice growth was higher than those of target weed species. Moreover, the root length was more tolerant to leaf methanol extract in comparison to the other plant parameters measured. A greater reduction was observed in chlorophyla content compared to chlorophylb and carotenoid. The results revealed that the reduction of transpiration volume closely coincided with the magnitude of growth inhibition of tested plants. Ultra-fast liquid chromatography analysis revealed 11 of 32 peaks in chemical proifle, including benzoic acid, caffeic acid, chlorogenic acid, isoorientin, isovitexin, orientin,p-anisic acid, syringic acid,trans-cinnamic acid,trans-ferulic acid, and vitexin have the same retention time with those peaks of the extract. The amount of compounds was present in the range of between 4817 and 115.5 mg kg–1 dry weight （DW）. The concentration-response bioassay of al 11 individual compounds and their equimolar mixture against the seeds of barnyardgrass revealed their contribution in the alelopahic activity ofT. tuberculata leaf extract. The examined compounds and their combination exhibited various degrees of growth inhibitory effects on the early growth of barnyardgrass. Therefore, the speciifc number, concentration, combination and inhibitory activity of bioactive compounds leads to alelopathy activity ofT. tuberculata leaves which could be employable directly as a natural herbicide and its growth inhibitor compounds can be used as a template for producing new herbicides.

Hippo-YAP/TAZ signaling in breast cancer:Reciprocal regulation of microRNAs and implications in precision medicine

Breast cancer is a molecularly heterogeneous disease and the most common female malignancy.In recent years,therapy approaches have evolved to accommodate molecular diversity,with a focus on more biologically based therapies to minimize negative consequences.To regulate cell fate in human breast cells,the Hippo signaling pathway has been associated with the alpha subtype of estrogen receptors.This pathway regulates tissue size,regeneration,and healing,as well as the survival of tissue-specific stem cells,proliferation,and apoptosis in a variety of organs,allowing for cell differentiation.Hippo signaling is mediated by the kinases MST1,MST2,LATS1,and LATS2,as well as the adaptor proteins SAV1 and MOB.These kinases phosphorylate the downstream effectors of the Hippo pathway,yes-associated protein(YAP),and transcriptional coactivator with PDZ-binding motif(TAZ),suppressing the expression of their downstream target genes.The Hippo signaling pathway kinase cascade plays a significant role in all cancers.Understanding the principles of this kinase cascade would prevent the occurrence of breast cancer.In recent years,small noncoding RNAs,or microRNAs,have been implicated in the development of several malignancies,including breast cancer.The interconnections between miRNAs and Hippo signaling pathway core proteins in the breast,on the other hand,remain poorly understood.In this review,we focused on highlighting the Hippo signaling system,its key parts,its importance in breast cancer,and its regulation by miRNAs and other related pathways.

Niosome-encapsulated auraptene reduced the mRNA expression of VEGF-A and PDGFs genes in human retinaderived RPE cell line

AIM:To evaluate the effect of auraptene(AUR)treatment in forms of free and encapsulated in niosome nanoparticles by investigating the mRNA expression level of vascular endothelium growth factor(VEGF)-A and platelet-derived growth factors(PDGFs)in human retinal pigment epithelium(RPE)cell line.METHODS:Niosome nanocarriers were produced using two surfactants Span 60 and Tween 80.RPE cell line was treated with both free AUR and niosome-encapsulated.Optimum dosage of treatments was calculated using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT)assay.Expression of VEGF-A and PDGF-A,PDGF-B,PDGF-C,PDGF-D genes was measured after total RNA extraction and cDNA synthesis,using real-time polymerase chain reaction(RT-PCR).RESULTS:The highest entrapment efficiency(EE)was achieved by Span 60:cholesterol(1:1)with 64.3%.The half maximal inhibitory concentration(IC50)of free and niosome-encapsulated AUR were 38.5 and 27.78µg/mL,respectively.Release study revealed that niosomal AUR had more gradual delivery to the cells.RT-PCR results showed reduced expression levels of VEGF-A,PDGF-A,PDGF-B,PDGF-C,and PDGF-D after treatment with both free and niosomal AUR.CONCLUSION:Niosomal formulation of Span 60:cholesterol(1:1)is an effective drug delivery approach to transfer AUR to RPE cells.VEGF-A,PDGF-A,PDGF-B,PDGF-C,and PDGF-D are four angiogenic factors,inhibiting which by niosomal AUR may be effective in age-related macular degeneration.

AIM:To investigate the impact of niosome nanoparticles carrying umbelliprenin(UMB),an anti-angiogenic and anti-inflammatory plant compound,on the expression of vascular endothelial growth factor(VEGF-A)and connective tissue growth factor(CTGF)genes in a human retinal pigment epithelium(RPE)-like retina-derived cell line.METHODS:UMB-containing niosomes were created,optimized,and characterized.RPE-like cells were treated with free UMB and UMB-containing niosomes.The IC_(50)values of the treatments were determined using an MTT assay.Gene expression of VEGF-A and CTGF was evaluated using real-time polymerase chain reaction after RNA extraction and cDNA synthesis.Niosomes’characteristics,including drug entrapment efficiency,size,dispersion index,and zeta potential were assessed.Free UMB had an IC_(50)of 96.2μg/mL,while UMB-containing niosomes had an IC_(50)of 25μg/mL.RESULTS:Treatment with UMB-containing niosomes and free UMB resulted in a significant reduction in VEGF-A expression compared to control cells(P=0.001).Additionally,UMB-containing niosomes demonstrated a significant reduction in CTGF expression compared to control cells(P=0.05).However,there was no significant reduction in the expression of both genes in cells treated with free UMB.CONCLUSION:Both free UMB and niosome-encapsulated UMB inhibits VEGF-A and CTGF genes expression.However,the latter demonstrates significantly greater efficacy,potentially due to the lower UMB dosage and gradual delivery.These findings have implications for anti-angiogenesis therapeutic approaches targeting age-related macular degeneration.

Development of hybrid optimization algorithm for structures furnished with seismic damper devices using the particle swarm optimization method and gravitational search algorithm

Previous studies about optimizing earthquake structural energy dissipation systems indicated that most existing techniques employ merely one or a few parameters as design variables in the optimization process,and thereby are only applicable only to simple,single,or multiple degree-of-freedom structures.The current approaches to optimization procedures take a specific damper with its properties and observe the effect of applying time history data to the building;however,there are many different dampers and isolators that can be used.Furthermore,there is a lack of studies regarding the optimum location for various viscous and wall dampers.The main aim of this study is hybridization of the particle swarm optimization(PSO) and gravitational search algorithm(GSA) to optimize the performance of earthquake energy dissipation systems(i.e.,damper devices) simultaneously with optimizing the characteristics of the structure.Four types of structural dampers device are considered in this study:(ⅰ) variable stiffness bracing(VSB) system,(ⅱ) rubber wall damper(RWD),(ⅲ) nonlinear conical spring bracing(NCSB) device,(iv) and multi-action stiffener(MAS) device.Since many parameters may affect the design of seismic resistant structures,this study proposes a hybrid of PSO and GSA to develop a hybrid,multi-objective optimization method to resolve the aforementioned problems.The characteristics of the above-mentioned damper devices as well as the section size for structural beams and columns are considered as variables for development of the PSO-GSA optimization algorithm to minimize structural seismic response in terms of nodal displacement(in three directions) as well as plastic hinge formation in structural members simultaneously with the weight of the structure.After that,the optimization algorithm is implemented to identify the best position of the damper device in the structural frame to have the maximum effect and minimize the seismic structure response.To examine the performance of the proposed PSO-GSA optimization method,it h

The cryptic lifestyle of most fungi necessitates molecular identification of the guild in environmental studies.Over the past decades,rapid development and affordability of molecular tools have tremendously improved insights of the fungal diversity in all ecosystems and habitats.Yet,in spite of the progress of molecular methods,knowledge about functional properties of the fungal taxa is vague and interpretation of environmental studies in an ecologically meaningful manner remains challenging.In order to facilitate functional assignments and ecological interpretation of environmental studies we introduce a user friendly traits and character database FungalTraits operating at genus and species hypothesis levels.Combining the information from previous efforts such as FUNGuild and FunFun together with involvement of expert knowledge,we reannotated 10,210 and 151 fungal and Stramenopila genera,respectively.This resulted in a stand-alone spreadsheet dataset covering 17 lifestyle related traits of fungal and Stramenopila genera,designed for rapid functional assignments of environmental stud-ies.In order to assign the trait states to fungal species hypotheses,the scientific community of experts manually categorised and assigned available trait information to 697,413 fungal ITS sequences.On the basis of those sequences we were able to summarise trait and host information into 92,623 fungal species hypotheses at 1%dissimilarity threshold.

Sensitivity Analysis of Pollutants and Pattern Factor in a Gas Turbine Model Combustor due to Changes in Stabilizing Jets Characteristics

In the present paper,a sensitivity analysis of pollutants and pattern factor in a model combustor due to changes in the geometrical characteristics of stabilizing jets has been carried out.The exhaust pollutants including NO_(x),CO and soot have been chosen due to their harmful effect on the environment.The pattern factor has been also considered owing to its impact on turbine blades.The geometrical characteristics comprise diameter,angle and position of stabilizing jets.Eulerian-Lagrangian approach has been employed to model liquid fuel injection and distribution,breakup and evaporation of droplets.For the analysis of reactive-spray flow characteristics,RANS approach,realizable k-εturbulence model,discrete ordinates radiative heat transfer model and steady flamelet combustion model together with the chemical reaction mechanism of diesel fuel(C_(10)H_(22))have been applied.NO_(x) modeling has been performed via post-processing.Sensitivity analysis is such that by making variations in the problem inputs(diameter,angle and position of jets)in an organized manner,the effects on the outputs(NO_(x),CO,soot and pattern factor)are predicted.The number and order of simulations are predicted by design of experiments and full factorial model.Results have been analyzed using analysis of variance.It has been observed that if interactions among the characteristics of jets are considered,it is possible to analyze the exhaust pollutants more accurately.In fact,by using the interactions,it is likely to find a point where all output parameters are improved.Results show that by considering interactions of stabilizing jet characteristics,the maximum values of NO_(x),CO,soot and pattern factor change from 13.927 ppm,11.198%mole fraction,2.877 ppm and 0.043 to 26.233 ppm,14.693%mole fraction,142.357 ppm and 0.060,respectively.Furthermore,the minimum values change from 5.819 ppm,7.568%mole fraction,0.013 ppm and 0.029 to 6.098 ppm,5.987%mole fraction,0.002 ppm and 0.027,respectively.

In this experimental study,the impact of Portland cement replacement by ground granulated blast furnace slag(GGBFS)and micronized rubber powder(MRP)on the compressive,flexural,tensile strengths,and rapid chloride migration test(RCMT)of concrete were assessed.In this study,samples with different binder content and water to binder ratios,including the MRP with the substitution levels of 0%,2.5%and 5%,and the GGBFS with the substitution ratios of 0%,20%and 40%by weight of Portland cement were made.According to the results,in the samples containing slag and rubber powder in the early ages,on average,a 12.2%decrease in the mechanical characteristics of concrete was observed,nonetheless with raising the age of the samples,the impact of slag on reducing the porosity of concrete lowered the negative impact of rubber powder.Regarding durability characteristics,the RCMT results of the samples were enhanced by using rubber powder because of its insulation impact.Moreover,adding slag into the MRP-included mixtures results in a 23%reduction in the migration rate of the chloride ion averagely.At last,four mathematical statements were derived for the mechanical and durability of concrete containing the MRP and GGBFS utilizing the genetic programming method.
