Targeted and activatable nanosystem for fluorescent and optoacoustic imaging of immune-mediated inflammatory diseases and therapy via inhibiting NF-κB/NLRP3 pathways

Immune-mediated inflammatory diseases(IMIDs)represent a diverse group of diseases and challenges remain for the current medications.Herein,we present an activatable and targeted nanosystem for detecting and imaging IMIDs foci and treating them through blocking NF-κB/NLRP3 pathways.A ROS-activatable prodrug BH-EGCG is synthesized by coupling a near-infrared chromophore with the NF-κB/NLRP3 inhibitor epigallocatechin-3-gallate(EGCG)through boronate bond which serves as both the fluorescence quencher and ROS-responsive moiety.BH-EGCG molecules readily form stable nanoparticles in aqueous medium,which are then coated with macrophage membrane to ensure the actively-targeting capability toward inflammation sites.Additionally,an antioxidant precursor N-acetylcysteine is co-encapsulated into the coated nanoparticles to afford the nanosystem BH-EGCG&NAC@MM to further improve the anti-inflammatory efficacy.Benefiting from the inflammation-homing effect of the macrophage membrane,the nanosystem delivers payloads(diagnostic probe and therapeutic drugs)to inflammatory lesions more efficiently and releases a chromophore and two drugs upon being triggered by the overexpressed in-situ ROS,thus exhibiting better theranostic performance in the autoimmune hepatitis and hind paw edema mouse models,including more salient imaging signals and better therapeutic efficacy via inhibiting NF-κB pathway and suppressing NLRP3 inflammasome activation.This work may provide perceptions for designing other actively-targeting theranostic nanosystems for various inflammatory diseases.

Highly dispersed Ni/MgO-mSiO_(2)catalysts with excellent activity and stability for dry reforming of methane

Highly dispersed Ni catalyst and alkaline promoters supported by mesoporous SiO_(2)nanospheres were synthesized and applied as an active and stable catalyst for dry reforming of methane(DRM).The as-prepared Ni/MgO-mSiO_(2)catalyst showed stable conversions of CH4 and CO_(2)around 82%and 85%in 120 h of DRM reaction,which was superior in performance compared to similar catalysts in literatures.Based on the transmission electron microscope(TEM)images,energy-dispersive spectroscopy(EDS),CO-pulse adsorption,temperature programmed reduction of the oxidized catalysts by hydrogen(H_(2)-TPR),X-ray photoelectron spectroscopy(XPS),temperature-programmed desorption of CO_(2)(CO_(2)-TPD),and thermal gravitational analysis(TGA),the promotion effect of MgO on the Ni catalyst was systematically studied.The introduction of Mg^(2+)in synthesis enhanced the interaction between Ni^(2+)and mSiO_(2),which led to a high dispersion of active centers and a strong“metal–support”interactions to inhibit the sintering and deactivation of Ni at reaction temperatures.On the other hand,Ni and MgO nanoparticles formed adjacently on mSiO_(2),where the“Ni-MgO”interface not only improved the Ni0 distribution and promoted the cracking of CH_(4)but also promoted the activation of CO_(2)and the elimination of carbon deposits.A high and stable conversion of CH4 and CO_(2)were then achieved through the synergistic effect of Ni catalyst,MgO promoter,and mSiO_(2)support.

of main observation and conclusion In-situ overexpressed hydrogen peroxide could serve as a biomarker for inflammation and ischemic kidney injury.Herein,a nanoprobe was developed for assay of hydrogen peroxide.The nanoprobe(TA-TPABQ)was formed via the boronate ester groups between the hydrophilic tannic acid and the boric-acid-containing compound(TPABQ)as well as the hydrophobic interactions.The probe with good photostability shows good sensitivity and selectivity towards H2O2-The probe was adopted for identifying endogenous and exogenous H2O2 in living cells.Moreover,the probe was utilized for in vivo imaging experiments in acute abdomina and ankle inflammation mouse models as well as acute renal ischemia mouse model.

Temperature-responsive Behavior of Polymer Fluorescent System via Electrostatic Interaction Mediated Aggregation/Deaggregation

A simple and effective polymer fluorescent thermosensitive system was successfully developed based on the synergistic effect of excimer/monomer interconversion of pyrene derivatives and electrostatic interaction between polyelectrolyte and charged fluorophore. As for the system, the excimer-monomer conversion, thermosensitive behavior and thermo-responsive reversibility were investigated experimentally. Temperature variation and temperature-distribution induced fluorescence changes can be observed directly by naked eyes. Thus, this polymer system holds promise for serving as a fluorescent thermometer.

Cost-Benefit Analysis of the Wuxikou Integrated Flood Management Project Considering the Effects of Flood Risk Reduction and Resettlement

Evaluation of the economic costs and benefits of flood disaster risk management projects is crucial.However,current costbenefit analyses(CBA) often lack reliable estimates of the expected loss reduction from flood control measures and ignore quantitative assessments of resettlement.To address these limitations,this study incorporated a probabilistic risk analysis method and quantitative resettlement benefits assessment into the CBA framework,using the Wuxikou Integrated Flood Management Project(WIFMP) in Jiangxi Province,China,as a case study.The direct economic benefits of flood control were estimated by integrating hydrological statistics,numerical flood inundation simulation,and quantitative damage analysis with exposure and vulnerability data.Furthermore,the resettlement benefits were quantified by measuring the annual income growth of migrants based on assumptions about household employment.Our analysis shows that the total WIFMP investment is RMB 3546.1 million yuan(USD 1=RMB 6.976 yuan),including loan principal and interest of 244.4 million yuan,and operations and maintenance of 605.5 million yuan at 2020 prices.Annual project benefits are estimated at 351.3 million yuan in flood risk reduction,155.7-191.9 million yuan from increased resettlement income,and 42.7 million yuan in power and water revenues.Considering the costs and benefits across the entire project lifecycle,the internal rate of return ranges from 13.7 to 14.2%,and the net present value ranges from 31.8 to 352.6 billion yuan.Through improved benefit estimation methodology,this research enables a more reliable and holistic evaluation of costs and benefits for flood risk management projects.It provides insights for policymakers and practitioners involved in similar projects,contributing to more informed decision making and better allocation of resources in flood disaster risk management.

Grain size effect on the assembly quality of micro-scaled barrel formed by microforming

In this research,a method employing micro-extrusion was designed to produce the micro-scaled barrel-shaped parts with complex geometrical features to study the feasibility of the proposed microforming method and its grain size effect on the formability of the complicated internal features in terms of deformation behavior,material evolution,accuracy of dimensions and final components quality.The results reveal that the deformation behavior is highly affected by grain size and becomes unpredictable with increased grain size.In addition,assembly parameters including feature dimension,tolerance and coaxiality also vary with grain size,and the variation of grain size needs to be accommodated by different assembly types,viz.,clearance fit or transition fit.From the microstructural evolution aspect,it was identified there were two dead zones and four shear bands,and the formation of these deformation zones was barely affected by the variation in grain size.Though bulges,cracks,and fracture induced voids were observed on the surface of the final components,tailoring the microstructure of the working material with finer grains could significantly avoid these defects.This study advances the understanding of forming microparts by extrusion processes and provides guidance for microforming of similar microparts.

Monte Carlo Simulation-Based Robust Workflow Scheduling for Spot Instances in Cloud Environments

When deploying workflows in cloud environments,the use of Spot Instances(SIs)is intriguing as they are much cheaper than on-demand ones.However,Sls are volatile and may be revoked at any time,which results in a more challenging scheduling problem involving execution interruption and hence hinders the successful handling of conventional cloud workflow scheduling techniques.Although some scheduling methods for Sls have been proposed,most of them are no more applicable to the latest Sls,as they have evolved by eliminating bidding and simplifying the pricing model.This study focuses on how to minimize the execution cost with a deadline constraint when deploying a workflow on volatile Sls in cloud environments.Based on Monte Carlo simulation and list scheduling,a stochastic scheduling method called MCLS is devised to optimize a utility function introduced for this problem.With the Monte Carlo simulation framework,MCLS employs sampled task execution time to build solutions via deadline distribution and list scheduling,and then returns the most robust solution from all the candidates with a specific evaluation mechanism and selection criteria.Experimental results show that the performance of MCLS is more competitive comparedwithtraditionalalgorithms.

On the Use of Recycled Asphalt and Trinidad Lake Asphalt(TLA)for the Preparation of High Modulus Asphalt Concrete

Rutting of asphalt pavement occurs earlier and is more serious under the increasingly heavy traffic load conditions that can be found in subtropical monsoon climate regions.High modulus asphalt concrete(HMAC)with excellent anti-rutting and anti-fatigue properties is generally used to mitigate this issue.Given the relatively high cost of the additives used in this type of asphalt,in this study the feasibility of using recycled asphalt mixture(RAP)and Trinidad lake asphalt(TLA)for the preparation of HMAC is considered.The mineral composition of the RAP is first analyzed,then the TLA modified asphalt with different levels of RAP content is used to assess the effect of RAP on the performance of the mixture.Finally,the high-temperature rheological properties of extracted bitumen and the adhesion characteristic between mineral and bitumen are analyzed.The results show that after RAP combustion,the yellowish and greyish minerals display different alkalinity values,while the greyish mineral exhibits an improved ability to combine with bitumen.Thus,RAP can significantly enhance the high-temperature rutting resistance of TLA modified asphalt mixtures.The aged bitumen in RAP makes the mixed binder harder and improves its rheological properties at high temperatures.

Chronic hepatitis B virus(HBV)infection remains a major public health concern globally,and T cell responses are widely believed to play a pivotal role in mediating HBV clearance.Accordingly,research on the characteristics of HBV-specific T cell responses,from activation to exhaustion,has advanced rapidly.Here,we summarize recent developments in characterizing T cell immunity in HBV infection by reviewing basic and clinical research published in the last five years.We provide a comprehensive summary of the mechanisms that induce effective anti-HBV T cell immunity,as well as the latest developments in understanding T cell dysfunction in chronic HBV infection.Furthermore,we briefly discuss current novel treatment strategies aimed at restoring anti-HBV T cell responses.

Electrochemical cutting of mortise-tenon joint structure by rotary tube electrode with helically distributed jet-flow holes

In aero-engines,mortise-tenon joint structures are often used to connect the blades to the turbine disk.The disadvantages associated with conventional manufacturing techniques mean that a low-cost,high-efficiency,and high-quality nickel-based mortise–tenon joint structure is an urgent requirement in the field of aviation engineering.Electrochemical cutting is a potential machining method for manufacturing these parts,as there is no tool degradation in the cutting process and high-quality surfaces can be obtained.To realize the electrochemical cutting of a mortise-tenon joint structure,a method using a tube electrode with helically distributed jet-flow holes on the side-wall is proposed.During feeding,the tube electrode rotates along its central axis.Flow field simulations show that the rotational speed of the tube electrode determines the direct spraying time of the high-speed electrolyte ejected from the jet-flow holes to the machining area,while the electrolyte pressure determines the flow rate of the electrolyte and the velocity of the electrolyte ejected from the jet-flow holes.The machining results using the proposed method are verified experimentally,and the machining parameters are optimized.Finally,mortise and tenon samples are successfully machined using 20 mm thick Inconel 718 alloy with a feeding rate of 5μm/s.
