TCM-HIN2Vec: A strategy for uncovering biological basis of heart qi deficiency pattern based on network embedding and transcriptomic ex.eriment

Objective: To elucidate the biological basis of the heart qi deficiency (HQD) pattern, an in-depth understanding of which is essential for improving clinical herbal therapy. Methods: We predicted and characterized HQD pattern genes using the new strategy, TCM-HIN2Vec, which involves heterogeneous network embedding and transcriptomic ex.eriments. First, a heterogeneous network of traditional Chinese medicine (TCM) patterns was constructed using public databases. Nex., we predicted HQD pattern genes using a heterogeneous network-embedding algorithm. We then analyzed the functional characteristics of HQD pattern genes using gene enrichment analysis and ex.mined gene ex.ression levels using RNA-seq. Finally, we identified TCM herbs that demonstrated enriched interactions with HQD pattern genes via herbal enrichment analysis. Results: Our TCM-HIN2Vec strategy revealed that candidate genes associated with HQD pattern were significantly enriched in energy metabolism, signal transduction pathways, and immune processes. Moreover, we found that these candidate genes were significantly differentially ex.ressed in the transcriptional profile of mice model with heart failure with a qi deficiency pattern. Furthermore, herbal enrichment analysis identified TCM herbs that demonstrated enriched interactions with the top 10 candidate genes and could potentially serve as drug candidates for treating HQD. Conclusion: Our results suggested that TCM-HIN2Vec is capable of not only accurately identifying HQD pattern genes, but also deciphering the basis of HQD pattern. Furthermore our finding indicated that TCM-HIN2Vec may be further ex.anded to develop other patterns, leading to a new approach aimed at elucidating general TCM patterns and developing precision medicine. © 2024 Beijing University of Chinese Medicine

Neuroprotective effects of Shaoyao Gancao decoction against ex.itatory damage in PC12 cells based on the Src-NR2-nNOS pathway

Objective: To ex.lore the neuroprotective effects of the Shaoyao Gancao decoction (SGD) against ex.itatory damage in PC12 cells and the role of the Src-NR2-nNOS pathway mediation by SGD in regulating γ-aminobutyric acid (GABA)-glutamate (Glu) homeostasis. Methods: N-Methyl-D-aspartic acid (NMDA) was used to establish a PC12 cell ex.itability injury model. To investigate the neuroprotective effect of SGD, a cell counting kit-8 (CCK-8) assay was used to determine PC12 cell viability, Annex.n V/Propidium Iodide (Annex.n V/PI) double staining was used to determine PC12 cell apoptosis, and Ca2+ concentration was observed using laser confocal microscopy. GABA receptor agonists and antagonists were used to analyze the neuroprotective interactions between γ-aminobutyric acid (GABA) and NMDA receptors. Additionally, molecular biology techniques were used to determine mRNA and protein ex.ression in the Src-NR2-nNOS pathway. We analyzed the correlations between the regulatory sites of GABA and NMDA interactions, ex.itatory neurotox.city, and brain damage at the molecular level. Results: NMDA ex.itotox.c injury manifested as a significant decrease in cell activity, increased apoptosis and caspase-3 protein ex.ression, and a significant increase in intracellular Ca2+ concentration. Administration of SGD, a GABAA receptor agonist (muscimol), or a GABAB receptor agonist (baclofen) decreased intracellular Ca2+ concentrations, attenuated apoptosis, and reversed NMDA-induced upregulation of caspase-3, Src, NMDAR2A, NMDAR2B, and nNOS. Unex.ectedly, a GABAA receptor antagonist (bicuculline) and a GABAB receptor antagonist (saclofen) failed to significantly increase ex.itatory neurotox.city. Conclusions: Taken together, these results not only provide an ex.erimental basis for SGD administration in the clinical treatment of central nervous system injury diseases, but also suggest that the Src-NR2A-nNOS pathway may be a valuable target in ex.itotox.city treatment. © 2024 Beijing Universit

High Temperature Ox.dation and Wear Resistance of Y-modified Hot Dipping Aluminized Coating on SCH12 Steel

Evaluating the Potential Economic Benefits of the New South Wales Node of Australia＇s Integrated Marine Observing System： [NSW-IMOS]

The Integrated Marine Observing System [IMOS] is an Australian national program for observing the oceans around Australia. As one of its important nodes, the New South Wales Integrated Marine Observing System （NSW-IMOS] aims to provide more accurate descriptions of the East Australian Current [EAC]. The purpose of this paper is to evaluate the potential economic benefits from NSW-IMOS. Six.related sectors which can potentially be among its main beneficiaries are considered： beach recreation, commercial fishing, recreational fishing, recreational boating, natural hazard predictions, and oil spill mitigation. The 1% constant percentage increase evaluation method is used to estimate the potential economic benefits to these six.beneficiaries. By using this method, our study shows that the total potential economic benefit for these sectors is estimated to be $ 6.07 million per year. We consider that this is indicative but not conclusive in demonstrating some of the potential economic benefits that can be provided from information gathered by NSW-IMOS facilities. We conclude with further evaluative approaches that could be used to provide more accurate estimates of potential economic benefits.

Impact of non-ideal UE hardware on cell-free massive MIMO network with centralized operation

This paper investigates the impact of non-ideal user equipment(UE) hardware on a cell-free(CF) massive MIMO(mMIMO) network with centralized operation under spatially correlated channels. The minimum mean-squared error(MMSE) estimator can be derived with the help of the generic non-ideal UE hardware model. It is demonstrated that even if the effective signal-to-noise ratio approaches infinity, pilot contamination and imperfect hardware can cause a non-zero estimation error floor. After that, a lower bound is determined for the ergodic uplink capacity of the centralized CF mMIMO network under non-ideal UE hardware. Moreover, the optimal receive combining vector is obtained to max.mize the uplink spectral efficiency(SE). The max.mum ratio(MR) and regularized zero-forcing(RZF) combining schemes are offered as alternatives in light of the computational complex.ty of the MMSE receiver. Comparing the RZF to the MMSE scheme under different levels of hardware impairments, our findings indicate that the RZF receiver suffers a negligible loss in total SE. For MR combining, a novel closed-form uplink achievable SE ex.ression is obtained based on the MMSE estimator and the use-and-then-forget bounding technique. This ex.ression gives vital insights into the achievable uplink performance with UE hardware impairments. Besides, for various hardware impairment factors, the impact of pilot sequence length on average sum SE is disclosed for different receive combining schemes. To increase the overall SE of the max.min fairness scheme, a heuristic fractional power control scheme with UE hardware impairments is developed, which can essentially avoid sacrificing the SE of other UEs while max.mizing the SE of the unluckiest UE in the whole network. Finally,our theoretical performance analysis and power control algorithm are validated by simulation results, and fundamental design guidelines are provided for selecting hardware satisfying the practical UE requirements.
