Antioxidant activity of Bios-p peptide analogue in HEK293T cells and three-dimensional structure prediction

studies had indicate that excessive production of reactive oxygen species (ROs) affect cellular signaling pathways, which is associated with pathological and physiological conditions such as cancer, diabetes and neurodegenerative diseases In this context, our laboratory has obtained the Bios-p, a ROs modulator, peptide analogue by sequencing from the seed of Bauhinia bauhinoides, which represents the active 12-amino acid, obtained from the inhibitor BbKI protease and we predicted the three-dimensional structure of Bios-p analogue peptide using homology modeling, being patented by the working group of Dr. Maria Luiza Vilela Oliva of UNIFEsP, Brazil (a member of our cluster). The protective effect on the viability and antioxidant capacity of Bios-p was studied in HEK 293T cells under oxidative stress induced by hydrogen peroxide (H2O2) using sYTOXGREEN/DHE and luminescence assay. The three-dimensional structure of Bios-p peptide analogue was predicted by homology-based modeling using Modeller9v8. The pretreatment with different concentrations of Bios-p (1 μM - 10 μM) showed an increase of 53.83% ± 3.86% the cellular viability in under oxidative stress compared to control. Furthermore, the results to indicate that HEK293T cells by incubating for 24 h with Bios-p shown a significant decreased of basal extracellular ROs on total cell population in 89.67% ± 0.76%, compared to control in the absence of the analogue. similarly it is observed that Bios-p has a significant antioxidant effect on extracellular ROs production when cells are subjected to oxidative stress induced by 200 μM H2O2 in 64.37% ± 4.63%, compared to control in absence of H2O2 and Bios-p. These results suggest that Bios-p has potential as antioxidant agent in cells HEK293T under H2O2-induced oxidative stress and that can protect the cells viability as concentration-dependent, and we propose a new biotechnological tool for modulate the ROs production.

A family of Ce-doped LaCoO_(3) perovskites are presented as possible catalysts for Cl-VOCs *** materials with different contents of Ce were obtained through the citrate and the reactive grinding *** insertion of Ce in the original perovskite structure favours the presence of Co^(2+)/Co^(3+) and Ce^(3+)/Ce^(4+) redox pairs and a higher content of oxygen vacancies that enhances the catalytic performance in chlorobenzene combustion based on differential kinetics *** family obtained by the grinding method presents a performance as high as that synthesized by citrate ***,the reactive grinding is a feasible green chemistry alternative to obtain a catalyst with the same performance as that obtained from traditional ***,the stability of samples was evaluated under total combustion reaction conditions showing an excellent activity during 45 h time on stream.

Characterization of Evolutionarily ConservedMotifs Involved in Activity and Regulation of theABA-INsENsITIVE （ABI） 4 Transcription Factor

In recent years, the transcription factor ABI4 has emerged as an important node of integration for externaland internal signals such as nutrient status and hormone signaling that modulates critical transitions during the growthand development of plants. For this reason, understanding the mechanism of action and regulation of this protein rep-resents an important step towards the elucidation of crosstalk mechanisms in plants. However, this understanding hasbeen hindered due to the negligible levels of this protein as a result of multiple posttranscriptional regulations. To betterunderstand the function and regulation of the ABI4 protein in this work, we performed a functional analysis of severalevolutionarily conserved motifs. Based on these conserved motifs, we identified ortholog genes of ABI4 in differentplant species. The functionality of the putative ortholog from Theobroma cacao was demonstrated in transient expres-sion assays and in complementation studies in plants. The function of the highly conserved motifs was analyzed aftertheir deletion or mutagenesis in the Arabidopsis ABI4 sequence using mesophyll protoplasts. This approach permitted usto immunologically detect the ABI4 protein and identify some of the mechanisms involved in its regulation. We identi-fied sequences required for the nuclear localization （AP2-associated motif） as well as those for transcriptional activationfunction （LRP motif）. Moreover, this approach showed that the protein stability of this transcription factor is controlledthrough protein degradation and subcellular localization and involves the AP2-associated and the PEsT motifs. We dem-onstrated that the degradation of ABI4 protein through the PEsT motif is mediated by the 26s proteasome in responseto changes in the sugar levels.

The most recent information on fascial tissue indicates that there are not fascial layers, but polyhedral microvacuoles of connective tissue, which connect the body systems and, by hosting specialized cells, permit several functions, such as motor, nervous, vascular and visceral. These microvacuoles（a repetition of polyhedral units of connective fibrils） under internal or external tension change shape and can manage the movement variations, regulating different body functions and ensuring the maintenance of efficiency of the body systems. Their plasticity is based on perfect functional chaos： it is not possible to determine the motion vectors of the different fibrils, which differ in behavior and orientation; this strategy confers to the fascial continuum the maximum level of adaptability in response to the changing internal and external conditions of the cell. The present commentary deals with this concept, providing clinical examples of different disease patterns, providing contrary examples in which this adaptability does not occur, and lastly suggesting considerations for the approach to manipulative therapy of the fascial tissue. The fascial continuum is like a flock of birds flying together without a predetermined logic and maintaining their individuality at the same time.

Involvement of Estrogen Receptors in the Anxiolytic-Like Effect of Phytoestrogen Genistein in Rats with 12-Weeks Postovariectomy

Phytoestrogens are natural compounds found in some vegetables, and they replicate many of the physiochemical and physiological properties of estrogens, including the regulation of mood. The phytoestrogen genistein exerts anxiolytic-like effects in rats with a chronic absence of ovarian hormones, but the mechanism involved in this effect remains to be explored. The present study explored the participation of estrogen receptor-β in the anxiolytic-like effect of genistein (1.0 mg/kg, i.p., for 4 days) in Wistar rats with 12-weeks postovariectomy, considered as experimental model of post-surgical menopause. In the light/dark test, a useful tool for anxiety study and for the screening of anxiolytic drugs, genistein reduced the latency to enter and increased the time spent in the light compartment and significantly increased the frequency and time spent exploring the light compartment compared with the control group, which is considered as an anxiolytic-like effect at experimental level. All behavioral effects produced by genistein in the light/dark test were blocked by previous tamoxifen administration (5.0 mg/kg, s.c., for 6 days), a non selective antagonist for estrogen receptor-β. The effects produced by genistein or tamoxifen in this test were not related to significant changes in general motor activity evaluated in the open field test. In conclusion, the specific contribution of present investigation was identify that estrogen receptor-β is involved in the anxiolytic-like effect produced by phytoestrogen genistein in rats with a long-term absence of ovarian hormones;supporting the hypothesis that estrogen receptor-β participates in the regulation of anxiety associated with low concentration of ovarian hormones and in the anxiolytic-like effects produced by natural estrogenic compounds such as phytoestrogens.

AIM To investigate the main current etiologies of cirrhosis in *** We performed a cross-sectional retrospective multicenter study that included eight hospitals in different areas of Mexico. These hospitals provide health care to people of diverse social classes. The inclusion criteria were a histological, clinical, biochemical, endoscopic, or imaging diagnosis of liver cirrhosis. Data were obtained during a 5-year period(January 2012-December 2017). REsULTs A total of 1210 patients were included. The mean age was 62.5 years(sD = 12.1), and the percentages of men and women were similar(52.0% vs 48.0%). The most frequent causes of liver cirrhosis were hepatitis C virus(HCV)(36.2%), alcoholic liver disease(ALD)(31.2%), and nonalcoholic steatohepatitis(23.2%), and the least frequent were hepatitis B virus(1.1%), autoimmune disorders(7.3%), and other conditions(1.0%).CONCLUsION HCV and ALD are the most frequent causes of cirrhosis in Mexico. However, we note that non-alcoholic fatty liver disease(NAFLD) as an etiology of cirrhosis increased by 100% compared with the rate noted previously. We conclude that NAFLD will soon become one of the most frequent etiologies of liver cirrhosis in Mexico.

Beneﬁcial microbes can improve soil health by promoting soil structure,nutrient cycling,and disease *** addition,a wide array of rhizospheric microbes are responsible for producing metabolically active compounds including various types of plant growth ***,microbial biodiversity studies could contribute to the improvement of agricultural practices in deprived areas,such as the Pampean semiarid *** vast majority of studies conducted on endophytic microorganisms have focused on intensive crop legume *** contrast,little attention has been paid to microorganisms of native legumes,whose ecology is not directly affected by human *** this study,endophytic microorganisms isolated from root nodules of a selected native legume of the genus Rhynchosia were *** isolates were studied with a focus on their plant growth-pro-moting rhizobacteria(PGPR)*** the edaphic characteristics of the Pampean semiarid region,the isolates obtained were evaluated for their ability to grow under three salt stress conditions(50,100,and 200 mM NaCl)and four different pH values(6,7,8,and 9).Based on their PGPR activities,the selected strains were phylogenetically grouped using *** results showed great variability among the isolates in terms of the characteristics *** legumes manifested a wide endophytic variability and remarkable perfor-mance in PGPR *** conclude that they could be used as potential bioinoculants for legume cultivation,an excellent alternative to the use of chemical fertilizers that currently pollute the environment.

Beneficial effects of nutritional supplements on intestinal epithelial barrier functions in experimental colitis models in vivo

Acute and chronic colitis affect a huge proportion of the population *** etiology of colitis cases can be manifold,and diet can significantly affect onset and outcome of *** many forms of acute colitis are easily treatable,chronic forms of colitis such as ulcerative colitis and Crohn’s disease(summarized as inflammatory bowel diseases)are multifactorial with poorly understood *** bowel diseases are characterized by exacerbated immune responses causing epithelial dysfunction and bacterial *** is no cure and therapies aim at reducing inflammation and restoring intestinal barrier ***,most drugs can have severe side *** in diet and inclusion of nutritional supplements have been extensively studied in cell culture and animal models,and some supplements have shown promising results in clinical *** of these nutritional supplements including vitamins,fatty acids and phytochemicals reduce oxidative stress and inflammation and have shown beneficial effects during experimental colitis in rodents induced by dextran sulphate sodium or 2,4,6-trinitrobenzene sulfonic acid,which remain the gold standard in pre-clinical colitis ***,we summarize the mechanisms through which such nutritional supplements contribute to epithelial barrier stabilization.
