Intrahepatic cholangiocarcinoma: Introducing the preoperative prediction score based on preoperative imaging

Background: Intrahepatic cholangiocarcinoma(ICC) still has a poor long-term outcome, even after complete resection. We investigated different parameters gathered in preoperative imaging and analyzed their influence on resectability, recurrence, and survival. Methods: All patients who underwent exploration due to ICC between January 2008 and June 2018 were analyzed retrospectively. Kaplan-Meier model, log-rank test and Cox regression were used. Results: Out of 184 patients, 135(73.4%) underwent curative intended resection. Median overall survival(OS) was 22.2 months with a consecutive 1-, 3-and 5-year OS of 73%, 29%, and 17%. Median recurrencefree survival(RFS) was 9.3 months with a consecutive 1-, 3-and 5-year RFS of 36%, 15%, and 11%. Site of tumor, parenchymal localization, tumor configuration/dissemination, and estimated tumor volume had significant influence on resectability. Univariate analyses showed that site of tumor, tumor configuration/dissemination, number of nodules, and estimated tumor volume had predictive values for OS and RFS. Together with tumor size the preoperative prediction(POP) score was created showing significance for OS and RFS(all P<0.001). In multivariate analysis, POP score(HR=1.779;95% CI: 1.26 8-2.4 95;P=0.001), T stage(HR=1.255;95% CI: 1.040-1.514;P=0.018) and N stage(HR=1.334;95% CI: 1.081-1.645;P=0.007) were the independent predictors for OS. For RFS, POP score(HR=1.733;95% CI: 1.30 0-2.311;P<0.0 01) and M stage(HR=3.036;95% CI: 1.376-6.697;P=0.006) were the independent predictors. Conclusions: The POP score showed to have a highly significant influence on OS and RFS. The score is easy to assess through preoperative imaging. For patients in the high risk group at least staging laparoscopy or preoperative chemotherapy should be evaluated, because they showed equal outcome compared to the irresectable group.

Giant cell tumors of bone(GCTB)are associated with massive bone destructions and high recurrence *** a previous study,we observed cytotoxic effects of three different compositions of bioactive glasses(BGs)towards GCTSC but not bone marrow derived stromal cells(BMSC)indicating that BGs represent promising candidates for the development of new therapeutic *** the current study we aimed to investigate the molecular mechanisms that are involved in BG induced *** observed,that BG treatment was not associated with any signs of apoptosis,but rather led to a strong induction of mitogen activated protein kinases(MAPK)and,as a consequence,upregulation of several transcription factors specifically in *** wide gene expression profiling further revealed a set of fifteen genes that were exclusively induced in GCTSC or induced significantly stronger in GCTSC compared to *** treatment further induced autophagy that was significantly more pronounced in GCTSC compared to BMSC and could be inhibited by MAPK *** with the known osteogenic properties of BGs our findings support the suitability of BGs as therapeutic agents for the treatment of ***,these data have to be verified under in vivo conditions.

Anopheles gambiae larvicidal and adulticidal potential of Phyllanthus amarus(Schumach and Thonn,1827)obtained from different localities of Nigeria

Objective:To screen phytochemicals in ethanolic leaf extracts of Phyllanthus amarus collected from three different geographical zones in Nigeria and evaluate their effects on larva and adult of Anopheles ***:The sample extracts of Phyllanthus amarus prepared with ethanol solvent were tested against Anopheles gambiae at two important developmental stages of its life cycle using slightly modified WHO ***:Alkaloids,saponins,tannins,flavonoids,glycosides,phenols,and terpenes were detected in each *** these samples,the extract from northwest exhibited the highest larvicidal activity(LC50=263.02 ppm),followed by southeast and southwest extracts(LC50=288.40 and 295.12 ppm,respectively after 48 h),while the extract from southwest exhibited the highest adulticidal activity(LC50=275.42 ppm),followed by northwest and southeast extract(LC50=301.99 and 316.22 ppm,respectively after 24 h).A 50%larva mortality was almost attained at 600 ppm after 48 h duration of exposure to the northwest ***:The tested samples possess strong larvicidal and adulticidal property against Anopheles gambiae which depends on their chemical composition and localities of *** studies are needed to explore the insecticidal activity against a wider range of mosquito species,and to identify active ingredient(s)of the extract responsible for such activity.

Proteome alterations in aqueous humour of primary open angle glaucoma patients

AIM: To unravel the primary open angle glaucoma(POAG) related proteomic changes in aqueous humour(AH).METHODS: Totally 35 patients listed for cataract surgery(controls: n=12, age: 67.4±13.6 y) or trabeculectomy for POAG(n=23, age: 72.5±8.3 y) were included. AH samples of those patients were obtained during cataract surgery or trabeculectomy. AH samples were subsequently pooled into the experimental groups under equal contribution in terms of protein amount of each individual patient. Protein samples were analyzed by a linear trap quadrupol Orbitrap Mass Spectrometry device with an upstream liquid chromatography system. The obtained raw data were analyzed using the Maxquant proteome software and compared. Proteins with a fold-change ratio higher than a cut-off of 2 were considered as noticeably ***: A total number of 175 proteins could be identified out of the AH from POAG and cataract by means of quantitative mass spectrometric analysis. Apolipoprotein D(fold change, 3.16 times), complement C3(2.96), pigment epithelium-derived factor(2.86), dickkopf-related protein 3(2.18) and wingless-related integration(Wnt) inhibitory factor 1(2.35) were significantly upregulated within the AH of glaucoma compared to cataract serving as ***: AH provides a tool to analyze changes in glaucoma and shows striking changes in Wnt signaling inhibitory molecules and other proteins.

This work investigated the degradation of tin – based gas-diffusion electrodes (GDE) and also a promising Bi2O3 GDE in electrochemical CO_(2) reduction in highly alkaline media which has not been studied before. The contributions of the electrode wetting (or flooding, if excessively) and catalyst leaching on the degradation were analyzed. Therefore, electrochemical impedance spectroscopy was used to monitor the wetted surface area of the GDE in combination with post-mortem analysis of the penetration depth by visualizing the electrolyte’s cation in the GDE cross-section. Furthermore, to reveal a possible degradation of the electrocatalyst, its distribution was mapped in the GDEs cross-section after operation while the catholyte was additionally analyzed via ICP-MS. The results clearly demonstrate that the SnO_(2) catalyst dissolves in the reaction zone inside the GDE and might be partially redeposited near the GDEs surface. Since the redeposition process occurs only partially a steady loss of catalyst was observed impeding a clear distinction of the two degradation phenomena. Nevertheless, the deterioration of the electrode performance measured as faraday efficiency (FE) of the parasitic hydrogen evolution reaction (HER) qualitatively correlates with the differential double layer capacitance (Cdl). A significant difference of the rate of increase for the hydrogen FE and Cdl can be ascribed to the superposition of both above-mentioned degradation mechanisms. The demonstrated instability of SnO_(2) contrasts with the behavior of Bi2O3 GDE which is stabilized during CO_(2) conversion by redeposition of the diluted dissolved species as metallic Bi which is active for the CO_(2) reduction reaction.

Exercise and antioxidant supplements in the elderly

Both exercise and aging increase reactive oxygen species （ROS）, which can result in damage to cells. Aging is the result of damage caused by ROS to the mitochondrial genome in post mitotic cells and numerous studies have demonstrated an increase in ROS or their byproducts with exercise. ROS can cause oxidative stress as they overwhelm the antioxidant cellular defenses. Therefore interventions aimed at limiting or inhibiting ROS production, such as supplementation with antioxidant vitamins, should be able to reduce fatigue during muscle contraction and the rate of formation of aging changes with a consequent reduction of the aging rate and disease pathogenesis. However, it has been shown that ROS are essential signaling molecules which are required to promote the health benefits of exercise and longevity. In young individuals, ROS are required for normal force production in skeletal muscle, for the development of training-induced adaptations in endurance performance, as well as for the induction of the endogenous defense systems. Thus, taking antioxidants during training, in young athletes, seems to be detrimental. However, antioxidant supplementation may be expected to be beneficial and is receiving growing attention in the active old population. In this manuscript we review the literature associated with the main areas of interest in this topic.

Regulation of the New Arabidopsis Imprinted Gene AtBMIIC Requires the Interplay of Different Epigenetic Mechanisms

Recently, it has been shown that plants contain homologs to the animal Polycomb repressive complex I （PRC1） components BM11 and RINGIA/B. In Arabidopsis, there are three BMIl-like genes, two of which, AtBMIIA and B, are required during post-embryonic plant growth to repress embryonic traits and allow cell differentiation. However, little is known about the third BMIl-like gene, AtBMIIC. In this work, we show that AtBMIIC is only expressed during endosperm and stamen development. AtBMIIC is an imprinted gene expressed from the maternal allele in the endosperm but bialleli- cally expressed in stamen. We found that the characteristic expression pattern of AtBMIIC is the result of a complex epigenetic regulation that involves CG DNA methylation, RNA-directed non-CG DNA methylation （RdDM）, and PcG activity. Our results show the orchestrated interplay of different epigenetic mechanisms in regulating gene expression throughout development, shedding light on the current hypotheses for the origin and mechanism of imprinting in plant endosperm.
