Targeted genome editing technology has been widely used in biomedical studies. The CRISPR- associated RNA-guided endonuclease Cas9 has become a versatile genome editing tool. The CRISPR/Cas9 system is useful for studying gene function through efficient knock-out, knock-in or chromatin modification of the targeted gene loci in various cell types and organisms. It can be applied in a number of fields, such as genetic breeding, disease treatment and gene functional investigation In this review, we introduce the most recent developments and applications, the challenges, and future directions of Cas9 in generating disease animal model. Derived from the CRISPR adaptive immune system of bacteria, the development trend of Cas9 will inevitably fuel the vital applications from basic research to biotechnology and bio- medicine.

AIM:To clarify the association between Helicobacter pylori(***)infection and the risk of esophageal carcinoma through a meta-analysis of published ***:Studies which reported the association between *** infection and esophageal cancer published up to June 2013 were *** odds ratios(ORs)and corresponding 95%CIs of *** on esophageal cancer with respect to health control groups were *** were extracted independently by two investigators and discrepancies were resolved by discussion with a third *** statistical software,STATA(version 12.0),was applied to investigate heterogeneity among individual studies and to summarize the studies.A meta-analysis was performed using a fixed-effect or random-effect method,depending on the absence or presence of significant ***:No significant association between *** infection and esophageal squamous cell carcinoma(ESCC)risk was found in the pooled overall population(OR=0.97,95%CI:0.76-1.24).However,significant associations between *** infection and ESCC risk were found in Eastern subjects(OR=0.66,95%CI:0.43-0.89).Similarly,cytotoxin-associated gene-A(CagA)positive strains of infection may decrease the risk of ESCC in Eastern subjects(OR=0.77,95%CI:0.65-0.92),however,these associations were not statistically significant in Western subjects(OR=1.26,95%CI:0.97-1.63).For esophageal adenocarcinoma(EAC)the summary OR for *** infection and CagA positive strains of infection were 0.59(95%CI:0.51-0.68)and 0.56(95%CI:0.45-0.70),***:*** infection is associated with a decreased risk of ESCC in Eastern populations and a decreased risk of EAC in the overall population.

Knowledge about the effects of vegetation types on soil properties and on water dynamics in the soil profile is critical for revegetation strategies in water-scarce regions, especially the choice of vegetation type and human management measures. We focused on the analysis of the effects of vegetation type on soil hydrological properties and soil moisture variation in the 0–400 cm soil layer based on a long-term（2004―2016） experimental data in the northern Loess Plateau region, China. Soil bulk density（BD）, saturated soil hydraulic conductivity（Ks）, field capacity（FC） and soil organic carbon（SOC） in 2016, as well as the volumetric soil moisture content during 2004–2016, were measured in four vegetation types, i.e., shrubland（korshinsk peashrub）, artificial grassland（alfalfa）, fallow land and cropland（millet or potato）. Compared with cropland, revegetation with peashrub and alfalfa significantly decreased BD and increased Ks, FC, and SOC in the 0–40 cm soil layer, and fallow land significantly increased FC and SOC in the 0–10 cm soil layer. Soil water storage（SWS） significantly declined in shrubland and grassland in the 40–400 cm soil layer, causing severe soil drought in the deep soil layers. The study suggested that converting cropland to grassland（alfalfa） and shrubland（peashrub） improved soil-hydrological properties, but worsened water conditions in the deep soil profile. However, natural restoration did not intensify deep-soil drying. The results imply that natural restoration could be better than revegetation with peashrub and alfalfa in terms of good soil hydrological processes in the semi-arid Loess Plateau region.

Distributed Lagrange Multiplier/Fictitious Domain （DLM/FD） method was employed to simulate the channel flow with polymer suspension. The polymer molecules were modeled as Finitely Extensible Nonlinear Elastic （FENE） chains and FENE chain cluster. The coiled stretched transition of FENE chains was examined and the change in configuration of FENE chains was presented. The average velocity profile of the fully developed channel flow with 64 FENE chains was given and fitted well with the power-law curve. The change of chain cluster configuration was also simulated. These simulations show that DLM/FD method is capable of simulating the motion of not only FENE chain, but also FENE chain cluster.

The water environment of swimming fish in nature is always complex which includes various vortices and fluctuations. In order to study the interaction between the fish and its surrounding complex flow, the physical model with a D-section cylinder placed at the front of a flapping foil is employed. The D-section cylinder is used to produce vortices to contact with the foil as well as the vortices shed from the foil. According to the experimental work of Gopalkrishnan et al., there are three interaction modes between vortices shed from the cylinder and the flapping foil, which are expanding wake, destructive interaction and constructive interaction. Here in this article, three of those typical cases are picked up to reproduce the vortices interaction modes with the modified immersed boundary methods and their hydrodynamic performances are studied further. Results show that, for expanding wake mode and destructive interaction mode, the incoming vortices contact with the foil strongly, inducing relative low pressure domains at the leading-edge of the foil and enlarging the thrust of foils. For constructive mode, the foil slalom between the shed vortices from the D-section cylinder do not contact with them obviously and the foil＇s thrust is only enlarged a little.

Thermodynamic analysis of refractory siderite ore during carbothermic reduction was conducted. Micro- structure characteristics and phase transformation of siderite ore during sodium-carbonate-added catalyzing carboth- ermic reduction were investigated. X-ray diffraction （XRD）, scanning electron microscopy and energy-dispersive analysis of X rays were used to characterize the reduced samples. Results indicate that the solid reaction between FeO and SiO2 is inevitable during carbothermic reduction and the formation of fayalite is the main hindrance to the rapid reduction of siderite. The phase transformation of present siderite ore can be described as： siderite-magnetite-metallic iron, complying with the formation of abundant fayalite. Improving the reduction temperature （-1050 -C ） and duration is helpful for the formation and aggregation of metallic iron. The iron particle size in the reduced ore was below 20 l-m, and fayalite was abundant in the absence of sodium carbonate. With 5% Na2CO3 addition, the iron particle size in the reduced ore was generally above 50μm, and the diffraction intensity associated with metallic iron in the XRD pattern increased. The Na2O formed from the dissociation of Na2 CO3 can catalyze the carbothermie reduction of the siderite. This catalytic activity may be mainly caused by an increase in the reducing reaction activity of FeO.

Chemical biology has been attracting a lot of attention because of the key roles of chemical methods and techniques in helping to decipher and manipulate biological systems. Although chemical biology encompasses a broad field, this review will focus on chemical biology aimed at using exogenous chemical probes to interrogate, modify and manipulate biological processes, at the cellular and organismal levels,in a highly controlled and dynamic manner. In this area, many advances have been achieved for cancer biology and therapeutics, from target identification and validation based on active anticancer compounds(forward approaches) to discoveries of anticancer molecules based on some important targets including protein-protein interaction(reverse approaches). Herein we attempt to summarize some recent progresses mainly from China through applying chemical biology approaches to explore molecular mechanisms of carcinogenesis. Additionally, we also outline several new strategies for chemistry to probe cellular activities such as proximity-dependent labeling methods for identifying protein-protein interactions, genetically encoded sensors, and light activating or repressing gene expression system.

The use of coal fly ash（Cfa）, municipal solid waste incinerator bottom ash（MSWIBA） and flue gas desulfurization residue（FGDR） in road construction has become very common owing to its economical advantages. However, these residues may contain toxic constituents that pose an environmental risk if they leach out and flow through the soil, surface water and ***, it is necessary to assess the ecotoxicity and groundwater impact of these residues before decisions can be made regarding their utilization for road construction. In this study,the physico-chemical characteristics, leaching and phytotoxicity of these residues were investigated. Specifically, multivariate analyses were used to evaluate the contributions of the leaching constituents of the Cfa, MSWIBA and FGDR leachates to the germination index of wheat seeds. B, Ba, Cr, Cu, Fe and Pb were found to be more toxic to the wheat seeds than the other heavy metals. Furthermore, the leached concentrations of the constituents from the Cfa, MSWIBA and FGDR were below the regulatory threshold limits of the Chinese identification standard for hazardous wastes. Analyses conducted using a numerical groundwater model（Wisc LEACH） indicated that the predicted field concentrations of metals from the Cfa, MSWIBA and FGDR increased with time up to about 30 years at the point of compliance, then decreased with time and distance. Overall, this study demonstrated that the risks resulting from MSWIBA, Cfa and FGDR leaching could be assessed before its utilization for road construction, providing crucial information for the adoption of these alternative materials.

We study the uncertainties of quantum mechanical observables, quantified by the standard deviation(square root of variance) in Haar-distributed random pure states. We derive analytically the probability density functions(PDFs) of the uncertainties of arbitrary qubit *** on these PDFs, the uncertainty regions of the observables are characterized by the support of the PDFs. The state-independent uncertainty relations are then transformed into the optimization problems over uncertainty regions, which opens a new vista for studying stateindependent uncertainty relations. Our results may be generalized to multiple observable cases in higher dimensional spaces.

Developing earth-abundant electrocatalysts for hydrogen evolution reaction(HER)is important for the sustainable energy ***,efficient and stable heterocatalysts consisting of crystalline-amorphous M@MN_(x)(M=Co,Fe,Ni)encapsulated in N-doped carbon layers supported with N-doped graphene sheets(denoted as M@MN_(x)@NC-NG)are synthesized by facile hydrothermal reaction and nitridation *** the nitriding process,metal ions in M(tzbc)_2(H_(2)O)_4(tzbc=4-(1H-1,2,4-triazol-1-yl)benzoic acid)complexes are reduced to crystalline M cores,accompanied by the formation of amorphous MN_(x)shells;the tzbc ligands are insitu carbonized to form outermost N-doped carbon(NC)layers that connect with inner MN_(x)via M-N-C motifs inherited from the complex precursors and inhibit the transition of MN_(x)from amorphous to crystalline *** Co@CoN_(x)@NC-NG catalyst exhibits excellent HER activity with small overpotentials of 45 and 64 mV at a cathode current density of 10 mA·cm^(-2)and low Tafel slopes of 40 and 85 mV·dec^(-1)in 0.5 mol·L^(-1)H_(2)SO_4 and1.0 mol·L^(-1)KOH electrolytes,*** Co@CoN_(x)@NC-NG retains 97%of the initial overpotential after 100,000 s in both acidic and alkaline *** outstanding HER performance originates from the crystalline-amorphous Co@CoN_(x)that redistributes electrons around the heterointerfaces,facilitating the conversion process of H^(+)/H_(2)O to hydrogen and thereby promoting HER *** outermost NC layers serve as the armor of Co@CoN_(x),and graphene nanosheets act as carriers of egg-like Co@CoN_(x)@NC and conduction paths for electron shuttles,ensuring stable and continuous electrocatalytic hydrogen production.
