Peripheral nerve injuries induce a severe motor and sensory deficit. Since the availability of autologous nerve transplants for nerve repair is very limited, alternative treatment strategies are sought, including the use of tubular nerve guidance conduits(tNGCs). However, the use of tNGCs results in poor functional recovery and central necrosis of the regenerating tissue, which limits their application to short nerve lesion defects(typically shorter than 3 cm). Given the importance of vascularization in nerve regeneration, we hypothesized that enabling the growth of blood vessels from the surrounding tissue into the regenerating nerve within the tNGC would help eliminate necrotic processes and lead to improved regeneration. In this study, we reported the application of macroscopic holes into the tubular walls of silk-based tNGCs and compared the various features of these improved silk^(+) tNGCs with the tubes without holes(silk^(–) tNGCs) and autologous nerve transplants in an 8-mm sciatic nerve defect in rats. Using a combination of micro-computed tomography and histological analyses, we were able to prove that the use of silk^(+) tNGCs induced the growth of blood vessels from the adjacent tissue to the intraluminal neovascular formation. A significantly higher number of blood vessels in the silk^(+) group was found compared with autologous nerve transplants and silk^(–), accompanied by improved axon regeneration at the distal coaptation point compared with the silk^(–) tNGCs at 7 weeks postoperatively. In the 15-mm(critical size) sciatic nerve defect model, we again observed a distinct ingrowth of blood vessels through the tubular walls of silk^(+) tNGCs, but without improved functional recovery at 12 weeks postoperatively. Our data proves that macroporous tNGCs increase the vascular supply of regenerating nerves and facilitate improved axonal regeneration in a short-defect model but not in a critical-size defect model. Identifying cryptic species poses a substantial challenge to both biologists and naturalists due to morphological *** tabaci is a cryptic species complex containing more than 44 putative species;several of which are currently among the world's most destructive crop *** and delimiting the evolution of this species complex has proved *** develop a comprehensive framework for species delimitation and identification,we evaluated the performance of distinct data sources both individually and in combination among numerous samples of the *** species complex acquired *** datasets include full mitogenomes,single-copy nuclear genes,restriction site-associated DNA sequencing,geographic range,host speciation,and reproductive compatibility ***,our well-supported topologies generated from three dense molecular markers highlighted the evolutionary divergence of species of the *** complex and suggested that the nuclear markers serve as a more accurate representation of *** species *** compatibility datasets facilitated the identification of at least 17 different cryptic species within our *** geographic range information provides a complementary assessment of species recognition,while the host range datasets provide low rate of delimiting *** further summarized different data performances in species classification when compared with reproductive compatibility,indicating that combination of mtCOI divergence,nuclear markers,geographic range provide a complementary assessment of species ***,we represent a model for understanding and untangling the cryptic species complexes based on the evidence from this study and previously published articles.

Over the past decades,a number of water sciences and management programs have been developed to better understand and manage the water cycles at multiple temporal and spatial scales for various purposes,such as ecohydrology,global hydrology,sociohydrology,supply management,demand management,and integrated water resources management(IWRM).At the same time,rapid advancements have also been taking place in tracing,mapping,remote sensing,machine learning,and modelling technologies in hydrological *** those programs and advancements,a water crisis is intensifying *** missing link is effective interactions between the hydrological research and water resource management to support implementation of the UN Sustainable Development Goals(SDGs)at multiple spatial *** the watershed is the natural unit for water resources management,watershed science offers the potential to bridge this missing *** study first reviews the advances in hydrological research and water resources management,and then discusses issues and challenges facing the global water ***,it describes the core components of watershed science:(1)hydrological analysis;(2)water-operation policies;(3)governance;(4)management and *** framework takes into account water availability,water uses,and water quality;explicitly focuses on the storage,fluxes,and quality of the hydrological cycle;defines appropriate local water resource thresholds through incorporating the planetary boundary framework;and identifies specific actionable measures for water resources *** provides a complementary approach to the existing water management programs in addressing the current global water crisis and achieving the UN SDGs.

Perceptions of Sierra leonean Mango Farmers on the Pest Status and Current Management Practices for the Control of Fruit Fly in Sierra leone

Tephritid fruit flies are considered one of the world’s most notorious pests of horticultural crops, including mango (Mangefera indica l.) in Sierra leone, causing extensive direct and indirect damage. A survey was conducted among 60 mango farmers in 7 districts in Sierra leone between June and August, 2022, to assess their perceptions regarding fruit fly pest status and the current management options adopted for the control of this pest. Semi-structured questions designed in an open and closed-ended fashion were used for the study. The majority (83%) of the farmers were already aware of the fruit fly problem in the country with 62% perceiving it to be very severe. The majority (60%) of farmers, however, demonstrated poor knowledge of identifying fruit fly species, especially Bactrocera dorsalis, Ceratitis capitata, and Ceratitis cosyra. Farmers were more conversant about the direct damage symptoms to host fruits and the economic impact of fruit flies. A total of 32% of growers took no action to control fruit flies on their farms. Sixty-nine percent (69%) of the farmers adopted cultural control measures, like practicing prompt harvesting, collection and disposal of infested fruits, and weeding to maintain better sanitary conditions on their farms. Recommended fruit fly management strategies such as the use of botanicals and resistant varieties were either unknown or inaccessible to growers. A total of 52% applied chemicals that were not recommended for the control of fruit flies without considering their environmental and health risks. It is important to train fruit growers to improve their capabilities for fruit fly management through extension agents that are appropriate for helping them acquire basic knowledge of fruit fly pests and their management.

Immunotherapy and therapeutic vaccines in prostate cancer： an update on current strategies and clinical implications

In recent years, immunotherapy has emerged as a viable and attractive strategy for the treatment of prostate cancer. While there are multiple ways to target the immune system, therapeutic cancer vaccines and immune checkpoint inhibitors have been most successful in late-stage clinical trials. The landmark Food and Drug Administration approval of sipuleuceI-T for asymptomatic or minimally symptomatic metastatic prostate cancer set the stage for ongoing phase III trials with the cancer vaccine PSA-TRICOM and the immune checkpoint inhibitor ipilimumab. A common feature of these immune-based therapies is the appearance of improved overall survival without short-term changes in disease progression. This class effect appears to be due to modulation of tumor growth rate kinetics, in which the activated immune system exerts constant immunologic pressure that slows net tumor growth. Emerging data suggest that the ideal population for clinical trials of cancer vaccines is patients with lower tumor volume and less aggressive disease. Combination strategies that combine immunotherapy with standard therapies have been shown to augment both immune response and clinical benefit.

Currently,the prognosis of patients with metastatic prostate cancer is most commonly defined by clinical factors including performance status,presence of visceral metastasis,lactate dehydrogenase level,opioid analgesic use,albumin level,presence of anemia,prostate-specific antigen level,and alkaline phosphatase level(i.e.,Halabi model).1 This model is most commonly used in the current prospective clinical trials ***,it is only an inference of the underlying biology and not a direct measurement of that biology.
