Objective Viral encephalitis is an infectious disease severely affecting human *** is caused by a wide variety of viral pathogens,including herpes viruses,flaviviruses,enteroviruses,and other *** laboratory diagnosis of viral encephalitis is a worldwide ***,high-throughput sequencing technology has provided new tools for diagnosing central nervous system ***,In this study,we established a multipathogen detection platform for viral encephalitis based on amplicon *** We designed nine pairs of specific polymerase chain reaction(PCR)primers for the 12 viruses by reviewing the relevant *** detection ability of the primers was verified by software simulation and the detection of known positive *** sequencing was used to validate the samples,and consistency was compared with Sanger *** The results showed that the target sequences of various pathogens were obtained at a coverage depth level greater than 20×,and the sequence lengths were consistent with the sizes of the predicted *** sequences were verified using the National Center for Biotechnology Information BLAST,and all results were consistent with the results of Sanger *** Amplicon-based high-throughput sequencing technology is feasible as a supplementary method for the pathogenic detection of viral *** is also a useful tool for the high-volume screening of clinical samples.

脑梗死继发癫痫患者血清NRG1、VILIP-1水平及临床意义

目的检测脑梗死继发癫痫患者的血清神经调节蛋白1(NRG1)、视锥蛋白样蛋白-1(VILIP-1)水平并探讨其临床意义。方法选取2019年1月至2023年4月期间安阳市第三人民医院收治的100例脑梗死继发癫痫患者作为研究组,并依照发病时间分为早发型癫痫组(n=53)和晚发型癫痫组(n=47)。选取同期脑梗死未继发癫痫患者60例作为对照组。比较早发型癫痫组和晚发型癫痫组患者的血清NRG1、VILIP-1水平以及研究组和对照组患者的临床资料,采用Pearson相关分析血清NRG1与VILIP-1的关系,采用多因素Logistic回归分析脑梗死继发癫痫的影响因素,绘制受试者工作特征曲线(ROC)分析NRG1、VILIP-1对脑梗死继发癫痫的预测价值。结果早发型癫痫患者的血清NRG1、VILIP-1水平分别为(23.72±3.47)pg/mL、(627.49±72.29)pg/mL,明显高于晚发型癫痫的(20.38±3.60)pg/mL、(572.52±61.49)pg/mL,差异均有统计学意义(P<0.05);研究组患者入院时的美国国立卫生研究院卒中量表(NIHSS)评分、血清神经元特异性烯醇化酶(NSE)、NRG1、VILIP-1、同型半胱氨酸(Hcy)水平分别为(12.58±2.86)分、(8.87±1.54)k U/L、(22.15±3.53)pg/mL、(601.65±67.21)pg/mL、(31.53±4.08)μmol/L,明显高于对照组的(11.66±2.12)分、(7.76±1.67)k U/L、(18.72±2.83)pg/mL、(542.74±61.71)pg/mL、(31.53±4.08)μmol/L,而脑源性神经生长因子(BDNF)为(4.18±0.89)μg/mL,明显低于对照组的(5.02±1.10)μg/mL,差异均有统计学意义(P<0.05);Pearson相关分析结果显示,血清NRG1与VILIP-1表达水平呈正相关(r=0.553,P<0.05);多因素Logistic回归分析结果显示,NSE、NRG1、VILIP-1、Hcy均是脑梗死继发癫痫的危险因素(P<0.05),BDNF是脑梗死继发癫痫的保护因素(P<0.05);ROC分析结果显示,血清NRG1、VILIP-1预测脑梗死继发癫痫的AUC分别为0.844、0.768,NRG1联合VILIP-1预测的AUC为0.883,两者联合预测脑梗死继发癫痫的AUC优于NRG1(Z_(两者联合-NRG1)=2.058)、VILIP-1(Z_(两者联合-VILIP-1)=3.820)单独指标(P<0.05)。结论脑梗死继发癫痫患者血清NRG1、VILIP-1水平异常高表达,是患者脑梗死后继发癫痫的危险因素,两者联合检测对预测脑梗死后继发癫痫具有较高的临床价值。

目的构建耐核酸酶的含乙型脑炎病毒(Japanese encephalitis virus,JEV)部分非结构蛋白1(Non-structural protein 1,NS1)基因的假病毒,为JEV的实验室核酸检测提供质控品,提高检测质量。方法将JEV部分NS1基因、蚊18S RNA基因和人GAPDH基因片段次序连接克隆到该质粒上,构建重组载体pET-MS2-JEV;转化大肠杆菌后表达纯化假病毒,并对其进行耐受性和稳定性评估。结果构建了重组载体pET-MS2-JEV,表达获得的假病毒具有耐核酸酶、耐物理化学因子的特性,可在4℃和-20℃条件下稳定保存。结论本研究获得了稳定性好的含JEV部分NS1基因的假病毒,可作为JEV实验室核酸检测的质控品。

Significantly Enhanced Melt Memory Effect of Metallocene-made Isotactic Polypropylene Containing Talc

The melt memory effect is a widely observed phenomenon in semi-crystalline polymers. In practical applications, various additives are usually introduced into polymers, which may affect their melt memory behavior. In this work, the effect of talc on the melt memory effect of metallocene-made isotactic polypropylene(M-PP) was investigated in detail by using the differential scanning calorimetry. The results indicated that the introduction of talc significantly strengthened the melt memory effect of M-PP. Specifically, the upper limit temperature of Domain II increased from 161 ℃ to 174 ℃, resulting in a substantial widening of the temperature range of Domain IIa from 1 ℃ to 14 ℃. Analysis of the crystal orientation of the M-PP containing talc cooled from various Ts suggested that the remarkably enhanced melt memory effect could be ascribed to the stabilization of oriented nuclei facilitated by talc. This stabilizing effect was likely attributable to the prefreezing effect or the sorption interaction between talc and the M-PP chains.

Photothermal material applied in environmental governance has attracted growing *** combining the Stober method and dopamine-triggered coating strategy,Co-Mn precursor was in situ incorporated into the poly dopamine(PDA)layer over the surface of silica ***,a unique photothermal nanosphere with SiO_(2)core and thin carbon layer and dual Co-Mn oxides shell was allowed to form by sequential heat treatment in the inert atmosphere(SiO_(2)@CoMn/C).The bimetallic fraction of Co/Mn in the carbon layer and post-treatment calcination temperature was comprehensively tuned to optimize the peroxymonosulfate(PMS)activation performance of the *** state of bimetallic species was studied including their physical distribution,chemical valence,and interplay by various ***,Co oxides appear as dominant monodispersed nanoparticles(~10 nm),while Mn with cluster-like morphology is observed to uniformly distribute over thin-layer carbon and adhered to the surface of SiO_(2)nanospheres(~250 nm).The calcined temperature could tune the oxidized state of Co species,leading to the optimization of the catalytic performance of introduced dual metal *** a result,this obtained optimal catalyst integrated the advantages of exposed bimetallic CoMn species and N-doped thin carbon to deliver excellent catalytic PMS activation performance and photothermal synergetic catalytic mineralization ability for diversiform *** reactions condition controls and anion interference studies were conducted to identify the adaptability of the optimal ***,the application of solar-driven interfacial water evaporation using optimal SiO_(2)@Co_3Mn_1/C-600 catalyst was explored,showing a high water evaporation rate of 1.48 kg·m^(-2)·h^(-1)and an efficiency of 95.2%,further revealing a comprehensive governance functionality of obtained material in the complex pollution condition.
