The Strawberry fruit fra a Allergen functions in flavonoid Biosynthesis

The strawberry fra a I allergen is a homolog of the major birch pollen allergen Bet v 1. It is synthesized by red ripe fruits of fragaria x ananassa while white fruits of a mutant genotype, which is known to be tolerated by individuals affected by allergy, are devoid of it. Proteomic analyses have shown that fra a 1 is down-regulated in the tolerated white- fruited genotype along with enzymes of the anthocyanin pigment pathway. In this study, we report the spatial and tem- poral expression of three fra a genes that encode different isoforms, and the transient RNAi-mediated silencing of the fra a genes in strawberry fruits of the red-fruited cultivar Elsanta with an ihpRNA construct. As a consequence of reduced levels of fra a mRNAs, fruits were obtained that produced significantly decreased levels of anthocyanins and upstream metabolites. This effect is consistent with the parallel down-regulation of the phenylalanine ammonia lyase （faPAL） and to a lesser extent of the chalcone synthase （faCHS） transcript levels also found in these fruits. In naturally occurring white- fruited genotypes of f. chiloensis and f. vesca, fra a transcript levels are higher than those of the red-fruited varieties, likely to compensate for the low expression levels of faPAL and faCHS in these mutant genotypes. The results demonstrate that fra a expression is directly linked to flavonoid biosynthesis and show that the fra a allergen has an essential biological function in pigment formation in strawberry fruit.

Differential selection in HIV-1 gp120 between subtype B and East Asian variant B'

Aerial spraying plays an important role in promoting agricultural production and protecting the biological environment due to its flexibility,high effectiveness,and large operational area per unit of *** order to evaluate the performance parameters of the spraying systems on two fixed wing airplanes M-18B and Thrush 510G,the effective swath width and uniformity of droplet deposition under headwind flight were tested while the planes operated at the altitudes of 5 m and 4 *** results showed that although wind velocities varied from 0.9 m/s to 4.6 m/s,and the directions of the atomizer switched upward and downward in eight flights,the effective swath widths were kept approximately at 27 m and 15 m for the M-18B and Thrush 510G,respectively,and the latter was more *** addition,through analyzing the coefficients of variation(CVs)of droplet distribution,it was found that the CVs of the M-18B were 39.57%,33.54%,47.95%,and 59.04% at wind velocities of 0.9,1.1,1.4 and 4.6 m/s,respectively,gradually enhancing with the increasing of wind speed;the CVs of Thrush 510G were 79.12%,46.19%,14.90%,and 48.69% at wind velocities of 1.3,2.3,3.0 and 3.4 m/s,respectively,which displayed the irregularity maybe due to change of instantaneous wind ***,in terms of the CVs and features of droplet distribution uniformity for both airplanes in the spray swath,choosing smaller CV(20%-45%)as the standard of estimation,it was found that the Thrush 510G had a better uniform droplet distribution than the *** results provide a research foundation for promoting the development of aerial spraying in China.

Ustekinumab: “Real-world” outcomes and potential predictors of nonresponse in treatment-refractory Crohn’s disease

BACKGROUND Ustekinumab was approved in Europe for the treatment of adults with moderate to severe Crohn's disease(CD)in 2016,and there is an urgent need for data on its everyday *** To obtain data on the daily use of *** This is a retrospective monocentric *** with moderate to severe CD who began ustekinumab therapy at the inflammatory bowel diseases outpatient clinic of the Heidelberg University Hospital between December 2016 and March 2018 were selected based on electronic patient *** primary study endpoint was combined steroid-free clinical remission or steroid-free clinical response at 24±6 wk of ustekinumab *** study endpoints were:achievement of mucosal healing,sonographic and magnetic resonance imaging response,biochemical response,the need for intestinal surgery within 24±6 wk after treatment initiation,the occurrence of adverse events,treatment discontinuation due to nonresponse or adverse events,improvement of extraintestinal manifestations,clinical response at 48±6 wk of therapy,and association of response with nucleotid oligodimerisation domain 2 *** fifty-seven patients with CD(5.3%anti-tumour necrosis factorαnaive,63.2%having undergone at least one intestinal surgery)were included in the *** patients(35.1%)achieved steroid-free clinical remission,6(10.5%)steroid-free clinical response and 31(54.4%)were *** discontinuation due to adverse events occurred in two patients(3.5%).Male sex,the presence of extraintestinal manifestations and the use of steroids at baseline were predictors of nonresponse to ustekinumab *** In a“real-world”treatment-refractory cohort of patients with CD,ustekinumab appeared efficacious and safe.

A novel detection method of spray droplet distribution based on LIDARs

During the process of plant protection in agriculture,the distribution and deposition of droplets or fog fields could directly influence the effectiveness and efficiency of *** traditional method of measurement of the distribution of droplets mainly used water sensitive papers,glass containers or flour to collect data and inverse results,while a new method of measurement based on the principle of reflection of LIDAR was *** were the major targets of the study,and four important algorithms were primarily developed,including the recognition and extraction of targets,the superposition in time-domain,the calculation of effective ranges of distribution,and the development of 3D distribution *** with these algorithms,in order to eliminate the environmental noise,the methods of fuzzy Environment Matching and Secondary filter were created and ***,the statistics was used for analysis of the duration of scanning as well as computation of the distribution,with enough datasets but the minimum length of *** results of the experiments showed that the relative error of measurement was less than 7%and Relative Standard Deviation was less than 16%,compared with the values of manual ***,the 3D models were accurate and clarified in the wind-tunnel *** completed system based on this method could adapt to the requirements of both indoor and outdoor ***,it is capable of the quantized detection of droplet distribution,providing an effective way of tests for spray technique,especially for the research of the application of plant protection by UAVs.
