This paper proposes an investigation of the effects of laser engineering surface with dimple operating under lubrication. Dimples with semi-spherical geometry, with 100 μm in diameter and 25 μm in depth, were Laser-machined on the surfaces of high speed steel AISI H 13 discs, with three different distribution densities applied over the surface. Pin-on-disc experiments were performed by using counter bodies made with AISI 440c and Tungsten carbide ISO K20 pins. The experiments provided data information on the influence of dimple distribution on the coefficient of friction. Numerical simulations were performed to understand the influence of the pin and disc surface roughness on the contact problem. The results suggest that the design of dimple-structured surfaces rely on nine main parameters： material contact pair and its respective hardness, Young Modulus, the roughness of the surfaces, the dimple design, the dimple distribution, the load applied, the velocity and the type of lubricant. A better performance can be obtained from a compromise solution over these parameters, and the dimple concentration has a dominance over the other parameters. The best results were obtained with a lower concentration of dimples, under higher speed and load. Numerical simulations can also be used as a design tool, supporting decisions regarding shape and distribution of the dimples and material selections for the contact pair.

The mixture of Erigeron floribundus and Tragia benthamii (AEMEFTB) is traditionally used against pelvic pain, dysmenorrhea and female sexual dysfunctions. In a recent study, we showed that the aqueous extract of the mixture of AEMEFTB suppresses the endometrium growth in rats with experimental endometriosis. The present study was aimed at investigating the effects of AEMEFTB on estrogen’s sensitive sexual organs growth and architecture. Immature gonado-intact female rats were randomly distributed into 7 groups of 5 animals each and daily treated during one week with either distilled water (10 ml/kg), refined palm oil (1 ml/kg) or 17β-estradiol (1 μg/day). Objective:To evaluate the influence of dietary inclusions of Lactobacillus plantarum(***)on growth response,gut morphometry and intestinal microbial profile of clarias gariepinus(***)fingerlings was carried out using a total of 150 *** fingerlings(2.35±0.48 g/fish)by selecting at random into five treatments groups of 10 fish in 3 three replicates ***:*** isolated from corn slurry was cultured using standard *** isonitrogenous diets were prepared at 35%crude protein(T_(0),T_(1),T2,T_(3)and T_(4))with *** at inclusion rate of 0.0%,0.5%,1.0%,1.5%and 2.0%*** fish were fed at 5%body weight per day for 12 weeks twice ***:T_(4)recorded the highest mean weight gain and specific growth rate while the lowest was obtained in T_(1).T_(4)(1.97)when compared with other treatments had marginally lower feed conversion *** area was most significantly higher in T_(3)and T_(4)group when compared to the control(T_(0))and other lower dietary probiotic inclusion *** depth was highest in T_(4)with significant difference which also gave the maximum enterobacteriaceae count while T_(0)recorded the least ***:From these indications,*** fortified diets may enhance the growth of cultured *** fingerlings at 2.0%inclusion rate as it was observed to improve body weight gain,feed conversion ratio with increment in the absorptive area and the microbial count in the gut.

Genetic diversity evaluation of mutant lines is essential to facilitate their conservation and utility in breeding programs. characterization of plant genotypes using morphological markers has limitations which make the procedure inefficient. Application of molecular tools for characterization and diversity assessment has been found useful to complement phenotypic evaluation of plant population. Therefore genetic diversity of some cowpea mutant lines was studied using simple sequence repeats (SSr) markers. DNA barcoding marker, ribulose-1,5-bisphosphate carboxylase(rbcL) of the chloroplast DNA (cpDNA) was also used for characterization and identification of the mutants to species level. The mean polymorphic information content (0.51) obtained from the microsatellites showed high polymorphism in accessing wide genetic diversity among the mutants and their parents. Dendrogram generated revealed 8 groups with most mutants clustered separately from their parents. Sequence analysis revealed insertions/deletions (InDels) and base substitutions as the two main classes of mutations induced in the plastid DNA of the mutants studied. The nucleotide frequencies were 26.95% (A), 34.43% (T), 24.09% (c) and 14.53% (G). A total of 61.38% AT rich region was identified, while Gc rich region was found to be 38.62%. Highest rate of mutations were observed in region 3 - 4 indicating that the region is less conserved in cowpea rbcL gene. The present study proved that SSr markers are useful for the genetic diversity assessment of cowpea mutants. It also proved the efficiency of rbcL markers in mutants’ identification. The results indicate that the mutants are valuable genetic resources that have been developed to widen cowpea genetic base.

为了提高肿瘤治疗效果并最大程度降低其毒性,研究人员针对多种类型肿瘤正在研究如何根据患者个体情况化进行适应性治疗。随着现代辐照技术(如调强放疗)的引入,结合功能显像技术能够更精准地描绘治疗靶区的性能,为治疗适应创造了多种机会。根据功能性cT、MrI和PET显像结果,在治疗过程中随时调整放疗靶区,或明确原发肿瘤内部的亚区以优化放疗剂量策略。此外,借助解剖或功能显像还能够预测正常组织发生并发症的概率,如使用cT或PET显像来预测放射性肺炎。PET显像除了聚焦、监测实体肿瘤以辅助放疗,并调整治疗策略外,还能够验证质子束疗法的疗效。本文主要讨论放射肿瘤学中基于影像学的治疗适应的现状及其所面临的挑战。

Modular protein engineering is suited to recruit complex and multiple functionalities in single-chain polypeptides. Although still unexplored in a systematic way, it is anticipated that the positioning of functional domains would impact and refine these activities, including the ability to organize as supramolecular entities and to generate multifunctional protein materials. To explore this concept, we have repositioned functional segments in the modular protein T22-GFP-H6 and characterized the resulting alternative fusions. In T22-GFP-H6, the combination of T22 and H6 promotes selfassembling as regular nanoparticles and selective binding and internalization of this material in cXcr4-overexpressing tumor cells, making them appealing as vehicles for selective drug delivery. The results show that the pleiotropic activities are dramatically affected in module-swapped constructs, proving the need of a carboxy terminal positioning of H6 for protein self-assembling, and the accommodation of T22 at the amino terminus as a requisite for cXcr4^+ cell binding and internalization. Furthermore, the failure of self-assembling as regular oligomers reduces cellular penetrability of the fusions while keeping the specificity of the T22-cXcr4 *** these data instruct how multifunctional nanoscale protein carriers can be designed for smart, protein-driven drug delivery, not only for the treatment of cXcr4^+ human neoplasias, but also for the development of anti-HIV drugs and other pathologies in which cXcr4 is a relevant homing marker.

海参加工废弃物中多糖的理化性质及减肥活性研究

海参含有丰富的生物活性物质,具有低糖、低脂、高蛋白质、高膳食纤维的营养特性,并具有多种生物活性,近年来备受研究者关注。海参加工会产生大量废弃物,其中含有大量活性成分,但由于缺乏研究,没有得到充分利用,造成了宝贵资源的浪费和环境污染。基于此,本论文以海参加工废弃物为原料,提取分离海参多糖,并对其结构、理化性质和减肥活性进行了研究。采用单因素实验和响应面实验研究了海参多糖的最优提取条件,之后利用纤维素DEAE-52色谱柱和Sepharose 4B色谱柱对海参多糖进行了分离纯化。结果表明,最优提取条件为:81.7℃提取2次,每次提取1.16 h,提取得率为4.26%±0.11%;色谱柱分离纯化后得到三种多糖组分,分别为海参多糖I(ScIP-I)、海参多糖(ScIP-II)和海参多糖III(ScIP-III),其中ScIP-III占比最高。采用化学分析方法,结合HPLc、Gc、FT-Ir、NMr、SEM、AFM、TGA、DSc和微流变仪等仪器分析研究了ScIP-III的组成、结构和理化性质。结果显示,ScIP-III的总糖和蛋白质含量分别为82.20%和6.34%,由甘露糖,阿拉伯糖,半乳糖,葡萄糖和岩藻糖组成(摩尔百分比为22.03:19.31:11.78:3.22:42.57),Mw为4.91×10Da,多分散指数(Mw/Mn)为1.02;单糖残基之间以β型糖苷键连接;ScIP-III在SEM图像中为椭圆形颗粒,并与它由不同大小、球形结构和聚集的分子组成,AFM检测结果一致;DSc检测结果显示ScIP-III有两个峰,第一个峰是失水峰,Tp为89.12℃,总焓(ΔH)为-68.18,第二个峰是由于多糖的分子结构破坏导致,Tp为193.87°c,总焓(ΔH)为-70.84 J/g;热重分析结果中,30-130℃存在结合水和自由水损失的失重峰,200-400℃存在多糖糖苷键、糖环结构破坏的失重峰;微流变结果表明ScIP-III溶液的流动性、弹性和黏度随多糖浓度的增加而减小。采用高脂饲料建立肥胖小鼠模型,研究了海参多糖(ScIPs)的减肥作用。结果显示,ScIPs组能明显抑制肥胖小鼠体重增加,显著降低肥胖小鼠的脂肪指数、脂肪细胞大小和血脂水平(pcIPs可显著提高小鼠肠道短链脂肪酸含量(pcc1)、白细胞介素-6(Il-6)的m rNA含量(pcIPs可以改变厚壁菌门、变形菌门和拟杆菌门的相对丰度。厚壁菌门和拟杆菌门的比例从Nc组的1.2增加到HFD组(肥胖小鼠)的1.45。海参多糖干预后,LScIP、MScIP和HScIP组的厚壁菌门/拟杆菌门比例分别为0.87、1.3和0.95。ScIPs干预可以降低脱硫弧菌、双歧杆菌、细小杆菌和肠杆菌等有害厚壁细菌的比例,增加拟杆菌属、乳杆菌属、粪杆菌属和毛螺菌科等有益肠道菌群的多样性,有助于膳食未消化的生物活性脂质的发酵。上述结果证明ScIPs可通过调节与脂质代谢相关的酶、细胞因子的表达和肠道菌群组成实现降脂减肥的作用。
