Motor neurons are highly polarized cells,with long axons that extend to more than 1 m in the adult *** axons further arborize into a specialized synaptic compartment,the motor unit,containing up to 2000 neuromuscular junctions(NMJs).While the size of other neuronal synapses can be up to 1μm,the NMJ is much larger and can reach 10–30μm(Jones et al.,2017).The vast size of the motor unit requires motor neurons to evolutionally adapt and supply this distal portion with a sufficient amount of ATP,as well as to replenish the axonal protein pool in order to maintain their synapses.

Embryonic stem cells(ESCs)exhibit unique chromatin features,including a permissive transcriptional program and an open,decondensed chromatin *** pluripotent stem cells(iPSCs),which are very similar to ESCs,hold great promise for therapy and basic ***,the mechanisms by which reprogramming occurs and the chromatin organization that underlies the reprogramming process are largely *** we characterize and compare the epigenetic landscapes of partially and fully reprogrammed iPSCs to mouse embryonic fibroblasts(MEFs)and ESCs,which serves as a standard for *** immuno-fluorescence and biochemical fractionations,we analyzed the levels and distribution of a battery of histone modifications(H3ac,H4ac,H4K5ac,H3K9ac,H3K27ac,H3K4me3,H3K36me2,H3K9me3,H3K27me3,and gH2AX),as well as HP1a and lamin *** find that fully reprogrammed iPSCs are epigenetically identical to ESCs,and that partially reprogrammed iPSCs are closer to ***,combining both time-course reprogramming experiments and data from the partially reprogrammed iPSCs,we find that heterochromatin reorganization precedes Nanog expression and active histone ***,these data delineate the global epigenetic state of iPSCs in conjunction with their pluripotent state,and demonstrate that heterochromatin precedes euchromatin in reorganization during reprogramming.

Rabies virus （RABV） of the rhabdoviridae family is a prototype neurotropic virus that causes a fatal disease, and is still a major risk mostly in developing countries. A key step in the RABV infection process is its arrival into the central nervous system （CNS）, for which it uses the cellular transport machinery. Familial dysautonomia （FD） is a rare children neurodegenerative disease caused due to a point mutation in the IKBKAP gene that results in decreased IKK complex-associated protein （IKAP） protein production. The disease affects mostly the dorsal root ganglion （DRG） and the sympathetic ganglion. Recently, we found that the molecular mechanisms underlying neurodegeneration in FD patients are defects in axonal transport of nerve growth factors and microtubule stability in the DRG. Neurons are highly polarized cells with very long axons. In order to survive and maintain proper function, neurons depend on transport of proteins and other cellular components from the neuronal body along the axons. We further demonstrated that IKAP is necessary for axon maintenance and showed that phosphatidylserine acts as an HDAC6 inhib- itor to rescue neuronal function in FD cells. In this review, we will highlight our latest research findings.

Oral mixture of autologous colon-extracted proteins for the Crohn's disease: A double-blind trial

AIM: To evaluate the safety and efficacy of oral administration of AlequelTM, an autologous proteincontaining colon extract.a randomized,placebo-controlled,double-blind *** were orally administered with autologous protein-containing colon extract three doses of autologous study drug per week for 15 wk,for a total of45 *** were followed for safety *** was defined as a Crohn’s disease activity index(CDAI)score of less than or equal to *** patients were followed for changes in subsets of T cells by fluorescence-activated cell sorting ***:Analysis was performed on a total number of evaluable patients of 14 in the study drug group and15 in the placebo *** was well tolerated by all *** major treatment-related adverse events were reported or observed in any of the treated patients during the feeding or follow-up *** weeks 6 and 9 of the study,six of the 14(43%)evaluable subjects who received the study drug achieved a CDAI of 150 or *** contrast,five of the 15(33%)evaluable subjects in the placebo group achieved *** weeks 9 and 12,the remission rates were 50%and 33%for the drug group and placebo group,*** the drug-treated subjects who achieved remission,the effect of the drug was judged as stable in eight of the 14 subjects as measured by at least two CDAI scores indicating remission in the 15-wk treatment period.A decreased percentage of peripheral natural killer T regulatory cells(a decrease of 28%vs an increase of 16%)and an increased ratio of CD4+/CD8+T lymphocytes(an increase of 11%vs a decrease of9%)were noted in subjects with a significant clinical ***:Oral administration of the autologous colonic extract could be a safe and effective for the treatment of patients with moderate to severe Crohn’s disease.

A diagnosis of primary aortoenteric fistula is difficult to make despite a high level of clinical suspicion. It should be considered in any elderly patient who presents with upper gastrointestinal bleeding in the context of a known abdominal aortic aneurysm. We present the case of young man with no history of abdominal aortic aneurysm who presented with massive upper gastrointestinal bleeding. Initial misdiagnosis led to a delay in treatment and the patient succumbing to the illness. This case is unique in that the fistula formed as a result of complex atherosclerotic disease of the abdominal aorta, and not from an aneurysm.
