Mercury bioaccumulation in aquatic biota along a salinity gradient in the Saint John River estuary

Although estuaries are critical habitats for many aquatic species, the spatial trends of toxic methylmercury（MeHg） in biota from fresh to marine waters are poorly understood. Our objective was to determine if MeHg concentrations in biota changed along a salinity gradient in an estuary. Fourspine Stickleback（Apeltes quadracus）, invertebrates（snails,amphipods, and chironomids）, sediments, and water were collected from ten sites along the Saint John River estuary, New Brunswick, Canada in 2015 and 2016, with salinities ranging from 0.06 to 6.96. Total mercury（proxy for MeHg） was measured in whole fish and MeHg was measured in a subset of fish, pooled invertebrates, sediments, and water. Stable sulfur（δ34S）, carbon（δ13 C）, and nitrogen（δ15N） isotope values were measured to assess energy sources（S, C） and relative trophic level（N）. There were increases in biotic δ13C and δ34S from fresh to more saline sites and these measures were correlated with *** aqueous MeHg was higher at the freshwater than more saline sites, only chironomid MeHg increased significantly with salinity. In the Saint John River estuary, there was little evidence that MeHg and its associated risks increased along a salinity gradient.

Background: The benefits of using the oxidized oils from rendering and recycling as an economic source of lipids and energy in animal feed always coexist with the concerns that diverse degradation products in these oxidized oils can negatively affect animal health and performance. Therefore, the quality markers that predict growth performance could be useful when feeding oxidized oils to non-ruminants. However, the correlations between growth performance and chemical profiles of oxidized oils have not been well examined. In this study, six thermally oxidized soybean oils(OSOs) with a wide range of quality measures were prepared under different processing temperatures and processing durations, including 45 °C-336 h;67.5 °C-168 h;90 °C-84 h;135 °C-42 h;180 °C-21 h;and225 °C-10.5 h. Broilers and nursery pigs were randomly assigned to diets containing either unheated control soybean oil or one of six OSOs. Animal performance was determined by measuring body weight gain, feed intake,and gain to feed ratio. The chemical profiles of OSOs were first evaluated by common indicative tests, including peroxide value, thiobarbituric acid reactive substances, p-anisidine value, free fatty acids, oxidized fatty acids,unsaponifiable matter, insoluble impurities, and moisture, and then analyzed by the liquid chromatography-mass spectrometry-based chemometric ***: Among common quality indicators, p-anisidine value(An V), which reflects the level of carbonyl compounds,had the greatest inverse correlation with the growth performance of both broilers and pigs, followed by free fatty acids and oxidized fatty acids. Fast and sensitive antigen detection is important in biomedical research and development. Despite, being invented 48 years ago, the enzyme-linked immunosorbent assay(ElISA) remains one of the most successful and widely employed bioanalytical techniques in research and clinical diagnostics due to its reliability and simplistic design. Recently, nanotechnology has offered efficient signal reporting. In spite of some improvements in these systems, there are typically increased material costs involved and the need for expensive equipment or complicated chemical processes, thus negating any possible benefits over ElISA. Herein, we communicate a simple Cu-DNAzyme system for signal transduction of a CuO nanoparticle-labeled immunoassay. The reported immunoassay amplifies signal generation similar to traditional ElISA and is fast, simple, cost-effective, and sensitive, holding promise for biomedical applications and point-of-care testing.

We investigated the effects of land-use changes on soil carbon storage and soil CO2 flux by comparing soils from mature cloud forest and 31-year-old secondary forest, both in the Santa Elena Forest Reserve, a municipallyowned reserve at an elevation of 1600 to 1700 m near the town of Monteverde, and a clear-cut pasture near the reserve. Soils in the mature forest exhibit only weak horizonation but typically thick A horizons;they also consistently yield the highest carbon contents in the upper 30 cm. Soil CO2 flux was the highest in these soils, but also displayed the highest spatial variability. Secondary forest soils contain substantially less soil carbon than mature forest soils, but more than pasture soils. CO2 flux in the secondary forest soils was more similar to that of the mature forest, but displayed lower spatial variability. The pasture soils contain less soil carbon and produced lower CO2 flux levels than either of the forest soils. The pasture soils typically contain a well-defined coarse sandy layer 10 to 20 cm below the surface that we interpret as a sediment layer deposited across much of the landscape following a widespread erosion event, likely a consequence of the clear-cutting. Soil nitrogen concentrations are more than an order of magnitude lower than soil carbon concentrations, and display no trends between the different landscapes examined. Our preliminary results suggest that reforestation does restore soil carbon to clear-cut landscapes, but returning soil carbon levels to pre-land use levels occurs at a time scale of centuries, rather than decades.

The objective of this experiment was to determine the standardized ileal digestibility（SID） of amino acids（AA） in 3sources of distillers dried grains with solubles（DDGS） with different concentrations of *** growing barrows（initial body weight：76.1 ± 6.2 kg） were randomly allotted to a replicated 6×4 Youden square design with 6 diets and 4 *** fat content of the 3 sources of DDGS were 11.5,7.5,and 6.9%*** contained 60%DDGS and fat concentration of the diets were 7.5,52,and 5.2%,*** additional diets containing the 2sources of DDGS with 75 and 6.9%fat were also formulated,and corn oil was added to these diets to increase the concentration of fat in the diets to levels that were calculated to be similar to the diet containing conventional DDGS with 11.5%fat.A N-free diet was also formulated to calculate endogenous losses of crude protein（CP） and AA from the *** were fed experimental diets during four 7-d *** first 5 d of each period were an adaptation period and ileal digesta were collected on d 6 and 7 of each *** apparent ileal digestibililty（AID） and SID of CP and all indispensable AA,except AID Pro and SID of Trp,were greater（P 〈 0.01） in conventional DDGS than in the 2 sources of DDGS with reduced *** oil to the diets containing the 2 sources of DDGS with reduced fat did not consistently increase SID of *** conclusion,conventional DDGS has greater SID values for most AA compared with DDGS that contains less fat and inclusion of additional oil to diets containing low-fat DDGS does not increase AID or SID of *** lower AA digestibility in low-fat DDGS could not be overcome by the inclusion of additional fat to the diets.

Melanocytic nevi are a transient in vivo proliferation of melanocytes that after time undergo cellular senescence. Most nevi harbor B-Raf mutations, which appear to activate cellular mechanisms of senescence in melanocytes. Glycogen synthase kinase 3β (GSK-3β), a critical downstream effector of the AKT signaling pathway, is involved in the development of melanoma and has been associated with senescence in melanocytes. Our immunohistochemical and immunofluorescence studies revealed distinct, perinuclear, dot-like reactivity of GSK-3β in melanocytic nevi. Furthermore, our tissue microarray analysis demonstrated significant perinuclear dot-like sublocalization of GSK-3β in melanocytic nevi compared with the amount of GSK-3β observed in melanoma (P β in human nevi may contribute to senescence in melanocytes.
