Impact of cold on the immune system of burying beetle, Nicrophorus vespilloides （Coleoptera： Silphidae）

Insect overwintering is one of the most astonishing phases of the insect life cycle. Despite vast amounts of knowledge available about the physiological mechanisms of this phenomenon, the impact of stress factors on insect immune system functioning during the winter is still unknown. The aim of this study is to analyze how low temperatures influence the immune system of the beetle Nic＇rophorus vespilloides. The results show that the beetle＇s immune system is differently modulated by cold induced in laboratory settings than that which occurs in natural conditions, Among beetles cultured in conditions similar to summer, low temperatures, did not influence the number of circulating haemocytes, phenoloxidase activity, haemocytes morphology, and percentage ratio of haemocyte types. In these beetles, differences were noted only in the ability of haemocytes to perform phagocytosis. Individuals acclimated in natural conditions in autumn had a higher level of humoral response and a different percentage ratio of haemocyte types. During the winter period, the number of haemocytes in the beetles decreased, but the percentage ratio of phagocytic haemocytes increased. Furthermore, we noted an increase of phenoloxidase activity. Our study also showed mitotic divisions ofhaemocytes in haemolymph collected from burying beetles after cold exposure and from burying beetles collected from natural conditions during autumn and winter. Differences in response to low temperatures in laboratory conditions and the natural environment suggest that the simultaneous presence of other stress factors during winter such as desiccation and starvation have a significant influence on the activity of burying beetle＇s immune system.

The long-term immunological effects of alloferon and its analogues in the mealworm Tenebrio mofitor

The subject of this article is a search for the long-term immunological effects of alloferon and 3 structural analogues of alloferon, which were earlier characterized by the highest pro-apoptotic activity in Tenebrio molitor. The differences in the actions of these peptides on immune response were observed. Alloferon increased nodulation and significantly phenoloxidase activity in the hemolymph of experimentally infected T. molitor. However, [Phe（p-NH2）^1 ]- and [Phe（p-OMe） ^1 ]-alloferon strongly inhibited cellular and humoral defense of the mealworm against Staphylococcus aureus infection. One day after injection of these peptides, the specific biochemical and morphological hallmarks of apoptosis in bacteria-challenged hemocytes were visible; in contrast, 3 days after peptides injection in all hemocytes, caspase activation was not observed. However, these new, circulating hemocytes differed from the control and the peptide-untreated bacteria-challenged hemocytes. They had an increased adhesion that led to a separation of viable, anucleated fragments of hemocytes that retain the ability to adhere and to form long filopodia. The peptide-induced separation ofhemocyte fragments may resemble the formation ofplatelets in mammals and perhaps play a role in sealing wounds in insects. The results of in vivo studies may suggest a long half-life of studied peptides in the hemolymph of mealworm. Moreover, we showed the importance of the N-terminal histidine residues at position one of the alloferon molecule for its immunological properties in insects. The results obtained here show that alloferon plays pleiotropic functions in insects.

The change in the distribution of organisms in freshwater ecosystems due to natural or manmade processes raises the question of the impact of alien species on local *** most studies indicate a negative effect,the positive one is more difficult to discern,espe-cially in multispecies systems,including hosts and *** purpose of the study was to check whether the presence of an alien host,Potamopyrgus antipodarum,reduces the intensity of Echinoparyphium aconiatum metacercariae in a native host,Radix *** additionally tested the impact of water temperature and the biomass of the alien host on the dilution *** experimentally studied(1)the lifespan of echinostome cercariae in different temperatures,(2)the infectivity of cercariae toward the alien host and native host,and(3)the impact of different biomass of the alien host on the intensity of metacercariae in the native *** found that cercarial survival and infectivity were tem-perature ***,cercarial survival decreased with increasing temperature,contrary to cercarial *** cercariae entered the renal cavity of both the native host and alien host,and successfully transformed into *** number of metacercariae in the native host decreased with the increasing biomass of the alien *** results indicate that lymnaeids may benefit from the co-occur-rence with P antipodarum,as the presence of additional hosts of different origins may reduce the prevalence of parasites in native ***,the scale of the dilution effect depends not only on the increased spectrum of susceptible hosts but also on the other variables of the environment,including water temperature and host density.

In accordance with the World Health Organization data,cancer remains at the forefront of fatal *** upward trend in cancer incidence and mortality has been observed globally,emphasizing that efforts in developing detection and treatment methods should *** diagnostic path typically begins with learning the medical history of a patient;this is followed by basic blood tests and imaging tests to indicate where cancer may be located to schedule a needle *** initiation of diagnosis is crucial since delayed cancer detection entails higher costs of treatment and ***,there is a need for novel cancer detection methods such as liquid biopsy,elastography,synthetic biosensors,fluorescence imaging,and reflectance confocal *** therapeutic methods,although still common in clinical practice,pose many limitations and are ***,there is a dynamic advancement of clinical research and the development of more precise and effective methods such as oncolytic virotherapy,exosome-based therapy,nanotechnology,dendritic cells,chimeric antigen receptors,immune checkpoint inhibitors,natural product-based therapy,tumor-treating fields,and photodynamic *** present paper compares available data on conventional and modern methods of cancer detection and therapy to facilitate an understanding of this rapidly advancing field and its future *** evidenced,modern methods are not without drawbacks;there is still a need to develop new detection strategies and therapeutic approaches to improve sensitivity,specificity,safety,and ***,an appropriate route has been taken,as confirmed by the approval of some modern methods by the Food and Drug Administration.

Acyclic colorings of graphs with bounded degree

A k coloring(not necessarily proper) of vertices of a graph is called acyclic, if for every pair of distinct colors i and j the subgraph induced by the edges whose endpoints have colors i and j is acyclic. We consider some generalized acyclic k colorings, namely, we require that each color class induces an acyclic or bounded degree graph. Mainly we focus on graphs with maximum degree 5. We prove that any such graph has an acyclic 5 coloring such that each color class induces an acyclic graph with maximum degree at most 4. We prove that the problem of deciding whether a graph G has an acyclic 2 coloring in which each color class induces a graph with maximum degree at most 3 is NP complete, even for graphs with maximum degree 5. We also give a linear time algorithm for an acyclic t improper coloring of any graph with maximum degree d assuming that the number of colors is large enough.

Hybrid aqueous capacitors with improved energy/power performance

This work reports on a high-voltage,hybrid capacitor involving two separate redox *** solutions of Mg(NO3)2and KI have been used for negative and positive electrode,*** p H ? 2 for electrode( t) with KI solution and modifying Mg(NO3)2solution to p H ? 9 for negative side play a crucial role for a stable long-term operation of capacitor at enhanced voltage.A benefit from such a construction is a pseudocapacitive contribution from hydrogen sorption reaction on the negative electrode and high iodine/iodide activity on the positive electrode,enhancing the energy with no remarkable impact on the power *** solution allows a high voltage(1.8 V) to be reached and thereby high power and energy performance( 20 W h/kg at 1 k W/kg) to be *** long-term stability has been confirmed by floating and galvanostatic tests.

Acyclic improper colouring of graphs with maximum degree 4

A k-colouring （not necessarily proper） of vertices of a graph is called acyclic, if for every pair of distinct colours i and j the subgraph induced by the edges whose endpoints have colours i and j is acyclic. We consider acyclie k-eolourings such that each colour class induces a graph with a given （hereditary） property. In particular, we consider aeyclic k-eolourings in which each colour class induces a graph with maximum degree at most t, which are referred to as acyclic t-improper k-colourings. The acyelic t-improper chromatic number of a graph G is the smallest k for which there exists an acyclic t-improper k-colouring of G. We focus on acyclic colourings of graphs with maximum degree 4. We prove that 3 is an upper bound for the acyclic 3-improper chromatic number of this class of graphs. We also provide a non-trivial family of graphs with maximum degree 4 whose acyclic 3-improper chromatic number is at most 2, namely, the graphs with maximum average degree at most 3. Finally, we prove that any graph G with A（G） ≤ 4 can be acyelically coloured with 4 colours in such a way that each colour class induces an acyclic graph with maximum degree at most 3.

Phenotypic plasticity can explain evolution of sympatric polymorphism in the hairy snail Trochulus hispidus （Linnaeus, 1758）

Morphological variation of snails from the genus Trochulus is so huge that their taxonomy is unclear. The greatest variability concerns forms hispidus and sericeus/plebeius, which are often considered as separate species. To evidence the species barriers, we carried out crossbreeding ex- periments between these two sympatric morphs. Moreover, we compared the shell morphology of laboratory-bred offspring with their wild parents to test if the variation can be explained by the phenotypic plasticity model. We found that the two Trochulus morphs show no reproductive bar- riers. The fecundity rates, the mean clutch size, and F~ viability observed for all crosses were not significantly different. In hybrid crosses （in F2 generation）, we also recorded reproduction compati- bility, similar fecundity, and hatching success as in their parents. Accordingly, phylogenetic ana- lyses revealed the significant grouping of sequences from these different morphs and supported no constrains in reproduction between them. Comparison of shell morphology between wild and laboratory samples showed that various characters appeared highly plastic. The average shell shape of the hispidus morph changed significantly from flat with wide umbilicus to elevated with narrower umbilicus such as in the sericeus/plebeius morph. All these findings indicate that the examined morphs do not represent separate biological species and the evolutionary process is not advanced enough to separate their genetic pool. Therefore, phenotypic plasticity has played a sig- nificant role in the evolution of Trochulus shell polymorphism. The two morphs can evolve inde- pendently in separate phylogenetic lineages under the influence of local environmental conditions.

Drosophila melanogaster as a model system of aluminum toxicity and aging

The aim of this study was to investigate the toxic effects of aluminum （A1） on the model organism-Drosophila melanogaster. The study is especially concerned with the effects of aluminum on the fruit fly＇s development, life span, and circadian rhythm in rest and activity. Flies were exposed to aluminum in concentrations from 40 to 280 mg/kg in rearing media or the flies were raised on control medium. Moreover, the life span of insects exposed to aluminum containing 40, 120, or 240 mg/kg of A1 in the medium, only during their larval development, during the whole life cycle and only in their adult life was tested. To check if aluminum and aging cause changes in D. melanogaster behavior, the locomotor activity of flies at different ages was recorded. Results showed that aluminum is toxic in concentrations above 160 mg/kg in the rearing medium. Depending on A1 concentration and time of exposure, the life span of the flies was shortened. At intermediate concentrations （120 mg/kg）, however, A1 had a stimulating effect on males increasing their life span and level of locomotor activity. At higher concentration the aluminum exposure increased or decreased the level of locomotor activity ofD. melanogaster depending on age of flies. In addition, in the oldest insects reared on aluminum supplemented media and in mid-aged flies reared on the highest concentration of A1 the daily rhythm of activity was disrupted.

Import Determinants of Organelle-Specific and Dual Targeting Peptides of Mitochondria and Chloroplasts in Arabidopsis thaliana

Most of the mitochondrial and chloroplastic proteins are synthesized in the cytosol as precursor proteins carrying an N-terminal targeting peptide （TP） directing them specifically to a correct organelle. However, there is a group of proteins that are dually targeted to mitochondria and chloroplasts using an ambiguous N-terminal dual targeting peptide （dTP）. Here, we have investigated pattern properties of import determinants of organelle-specific TPs and dTPs combining mathematical multivariate data analysis （MVDA） with in vitro organellar import studies. We have used large datasets of mitochondrial and chloroplastic proteins found in organellar proteomes as well as manually selected data sets of experimentally confirmed organelle-specific TPs and dTPs from Arabidopsis thaliana. Two classes of organelle-specific TPs could be distinguished by MVDA and potential patterns or periodicity in the amino acid sequence contributing to the separation were revealed, dTPs were found to have intermediate sequence features between the organelle-specific TPs. Interestingly, introducing positively charged residues to the dTPs showed clustering towards the mitochondrial TPs in silico and resulted in inhibition of chloroplast, but not mitochondrial import in in vitro organellar import studies. These findings suggest that positive charges in the N-terminal region of TPs may function as an ＇avoidance signal＇ for the chloroplast import.
