Inflammatory bowel disease(IBD),which includes Crohn’s disease and ulcerative colitis,has a not yet completely defined aetiology and is characterized by a progressive chronic inflammation that involves nitroxidative stress and *** manifestations can occur and affect several organs,including the liver and bile ducts,joints,skin,eyes,and less frequently,the heart,brain,and kidneys,increasing the risk of morbidity and *** repercussions may be associated with the activity or severity of *** present review proposes to report and analyse the participation of dysbiosis and nitroxidative stress in the genesis of extraintestinal manifestations,aiming to contribute to a better understanding of the disease and to focus on the development of individualized preventive and therapeutic strategies.

Objective: Validate Digital Cervicography as adjunctive cervical cancer screening test to VIA and Cytology. Method: Women (1292) were submitted to both cytological and VIA tests to obtain a sample of 301 positive VIA cases (23.3% ) which have had the Digital Cervicography (DC) taken and evaluated. Just cases considered positive by DC (149/301 cases- 49.5% )- and/or positive Pap smear tests were referred to colposcopy and biopsy whenever indicated. Results: Cervical smear was positive in 5.4% , including LGSIL (4% ), HGSIL (1% ) and one case of invasive cancer (0.4% ), and 1.6% of ASCUS or *** identified 81 positive cases (74 LGSIL; 5 HGSIL; 2 cancers). The sensitivity of cytology was 14.8% , and the DC was 100% . Corresponding specificities were 95.4% and 69.1% , respectively. The NPV and PPV for DC were 100% and 54.4% . Conclusion: DC increases sensitivity, specificity and positive predictive value of VIA, becoming a valid adjunctive screening test for cervical cancer in low resource settings.

Aside from Polo and Aurora, a third but less studied kinase family involved in mitosis regulation is the never in mitosis-gene A(NIMA)-related kinases(Neks). The founding member of this family is the sole member NIMA of Aspergillus nidulans, which is crucial for the initiation of mitosis in that organism. All 11 human Neks have been functionally assigned to one of the three core functions established for this family in mammals:(1) centrioles/mitosis;(2) primary ciliary function/ciliopathies; and(3) DNA damage response(DDR). Recent findings, especially on Nek 1 and 8, showed however, that several Neks participate in parallel in at least two of these contexts: primary ciliary function and DDR. In the core section of this in-depth review, we report the current detailed functional knowledge on each of the 11 Neks. In the discussion, we return to the cross-connections among Neks and point out how our and other groups' functional and interactomics studies revealed that most Neks interact with protein partners associated with two if not all three of the functional contexts. We then raise the hypothesis that Neks may be the connecting regulatory elements that allow the cell to fine tune and synchronize the cellular events associated with these three core functions. The new and exciting findings on the Nek family open new perspectives and should allow the Neks to finally claim the attention they deserve in the field of kinases and cell cycle biology.

Virulence properties of a peptide hemolysin produced by enterococcus faecalis

The characterization and prevalence of virulence factors associated with enterococcal invasiveness and severity of disease are important areas to be investigated. Recently, we described the production of a heat-stable hemolysin by clinical isolates of enterococcus faecalis cultived in BHI-GA (BHI with glucose and L-arginine). Now, we purified the hemolysin from the culture supernatant by ultra-filtration (PM-10 membrane) and ethanol extraction followed by chromatography in a mBondapak C18 and Superdex Peptide columns. The hemolytic activity was not affected by the proteolytic enzymes. Cholesterol, phospholipids, eDTA and also bivalent ions did not inhibit the hemolytic activity. Among the various carbohydrates, only dextran 4 protected the erythrocytes against lyse. Scanning electron microscopy showed that lyse of erythrocytes occured at once after the exposure to the hemolysin. The mito-chondrial activity and the cell membrane integrity were significantly affected by the hemolysis, within 20 min of exposure and caused apoptosis after 12 h incubation, 51.92% in HeLa and 68% in Hep-2 cells, analyzed by flow cytometry. These results suggest that the heat-stable pore forming hemolysin might be a putative virulence factor in enterococci infections.
