BACKGROUND Sleep disturbance is a core feature of post-traumatic stress disorder(PTSD).Given the relationship between sleep disturbance and PTSD,there has been a relative paucity of studies examining the potential therapeutic impact of using pharmacotherapy to target sleep disturbance in patients with ***(ESZ)is a non-benzodiazepine y-aminobutyric acid-A receptor agonist indicated for the treatment of sleep and may affect sleep in patients with *** To evaluate the efficacy of ESZ vs placebo(PBO)for patients with PTSD and *** The study was a 12-wk,double blind,randomized controlled trial with 3 mg of ESZ(n=13)or PBO(n=12).RESULTS Patients in both arms experienced significant improvement in PTSD symptoms as assessed by the Clinician-Administered PTSD Scale for DSM-IV(CAPS):ESZ(t11=-3.12,P=0.005)and PBO(t11=-3.5,P=0.002)and by self-report with the Short PTSD Rating Interview(ESZ t11=-3.38,P=0.003 and PBO t11=-4.48,P=0.0005).There were no significant differences between treatments on the CAPS(t22=-0.13,P=0.70)or the Short PTSD Rating Interview(t22=-0.58,P=0.56).Similarly,both treated groups improved on sleep measures as assessed by the Pittsburgh Sleep Quality Index with PTSD Addendum(PSQI)and on total sleep time(TST)and sleep latency assessed by actigraphy with no significant differences between groups(PSQI t22=-0.24,P=0.81;total sleep time t10=0.13,P=0.90 and sleep latency t10=0.68,P=0.50).There was a significant correlation between improvement in sleep and overall improvement in PTSD as measured by change scores on the PSQI and CAPS,r(8)=0.79,P=0.01 for ESZ treated subjects,but not for those treated with PBO r(9)=0.16,P=*** events of ESZ were consistent with the known profile of the medication including dysgeusia(30%,mild),sedation(20%,mild)and headache(20%,moderate to severe).CONCLUSION Results do not support the hypothesis of a specific positive effect of ESZ compared to PBO for measures of PTSD and associated sleep disturban

Lysine succinylation(Ksucc),defined as a transfer of a succinyl group to a lysine residue of a protein,is a newly identified protein post-translational modification^*** chemical modification is reversible,dynamic,and evolutionarily conserved^4 where it has been comprehensively studied in both bacterial and mammalian cells^*** proteins involved in the regulation of various cellular and biological processes have been shown to be heavily succinylated^*** clinical data provides evidence that dysregulation of Ksucc is correlated with the development of several diseases,including cardiovascular diseases and cancer^***,an in-depth understanding of Ksucc and its regulation is important not only for understanding its physiological function but also for developing drug therapies and targeted agents for these *** this review,we highlight some of the recent advances in understanding the role of Ksucc and desuccinylation under physiological and pathological conditions.

The impact of iodinated X-ray contrast agents on formation and toxicity of disinfection by-products in drinking water

The presence of iodinated X-ray contrast media（ICM） in source waters is of high concern to public health because of their potential to generate highly toxic disinfection by-products（DBPs）. The objective of this study was to determine the impact of ICM in source waters and the type of disinfectant on the overall toxicity of DBP mixtures and to determine which ICM and reaction conditions give rise to toxic by-products. Source waters collected from Akron,Oh were treated with five different ICMs, including iopamidol, iopromide, iohexol,diatrizoate and iomeprol, with or without chlorine or chloramine disinfection. The reaction product mixtures were concentrated with XAD resins and the mammalian cell cytotoxicity and genotoxicity of the reaction mixture concentrates was measured. Water containing iopamidol generated an enhanced level of mammalian cell cytotoxicity and genotoxicity after disinfection. While chlorine disinfection with iopamidol resulted in the highest cytotoxicity overall, the relative iopamidol-mediated increase in toxicity was greater when chloramine was used as the disinfectant compared with chlorine. Four other ICMs（iopromide, iohexol, diatrizoate, and iomeprol） expressed some cytotoxicity over the control without any disinfection, and induced higher cytotoxicity when chlorinated. Only iohexol enhanced genotoxicity compared to the chlorinated source water.

Droplet digital PCR for quantification of ITGA6 in a stool mRNA assay for the detection of colorectal cancers

AIM To investigate the use of droplet digital polymerase chain reaction(dd PCR) for detecting host m RNA markers in stools as a non-invasive test for colorectal cancer *** dd PCR and quantitative PCR were compared side by side for their performance in the detection of ITGA6 and ITGA6 A transcripts in stool samples obtained from patients with various types of colorectal lesions(advanced adenomas and stage Ⅱ-Ⅳ colorectal cancers) and control(patients displaying no pathological findings) using duplex Taq Man reactions for both methods. ITGA6 and ITGA6 A were chosen for this proof-of-concept study based on their relative medium and low abundance in stool samples, respectively, as established in a previous *** We found that the dd PCR and q PCR methods per-formed equally well in this Taq Man duplex assay for the detection of ITGA6 and ITGA6 A transcripts in stools of patients with colorectal lesions. For ITGA6, receiver operating characteristic(ROC) curve analysis showed comparable areas under the curve of 0.91(P he prediction of advanced adenomas and colorectal cancers, respectively. ITGA6 A, which was detected at very low levels in control patients, was found to be significantly elevated(over 40 times) in stage Ⅱ and Ⅲ colorectal cancers(P he two sets of data revealed a strong correlation of the copy numbers obtained by dd PCR and q PCR for both ITGA6 and ITGA6 *** We found that ITGA6 and ITGA6 A detection in stools of patients with colorectal cancers with dd PCR is comparable to that of q PCR using Taq Man assays.

Differential effects of energy balance on experimentally-induced colitis

AIM： To characterize the influence of diet-induced changes in body fat on colitis severity in SlVlAD3-/- mice.METhODS： SMAD3-/- mice （6-8 wk of age） were ran- domly assigned to receive a calorie restricted （30% of control; CR）, control （CON）, or high fat （hF） diet for 20 wk and were gavaged with sterile broth or with helicobacter hepaticus （h. hepaticus） to induce colitis. Four weeks after infection, mice were sacrificed and the cecum and colons were processed for histological evaluation.RESULTS： Dietary treatment significantly influenced body composition prior to infection （P 〈 0.05）, with CR mice having less （14%±2%） and hF-fed mice more body fat （32% ± 7%） compared to controls （22%±4%）. Differences in body composition were associated with alterations in plasma levels of leptin （hF 〉 CON 〉 CR） and adiponectin （CON 〉 hF 〉/ CR）（P 〈 0.05）. There were no significant differences in colitis scores between CON and hF-fed mice 4 wk post-infection. Consistent with this, differences in proliferation and in- flammation markers （COX-2, iNOS）, and infiltrating cell types （CD3^＋ T lymphocytes, macrophages） were not observed. Unexpectedly, only 40% of CR mice survived infection with h. hepaticus, with mortality observed as early as I wk following induction of colitis.CONCLUSION： Increased adiposity does not influence colitis severity in SMAD3-/- mice. Importantly, caloric restriction negatively impacts survival following pathogen challenge, potentially due to an impaired immune response.

ELECTROACTIVE AND NANOSTRUCTURED POLYMERS AS SCAFFOLD MATERIALS FOR NEURONAL AND CARDIAC TISSUE ENGINEERING

Conducting polymer, polyaniline （PANI）, has been studied as a novel electroactive and electrically conductive material for tissue engineering applications. The biocompatibility of the conductive polymer can be improved by （i） covalently grafting various adhesive peptides onto the surface of prefabricated conducting polymer films or into the polymer structures during the synthesis, （ii） co-electrospinning or blending with natural proteins to form conducting nanofibers or films, and （iii） preparing conducting polymers using biopolymers, such as collagen, as templates. In this paper, we mainly describe and review the approaches of covalently attaching oligopeptides to PANI and electrospinning PANI-gelatin blend nanofibers. The employment of such modified conducting polymers as substrates for enhanced cell attachment, proliferation and differentiation has been investigated with neuronal PC-12 cells and h9c2 cardiac myoblasts. For the electrospun PANI- gelatin fibers, depending on the concentrations of PANI, h9c2 cells initially displayed different morphologies on the fibrous substrates, but after one week all cultures reached confluence of similar densities and morphologies. Furthermore, we observed, that conductive PANI, when maintained in an aqueous physiologic environment, retained a significant level of electrical conductivity for at least 100 h, even though this conductivity was decreasing over time. Preliminary data show that the application of micro-current stimulates the differentiation of PC-12 cells. All the results demonstrate the potential for using PANI as an electroactive polymer in the culture of excitable cells and open the possibility of using this material as an electroactive scaffold for cardiac and/or neuronal tissue engineering applications that require biocompatibility of conductive polymers.

Dynamics of Strigolactone Function and Shoot Branching Responses in Pisum sativum

Strigolactones （SLs）, or their metabolites, were recently identified as endogenous inhibitors of shoot branch- ing. however, certain key features and dynamics of SL action remained to be physiologically characterized. here we show that successive direct application of SL to axillary buds at every node along the stem can fully inhibit branching. The SL inhibition of early outgrowth did not require inhibitory signals from other growing buds or the shoot tip. In add- ition to this very early or initial suppression of outgrowth, we also found SL to be effective, up to a point, at moderating the continuing growth of axillary branches. The effectiveness of SL at affecting bud and branch growth correlated with the ability of SL to regulate expression of PsBRC1. PsBRC1 is a transcription factor that is expressed strongly in axillary buds and is required for SL inhibition of shoot branching. Consistent with a dynamic role of the hormone, SL inhibition of bud growth did not prevent buds from later responding to a decapitation treatment, even though SL treatment immediately after decapitation inhibits the outgrowth response. Also, as expected from the hypothesized branching control network in plants, treatment of exogenous SL caused feedback down-regulation of SL biosynthesis genes within 2 h. Altogether, these results reveal new insights into the dynamics of SL function and support the premise that SLs or SL-derived metabolites function dynamically as a shoot branching hormone and that they act directly in axillary buds.
