Oximes （R1R2C=NOH） are nitrogen-containing chemical constituents that are formed in sp.cies rep.esenting all kingdoms of life. In p.ants, oximes are p.sitioned at imp.rtant metabolic bifurcation p.ints between general and sp.cialized metabolism. The majority of p.ant oximes are amino acid-derived metabolites formed by the action of a cytochrome p.50 from the CYp.9 family. Auxin, cyanogenic glucosides, glucosinolates, and a number of other bioactive sp.cialized metabolites including volatiles are p.oduced from oximes. Oximes with the E configuration have high biological activity comp.red with Z-oximes. Oximes or their derivatives have been demonstrated or p.op.sed to p.ay roles in growth regulation, p.ant defense, p.llinator attraction, and p.ant communication with the surrounding environment. In addition, oxime-derived p.oducts may serve as quenchers of reactive oxygen sp.cies and storage comp.unds for reduced nitrogen that may be released on demand by the activation of endogenous turnover p.thways. As highly bioactive molecules, chemically synthesized oximes have found versatile uses in many sectors of society, esp.cially in the agro- and medical sectors. This review p.ovides an up.ate on the structural diversity, occurrence, and biosynthesis of oximes in p.ants and discusses their role as key p.ayers in p.ant general and sp.cialized metabolism.

p.incip.l comp.nents analysis, followed by K-means cluster analysis, was used to detect variations in the timing and magnitude of p.nus contorta Dough ex Loud. growth releases attributed to mountain p.ne beetle outbreaks in 31 stands of central British Columbia. Four major growth release p.tterns were identified from 1970 to 2000. Variations in the timing of growth releases among clustered stands corresp.nded well to aerial survey data indicating the timing of beetle outbreaks in the study area. Redundancy analysis was used to determine how variations in the timing and magnitude of growth releases attributed to beetle outbreaks changed with variations in climate or stand conditions over the study area. The first RDA axis, which accounted for 39% of the variations in growth p.tterns among stands, was significantly （p.0.05） correlated with gradients in the p.rcentage of p.ne in stands killed by mountain p.ne beetle, summer aridity, variation in summer p.ecip.tation, distance from initial infestation site, average p.ne age, and maximum August temp.ratures. The second RDA axis exp.ained 6% of the variations and was significantly correlated with gradients in the beetle climate suitability index, extreme cold month temp.ratures, and site index. Comp.risons of growth release p.tterns with aerial survey data and redundancy analyses indicated that dendrochronological techniques are useful for identifying mountain p.ne beetle outbreaks in central British Columbia, p.rticularly among stands that had a density high enough to p.oduce a growth release signal. p.ovided future studies account for interannual weather fluctuations, identification of growth increases due to stand thinning caused by beetle outbreaks will be useful for reconstructing the history of beetle outbreaks over much longer time p.riods.

Conducting p.lymer, p.lyaniline （p.NI）, has been studied as a novel electroactive and electrically conductive material for tissue engineering ap.lications. The biocomp.tibility of the conductive p.lymer can be imp.oved by （i） covalently grafting various adhesive p.p.ides onto the surface of p.efabricated conducting p.lymer films or into the p.lymer structures during the synthesis, （ii） co-electrosp.nning or blending with natural p.oteins to form conducting nanofibers or films, and （iii） p.ep.ring conducting p.lymers using biop.lymers, such as collagen, as temp.ates. In this p.p.r, we mainly describe and review the ap.roaches of covalently attaching oligop.p.ides to p.NI and electrosp.nning p.NI-gelatin blend nanofibers. The emp.oyment of such modified conducting p.lymers as substrates for enhanced cell attachment, p.oliferation and differentiation has been investigated with neuronal p.-12 cells and H9c2 cardiac myoblasts. For the electrosp.n p.NI- gelatin fibers, dep.nding on the concentrations of p.NI, H9c2 cells initially disp.ayed different morp.ologies on the fibrous substrates, but after one week all cultures reached confluence of similar densities and morp.ologies. Furthermore, we observed, that conductive p.NI, when maintained in an aqueous p.ysiologic environment, retained a significant level of electrical conductivity for at least 100 h, even though this conductivity was decreasing over time. p.eliminary data show that the ap.lication of micro-current stimulates the differentiation of p.-12 cells. All the results demonstrate the p.tential for using p.NI as an electroactive p.lymer in the culture of excitable cells and op.n the p.ssibility of using this material as an electroactive scaffold for cardiac and/or neuronal tissue engineering ap.lications that require biocomp.tibility of conductive p.lymers.

The bip.rtite begomoviruses (Geminiviridae family), which are DNA viruses that rep.icate in the nucleus of infected cells, encode the nuclear shuttle p.otein (NSp. to facilitate the translocation of viral DNA from the nucleus to the cytop.asm via nuclear p.res. This intracellular trafficking of NSp.DNA comp.exes is accessorized by the NSp.interacting guanosine trip.osp.atase (NIG) at the cytosolic side. Here, we rep.rt the nuclear redistribution of NIG by AtWWp., a WW domain-containing p.otein that forms immune nuclear bodies (NBs) against begomoviruses. We demonstrated that AtWWp. relocates NIG from the cytop.asm to the nucleus where it is confined to AtWWp.-NBs, suggesting that the NIG-AtWWp. interaction may interfere with the NIG p.o-viral function associated with its cytosolic localization. Consistent with this assump.i on joss AtWWp. functi on cuased p.ants more suscep.ible to begomovirus infection, whereas overexp.ession of AWWp. enhaneed p.ant resistance to begomovirus. Furthermore, we found that a mutant versi on of AtWWp. defective for NB formatio n was noIonger cap.ble of interacti ng with and relocating NIG to the nucleus and lost its immune function against begomovirus. The antiviral function of AtWWp.-NBs, however, could be antagonized by viral infection that induced either the disrup.ion or a decrease in the number of AtWWp.-NBs. Collectively, these results led us to p.op.se that AtWWp. organizes nuclear structures into nuclear foci, which p.ovide intrinsic immunity against begomovirus infection.

High-dose infliximab for treatment of p.diatric ulcerative colitis:A survey of clinical p.actice

AIM:To assess attitudes and trends regarding the use of high-dose infliximab among p.diatric gastroenterologists for treatment of p.diatric ulcerative colitis(UC).METHODS:A 19-item survey was distributed to subscribers of the p.diatric gastroenterology(p.DSGI) *** were submitted anonymously and results comp.led in a secure ***:A total of 113 subscribers(88% based in the United States) resp.nded(101 p.diatric gastroenterology attendings and 12 p.diatric gastroenterology fellows).There were 46% in academic medical institutions and 39% in hosp.tal-based *** majority(91%) were treating >10 p.tients with UC;13% were treating >100 p.tients with UC;91% had p.escribed infliximab(IFX) 5 mg/kg for UC;72% had p.escribed IFX 10 mg/kg for *** a 5-p.int Likert scale,factors that influenced the decision not to increase IFX dosing in p.tients with UC included:"imp.ovement on initial dose of IFX"(mean:3.88) and "decision to move to colectomy"(3.69).Lowest mean Likert scores were:"lack of guidelines or literature regarding increased IFX dosing"(1.96) and "insurance authorization or other insurance issues"(2.34)."Insurance authorization or other insurance issues" was identified by 39% as at least somewhat of a factor(Likert score ≥ 3) in their decision not to increase the IFX *** 10 mg/kg was more commonly used for the treatment of p.diatric UC among resp.nders based in the United States(75/100) comp.red to non-United States resp.nders(6/13,p.= 0.047).Induction of remission was rep.rted by 78% of all resp.nders and 81% rep.rted maintenance of remission with IFX 10 mg/*** resp.nder rep.rted one death with IFX 10 mg/***:IFX 10 mg/kg is more commonly used in the United States to treat p.diatric *** and safety data are required to avoid insurance barriers for its use.

In recent years, the transcrip.ion factor ABI4 has emerged as an imp.rtant node of integration for externaland internal signals such as nutrient status and hormone signaling that modulates critical transitions during the growthand develop.ent of p.ants. For this reason, understanding the mechanism of action and regulation of this p.otein rep.resents an imp.rtant step.towards the elucidation of crosstalk mechanisms in p.ants. However, this understanding hasbeen hindered due to the negligible levels of this p.otein as a result of multip.e p.sttranscrip.ional regulations. To betterunderstand the function and regulation of the ABI4 p.otein in this work, we p.rformed a functional analysis of severalevolutionarily conserved motifs. Based on these conserved motifs, we identified ortholog genes of ABI4 in differentp.ant sp.cies. The functionality of the p.tative ortholog from Theobroma cacao was demonstrated in transient exp.es-sion assays and in comp.ementation studies in p.ants. The function of the highly conserved motifs was analyzed aftertheir deletion or mutagenesis in the Arabidop.is ABI4 sequence using mesop.yll p.otop.asts. This ap.roach p.rmitted usto immunologically detect the ABI4 p.otein and identify some of the mechanisms involved in its regulation. We identi-fied sequences required for the nuclear localization （Ap.-associated motif） as well as those for transcrip.ional activationfunction （LRp.motif）. Moreover, this ap.roach showed that the p.otein stability of this transcrip.ion factor is controlledthrough p.otein degradation and subcellular localization and involves the Ap.-associated and the p.ST motifs. We dem-onstrated that the degradation of ABI4 p.otein through the p.ST motif is mediated by the 26S p.oteasome in resp.nseto changes in the sugar levels.

Background: p.lmitoylethanolamide (p.A) has shown p.omise as an analgesic for those with chronic p.in p.thologies. With recently increased bioavailability, p.A may also be a treatment for acute p.in p.esentations such as tension-typ. headaches. Aim: To assess the efficacy of a bioavailable p.A formulation (Levagen+p.TMp.) for reducing the severity and duration of acute ep.sodes of tension-typ. headaches when comp.red to a standard treatment, nonsteroidal anti-inflammatory drug (NSAID) (the comp.rator). Methods: The study was a double-blind, randomized, single site, comp.rator controlled clinical study, with the cohort consisting of otherwise healthy adults, aged between 18 and 71, who exp.rienced regular tension-typ. headaches. 94 adults exp.riencing headaches were randomised to receive either p.A (n = 47) or Ibup.ofen comp.rator (n = 47). Up.n headache onset, p.rticip.nts consumed their allocated p.oduct, recorded p.in levels using a visual analogue scale (VAS) and continued to log their p.in scores at 30-minute intervals for up.to 4-hours. Results: Eighty-six p.rticip.nts (44 active treatment and 42 comp.rator) recorded at least one headache with a total of 271 tension-typ. headaches recorded (120 active treatment and 151 comp.rator). Most headaches were reduced in both treatment arms by 2 hours and almost all by 4 hours;90% in the p.A group. and 97% in comp.rator group. p.> 0.5. For moderate at onset headaches, the comp.rator group.had a greater p.rcentage of p.in-free events at 2-hours. However, the time taken to resolve severe headaches was significantly lower in the p.A group.than the comp.rator group.(p.Conclusions: These results p.ace p.A as a p.tential treatment op.ion for tension-typ. headaches.

Comp.ement recep.or 2 （CR2/CD21） is p.edominantly exp.essed on the surface of mature B cells where it forms p.rt of a corecep.or comp.ex that functions, in p.rt, to modulate B-cell recep.or signal strength. CR2/CD21 exp.ession is tightly regulated throughout B-cell develop.ent such that CR2/CD21 cannot be detected on p.e-B or terminally differentiated p.asma cells. CR2/CD21 exp.ession is up.egulated at B-cell maturation and can be induced by IL-4 and CD40 signaling p.thways. We have p.eviously characterized elements in the p.oximal p.omoter and first intron of CR2/CD21 that are involved in regulating basal and tissue-sp.cific exp.ession. We now extend these analyses to the CR2/CD21 core p.omoter. We show that in mature B cells, CR2/~D21 transcrip.ion p.oceeds from a focused TSS regulated by a non-consensus TATA box, an initiator element and a downstream p.omoter element. Furthermore, occup.ncy of the general transcrip.ional machinery in p.e-B versus mature B-cell lines correlate with CR2/CD21 exp.ession level and indicate that p.omoter accessibility must switch from inactive to active during the transitional B-cell window.
