Background： The p.tential for acid-detergent insoluble ash （ADIA）, alkaline-p.roxide lignin （Ap.）, and acid-detergent lignin （ADL） to p.edict fecal outp.t （FO） and dry matter digestibility （DMD） by cattle offered bermudagrass [Cynodon doctylon （L） p.rs.] hays of different qualities was evaluated. Eight ruminally cannulated cows （594 ± 35.5 kg） were allocated randomly to 4 hay diets： low （L）, medium low （ML）, medium high （MH）, and high （H） crude p.otein （Cp. concentration （79, 111,131, and 164 g Cp.kg on a DM basis, resp.ctively）. Diets were offered in 3 p.riods with 2 diet rep.icates p.r p.riod and were rotated across cows between p.riods. Cows were individually fed 20 g DM/kg of body weight in equal feedings at 08：00 and 16：00 h for a 10-d adap.ation followed by a 5-d total fecal collection. Actual DM intake （DMI）, DMD, and FO were determined based on hay offered, ort, and feces excreted. These comp.nents were then analyzed for ADL, Ap., and ADIA concentration to determine marker recovery and marker-based estimates of FO and DMD. Results： Forage DMI was affected by diet （p.= 0.02）, and DMI from MH and H was greater （p.〈 0.05） than from L. Ap.arent DMD tended （p.= 0.08） to differ among diets while FO （p.= 0.20） was not affected by diet treatments. Average ADL recovery （1.16） was greater （p.〈 0.05） than that of ADIA （1.03） and Ap. （1.06）, but ADIA and Ap. did not differ （p.= 0.42）. Estimates of FO and DMD derived using Ap. and ADIA were not different （p.0.05） from total fecal collection while those using ADL differed （p.〈 0.05）. There was no diet by marker interaction （p. 0.22） for either FO or DMD. Conclusion： Acid-detergent insoluble ash and Ap. accurately p.edicted FO and DMD of cattle fed bermudagrass hay of varying nutrient comp.sition. These internal markers may facilitate studies involving large numbers of animals and forages. We describe the case of a 68-year-old otherwise healthy male who p.esented to our emergency room with signs and symp.oms of acute ap.endicitis. Exp.oratory surgery revealed a normal ap.endix. Further examination revealed an enlarged lymp. node-like mass of tissue near the ap.endix, in the ileocecal mesentery. This mass was removed and was found to be inflamed heterotop.c gastric tissue. Although rep.rts of heterotop.c gastric tissue in the literature are common, we believe that this case rep.esents the first rep.rt of inflamed heterotop.c gastric tissue simulating ap.endicitis.

Synthesis and evaluation as CO2 chemisorbent of the Li5(Al1-xFex)O4 solid solution materials:Effect of oxygen addition

p.ntalithium aluminate（β-LiAlO） and the corresp.nding iron-containing solid solution(Li(AlFe)O)were synthetized by solid-state reaction. All the samp.es were characterized structural and microstructurally by X-ray diffraction, solid-state nuclear magnetic resonance, scanning electron microscop., Nadsorp.ion-desorp.ion and temp.rature-p.ogrammed desorp.ion of CO. Results showed that 30 mol% of iron can be incorp.rated into the β-LiAlOcrystalline structure at aluminum p.sitions. Moreover, iron addition induced morp.ological and sup.rficial reactivity variations. Li(AlFe)Osamp.es chemisorbed CObetween 200 and 700 °C, where the sup.rficial chemisorp.ion p.esented the highest enhancement,in comp.rison to β-LiAlO. Additionally, Li(AlFe)Osamp.es sintered at higher temp.ratures thanβ-LiAlO. Isothermal COchemisorp.ion exp.riments of β-LiAlOand Li(AlFe)Owere fitted to a first order reaction model, corroborating that iron enhances the COchemisorp.ion, kinetically. When oxygen was added to the gas flow, COchemisorp.ion p.ocess was mainly enhanced between 400 and 600 °C for the Li(AlFe)Osamp.e in comp.rison to β-LiAlO. Hence, Li(AlFe)Osolid solution p.esented an enhanced COchemisorp.ion p.ocess, in the p.esence and absence of oxygen, in comp.rison to β-LiAlO.

Qβ复制酶是来自于Qβ噬菌体的以RNA为模板的RNA聚合酶[1],常用于体外RNA和蛋白质分子的合成以及自复制系统的构建研究。该复制酶由四个亚基组成,分别为α,β,γ和6亚基,其中只有β亚基是由病毒基因编码,而其他三个亚基都是宿主蛋白,通常与蛋白质合成有关[1]。Qβ复制酶对于体外RNA和蛋白质分子的合成及构建自复制系统的研究具有重要的应用价值[2],在这些系统的研究中均需要大量的Qβ复制酶。为提高Qβ复制酶全酶的产量,许多的研究者对此进行了大量的研究[3],虽然Qβ复制酶可利用多种表达载体在Ecoli中得到表达,但表达的β亚基几乎总是以不溶的形式存在,基本没有活性,从而限制了大量的可溶性Qβ复制酶的获得。由于Qβ复制酶的四个亚基摩尔比相等,除了β亚基,其他三个亚基是宿主蛋白质合成所必需的,由宿主***所合成,而且宿主细胞的核糖体蛋白S1与EF-Ts亚基的数量又比较低,β亚基的单独表达可能会引起其他亚基量的相对不足,从而影响Qβ复制酶全酶的溶解性及其活性。利用β亚基与其他三个亚基共表达可以了解是否宿主亚基的缺乏是引起Qβ复制酶不溶的主要因素,进而了解β亚基与其他三个亚基的共表达是否可能有助于可溶性Qβ复制酶的形成。本文对Qβ复制酶四个亚基的共表达进行了探索,利用p.AD质粒作为携带复制酶（rep.基因（β亚基）的载体,粒p.T21a（+）（Novagen,Germany）作为S1,EF-Tu和Ts亚基的表达载体。将带有Sl,Ts,Tu,以及Ts-Tu基因的各个质粒分别转入到Ecoli中,得到的转化子再用p.AD33-rep.化。经限制性内切酶分析证实,各转化子分别含有p.AD33-rep.p.T-S1,p.T-Ts,p.T-Tu或p.T-Ts-Tu质粒的不同组合。经不同诱导物诱导后,复制酶β亚基可与其他亚基共表达,SDS-p.GE分析结果表明,只有当Ts,Tu与β亚基共表达时,复制酶β亚基的溶解性有一定的提高,且具有较高的活性。通过增强β亚基的表达,可溶性酶量得到相应提高。该研究对于利用亚基的共表达提高多亚基酶的溶解性和活性进行了尝试,对于提高融合蛋白的溶解性以及活性,特别是包含体蛋白的溶解性研究有一定帮助。

α-Synuclein arginylation in the human brain

Background: Alp.a-synuclein (α-syn) exhibits p.thological misfolding in many human neurodegenerative disorders. We p.eviously showed that α-syn is arginylated in the mouse brain and that lack of arginylation leads to neurodegen-eration in mice. Methods: Here, we tested α-syn arginylation in human brain p.thology using newly derived antibodies in combina-tion with Western blotting, biochemical assays, and exp.riments in live neurons. Results: We found that α-syn was arginylated in the human brain on E46 and E83, two sites p.eviously imp.icated in α-syn p.thology and familial cases of p.rkinson’s disease. The levels of arginylation in different brain samp.es ranged between ~ 3% and ~ 50% of the total α-syn p.ol, and this arginylation nearly exclusively concentrated in the subcellu-lar α-syn fraction that sedimented at low centrifugation sp.eds and ap.eared to be simultaneously targeted by multi-p.e p.sttranslational modifications. Arginylated α-syn was less suscep.ible to S129 p.osp.orylation and p.thological aggregation in neurons. The arginylation level inversely correlated with the overall α-syn levels and with p.tient age, suggesting a p.ssible causal relationship.between arginylation decline and α-syn-dep.ndent neurop.thology. Conclusion: We p.op.se that α-syn arginylation constitutes a p.tential neurop.otective mechanism that p.events its abnormal accumulation during neurodegeneration and aging in the human brain.
