Reduced expression ofβ-catenin inhibitor Chibby in colon carcinoma cell lines

AIM： To analyse the Chibby expression and its function in colon carcinoma cell lines and colorectal carcinoma （CRC）. METHODS： Chibby expression levels were investigated by quantitative RT-PCR in a panel of seven different colon carcinoma cell lines. By sequencing, we analysed mutational status of Chibby. To test whether Chibby exhibited effects on β-catenin signalling in colon carcinoma cells, we transfected SW480 cells with Chibby expression plasmid and, subsequently, analysed activity of β-catenin and tested for alterations in cellular phenotype. In addition, we examined Chibby mRNA levels in samples of colorectal carcinomas and adjacent normal tissues by using quantitative RT-PCR and hybridised gene chips with samples from CRC and normal tissues. RESULTS： Chibby mRNA expression was strongly downregulated in colon carcinoma cell lines in comparison to normal colon epithelial cells and no mutation in any of the examined colon carcinoma cell lines was found. Further, we could show that Chibby inhibited β-catenin activity in TOPflash assays when over-expressed in SW480 cells. Proliferation and invasion assays with Chibby transfected SW480 cells did not reveal profound differences compared to control cells. In contrast to these in vitro data, quantitative RT-PCR analyses of Chibby mRNA levels in CRC tumor samples did not show significant differences to specimens in adjacent non-cancerous tissue. Consistent with these findings, gene chips analysing tissue samples of tumors and corresponding normal tissue did not show altered Chibby expressionCONCLUSION： Altered Chibby expression might be observed in vitro in different colon carcinoma cell lines. However, this finding could not be confirmed in vitro in CRC tumors, indicating that Chibby is not likely to promote CRC tumor development or progression. The Role of Stationary and Transient Planetary Waves in the Maintenance of Stratospheric Polar Vortex Regimes in Northern Hemisphere Winter

Using 1958-2002 NCEPNCAR reanalysis data, we investigate stationary and transient planetary wave propagation and its role in wave-mean flow interaction which influences the state of the polar vortex （PV） in the stratosphere in Northern Hemisphere （NH） winter. This is done by analyzing the Eliassen-Palm （E-P） flux and its divergence. We find that the stationary and transient waves propagate upward and equatorward in NH winter, with stronger upward propagation of stationary waves from the troposphere to the stratosphere, and stronger equatorward propagation of transient waves from mid-latitudes to the subtropics in the troposphere. Stationary waves exhibit more upward propagation in the polar stratosphere during the weak polar vortex regime （WVR） than during the strong polar vortex regime （SVR）. On the other hand, transient waves have more upward propagation during SVR than during WVR in the subpolar stratosphere, with a domain of low frequency waves. With different paths of upward propagation, both stationary and transient waves contribute to the maintenance of the observed stratospheric PV regimes in NH winter.

The Interannual Variability of East Asian Winter Monsoon and Its Relation to the Summer Monsoon

Based on the NCEP/ NCAR reanalysis data the interannual variability of the East Asian winter mon-soon (EAWM) is studied with a newly defined EAWM intensity index. The marked features for a strong (weak) winter monsoon include strong (weak) northerly winds along coastal East Asia, cold (warm) East Asian continent and surrounding sea and warm (cold) ocean from the subtropical central Pacific to the trop-ical western Pacific, high (low) pressure in East Asian continent and low (high) pressure in the adjacent ocean and deep (weak) East Asian trough at 500 hPa. These interannual variations are shown to be closely connected to the SST anomaly in the tropical Pacific, both in the western and eastern Pacific. The results suggest that the strength of the EAWM is mainly influenced by the processes associated with the SST anom-aly over the tropical Pacific. The EAWM generally becomes weak when there is a positive SST anomaly in the tropical eastern Pacific (El Ni?o), and it becomes strong when there is a negative SST anomaly (La Ni?a). Moreover, the SST anomaly in the South China Sea is found to be closely related to the EAWM and may persist to the following summer. Both the circulation at 850 hPa and the rainfall in China confirm the connection between the EAWM and the following East Asian summer monsoon. The possible reason for the recent 1998 summer flood in China is briefly discussed too. The Influence of High Temperature Exposure on the Wear Performance of a Ni Based Alloy PTA Coating

Degradation phenomena like wear and corrosion, may have their effects accelerated as components operate at high temperature. The aim of this work is to make an indirect evaluation of the influence of high temperature exposure on wear behavior of Ni coatings. A Ni-Cr-Mo-W alloy, was deposited with Plasma Transferred Arc (PTA) process. An indirect approach based on the Ahrrenius relationship was used to evaluated long time exposures at high temperatures. To simulate longer exposures at lower temperatures, coatings were exposed to higher temperatures for the same time interval. So coatings were soaked in an air furnace at 650°C, 1000°C and 1200°C. Metal/metal wear behavior was evaluated by pin-on-disc tests. Pins were removed from the coatings by eletrodischarge machining and rubbed against a VC31 quenched and tempered tool steel. Two sets of wear tests were run at 2m/s, for different loads (0.5, 1.0, 2.0 and 3.0Kg) and for different sliding distances (120, 720, 1080, 1440 and 1800m).Coatings characterization was done by scanning electronic microscopy and Vickers microhardness. Results showed as temperature is increased, coatings wear performance is altered, and this is associated with microstructural changes.
