Tityus serrulatus venom and its toxins Ts1 and Ts5 increase cytosolic ca^(2+)concentration in isolated vascular smooth muscle cells

Voltage-gated Na+ channel (Nav channel) scorpion toxins are classified as α- and β-neurotoxins. Ts5 (α-neurotoxin) and Ts1 (β-neurotoxin) from Tityus serrulatus venom (TsV) interact with Nav channels, increasing Na+ influx and activating voltage-dependent ca2+ channels. This study aimed to investigate the effect of TsV, Ts1 and Ts5 on the cytosolic ca2+ concentration ([ca2+]c) in rat aortic smooth muscle cells. Toxins were isolated by ion exchange chromatography (Ts1) followed by RP-HPLc (Ts5). The rat aortic smooth muscle cells were isolated in Hanks buffer pH 7.4 and loaded with 5 μmol/L of Fura-2AM (45 minutes at 37℃), in order to measure [ca2+]c by fluorescence of Fura-2/AM (ratio 340/380 nm). The fluorescence was measured in one single cell (excitation: 340 and 380 nm;emission: 510 nm). TsV (100 and 500 mg/mL) and its toxins Ts1 and Ts5 (50 and 100 mg/mL each) led to a concentration-dependent increase in [ca2+]c. Tetrodotoxin (1 mmol/L), a Nav channel blocker, and verapamil (1 mmol/L), a voltage-operated ca2+ channel blocker, inhibited the increase in [ca2+]c induced by TsV (500 mg/mL). In conclusion, TsV and its toxins induce a concentration-dependent increase in [ca2+]c that probably occurs through interaction with Nav channels, thus inducing depolarization and consequent ca2+ influx. This assumption is based on the fact that this effect is inhibited by tetrodotoxin and verapamil, showing a direct action of TsV toxins on aorta smooth muscle cells.

Modeling the Kinetics of Moisture Desorption from Safou Pulp [Dacryodes edulis (G. Don) ***] before the Oil Extraction Step

The aim of this work is to model the drying kinetics of Safou pulp with or without endocarp using a phenomenological approach. Oven-drying kinetics at 70˚c, 90˚c and 105˚c were monitored using the curves given by the reduced mass as a function of time, which are modeled according to the Avrami/page, Fick and Peleg models using OringinPro 2018 software. The results showed that parameters k and n of the Avrami/Page model vary very little with fruit size and drying temperature (0.0018 ± 0.0002 k n k (Avrami model/page) were virtually identical, while b (Fick model) and n (Avrami model/page) were virtually identical for the same sample. For the Peleg model, the parameter a, varies from 0.0018 ± 0.0002 to 0.03328 ± 0.0079, with a ratio of 18.6 for all experimental conditions studied. However, with 0.977 R2 χ2 ch ranges from 0.82 ± 0.05 to 1.21 ± 0.02. Thus, drying modeling by these three models can be used to describe and predict the progress of oven-drying of safou pulp.

Beans (Phaseolus vulgaris L.) are widely grown in cameroon and play a key role in the fight against food insecurity, malnutrition and poverty. However, its cultivation encounters problems due to abiotic and biotic stresses, which leads to the use of synthetic fertilizers and pesticides, which cause significant damage to the environment and human health due to the presence of synthetics residues in the seeds, pods and in the leaves that are eaten. Promoting the use of natural products is becoming a necessity for organic and eco-responsible agriculture that limits contamination problems and improves people’s purchasing power. This study aims to assess the effect of biostimulants based on natural products on the growth and nutritional value of common bean (Phaseolus vulgaris L.). Bean seedlings from white variety (MEX-142) and red variety (DOR-701) were treated every seven days in the field from their pre-emergence, emergence and growth to their maturation under a randomized block experimental design. Six treatments and three repetitions with the biostimulants based on natural products and controls were thus performed and the agromorphological parameters were measured. After 120 days, the contents of growth biomarkers and defense-related enzymes were evaluated in leaves, while the contents of macromolecules, minerals and antinutrients were evaluated in seeds. These biostimulants significantly increased (P P cluding oxalates, phytates, tannins and saponins in seeds compared to controls (T+ and T−). Treatment with biostimulants, in particular BS4, improves the performance of bean plants in the field as well as the biofortification of seeds regardless of the variety.

The interferon inducing pathways and the hepatitis c virus

The innate immune response is triggered by a variety of pathogens, including viruses, and requires rapid induction of typeⅠ?interferons (IFN), such as IFNβ and IFNα. IFN induction occurs when specific pathogen motifs bind to specific cellular receptors. In non-professional immune, virally-infected cells, IFN induction is essentially initiated after the binding of dsRNA structures to TLR3 receptors or to intracytosolic RNA helicases, such as RIG-Ⅰ/MDA5. This leads to the recruitment of specific adaptors, such as TRIF for TLR3 and the mitochondrial-associated IPS-1/VISA/MAVS/cARDIF adapter protein for the RNA helicases, and the ultimate recruitment of kinases, such as MAPKs, the canonical IKK complex and the TBK1/IKKε kinases, which activate the transcription factors ATF-2/ c-jun, NF-κB and IRF3, respectively. The coordinated action of these transcription factors leads to induction of IFN and of pro-inflammatory cytokines and to the establishment of the innate immune response. HcV can cleave both the adapters TRIF and IPS-1/VISA/MAVS/ cARDIF through the action of its NS3/4A protease. This provokes abrogation of the induction of the IFN and cytokine pathways and favours viral propagation and presumably HcV chronic infection.

What is the most effective exercise protocol to improve cardiovascular fitness in overweight and obese subjects？

Background: Increased peak oxygen consumption(VO2 peak) can reduce cardiovascular risks associated with obesity. Our aim was to analyze the effect of a weight loss program on cardiovascular fitness in overweight(W) and obese(O) ***: One hundred and sixty-seven subjects(77 males and 90 females), aged 18–50 years, performed a modified Bruce protocol before(pre)and after(post) a weight loss program of 24 weeks. This program combined physical training(strength, S; endurance, E; combined strength + endurance, SE; or physical activity recommendation, PA) 3 times per week, with a 25%–30% caloric restriction ***: VO2 peak improved in overweight and obese males(pre and post values in L/min, respectively; W = 3.2 ± 0.6 vs. 3.7 ± 0.5, p creased for all 4 interventions in males(p cept for S in the obese group(1.6 ± 0.2 vs. 1.7 ± 0.3, p = 0.141). In females, it increased in E(0.9 ± 0.2 vs. 1.4 ± 0.3, p cts except for females in PA group(15.7 ± 0.3 min vs. 15.9 ± 0.3 min, p = 0.495).conclusion: Our results suggest that all methods, including the recommendation of physical activity, can improve cardiovascular fitness in overweight subjects and obese males.

The Influence of Public Policies on Urban Mobility： A comparative Study between Porto Alegre （Brazil） and Washington D.c. （United States）

Increasing urbanization rates observed in the last decades have significantly contributed to a higher number of travelers to urban environments. The objective of this study was to identify measures and factors that encourage the use of alternative transportation in the cities of Washington D.c., USA and Porto Alegre, Brazil. The methodology used to compare these two cities was an evaluation of their urban mobility plans, including strategic analyses and measures to promote the use of public transport. This approach was based on factors described in the following document：＂Urban Sustainability： Impacts of Economic Development and Their consequences on the Process of Urbanization in Emerging countries＂, prepared by the United Nations. The results indicate different planning strategies, ranging from focusing on tax integration policies in order to become more financially attractive to the user, to using different modes of mass transportation integrated with active mobility. In Washington D.c., an innovative strategy was developed in preparation to forecast scenarios, as the measures comprised projection for the year 2040, which will respond to mobility demands in the coming decades.

Effects of Dexamethasone, clonidine, Tramadol and Nalbuphine on Fentanyl-Induced Hyperalgesia in Rats

Opioids are drugs used to alleviate pain. However, studies have demonstrated that these drugs can cause an increase in pain sensitivity, which is called opioid-induced hyperalgesia. The objective of this study was to describe the effects of dexamethasone, clonidine, tramadol and nalbuphine on fentanyl-induced hyperalgesia in rats. After obtaining approval from the committee for the Ethical Use of Animals (cEUA), 36 male Wistar rats were divided into 6 groups: Group 1 (GcSSL) wherein the rats received 1 ml 0.9% saline solution in two injections;Group 2 (GFTSL), received fentanyl at a dose of 100 ug·kg-1 followed by 1 ml 0.9% saline solution via intraperitoneal;the remaining groups (3, 4, 5, 6) received fentanyl at a dose of 100 ug·kg-1 following doses via intraperitoneal: Group 3 (GFTDX), dexamethasone at a dose of 1.0 mg·kg-1;Group 4 (GFTcL), clonidine at a dose of 20 mg·kg-1;Group 5 (GFTTR), tramadol at a dose of 50 mg·kg-1, and Group 6 (GFTNB), nalbuphine at a dose of 5 mg·kg-1. Under general anestesia using isoflurane, the animals were submitted to a surgical incision. Hyperalgesia was evaluated by applying Von Frey filaments at 2 hours after the incision and on the 1st, 3rd and 5th days afterward. At 2 hours after the surgical procedure, there was lower intensity of pain in the fentanyl group (GFTSL) compared to the other groups, and on the fifth day there were no significant differences for pain intensity between groups. The results suggest the presence of fentanyl-induced hyperalgesia and efficacy in its reduction by dexamethasone, clonidine, tramadol and nalbuphine.

Physiological conditioning of Alibertia edulis(Rich)Seeds

Physiological conditioning of seeds has been shown to increase the uniformity of seedlings;thus, it has been useful for propagating native tree species from the Brazilian cerrado which, otherwise, are difficult to propagate successfully. The objective of this work was to evaluate the effect of physiological conditioning of Alibertia edulis seeds by soaking for 5 or 10 days in solutions of varying osmotic potential. After conditioning, seeds were dried down to original water content, sown on sheets of germitest paper inside gerbox plastic boxes, and incubated at 25°c. We evaluated the effect of conditioning by studying seed germination and vigor. Seed conditioning by osmotic pretreatment showed positive effects;however, germination and growth of seedlings from seeds conditioned at osmotic potentials of -0.3 to -0.7 MPa were reduced. Osmoconditioning for 10 days at -0.7 MPa resulted in increased percent germination, indicating that the longest imbibition period in the osmotic solution of the lowest osmotic potential (-1.3 MPa) favored the seed germination process. A. edulis seeds did not require conditioning to attain high germination rates;nonetheless, osmotic conditioning reduced average seed-germination time.
