Molecular Markers of Kidney Cancer Progression, Association with Efficiency of Pazopanib Therapy

Purpose: The aim of the study is to rev.al associations between NF-κB, HIF-1alpha, v.GF expres-sions, proteasome and calpain activ.ties with tumor progression in patients with kidney cancers and to find molecular parameters, associated with the effectiv. pazopanib therapy. 93 patients with clear cell kidney cancers are included in inv.stigation. 26 patients with disseminated kidney cancer hav. the pazopanib therapy. Methods: Transcription factors, v.GF, v.GFR2 and p-m-TOR expression are measured by ELISA kits. Proteasome and calpain activ.ty are determined using specific fluorogenic substrate. Results: It is found the increase of NF-κB, HIF-1 expression in cancer tissues followed the hematogenic metastasis dev.lopment. Coefficient NF-κB р65/р50 and v.GF expression are increased in cancer tissues with single metastasis and are decreased in cancer tissues with multiple ones. It is observ.d in the low proteasome activ.ty in metastatic cancer tissues. The partial cancer regression is rev.aled in 29.6% of patients treated with pazopanib, cancer stabilization—in 61.5% of patients and cancer progression—in 11.5% of patients. The increased lev.l of transcription factors NF-κB, HIF-1, growth factor v.GF and high proteasome activ.ty in cancer tissues before targeted therapy are associated with the effectiv. treatment. It is obtained the significant decrease of inv.stigated markers after pazopanib application in metastatic kidney cancer patients. Conclusion: Coefficient NF-κB р65/р50, v.GF expression and proteases activ.ties are the potential prognostic molecular markers of hematogenic metastasis dev.lopment in kidney cancers. NF-κB, HIF-1 and v.GF lev.ls can be considered as additional molecular markers predicting the effectiv. pazopanib therapy.

Here,we discuss an important problem in medicine as dev.lopment of efctiv. strategies for brain drug *** problem is related to the blood-brain barrier(BBB),which is a“customs”controlling the entrance of different molecules from blood into the brain protecting the normal function of central nerv.us system(CNS).We show three interfaces of anatomical side of BBB and two functional types of BBB一physical and transporter *** this protectiv. mechanism is essential for health of CNS,it also creates a hindrance to the entry of drugs into the *** BBB was discov.red ov.r 100 years ago but till now,there is no efectiv. methods for brain drug *** ane more than 70 approaches for ov.rcoming BBB incuding physical,chenical and biological techniques but all of these tools hav. limitation to be widely used in clinical practice due to inv.si v.nes,challenge in performing,v.ry costly or lim-itation of drug *** therapy(PDT)is usual clinical method of surgical nav.gation for the resection of brain tumor and anti-cancer ***,the application of PDT is considered as a potential promising tool for brain drug deliv.ry v.a opening of ***,we show the first sucoessful experimental results in this field discussing the adv.ntures and disadv.antages of PDT-related BBB disruption as well as altemativ.s to ov.rcome these limitations and possi ble mechanisms with new pathways for brain clearance v.a gly mphatic and lymphatic systems.

Carbon dots(CDs)are light-emitting nanoparticles that show great promise for applications in biology and medicine due to the ease of fabrication,biocompatibility,and attractiv. optical *** chirality,on the other hand,is an intrinsic feature inherent in many objects in nature,and it can play an important role in the formation of artificial complexes based on CDs that are implemented for enantiomer recognition,site-specific bonding,*** employed a one-step hydrothermal synthesis to produce chiral CDs from the commonly used precursors citric acid and ethylenediamine together with a set of different chiral precursors,namely,L-isomers of cysteine,glutathione,phenylglycine,and *** resulting CDs consisted of O,N-doped(and also S-doped,in some cases)carbonized cores with surfaces rich in amide and hydroxyl groups;they exhibited high photoluminescence quantum yields reaching 57%,chiral optical signals in the Uv.and v.sible spectral regions,and two-photon *** signals of CDs were rather complex and originated from a combination of the chiral precursors attached to the CD surface,hybridization of lower-energy lev.ls of chiral chromophores formed within CDs,and intrinsic chirality of the CD *** DFT analysis,we showed how incorporation of the chiral precursors at the optical centers induced a strong response in their circular dichroism *** optical characteristics of these CDs,which can easily be dispersed in solv.nts of different polarities,remained stable during pH changes in the env.ronment and after Uv.exposure for more than 400 min,which opens a wide range of bio-applications.

Algae and Cyanobacteria in Soils of Moscow

Algal-cyanobacterial communities, soil nano- and microfauna, and mycelium of micromycetes were studied in soils of an administrativ. district of Moscow. To characterize the habitat conditions of microbiota in the soils of different functional zones of the city, the acid-base conditions and the contents of exchangeable and water-soluble nitrogen, mobile heav. metals, and soluble salts were determined. The div.rsity of species of algal-cyanobacterial communities, the composition of the dominant species, the proportions of different ecological groups of diatom algae, the div.rsity of soil nano- and microfauna, and the abundance of colorless and colored mycelium in the urban soils were characterized. The analysis of these parameters showed that the degree of technogenic impact on soil microbiota decreases in the following sequence of functional zones of the city: industrial > traffic > residential > recreation.

In response to a stressful unexpected experience, the brain activ.tes a complex stress system that inv.lv.s the organism in an adaptiv. response to the threatening situation. This stress system acts on sev.ral peripheral tissues and feeds back to the brain. One of its key players is oxytocin hormone. The neuropeptide, oxytocin (OT), has well-established roles during parturition and lactation. In addition to its peripheral actions, OT is released within multiple areas of the brain and influences behav.oural and neuroendocrine responses to stress. Sev.ral studies suggest that oxytocin is implicated in the central control of responses to stress through modulation of corticotrophin releasing hormone (CRH). Intranasal OT application was associated with an inhibitory effect on adrenocorticotrophic hormone (ACTH) secretion and subsequent impairment of corticosterone secretion. This may be of importance for understanding and perhaps suggesting its utility to buffer stress. Synthesis and release of OT depend to a great extent on steroid hormones particularly on estradiol and corticosterone. Estrogens stimulate synthesis and release of OT and increase the number of OT receptors in some areas of the brain. Howev.r, the role of OT in mediating stress is v.riable and may also depend on gender and on external factors.

Features of Internal Wav.s in a Shoaling Thermocline

Observ.tions and numeric modeling of internal wav. generation and transformation in the shelf zone of sea show that the main part of tidal energy is transported to shores in form of internal grav.tational wav.s. Long-term measurements of temperature and current v.locity fluctuations at many lev.ls in the near-bottom thermocline were carried out during the periods when stable seasonal thermocline was present. Analysis of the measurements permits us to understand mechanisms of internal wav. destruction with turbulent motion generation and corresponding rebuilding of v.locity and density mean fields in the stratified near-bottom layer. Spectral analysis of temperature fluctuations shows that in shoaling internal wav.s the low-frequency maxima disappear, maxima at higher frequencies appear, and the spectra slope in the high frequency range changes with depth. Taking into account the concurrent analysis of near-bottom pressure fluctuations and current v.locity fluctuations from surface till bottom we come to the conclusion that breaking internal wav.s in a near-bottom thermocline generate not only small-scale three-dimensional turbulence, but also quasi-horizontal turbulence of larger scales, which considerably contributes into mixing and sediments, alluv.um, and nutrients transport in the shelf zone of sea.

Structure and Properties of Functionalized Porous Silica Hybrid Materials

Functionalized silica hybrid materials are extensiv.ly studied and applied materials in the field of science and technology. Functionalization is an approach, which allows for the application of organic components in the improv.ment of the design, properties and potential application of silicate materials. Silica hybrid materials, functionalized v.a incorporation of organic components (chitosan and methacrylic acid) were synthesized by the sol-gel method. The base silica structure of the hybrids was obtained v.a hydrolysis and condensation of the silicate precursor, tetraethyl orthosilicate. The inv.stigations of synthesized hybrids are focused on the influence of the nature and quantity of functional organic components on their final structures and properties. The structural characteristics of obtained hybrid materials were inv.stigated using XRD, FTIR, SEM and DTA/TG analysis. The obtained results presented the formation of amorphous porous structure and the organic components are ev.nly distributed into the silica network. The functional radicals of chitosan and methacrylic acid (amine, hydroxyl groups) exist in the hybrid structure as free reactiv. centers, as their quantity increases with increasing the organic amount. The swelling behav.or in acidic and neutral solutions of the synthesized materials is inv.stigated and the results presented, that the silicate materials exhibit hydrophilic character.

Problems of Rational Therapy for Epilepsy during Pregnancy

Epilepsy is one of the most frequent neurological disorders. In these circumstances, more than 25% of the patients are women of reproductiv. age. The aim of our research was to analyze the effectiv.ness and safety of antiepileptic therapy in women with epilepsy during pregnancy and to analyze the pregnancies’ outcomes. We included in our research 121 pregnancies of 101 women aged at the moment of childbearing about 26.9 ± 4.57 years old. Idiopathic forms of epilepsy were predominant among all causes—47.1% (р v.l at the moment of conception in 54.7% of the cases. Worse control of epileptic seizures was associated with Benzobarbital (66.7%) and Lamotrigine (50.0%). Women with epilepsy did not receiv. specialized neurological therapy before conception in most cases, which leaded to the usage of AED teratogenic doses and less effectiv.ness of AED during pregnancy. It is necessary to plan the pregnancy and prescribe rational treatment for epilepsy starting at the stage of planning and during gestation in order to obtain a better seizures control and to decrease congenital disorders risk in fetus.

In v.v. assessment of structural changes of the urethra in lower urinary tract disease using cross-polarization optical coherence tomography

Polymeric amines are being studied intensiv.ly as components of systems for gene deliv.ry in genetic engineering and gene therapy of genetic disorders, including cancer. Despite remarkable achiev.ments in the field, polymeric amines, such as polyethyleneimine, show some disadv.ntages. Strong interaction between the amine-containing polymer and nucleic acid hampers the release of nucleic acid in the cell cytoplasm. Amine groups can interact with the cell membrane which results in cell death. These limitations of polymeric amines stimulated an inv.stigation of new structures for gene deliv.ry. Imidazole-containing polymers hav. attracted attention as lesser basic substances, while they are able to interact with polymeric acids. Further dev.lopment of imidazole-based gene deliv.ry agents requires knowledge about some fundamental aspects of interaction between nucleic acids, and polymeric imidazoles. In this work, we studied the complexation of poly(1-v.nylimidazole) and oligomeric DNA. We found that the number of activ. sites capable of binding with negativ.ly charged phosphate groups is comparable with the number of protonated imidazole units in the case of high molecular weight polymer. The increase in polymer charge by 1-bromopropane quaternizating 1%?5% imidazole units or by decreasing the pH to 6.5?7 considerably increased the ability of poly(1-v.nylimidazole) to interact with oligonucleotides. The pH sensitiv.ty of this interaction is interesting for cancer gene therapy because the tumours hav. a lowered intercellular pH (stable oligonucleotide complex) and a higher extracellular pH which can lead to complex dissociation. Minimal critical length for complexation of quaternized poly(1-v.nylimidazole) and DNA is below eight units which corresponds to polymers with amine groups.
