Effect of Aspergillus niger prolyl endopeptidase in patients with celiac disease on a long-term gluten-free diet

BACKGROUND The gluten-free diet(GfD)has limitations,and there is intense research in the development of adjuvant therapies.AIM To examine the effects of orally administered Aspergillus niger prolyl endopeptidase protease(AN-PEP)on inadvertent gluten exposure and symptom prevention in adult celiac disease(CeD)patients following their usual GfD.METHODS This was an exploratory,double-blind,randomized,placebo-controlled trial that enrolled CeD patients on a long-term GfD.After a 4-wk run-in period,patients were randomized to 4 wk of two AN-PEP capsules(GliadinX;AVI Research,LLC,United States)at each of three meals per day or placebo.Outcome endpoints were:(1)Average weekly stool gluten immunogenic peptides(GIP)between the run-in and end of treatments and between AN-PEP and placebo;(2)celiac symptom index(CSI);(3)CeD-specific serology;and(4)quality of life.Stool samples were collected for GIP testing by ELISA every Tuesday and friday during run-ins and treatments.RESULTS forty patients were randomized for the intention-to-treat analysis,and three were excluded from the per-protocol assessment.Overall,628/640(98.1%)stool samples were collected.GIP was undetectable(f samples,and no differences between treatment arms were detected.Only 0.5%of samples had GIP concentrations sufficiently high(>0.32μg/g)to potentially cause mucosal damage.Median GIP concentration in the AN-PEP arm was 44.7%lower than in the run-in period.One-third of patients exhibiting GIP>0.08μg/g during run-in had lower or undetectable GIP after AN-PEP treatment.Compared with the run-in period,the proportion of symptomatic patients(CSI>38)in the AN-PEP arm was significantly lower(Pfic serologies.CONCLUSION This exploratory study conducted in a real-life setting revealed high adherence to the GfD.The AN-PEP treatment did not significantly reduce the overall GIP stool concentration.However,given the observation of a significantly lower prevalence of patients with severe symptoms in the AN-P

Opinion Dynamics in Two-Layer Networks with Hypocrisy

We propose a general concealed voter model(GCVM),in which individuals interact in two layers and can exchange their opinions in the internal layer.This interaction is not allowed in a CVM.By exchanging opinions in the internal layer,we mean that individuals share their real or internal opinions with their close friends.The process of opinion formation in GCVM is presented in the paper.We make the series of numerical simulations of GCVM with different network structures(both external and internal)and get some counterintuitive conclusions.for instance,we find out that sometimes with a relatively simple network structure of an external layer the consensus within the individuals’opinions cannot be reached,and if individuals in the network are not good at expressing their opinions publicly(in an external layer),exchanging opinions with their close friends(in an internal layer)is almost useless.

Strong Subgame Consistency of the Core in Stochastic Network formation Games

We consider a model of network formation as a stochastic game with random duration proposed initially in Sun and Parilina(Autom Remote Control 82(6):1065–1082,2021).In the model,the leader first suggests a joint project to other players,i.e.,the network connecting them.Second,the players are allowed to form fresh links with each other updating the initially proposed network.The stage payoff of any player is defined depending on the network structure.There are two types of randomness in the network formation process:(i)links may fail to be formed with different probabilities although players intend to establish them,(ii)the game process may terminate at any stage or transit to the next stage with a certain probability distribution.finally,a network is formed as a result of players’decisions and realization of random variables.The cooperative version of the stochastic game is investigated.In particular,we examine the properties of subgame consistency as well as strong subgame consistency of the core.We provide a payment mechanism or regularization of the core elements to sustain its subgame consistency and avoid the player’s deviations from the cooperative trajectory.In addition,the distribution procedure of the core elements is regularized in case there are negative payments to achieve only nonnegative payments to the players at any stage.The sufficient condition of a strongly subgame consistent core is also obtained.We illustrate our theoretical results with a numerical example.

SIRT2 as a potential new therapeutic target for Alzheimer's disease

Alzheimer's disease is the most common cause of dementia globally with an increasing incidence over the years,bringing a heavy burden to individuals and society due to the lack of an effective treatment.In this context,sirtuin 2,the sirtuin with the highest expression in the brain,has emerged as a potential therapeutic target for neurodegenerative diseases.This review summarizes and discusses the complex roles of sirtuin 2 in different molecular mechanisms involved in Alzheimer's disease such as amyloid and tau pathology,microtubule stability,neuroinflammation,myelin formation,autophagy,and oxidative stress.The role of sirtuin 2 in all these processes highlights its potential implication in the etiology and development of Alzheimer's disease.However,its presence in different cell types and its enormous variety of substrates leads to apparently contra dictory conclusions when it comes to understanding its specific functions.further studies in sirtuin 2 research with selective sirtuin2 modulators targeting specific sirtuin 2 substrates are necessary to clarify its specific functions under different conditions and to validate it as a novel pharmacological target.This will contribute to the development of new treatment strategies,not only for Alzheimer's disease but also for other neurodegenerative diseases.

Mid-gastrointestinal bleeding accounts for approximately 5%-10%of all gastrointestinal bleeding cases,and vascular lesions represent the most frequent cause.The rebleeding rate for these lesions is quite high(about 42%).We hereby recommend that scheduled outpatient management of these patients could reduce the risk of rebleeding episodes.

BACKGROUND Gastroduodenal endoscopy and biopsy following positive specific serology is considered the gold standard to diagnose celiac disease(CeD)in adults.Whether upper endoscopy helps detect comorbid conditions is unknown.AIM To investigate the prevalence of non-celiac endoscopic findings in patients in whom endoscopy was performed to confirm CeD diagnosis.METHODS This is an observational,descriptive,multicenter,retrospective study that reports endoscopic findings obtained in adult patients enrolled in local registries from four tertiary centers.We collected data reported on first endoscopy,indicated for investigation of CeD.Diagnosis of CeD was performed by histology(≥Marsh 2type mucosal damage)and specific serology.Two European and one North American center included biopsy-confirmed CeD following positive serology.A fourth center(South America)included symptomatic patients undergoing endoscopy,irrespective of CeD serology.The latter cohort included a non-CeD control group.RESULTS A total of 1328 patients(80%female;35 years median age)were enrolled,of whom 95.6%had positive specific serology.In 135 patients,endoscopy revealed 163 abnormalities unrelated to CeD(prevalence:10.1%).Erosive reflux esophagitis(6.4%),gastric erosions(2.0%),and suspicion of esophageal metaplasia(1.2%)were the most common findings.Biopsy-confirmed Barrett’s esophagus was infrequent(0.2%).No endoscopic cancer was detected.Older patients(≥51 years of age)had a higher prevalence of endoscopic findings than those≤50(Pf comorbid endoscopic findings compared with controls(29.1%;Pf this cohort,having CeD was associated with a 72%reduction in the risk of any endoscopic abnormality(Pf finding at least one endoscopic lesion(Pfew comorbid endos

Techniques for high-definition micromanipulations,such as optical tweezers,hold substantial interest across a wide range of disciplines.However,their applicability remains constrained by material properties and laser exposure.And while microfluidic manipulations have been suggested as an alternative,their inherent capabilities are limited and further hindered by practical challenges of implementation and control.Here we show that the iterative application of laser-induced,localized flow fields can be used for the relative positioning of multiple micro-particles,irrespectively of their material properties.Compared to the standing theoretical proposal,our method keeps particles mobile,and we show that their precision manipulation is non-linearly accelerated via the multiplexing of temperature stimuli below the heat diffusion limit.The resulting flow fields are topologically rich and mathematically predictable.They represent unprecedented microfluidic control capabilities that are illustrated by the actuation of humanoid micro-robots with up to 30 degrees of freedom,whose motions are sufficiently well-defined to reliably communicate personal characteristics such as gender,happiness and nervousness.Our results constitute high-definition micro-fluidic manipulations with transformative potential for assembly,micro-manufacturing,the life sciences,robotics and optohydraulically actuated micro-factories.

Witness for resolution: post-conflict quadratic affiliation in semi-free ranging pigs

In social mammals,post-conflict resolution can involve the reunion of former opponents(reconciliation),spontaneous/solicited post-conflict affliation of a third party with either opponent(triadic contacts),and affiliation between other individuals(hereafter bystanders;quadratic contacts).Quadratic contacts-possibly informing complex cognitive abilities-have been neglected in post-conflict studies.We investi-gated quadratic affliation in semi-free ranging pigs Sus scrofa,at the ethical farm Parva-Domus(Cavagnolo,Italy).Kinship was known.We collected behavioral data on adult pigs(n=104)via video recordings(43 h)followed by video analyses.Affiliative and anxiety behaviors between bystanders were collected under post-conflict(PC;following a conflict between non-bystanders)and matched-control(MC;no conflict)conditions.Quadratic affiliation was present in pigs,as bystanders affliated more in PC than MC,and such affiliation was followed by a decrease in the anxiety behaviors of both the interacting bystanders.Thus,quadratic contacts may be partly aimed at reducing one's own anxiety(intrinsic regulation).Quadratic affiliation was highest between closely related bystanders,which suggests that such affiliation may be most effective when close kin is involved.Quadratic affiliation was lowest after reconciliation and spontaneous triadic contacts.This suggests that direct peacemaking between opponents and spontaneous triadic contacts with close kin may most likely replace quadratic affiliation.Hence,pigs can be influenced by the negative events that affect other pigs-but not themselves-and their response may be modulated by social factors.Such non-random quadratic affiliation may point toward the presence of elements of social appraisal abilities inpigs.

Effect of the anionic composition of sulfolane based electrolytes on the performances of lithium-sulfur batteries

In lithium-sulfur batteries,cell design,specifically electrolyte design,has a key impact on the battery performance.The effect of lithium salt anion donor number（DN）(DN[Pf6]-=2.5,DN[N（SO2Cf3）2]-=5.4,DN[ClO4]-=8.4,DN[SO3Cf3]-=16.9,and DN[NO3]-=21.1) on the patterns of lithium-sulfur batteries and lithium metal electrode performances with sulfola ne-based electrolytes is investigated.An increase in DN of lithium salt anions leads to an increase in the depth and rate of electrochemical reduction of sulfur and long-chain lithium polysulfides and to a decrease in those for medium-and short-chain lithium polysulfides.DN of lithium salt anions has weak effect on the discharge capacity of lithium-sulfur batteries and the Coulomb efficiency during cycling,with the exception of LiSO3Cf3and LiNO3.An increase in DN of lithium salt anions leads to an increase in the cycling duration of lithium metal anodes and to a decrease in the presence of lithium polysulfides.In sulfolane solutions of LiNO3and LiSO3Cf3,lithium polysulfides do not affect the cycling duration of lithium metal anodes.

Influence of Bottom Inclination on the flow Structure in a Rotating Convective Layer

The formation of convective flows in a rotating cylindrical layer with an inclined bottom and free surface is studied.Convection is driven by localized cooling at the center of the upper free surface and by rim heating at the bottom near the sidewall.The horizontal temperature difference in a rotating layer leads to the formation of a convective flow with a complex structure.The mean meridional circulation,consisting of three cells,provides a strongly non-uniform differential rotation.As a result of the instability of the main cyclonic zonal flow,the train of baroclinic waves appears in the upper layer.The baroclinic waves provide most of the heat transfer in the middle radii and are responsible for strong temperature and velocity fluctuations.It is shown that the inclination of the bottom is a crucial factor for the structure of the convective cells and the dynamics of the baroclinic waves.The increase in the inclination angle leads to a significant increase in the energy of the waves.The obtained results may be important for heat and mass transfer in various geophysical and industrial systems,including transport of various additives and impurities in rotating crucibles,and crystallization processes.
