Advances in small animal mesentery models for in vivo flow cytometry,dynamic microscopy,and drug screening

Using animal mesentery with intravital optical microscopy is a well-established experimental model for studying blood and lymph microcirculation in vivo. Recent advances in cell biology and optical techniques provide the basis for extending this model for new applications, which should generate significantly improved experimental data. This review summarizes the achievements in this specific area, including in vivo label-free blood and lymph photothermal flow cytometry, super-sensitive fluorescence image cytometry, light scattering and speckle flow cytometry, microvessel dynamic microscopy, infrared (IR) angiography, and high-speed imaging of individual cells in fast flow. The capabilities of these techniques, using the rat mesentery model, were demonstrated in various studies; e.g., real-time quantitative detection of circulating and migrating individual blood and cancer cells, studies on vascular dynamics with a focus on lymphatics under normal conditions and under different interventions (e.g. lasers, drugs, nicotine), assessment of lymphatic disturbances from experimental lymphedema, monitoring cell traffic between blood and lymph systems, and high-speed imaging of cell transient deformability in flow. In particular, the obtained results demonstrated that individual cell transportation in living organisms depends on cell type (e.g., normal blood or leukemic cells), the cell’s functional state (e.g., live, apoptotic, or necrotic), and the functional status of the organism. Possible future applications, including in vivo early diagnosis and prevention of disease, monitoring immune response and apoptosis, chemo- and radio-sensitivity tests, and drug screening, are also discussed.

Rapid Multi-Wavelength Optical Assessment of Circulating Blood volume Without a Priori Data

The measurement of circulating blood volume （CBv） is crucial in various medical conditions including surgery, iatrogenic problems, rapid fluid administration, transfusion of red blood ceils, or trauma with extensive blood loss including battlefield injuries and other emergencies. Currently, available commercial techniques are invasive and time-consuming for trauma situations. Recently, we have proposed high-speed multi-wavelength photoacoustic/photothermal （PA/PT） flow cytometry for in vivo CBv assessment with multiple dyes as PA contrast agents （labels）. As the first step, we have characterized the capability of this technique to monitor the clearance of three dyes （indocyanine green, methylene blue, and trypan blue） in an animal model. However, there are strong demands on improvements in PA/PT flow cytometry. As additional verification of our proof-of-concept of this technique, we performed optical photometric CBv measurements in vitro. Three label dyes--methylene blue, crystal violet simultaneous photometric determination of the and, partially, brilliant green--were selected for components of their two-dye mixtures in the circulating blood in vitro without any extra data （like hemoglobin absorption） known a priori. The tests of single dyes and their mixtures in a flow system simulating a blood transfusion system showed a negligible difference between the sensitivities of the determination of these dyes under batch and flow conditions. For individual dyes, the limits of detection of 3×10-6M-3×10-5M in blood were achieved, which provided their continuous determination at a level of 10-SM for the CBv assessment without a priori data on the matrix. The CBv assessment with errors no higher than 4% were obtained, and the possibility to apply the developed procedure for optical photometric （flow cytometry） with laser sources was shown.

Free-Standing α-MoO_(3)/Ti_(3)C_(2) MXene Hybrid Electrode in Water-in-Salt Electrolytes

While transition-metal oxides such as α-MoO_(3)provide high capacity,their use is limited by modest electronic conductivity and electrochemical instability in aqueous ***-dimensional(2D)MXenes,offer metallic conductivity,but their capacitance is limited in aqueous *** of partially solvated cations into Ti_(3)C_(2)MXene from lithium-based water-in-salt(WIS)electrolytes enables charge storage at positive potentials,allowing a wider potential window and higher ***,we demonstrate that α-MoO_(3)/Ti_(3)C_(2)hybrids combine the high capacity of α-MoO_(3)and conductivity of Ti_(3)C_(2)in WIS(19.8 m LiCI)electrolyte in a wide1.8 v voltage *** voltammograms reveal multiple redox peaks from α-MoO_(3)in addition to the well-separated peaks of Ti_(3)C_(2)in the hybrid *** leads to a higher specific charge and a higher rate capability compared to a carbon and binder containing α-MoO_(3)*** results demonstrate that the addition of MXene to less conductive oxides eliminates the need for conductive carbon additives and binders,leads to a larger amount of charge stored,and increases redox capacity at higher *** addition,MXene encapsulated α-MoO_(3)showed improved electrochemical stability,which was attributed to the suppressed dissolution of α-MoO_(3).The work suggests that oxide/MXene hybrids are promising for energy storage.

Age of maturation and behavioral tactics in male yellow ground squirrel Spermophilus fulvus during mating season

Life-history theory predicts that in hibernators age of maturation is related positively to body size and negatively to the duration of active season aboveground. Yellow souslik is a large-sized ground squirrel with long hibernation, which suggests late maturation. We used four-year field observations of marked individuals to determine the age of maturation in males through analysis of age-dependent variation in body size, mass, androgen status, timing of spring emergence, ranging patterns and social behavior during the mating season. Yearling males were smaller, lighter, had lower level of fecal testosterone, emerged later and had smaller home ranges than older males. Social activity and the number of females encountered did not differ between age classes. After the second hibernation none of the studied parameters varied with age. Cluster analysis revealed two behavioral tactics: “active” males (adults only) emerged earlier, ranged more widely, initiated more contacts, encountered more females and were heavier than “passive” males (both yearling and adult). Thus, males ofS. fulvusreached adult size and matured after two hibernations, which is relatively fast for such a big species with short active period. Indirect evidence for copulations and high variation among yearlings in all parameters suggest that some of them might successfully compete with adults. Active tactic of wandering and searching for females is energetically costly, and probably only adult males in good physical condition can afford it, whereas passive tactic of residing is energy saving and good for adults in poor condition and for yearlings that are continuing to grow.

To pursue vNIIEF–vNIITF joint investigations,this paper briefly describes the experimental setup and provides numerical 3D-computation results(LEGAK-3D technique)on special features in the convergence dynamics of steel shells under their quasi-spherical explosive loading in the system with the 40-mm outer radius of the explosive *** computation results were compared with the data experimentally registered for shells of the 30KhGSA steel,both as-received and quenched to HRC 35...40,and the austenitic 12Kh18N10T stainless *** comparison was also made with laserinterferometry results obtained directly under explosive loading,as well as with gammatomography and scanning electron microscopy investigations of the recovered shells.

Photocatalytic Treatment of Waste Water from Rice Husk Alkaline Hydrolysate

SUMMARY We carried out photocatalytic treatment of a lignin-free solution of rice husk alkaline hydrolysate [pH of 2, chemical oxygen demand (COD) of 12.8 g/L] using photo-Fenton process in the ultraviolet (Uv, wavelength of 365 nm) and visible spectrum. Oxidation of lignin was performed at a constant mass ratio of COD:H2O2 as 1:2 and Fe^2+ concentration of 80 mg/L. It was established that for the destruction of alkaline lignin, exposure to visible or Uv radiation was necessary. Decolorization efficiency of a lignin-free solution depends on the dilution rate of the solution. The advanced oxidative photo-Fenton process (Uv/Н2О2/Fe^2+) made it possible to reduce the color of lignin-free solution at a dilution ratio of 1:5 by 94% of decolorization efficiency, 87% of phenolic compound and 79% of COD. With the help of gas chromatography, the products of lignin destruction were low-toxic and usable.

Traditionally,urbanization has been seen as a negative phenomenon for ***,changes in the environmental parameters induced by urbanization might be favorable for some *** the past half-century,the common hamster has actively populated cities,establishing populations in some European,Russian and Kazakhstan *** on integrative methods,we investigated free-range common hamsters inhabiting Simferopol from 2015 to 2018 to reveal possible adaptations to the urbanized environment across several parameters,including lifespan,hibernation period,reproductive activity and body *** show that in urban areas,the common hamster demonstrates an extremely short hibernation period compared to other localities,possibly due to enhanced food resources from urban forestry(walnuts,locus and hazelnut),allowing the species to start breeding very early(February)and finish as late as *** present the first evidence of polyandry for this species:mating of receptive females with several males and subsequent confirmation of multiple *** high reproductive potential,the lifespan of the common hamster in urban conditions is generally very short(less than 1 year).We speculate that in the process of synurbization,the common hamster’s innate plasticity across many life history traits permits it to successfully colonize throughout a wide range of habitats,with the ability to form novel adaptations to urban environments.

Jasmonic Acid Oxidase 2 Hydroxylates Jasmonic Acid and Represses Basal Defense and Resistance Responses against Botrytis cinerea Infection

Jasmonates （JAs） orchestrate immune responses upon wound/herbivore injury or infection by necrotro- phic pathogens. Elucidation of catabolic routes has revealed new complexity in jasmonate metabolism. Two integrated pathways attenuate signaling by turning over the active hormone jasmonoyl-isoleucine （JA-Ile） through w-oxidation or deconjugation, and define an indirect route forming the derivative 12OH-JA. Here, we provide evidence for a second 12OH-JA formation pathway by direct jasmonic acid （JA） oxidation. Three jasmonic acid oxidases （JAOs） of the 2-oxoglutarate dioxygenase family catalyze spe- cific oxidation of JA to 12OH-JA, and their genes are induced by wounding or infection by the fungus Botrytis cinerea. JA02 exhibits the highest basal expression, and its deficiency in jao2 mutants strongly enhanced antifungal resistance. The resistance phenotype resulted from constitutive expression of antimi- crobial markers rather than from their higher induction in infected jao2 plants and could be reversed by ectopic expression of any of the three JAOs injao2. Elevated defense injao2 was dependent on the activity of JASMONATE RESPONSE 1 （JAR1） and CORONATINE-INSENSITIvE 1 （COI1） but was not correlated with erihanced JA-Ile accumulation. Instead, jao2 mutant lines displayed altered accumulation of several JA species in healthy and challenged plants, suggesting elevated metabolic flux through JA-Ile. Collectively, these data identify the missing enzymes hydroxylating JA and uncover an important metabolic diversion mechanism for repressing basal JA defense responses.
