Drought is a major environmental factor limiting cotton (Gossypium hirsutum L.) productivity worldwide and projected climate changes could increase their negative effects in the future. Thus, targeting the molecular mechanisms correlated with drought tolerance without reducing productivity is a challenge for plant breeding. In this way, we evaluated the effects of water deficit progress on AtDREB2A-CA transgenic cotton plant responses, driven by the stress-inducible rd29 promoter. Besides shoot and root morphometric traits, gas exchange and osmotic adjustment analyses were also included. Here, we present how altered root traits shown by transgenic plants impacted on physiological acclimation responses when submitted to severe water stress. The integration of AtDREB2A-CA into the cotton genome increased total root volume, surface area and total root length, without negatively affecting shoot morphometric growth parameters and nor phenotypic evaluated traits. Additionally, when compared to wild-type plants, transgenic plants (17-T0 plants and its progeny) highlighted a gradual pattern of phenotypic plasticity tosome photosynthetic parameters such as photosynthetic rate and stomatal conductance with water deficit progress. Transgene also promoted greater shoot development and root robustness (greater and deeper root mass) allowing roots to grow into deeper soil layers. The same morpho-physiological trend was observed in the subsequent generation (17.6-T2). Our results suggest that the altered root traits shown by transgenic plants are the major contributors to higher tolerance response, allowing the AtDRE2A-CA-cotton plants to maintain elevated stomatal conductance and assimilate rates and, consequently, reducing their metabolic costs involved in the antioxidant responses activation. These results also suggest that these morpho-physiological changes increased the number of reproductive structures retained per plant (26% higher) when compared with its non-transgenic cou

Production of Lettuce Seedlings with Association of Diazotrophic Bacteria and Humic Acids

The objective of this work was to verify the responses in agronomic and physiological characters of lettuce seedlings when submitted to different doses of humic acids and the presence or absence of inoculation of the bacterium Azospirillum brasilense. The experiment was carried out at the Protected Cultivation Station and Biological Control Teacher Mário César Lopes, belonging to the Experimental Stations Nucleus of the State University of the West of Paraná. The experimental design was a randomized block design, in a 5 × 2 factorial scheme, with four repetitions, totaling 40 plots. The first factor consisted of five solutions containing humic acid (100%, 75%, 50%, 25% and 0%) and the second factor was the presence or absence of the diazotrophic bacterium Azospirillum brasilense obtained by commercial product. Germination tests were carried out in addition to the determination of agronomic characters such as the number of fully expanded leaves, plant height, stem diameter, main root length, shoot dry mass, root dry mass, total dry mass, and Dickson quality index. The association of diazotrophic bacteria and humic acids did not influence the emergency rate, mean emergence time, mean speed of emergency, emergence speed index, plant height, and root length of lettuce seedlings. The dilutions of humic acids, regardless of inoculation with bacteria diazotrophic favored the development of the number of leaves, stem diameter and Dickson quality index. Inoculation of lettuce seeds with diazotrophic bacteria associated with the addition of humic acids, favored the development of the dry mass of root and total dry matter.

Raman Spectroscopy Mapping of Plasma Thermally Sprayed Silicon Sheet for Solar Cell Substrate

High purity semiconductor films commit 50% of crystalline photovoltaic cell cost and current efforts focus on photovoltaic alternative devices, known as wafer equivalents, based on reduction of substrate and active light absorbing layer thickness and use of foreign substrates instead of traditional silicon wafers. In this way, knowing the substrate crystalline fraction, roughness and grain size distribution is relevant for thin film epitaxial growth. This work presents a semi-quantitative crystalline fraction analysis measured by Raman mapping microscopy of pc-Si sheets prepared by Atmospheric Thermal Plasma Spray (ATPS) as initial reference for solar cell substrates. Results showed a structural mixed silicon sheet with different values of crystalline fraction, specific regions of amorphous structure contribution, transition regions and a range of crystallite size. The results suggests the need for better ATPS substrate temperature control for higher and homogeneous crystalline fraction values to attend to requirements of a thin film solar cell substrate.
