Lignin emerged around 450 million years ago in early tracheophytes and played a pivotal role in the colonization of terrestrial ecosystems by plants.This aromatic biopolymer provides mechanical strength to supportive tissues and hydrophobicity to water-transporting vasculature and acts as a physical barrier against herbivores and pathogens(Renault et al.,2019).The deposition of lignin in cell walls for proper cellular functionalization involves several molecular steps,including the tight regulation of tissue-or cell-type-specific expression of lignin biosynthetic genes,the biosynthesis of distinct lignin monomers and their transportation to the apoplast,and the polymerization of monomer radicals to produce tissue-or cell-type-dependent polymers with distinct physico-chemical properties.

Among the impacts of climate change,there is the intensification of phenomena such as the El Niño South-ern Oscillation(ENSO)responsible for El Niño and La Niña.However,understanding their effects on the functional pro-cesses of forests is limited.Therefore,this study evaluated the effects of ENSO on litter stock and water holding capac-ity(WHC)in a successional forest in eastern Amazonia.Evaluations occurred in periods with the most rainfall in El Niño(2019)and least in La Niña(2021)years.Twelve permanent plots were used to sample litter.ENSO effects were evident for WHC,higher during El Niño.However,this influence was not clear for litter,as only in the rainy season effects were found.There was a positive correlation of WHC with precipitation and humidity,while litter stocks were negatively correlated with temperature and wind speed.Although the subject of this study requires long-term assessments,preliminary results suggests that,depending on the intensity of ENSO,forest functional processes can be strongly impacted and altered.The conclusion reinforces warnings by the scientific community about the impacts of climate change on the maintenance of litter stocks,decomposition and,consequently,the biogeochemical cycle and essential ecosystem services for the maintenance of Amazonia biodiversity.The need to develop long-term research to understand the effects of climatic change on litter stocks and water holding capacity is highlighted,especially in Amazonia.

Integrated grain cropping systems promote soil health(SH)and sustainability.microbial biomass and activity(mBA)and community structure(mCS)are key to SH.Integration of maize with Urochloa pastures strongly impacts mBA and mCS.mBA is more sensitive than mCS to shifts in grain cropping systems.Systems under continuous Urochloa increased microbial activity and AmF abundance.Tropical soils are prone to degradation.Adoption of conservation agricultural practices is essential to improve soil health,which is influenced by soil microbes.In this study we analyzed shifts in microbial biomass and activity(mBA)and microbial community structure(mCS)based on fatty acid methyl esthers(FAmEs)between five no-till agricultural practices:maize monoculture(mm);maize annualy intercropped with Urochloa decumbens(m/Ud);m/Ud with soybean rotation every other year(m/Ud-S);m/Ud keeping the pasture for the next two years(m/Ud-Ud-Ud);and maize intercropped with U.ruziziensis keeping the pasture for the next two years(m/Ur-Ur-Ur).Results indicated that mBA was affected by the inclusion of Urochloa intercropping and by rotation with soybean.Systems under a longer residence time with Urochloa in the field had higherβ-glucosidase activity and soil basal respiration,indicating a greater microbial activity.mCS was less affected than mBA by the investigated cropping systems.mCS changed only in the continuous pasture systems,which were enriched in arbuscular mycorrhyzal fungi(AmF).Additionally,the continuous pasture systems had lower microbial stress ratios than the other agricultural practices.In sum,our study showed that utilization of Urochloa spp.under longer periods in no-till agricultural practices contributes to increase microbial activity,AmF abundance and decrease microbial stress ratio.These changes are primarily beneficial for soil health.

With the increase in antimicrobial resistance,it has become necessary to explore alternative approaches for combating and preventing diseases.DB-cinnamaldehyde(CNm)and Benzyl4-amino(B4Am)are bioactive compounds derived from chalcones but with restricted solubility in aqueous media.Nanoemulsions can enhance the solubility of compounds and can be a promising alternative in the development of novel antimicrobials,with reduced side effects and prolonged release.The objective of this study was to evaluate the stability of oil-in-water nanoemulsions loaded with two distinct types of chalcones at two different dosages,to propose a stable formulation with antimicrobial properties.Results showed that nanoemulsions presented high encapsulation efficiency,low polydispersity index(PDI)and particle size below 200 nm,indicating that emulsification was a suitable method for nanoemulsion preparation.Nanoemulsions with higher dosages exhibited significant antimicrobial effects when compared to free chalcones and positive controls.Notably,B4Am nanoemulsions at higher dosages showed expressive activity against Salmonella minnesota,with a 420%greater inhibitory response compared to the free form and showing equivalence to the positive control.CNm nanoemulsions showed excellent inhibitory activity at the highest dosage,equivalent to the positive control against S.minnesota and Staphylococcus aureus.The greater number of conjugated bonds in CNm increased the antimicrobial activity in comparison with B4Am,and the formation of nanometric domains enhanced the bioavailability,being a promising alternative for antimicrobial applications.

BACKGROUND Prevalence of hepatocellular carcinoma(HCC)is increasing,especially in patients with metabolic dysfunctionassociated steatotic liver disease(mASLD).AIm To investigate rifaximin(RIF)effects on epigenetic/autophagy markers in animals.mETHODS Adult Sprague-Dawley rats were randomly assigned(n=8,each)and treated from 5-16 wk:Control[standard diet,water plus gavage with vehicle(Veh)],HCC[high-fat choline deficient diet(HFCD),diethylnitrosamine(DEN)in drinking water and Veh gavage],and RIF[HFCD,DEN and RIF(50 mg/kg/d)gavage].Gene expression of epigenetic/autophagy markers and circulating miRNAs were obtained.RESULTS All HCC and RIF animals developed metabolic-dysfunction associated steatohepatitis fibrosis,and cirrhosis,but three RIF-group did not develop HCC.Comparing animals who developed HCC with those who did not,miR-122,miR-34a,tubulin alpha-1c(Tuba-1c),metalloproteinases-2(mmp2),and metalloproteinases-9(mmp9)were significantly higher in the HCC-group.The opposite occurred with Becn1,coactivator associated arginine methyltransferase-1(Carm1),enhancer of zeste homolog-2(Ezh2),autophagy-related factor LC3A/B(map1 Lc3b),and p62/sequestosome-1(p62/SQSTm1)-protein.Comparing with controls,map1 Lc3b,Becn1 and Ezh2 were lower in HCC and RIF-groups(Pm1 was lower in HCC compared to RIF(Pmmp9 was higher in HCC in relation to the control;the opposite was observed for p62/Sqstm1(Pm1 protein was lower in the RIF-group compared to the control(P=0.024).There was no difference among groups for Tuba-1c,Aldolase-B,alpha-fetoprotein,and mmp2(P>0.05).miR-122 was higher in HCC,and miR-34a in RIF compared to controls(PmiR-26b was lower in HCC compared to RIF,and the inverse was observed for miR-224(Pmong groups regarding miR-33a,miR-143,miR-155,miR-375 and miR-21(P>0.05).CONCLUSION RIF might have a possible beneficial effect on preventing/delaying liver carcinogenesis through epigenetic modulation in a rat model of mASLD

Cell walls in plants,particularly forest trees,are the major carbon sink of the terrestrial ecosystem.Chemical and biosynthetic features of plant cell walls were revealed early on,focusing mostly on herbaceous model species.Recent developments in genomics,transcriptomics,epigenomics,transgenesis,and associated analytical techniques are enabling novel insights into formation of woody cell walls.Here,we review multilevel regulation of cell wall biosynthesis in forest tree species.We highlight current approaches to engineering cell walls as potential feedstock for materials and energy and survey reported field tests of such engineered transgenic trees.We outline opportunities and challenges in future research to better understand cell type biogenesis for more efficient wood cell wall modification and utilization for biomaterials or for enhanced carbon capture and storage.

Antimicrobial resistance(AmR)poses a critical threat to global health and development,with environmental factors—particularly in urban areas—contributing significantly to the spread of antibiotic resistance genes(ARGs).However,most research to date has been conducted at a local level,leaving significant gaps in our understanding of the global status of antibiotic resistance in urban environments.To address this issue,we thoroughly analyzed a total of 86,213 ARGs detected within 4,728 metagenome samples,which were collected by the meta SUB International Consortium involving diverse urban environments in 60 cities of 27 countries,utilizing a deep-learning based methodology.Our findings demonstrated the strong geographical specificity of urban environmental resistome,and their correlation with various local socioeconomic and medical conditions.We also identified distinctive evolutionary patterns of ARG-related biosynthetic gene clusters(BGCs)across different countries,and discovered that the urban environment represents a rich source of novel antibiotics.Our study provides a comprehensive overview of the global urban environmental resistome,and fills a significant gap in our knowledge of large-scale urban antibiotic resistome analysis.

Background Schistosomiasis is highly endemic in sub-Saharan Africa and frequently imported to Europe. male urogenital manifestations are often neglected. We aimed to ascertain the prevalence of genitourinary clinical signs and symptoms among long-term African migrants in a non-endemic European country using a serology test.methods We carried out a prospective, community-based cross-sectional study of adult male migrants from sub-Saharan Africa living in Spain.Schistosoma serology tests and microscopic urine examinations were carried out, and clinical data were obtained from an electronic medical record search and a structured questionnaire.Results We included 388 adult males, mean age 43.5 years [Standard Deviation (SD) = 12.0, range: 18–76]. The median time since migration to the European Union was 17 [Interquartile range (IQR): 11–21] years. The most frequent country of origin was Senegal (N = 179, 46.1%). Of the 338, 147 (37.6%) tested positive forSchistosoma. Parasite eggs were present in the urine of only 1.3%. Nine genitourinary clinical items were significantly associated with positiveSchistosoma serology results: pelvic pain (45.2%;OR = 1.57, 95%CI: 1.0–2.4), pain on ejaculation (14.5%;OR = 1.85, 95%CI: 1.0–3.5), dyspareunia (12.4%;OR = 2.45, 95%CI: 1.2–5.2), erectile dysfunction (9.5%;OR = 3.10, 95%CI: 1.3–7.6), self-reported episodes of infertility (32.1%;OR = 1.69, 95%CI: 1.0–2.8), haematuria (55.2%;OR = 2.37, 95%CI: 1.5–3.6), dysuria (52.1%;OR = 2.01, 95%CI: 1.3–3.1), undiagnosed syndromic STIs (5.4%), and orchitis (20.7%;OR = 1.81, 95%CI: 1.0–3.1). Clinical signs tended to cluster.Conclusions Urogenital clinical signs and symptoms are prevalent among male African long-term migrants with a positiveSchistosoma serology results. Genital involvement can be frequent even among those with long periods of non-residence in their sub-Saharan African countries of origin. Further research is needed to develop diagnostic tools and validate therapeutic approaches to chronic schistosomiasis.

The accelerated growth of the vehicular fleet, the modernization of large urban centers, and the few adjustments to the road network in Fortaleza have intensified the problems of traffic and emissions of atmospheric pollutants, highlighting the necessity for strategic urban planning initiatives to address the escalating issues of traffic and pollution. With the objective of analyzing the indices of concentrations of atmospheric pollutants and estimating how these levels can affect human health, this work consists of a study of the analysis of air quality in the intense trade region of Fortaleza. For this, the analysis zone was divided into three perimeters (major - medium - minor), where each perimeter was analyzed at 7 am, 12 noon and 5 pm. Concentrations of the type of O3, particulate matter (Pm2.5 and Pm10), CO2 and HCHO were collected. Our results demonstrate that most of the analyses are within the limits of current legislation;however, at certain times and perimeters, the analyses of CO2 and HCHO exceeded the established limits. In view of the above, we conclude that public policies to control air quality are necessary to reduce the damage to human health and the environment caused by pollutants.
