Immunotherapy becomes a promising line of treatment for breast cancer(BC)however,its success rate is still ***:The study was designed to optimize the condition for producing an effective dendritic cell(DCs)based immunotherapy by using DCs and T lymphocytes together with tumor-inﬁltrating lymphocytes(TILs)and tumor-inﬁltrating DCs(TIDCs),treated with anti-PD1 and anti-CTLA4 monoclonal *** mixture of immune cells was co-cultured with autologous breast cancer cells(BCCs)isolated from 26 BC ***:There was a signiﬁcant upregulation of CD86 and CD83 on DCs(p=0.001 and 0.017,respectively),similarly upregulation of CD8,CD4 and CD103 on T cells(p=0.031,0.027,and 0.011,respectively).While there was a signiﬁcant downregulation of FOXP3 and combined ***8 expression on regulatory T cells(p=0.014 for both).Increased CD8/Foxp3 ratio(penase(LDH,p=0.02),and a signiﬁcant decrease in vascular endothelial growth factor(VEGF,pmmed cell death ligand 1(PDL-1)were downregulated in BCCs(pmilarly cytotoxic T lymphocyte antigen-4(CTLA4,p=0.02),Programmed cell death 1(PD-1,pmmune cells(DCs,T cells,TIDCs,and TILs)with immune checkpoint inhibitors could produce a potent and effective BC ***,these data should be validated on an experimental animal model to be transferred to the clinical setting.

Ensemble machine Learning to Enhance Q8 Protein Secondary Structure Prediction

Protein structure prediction is one of the most essential objectives practiced by theoretical chemistry and bioinformatics as it is of a vital importance in medicine,biotechnology and *** secondary structure prediction(PSSP)has a significant role in the prediction of protein tertiary structure,as it bridges the gap between the protein primary sequences and tertiary structure *** secondary structures are classified into two categories:3-state category and 8-state *** the 3 states and the 8 states of secondary structures from protein sequences are called the Q3 prediction and the Q8 prediction problems,*** 8 classes of secondary structures reveal more precise structural information for a variety of applications than the 3 classes of secondary structures,however,Q8 prediction has been found to be very challenging,that is why all previous work done in PSSP have focused on Q3 *** this paper,we develop an ensemble machine Learning(mL)approach for Q8 PSSP to explore the performance of ensemble learning algorithms compared to that of individual mL algorithms in Q8 *** ensemble members considered for constructing the ensemble models are well known classifiers,namely SVm(Support Vector machines),KNN(K-Nearest Neighbor),DT(Decision Tree),RF(Random Forest),and NB(Naïve Bayes),with two feature extraction techniques,namely LDA(Linear Discriminate Analysis)and PCA(Principal Component Analysis).Experiments have been conducted for evaluating the performance of single models and ensemble models,with PCA and LDA,in Q8 *** novelty of this paper lies in the introduction of ensemble learning in Q8 PSSP *** experimental results confirmed that ensemble mL models are more accurate than individual mL *** also indicated that features extracted by LDA are more effective than those extracted by PCA.

Therapeutic potential of Calotropis gigantea extract against invasive pulmonary aspergillosis:In vitro and in vivo study

Resveratrol and fenofibrate ameliorate fructose-induced nonalcoholic steatohepatitis by modulation of genes expression

AIm: To evaluate the effect of resveratrol, alone and in combination with fenofibrate, on fructose-induced metabolic genes abnormalities in ***: Giving a fructose-enriched diet (FED) to rats for 12 wk was used as a model for inducing hepatic dyslipidemia and insulin resistance. Adult male albino rats (150-200 g) were divided into a control group and a FED group which was subdivided into 4 groups, a control FED, fenofibrate (FENO) (100 mg/kg), resveratrol (RES) (70 mg/kg) and combined treatment (FENO + RES) (half the doses). All treatments were given orally from the 9th week till the end of experimental period. Body weight, oral glucose tolerance test (OGTT), liver index, glucose, insulin, insulin resistance (HOmA), serum and liver triglycerides (TGs), oxidative stress (liver mDA, GSH and SOD), serum AST, ALT, AST/ALT ratio and tumor necrosis factor-α (TNF-α) were measured. Additionally, hepatic gene expression of suppressor of cytokine signaling-3 (SOCS-3), sterol regulatory element binding protein-1c (SREBP-1c), fatty acid synthase (FAS), malonyl CoA decarboxylase (mCD), transforming growth factor-β1 (TGF-β1) and adipose tissue genes expression of leptin and adiponectin were investigated. Liver sections were taken for histopathological examination and steatosis area were ***: Rats fed FED showed damaged liver, impairment of glucose tolerance, insulin resistance, oxidative stress and dyslipidemia. As for gene expression, there was a change in favor of dyslipidemia and nonalcoholic steatohepatitis (NASH) development. All treatment regimens showed some benefit in reversing the described deviations. Fructose caused deterioration in hepatic gene expression of SOCS-3, SREBP-1c, FAS, mDA and TGF-β1 and in adipose tissue gene expression of leptin and adiponectin. Formulation and evaluation of anti-asthmatic drug montelukast in mucoadhesive buccal patches

Objective: To formulate and evaluate anti-asthmatic drug montelukast in mucoadhesive buccal patches. methods:Buccal patches were formulated by using different hydrophilic polymers by solvent casting technique. Buccal patches were evaluated by seven physical appearances, in addition toin vitro drug release study. Results: All patches were uniform and translucent, and had smooth surface. In vitro release studies were conducted for montelukast buccal patches proved that release in the range of 75.26%-92.30% in 8 h. Emission of montelukast from all patches simulated zero order and diffusion mechanism. Finally it can be concluded that F3, F15 and F16 are the best formulation. Conclusions: The investigation concluded that patch of 5 mg of montelukast sodium were formulated by using sodium alginate with sodium carboxy methyl cellulose, hydroxy propyl methyl cellulose K100m with sodium carboxy methyl cellulose, and hydroxy propyl methyl cellulose K100m with sodium alginate (F3, F15 and F16 formulations) were the best formulations.

CDYL1 fosters double-strand break-induced transcription silencing and promotes homology-directed repair

Cells have evolved DNA damage response （DDR） to repair DNA lesions and thus preserving genomic stability and impeding carcinogenesis. DNA damage induction is accompanied by transient transcription repression. Here, we describe a previously unrecognized role of chromodomain Y-like （CDYL1） protein in fortifying double-strand break （DSB）-induced transcription repression and repair. We showed that CDYL1 is rapidly recruited to damaged euchromatic regions in a poly （ADP-ribose） polymerase 1 （PARP1）-dependent, but ataxia telangiectasia mutated （ATm）-independent, manner. While the C-terminal region, containing the enoyl-CoA hydratase like （ECH） domain, of CDYL1 binds to poly （ADP-ribose）（PAR） moieties and mediates CDYL1 accumulation at DNA damage sites, the chromodomain and histone H3 trimethylated on lysine 9 （H3K9me3） mark are dispensable for its recruitment. Furthermore, CDYL1 promotes the recruitment of enhancer of zeste homolog 2 （EZH2）, stimulates local increase of the repressive methyl mark H3K27me3, and promotes transcription silencing at DSB sites. In addition, following DNA damage induction, CDYL1 depletion causes persistent G2/m arrest and alters H2AX and replication protein A （RPA2） phosphorylation. Remarkably, the ‘traffic-light reporter’ system revealed that CDYL1 mainly promotes homology-directed repair （HDR） of DSBs in vivo. Consequently, CDYL1-knockout cells display synthetic lethality with the chemotherapeutic agent, cisplatin. Altogether, our findings identify CDYL1 as a new component of the DDR and suggest that the HDR-defective ‘BRCAness’ phenotype of CDYL1-deficient cells could be exploited for eradicating cancer cells harboring CDYL1 mutations.

Corrosion Protection of 5083 AA in SalineWater by Polyacrylonitrile Nanofibers

Polymeric nanofibers are a promising technology to protect the metal surfaces from *** the literature search,the use of polyacrylonitrile nanofibres(PANNFs)as a corrosion inhibitor coating for aluminum alloys has not been *** work includes the development of a new,lightweight,high surface area and efficient coating of PANNFs that produced using electrospinning process to resist the corrosion of aluminum alloys(AA5083)which immersed in 0.6 m NaCl at alkaline medium(pH=12)and acidic medium(pH=1)at a range of temperatures(293–323)*** PANNFs coating was successfully deposited on AA 5083 specimens,where these samples were considered as a collector electrode in the electrospinning *** corrosion experiments of the aluminum alloys coated with PANNFs before and after immersion in both corrosive mediums were investigated using cyclic potential polarization(CPP).The results confirmed that the PANNFs coating was able to protect the surface of the aluminum specimens from corrosion,by reducing the corrosion current and increasing the surface polarization resistance,thus reducing the corrosion *** protection efficiency was found in the alkaline medium 98.8%while in the acidic medium 83.3%.So,it was in both mediums decreased with the increase in *** shape,distribution and size of the polymeric nanofibers that formed the coating were also examined using field emission scanning electron microscopy(FE-SEm)and the percentages of the structural components of these fibers were detected using the X-ray dispersion spectroscopy(EDS).The surface of aluminum specimens was completely covered by *** electrospun nanofibers have worn out and lined up spacing after immersion in the corrosive *** diameters average of PANNFs was found to be about 200 and 150 nm before and after immersion,respectively.

Changing dietary n-6:n-3 ratio using different oil sources affects performance,behavior,cytokines mRNA expression and meat fatty acid profile of broiler chickens

Typical formulated broiler diets are deficient in n-3 poly-unsaturated fatty acids(PUFA) due to widening n-6:n-3 PUFA ratio which could greatly affect performance,immune system of birds and,more importantly,meat *** study was conducted to evaluate the effect of modifying dietary n-6:n-3 PUFA ratio from plant and animal oil sources on performance,behavior,cytokine m RNA expression,antioxidative status and meat fatty acid profile of broiler ***(n=420) were fed 7 diets enriched with different dietary oil sources and ratios as follows: sunflower oil in control diet(C); fish oil(FO); 1:1 ratio of sunflower oil to FO(C1 FO1); 3:1 ratio of sunflower oil to fish oil(C3 FO1); linseed oil(LO); 1:1 ratio of sunflower oil to linseed oil(C1 LO1); 3:1 ratio of sunflower oil to linseed oil(C3 LO1),resulting in dietary n-6:n-3 ratios of approximately 40:1,1.5:1,4:1,8:1,1:1,2.5:1 and 5:1,*** best final body weight,feed conversion ratio as well as protein efficiency ratio of broilers were recorded in the C1 FO1 and C1 LO1 *** with the control group,the dressing percentage and breast and thigh yield were highest in the C1 FO1 and C1 LO1 *** the dietary n-6:n-3 ratio increased(P meat contents of eicosapentaenoic acid and docosahexaenoic acid increased(P m FO and LO clearly decreased(P m total cholesterol,triglycerides and very low-density lipoproteins and enhanced antioxidative *** feeding frequency was decreased(P mpared to plant oil,to broiler diets enhanced the relative m RNA expression of interferon gamma,interleukin-1 beta,interleukin-2 and

Validation of prognostic indices in Egyptian Budd-Chiari syndrome patients: A single-center study

AImTo compare predictive ability of Budd-Chiari syndrome (BCS) prognostic indices (PIs) for one-year survival and Transjugular intrahepatic portosystemic shunt (TIPS) *** retrospective study enrolled 194 Egyptian patients with primary BCS who presented to the Budd-Chiari Study Group of Ain Shams University Hospital. Calculation of the available PIs was performed using Child-Pugh and model for end-stage liver disease scores, BCS-specific PIs (Clichy, New Clichy and Rotterdam) for all patients, and BCS-TIPS PI only for patients who underwent TIPS. The overall one-year survival rate and the one-year shunt patency rate for TIPS were *** overall one-year survival rate was 69.6%, and the New Clichy PI revealed the best validity for its prediction at a cut-off value of 3.75, with sensitivity and specificity of 78% and 73.3%, respectively [area under receiver operating characteristic curve (AUC) = 0.806]. The one-year survival rate post-TIPS was 89.7%, and the BCS-TIPS score demonstrated validity for its prediction at a cut-off value of 3.92 (sensitivity and specificity were 71.4% and 64.5%, respectively) (AUC = 0.715). Logistic regression analysis revealed that the New Clichy PI (P = 0.030), high serum total bilirubin (P = 0.047) and low albumin (P mortality within one year. The one-year shunt patency rate in TIPS was 80.2%, and none of the PIs exhibited significant validity for its *** New Clichy score could independently predict the one-year survival in Egyptian BCS patients.
