Weighted gene co-expression network analysis identiffes potential regulators in response to Salmonella enteritidis challenge in the reproductive tract of laying ducks

Salmonella enteritidis(Se)is a zoonotic and vertically transmitted pathogen,often colonized in the reproductive tract of adult poultry,which can result in direct contamination of eggs and threaten human *** studies have revealed that some pattern recognition receptors and resistance genes were involved in regulating immune responses to Se invasion in ***,the role of these immune response genes was not independent,and the interactions among the genes remained to be further *** this study,Se burden and colonization were determined in reproductive tissue after the ducks were Se-infected,and RNA-sequencing was performed to construct co-expression networks by weighted gene co-expression network analysis(WGCNA).The result showed that Se could be isolated from 22% of infected-birds in any segment of the reproductive tract and the Se was readily colonized in the stroma,small follicle,isthmus,and vagina of the reproductive tracts in morbid *** top central,highly connected genes were subsequently identified three specific modules in the above four tissues at the defined cut-offs(Pew candidate regulators and 125 transcription ***,those 185 differentially expressed genes(DeGs)in these modules were ***,the hub genes(TRAF3,CXCR4 and IL13RA1)were identified to act with many other genes through immune response pathways including NF-kappaB,Toll-like receptor,steroid biosynthesis,and p53signaling *** data provide references that will understand the immune regulatory relationships during Se infection,but also assist in the breeding of Se-resistant lines through potential biomarkers.

Basin Groundwater Recharge and Discharge in the Gully erosion Prone Areas of Anambra, Nigeria

Anambra State of Nigeria is ravaged by ecological hazards of gully erosion and landslides. Over 450 active gully and landslide sites with depths ranging from 5 m to over 120 m are concentrated on both sides of the Awka-Nanka-Umuchu-Orlu cuesta. The Idemili River and Odo River form the major drainage basins of the area. The geomorphic characteristic of the cuesta/escarpment is a major controlling factor in groundwater recharge and discharge within the drainage basins. Rise in aquifer levels result in hydrogeotechnical implications of gully development and landslides. The calculated values of hydraulic properties of conductivity and transmissivity range from 1.20×10^-1 to 5.93×10^-1 cm/s and 1.15×10^5-13.05×10.3 m^2/s respectively. The values of groundwater velocity and groundwater discharge were 82 *** and 2.96×10^6 m^*** The plasticity index of the soil in the gully prone areas is relatively low ranging from 12.50-36.57%. The unique features of the geologic formations together with obtained aquifer characteristics favour gully/landslide hazards in the area.

Disinfection byproducts in chlorinated or brominated swimming pools and spas: Role of brominated DBPs and association with mutagenicity

Although the health benefits of swimming are well-documented, health effects such as asthma and bladder cancer are linked to disinfection by-products(DBPs) in pool water. DBPs are formed from the reaction of disinfectants such as chlorine(Cl) or bromine(Br) with organics in the water. Our previous study(Daiber et al., environ. Sci. Technol. 50, 6652;2016) found correlations between the concentrations of classes of DBPs and the mutagenic potencies of waters from chlorinated or brominated swimming pools and spas. We extended this study by identifying significantly different concentrations of 21 individual DBPs in brominated or chlorinated pool and spa waters as well as identifying which DBPs and additional DBP classes were most associated with the mutagenicity of these waters. Using data from our previous study, we found that among 21 DBPs analyzed in 21 pool and spa waters, the concentration of bromoacetic acid was significantly higher in Brwaters versus Cl-waters, whereas the concentration of trichloroacetic acid was significantly higher in Cl-waters. Five Br-DBPs(tribromomethane, dibromochloroacetic acid, dibromoacetonitrile, bromoacetic acid, and tribromoacetic acid) had significantly higher concentrations in Br-spa versus Cl-spa waters. Cl-pools had significantly higher concentrations of ClDBPs(trichloroacetaldehyde, trichloromethane, dichloroacetic acid, and chloroacetic acid), whereas Br-pools had significantly higher concentrations of Br-DBPs(tribromomethane, dibromoacetic acid, dibromoacetonitrile, and tribromoacetic acid). The concentrations of the sum of all 4 trihalomethanes, all 11 Br-DBPs, and all 5 nitrogen-containing DBPs were each significantly higher in brominated than in chlorinated pools and spas. The 8 Br-DBPs were the only DBPs whose individual concentrations were significantly correlated with the mutagenic potencies of the pool and spa waters. A theoretical and experimental study of the thermal decomposition of nitroguanidine(NQ) has been carried out. Various thermolysis channels were studied by quantum chemistry methods at the CCSD(or DLPNO-CCSD) level using the aug-cc-pVDZ basis set. It is shown that the lowest activation enthalpies(170-180kJ/mol) are characteristic of the reactions of NO2abstraction from the initial NQ and the reaction channel with the transfer of oxygen from the nitro group to carbon in the limiting stage. Additionally, the thermolysis of NQ was studied experimentally in a nonisothermal mode with heating rates from 1 to 10K/min. In these experiments, the weight loss of the sample, thermal effects, and mass spectra of the products were recorded. An analysis of the experimental data confirmed the results of a theoretical study of the mechanism of thermal decomposition of NQ. The main thermolysis products are N2O, HNCO, NH3, and NO2, which fully corresponds to quantum chemical calculations.
