Variations in Vascular e.dothe.ial Growth Factor (Ve.F) le.e.s we.e.prospe.tive.y e.aluate. in 18 young wome. unde.going in vitro fe.tilization tre.tme.ts according to the.“Long Protocol” and a typical patte.n of Ve.F le.e.s was re.orde.. A significant incre.se.in Ve.F conce.trations was obse.ve. only whe. the.follicle. re.che. a me.n diame.e. of 15.3 mm in concurre.ce.with mature.oocyte.re.rie.al. Since.an incre.se.in Ve.F le.e.s is re.ate. to follicular vascularity and oocyte.de.e.oppme.t, our study supports the.approach that oocyte.re.rie.al may be.pe.forme. whe. follicle. > 15 mm in diame.e. appe.r. Anticipating e.g re.rie.al in young patie.ts with an optimal ovarian re.e.ve.may de.re.se.the.incide.ce.of se.e.e.ovarian hype.stimulation, without compromising the.tre.tme.t re.ults.

＜正＞RNA inte.fe.e.ce.has be.n first use. in 1998 to show se.ue.ce.- spe.ific ge.e.sile.cing in C. e.e.ans. Today, se.e. ye.rs late., RNA inte.fe.e.ce.is an e.te.sive.y use. ge.omics tool. This approach to inhibit ge.e.e.pre.sion can be.applie. to virtually all ce.l type. and it may have.se.e.al advantage. ove. the.more.conve.tional approach of inhibiting ge.e.function by homologous re.ombination. He.e. we.pre.e.t data obtaine. using siRNA to study the.function of Dystroglycan （DG）, the.ce.tral compone.t of the.dystrophin associate. prote.n comple. （DAPC）. This comple. is situate. at the.plasmale.ma and conne.ts the.e.tra ce.lular matrix to the.cytoske.e.on. Disruption of this comple. cause. by mutations in a numbe. of its compone.ts has be.n shown to cause.a varie.y of muscular dystrophie.. Dystroglycan is the.ce.tral compone.t of the.DAPC, and is thought to have.a signaling function in addition to its structural role. Although various signaling mole.ule. have.be.n shown to associate.with DG and the.DAPC, a spe.ific signaling role.for the.comple. has not be.n pre.iously de.cribe.. DG null mice.die.during e.rly e.bryoge.e.is be.ause.of de.e.ts in e.tra - e.bryonic structure. and the.r association with the.e.tra - ce.lular matrix. Muscle.spe.ific targe.ing of DG has yie.de. ve.y little.information as to the.function of this prote.n. We.have.sile.ce. the.e.pre.sion of DG in C2C12 mouse.myoblasts using siRNA and have.shown that its pre.e.ce.is ne.e.sary for corre.t e.pre.sion of muscle.spe.ific prote.ns such as myoge.in, Me.-2, cave.lin-3 and MHC. This constitute. dire.t e.ide.ce.that DG is pivotal in te.minal muscle.diffe.e.tiation. It is of inte.e.t to note.that this e.ide.ce.was not uncove.e. by targe.ing e.pe.ime.ts. The.implication is that, at le.st in some.case., inve.tigating the.function of a give. prote.n by ge.e.sile.cing or somatic targe.ing are.approache. pre.e.able.to the.cre.tion of null mice.

Inte.ve.tion(s): Nine.y-e.ght IVF/e. cycle. with hypnosis during the.e. proce.ure.we.e.matche. with 96 re.ular IVF/e. cycle.. Main Outcome.Me.sure.: Comparison of clinical pre.nancy and implantation rate. be.we.n the.two groups. Re.ult(s): We.obtaine. 52 clinical pre.nancie. out of 98 cycle. (53.1% ) with an implantation rate.of 28% among hypnosis IVF/e. cycle., and 29 out of 96 (30.2% ) clinical pre.nancie. and an implantation rate.of 14.4% in the.control cycle.. Our ove.all IVF program pre.nancy rate.for the.same.pe.iod was 32.1% . Logistic re.re.sion analysis was pe.forme. e.phasizing the.positive.contribution of hypnosis to the.IVF/e. conce.tion rate.. Conclusion(s): This study sugge.ts that the.use.of hypnosis during e. may significantly improve.the.IVF/e. cycle.outcome.in te.ms of incre.se. implantation and clinical pre.nancy rate.. Furthe.more. it se.ms that the.patie.ts’ attitude.to the.tre.tme.t was more.favorable.

Although the.fre.ue.cy of CD56＋CD16-dNK among the.total CD45＋ le.kocyte. did not change.ove. this pe.iod, the.e.pre.sion of the.activating re.e.tors, NKp80 and NKG2D, was gre.tly upre.ulate.. We.obse.ve. a significantly highe. numbe. of e.travillous trophoblast ce.ls in proximity to the.dNK ce.ls in the.first trime.te. in comparison with the.se.ond trime.te. de.idua. NKG2D e.pre.sion by first trime.te. dNK ce.ls was de.re.se. whe. co-culture. with the.HTR-8 trophoblast ce.l line. In the.se.ond trime.te., functional marke.s of dNK activation, i.e., angioge.ic factor production （e.g., vascular e.dothe.ial growth factor, inte.le.kin-8, inte.fe.on-gamma）, re.aine. stable.de.pite.an incre.se.in NKp80 or NKG2D surface.e.pre.sion. Furthe.more. the.de.ranulation capacity of dNK ce.ls, as asse.se. by CD107a, was de.re.se. in the.se.ond trime.te.. We.sugge.t that in the.first trime.te., trophoblast-dNK inte.actions ge.e.ate.a population of dNK ce.ls with a suppre.se. activating phe.otype. In the.se.ond trime.te., the.loss of trophoblast-dNK inte.actions le. to the.inhibition of dNK ce.l function, although the.r activating re.e.tor e.pre.sion was incre.se..
